Salsolinol as an RNA m~6A methylation inducer mediates dopaminergic neuronal death by regulating YAP1 and autophagy

Salsolinol(1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline,Sal)is a catechol isoquinoline that causes neurotoxicity and shares structural similarity with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,an environmental toxin that causes Parkinson's disease.However,the mechanism by which Sal mediates dopaminergic neuronal death remains unclear.In this study,we found that Sal significantly enhanced the global level of N~6-methyladenosine(m~6A)RNA methylation in PC12 cells,mainly by inducing the downregulation of the expression of m~6A demethylases fat mass and obesity-associated protein(FTO)and alk B homolog 5