Risk of colon cancer in hereditary non-polyposis colorectal cancer patients as predicted by fuzzy modeling:Influence of smoking

AIM： To investigate whether a fuzzy logic model could predict colorectal cancer （CRC） risk engendered by smoking in hereditary non-polyposis colorectal cancer （HNPCC） patients. MeTHODS： Three hundred and forty HNPCC mismatch repair （MMR） mutation carriers from the Creighton University Hereditary Cancer Institute Registry were selected for modeling. Age-dependent curves were generated to elucidate the joint effects between gene mutation （hMLH1 or hMSH2）, gender, and smoking status on the probability of developing CRC. ReSULTS： Smoking significantly increased CRC risk in male hMSH2 mutation carriers （P 〈 0.05）. hMLH1 mutations augmented CRC risk relative to hMSH2 mutation carriers for males （P 〈 0.05）. Males had a significantly higher risk of CRC than females for hMLH1 non smokers （P 〈 0.05）, hMLH1 smokers （P 〈 0.1） and hMSH2 smokers （P 〈 0.1）. Smoking promoted CRC in a dose-dependent manner in hMSH2 in males （P 〈 0.05）. Females with hMSH2 mutations and both sexes with the hMLH1 groups only demonstrated a smoking effect after an extensive smoking history （P 〈 0.05）. CONCLUSION： CRC promotion by smoking in HNPCC patients is dependent on gene mutation, gender and age. These data demonstrate that fuzzy modeling may enable formulation of clinical risk scores, thereby allowing individualization of CRC prevention strategies.

Significant association between ABO blood group and pancreatic cancer

AIM:To evaluate whether the ABO blood group is related to pancreatic cancer risk in the general population of the United ***:Using the University of Pittsburgh's clinicalpancreatic cancer registry,the blood donor database from our local blood bank (Central Blood Bank),and the blood product recipient database from the regional transfusion service (Centralized Transfusion Service) in Pittsburgh,Pennsylvania,we identified 274 pancreatic cancer patients with previously determined serological ABO blood group *** ABO blood group frequency was compared between these patients and 708842 individual,community-based blood donors who had made donations to Pittsburgh's Central Blood Bank between 1979 and ***:The frequency of blood group A was statistically significantly higher amongst pancreatic cancer patients compared to its frequency amongst the regional blood donors [47.63% vs 39.10%,odds ratio (OR)=1.43,P=0.004].Conversely,the frequency of blood group O was significantly lower amongst pancreatic cancer patients relative to the community blood donors (32.12% vs 43.99%,OR=0.60,P=0.00007).There were limited blood group B (n=38) and AB (n=17) pancreatic cancer patients;the overall P trend value comparing patient to donor blood groups was ***:The ABO blood group is associated with pancreatic cancer *** studies should examine the mechanism linking pancreatic cancer risk to ABO blood group.

AIM： To test the hypothesis that calcium sensing receptor （CASR） polymorphisms are associated with chronic pancreatitis （CP）, and to determine whether serine protease inhibitor Kazal 1type （SPINK1） N34S or alcohol are necessary co-factors in its etiology. MeTHODS： Initially, 115 subjects with pancreatitis and 66 controls were evaluated, of whom 57 patients and 21 controls were predetermined to carry the high-risk SPINK1 N34S polymorphism. We sequenced CASR gene exons 2, 3, 4, 5 and 7, areas containing the majority of reported polymorphisms and novel mutations. Based on the initial results, we added 223 patients and 239 controls to analyze three common nonsynonymous single nucleotide polymorphisms （SNPs） in exon 7 （A986S, R990G, and Q1011e）. ReSULTS： The CASR exon 7 R990G polyrnorphism was significantly associated with CP （OR, 2.01; 95% CI, 1.12-3.59; P = 0.015）. The association between CASR R990G and CP was stronger in subjects who reported moderate or heavy alcohol consumption （OR, 3.12; 95% CI, 1.14-9.13; P = 0.018）. There was no association between the various CASR genotypes and SPINK1 N34S in pancreatitis. None of the novel CASR polymorphisms reported from Germany and India was detected. CONCLUSION： Our United States-based study confirmed an association of CASR and CP and for the first time demonstrated that CASR R990G is a significant risk factor for CP. We also conclude that the risk of CP with CASR R990G is increased in subjects with moderate to heavy alcohol consumption.

Applying a Context-based Method to Build a Knowledge Graph for the Blue Amazon

Knowledge graphs are employed in several tasks,such as question answering and recommendation systems,due to their ability to represent relationships between *** constructing such a graphs,however,remains an unresolved challenge within knowledge *** tackle this challenge,we propose CtxKG,a method specifically aimed at extracting knowledge graphs in a context of limited resources in which the only input is a set of unstructured text *** is based on OpenIe(a relationship triple extraction method)and BeRT(a language model)and contains four stages:the extraction of relationship triples directly from text;the identification of synonyms across triples;the merging of similar entities;and the building of bridges between knowledge graphs of different *** method distinguishes itself from those in the current literature(i)through its use of the parse tree to avoid the overlapping entities produced by base implementations of OpenIe;and(ii)through its bridges,which create a connected network of graphs,overcoming a limitation similar methods have of one isolated graph per *** compare our method to two others by generating graphs for movie articles from Wikipedia and contrasting them with benchmark graphs built from the OMDb movie *** results suggest that our method is able to improve multiple aspects of knowledge graph *** also highlight the critical role that triple identification and named-entity recognition have in improving the quality of automatically generated graphs,suggesting future paths for ***,we apply CtxKG to build BlabKG,a knowledge graph for the Blue Amazon,and discuss possible improvements.

Adoptive cell therapy(ACT)is an immunotherapy strategy for cancer that has seen widespread clinical *** ACT,patient-derived lymphocytes are stimulated with the antigen of interest ex vivo,proliferated,then returned to the patient to initiate an antigen-specific antitumor *** effective,this process is resource-intensive and logistically impossible for many *** artificial antigen presenting cells(aAPCs)offer a potential“off-the-shelf”alternative to ex vivo *** particulate aAPCs perform well in vitro,they have had limited success in vivo due to poor bioavailability after *** to bioavailability include rapid clearance,unfavorable biodistribution,and inadequate interactions with CD8+T cells at sites of *** properties such as elasticity have been shown to vastly impact the bioavailability and particle-cell interactions,but this has yet to be investigated in the context of aAPCs for in vivo T-cell *** literature likewise indicates that biomaterial properties,especially elasticity,can modulate T-cell activation in *** the goal of creating a more biomimetic,next-generation particulate aAPC,we developed a poly(ethylene)glycol hydrogel particle platform with tunable elasticity to investigate the impact of elasticity on antigen-specific T cell activation for in vivo adoptive *** this knowledge,we were able to gain more precise control over in vivo T cell activation and investigate possible mechanisms including the effects of aAPC elasticity on T cell binding,macrophage uptake,and the protein corona.

Objective:The aim of this study was to assess the relative efficacy of medications used following severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection on self-reported alterations in taste and/or smell ***:Seven hundred and fourteen persons with self-reported postcoronavirus disease 2019(post-COVID-19)chemosensory disorders were personally interviewed regarding specific medications they were administered following the acute phase of the *** dependent measure—self-reported total recovery of chemosensory symptoms—was subjected to stepwise logistic *** predictors included demographic and clinical variables,in addition to specific medications used to mitigate disease symptoms(i.e.,systemic corticosteroids,oseltamivir,vitamin C,ibuprofen,hydroxychloroquine,azithromycin,ivermectin,nitazoxanide,anticoagulants,and zinc).Results:The median time between COVID-19 symptom onset and the interviews was 81 days(interquartile range:60-104).Of the 714 subjects,249(34.9%)reported total recovery of their chemosensory function;437(61.2%)had at least one treatment since the beginning of the *** and those with more comorbidities had undergone more *** recovery rates of the treated and nontreated groups did not differ ***,respondents who had used nitazoxanide tended to have a higher rate of self-reported taste or smell *** who took oral zinc were less likely to ***:No medication employed during the first months after SARS-CoV-2 infection had a clear positive effect on returning self-reported smell or taste function to normal,although nitrazoxide trended in a positive *** zinc had a negative effect on the reported recovery of these senses.

Beyond Green Capitalism： Social-ecological Transformation and Perspectives of a Global Green-Left

Given certain reactions to the ecological crisis as part of the multiple crisis of capitalism （like the so-called energy transition in Germany）, a new mode of development might emerge which can be called green capitalism. This would shift the terrain of social critique and emancipatory social struggles. The paper introduces the debate on ＂social-ecological transformation＂ which emerged as a radical part of a more comprehensive debate on ＂great＂ or ＂societal transformation＂ and high- lights its core issues： the issue of a necessary attractive mode of production and living for all, the role of pioneers of change and changing political and economic institutions, the acknowledgment of shifting social practices, the requirement for alternative imaginaries or ＂stories＂ of a good life as part of a contested process which is called ＂futuring.＂ As an example for alternative imaginaries, the current debate on ＂degrowth＂ is outlined and evaluated. The second part of the paper focuses more concretely on issues around the formation and existence of a global green-left. After mentioning a crucial problem for any global alternative--i.e., the structural feature of economic and geopolitical competition which historically divided the global Left and pulled it into compromises at national or regional scales--four requirements or characteristics of a global green-left are highlighted： to weaken and change capitalistically driven competition and competitiveness, to push a social-ecological transformation in democratic ways and not at the back ofordinary people （like conservative and liberal proposals for transformation tend to do）, to link more systematically green issues with labor issues and, finally, to transform the overall dispositive of political action from a ＂distributive＂ to a ＂transformative Left.＂ One dimension of such an enhancement, it is concluded, is a broader understanding of the ＂economy＂ itself by acknowledging the demands and achievements of a ＂care revolution＂ which
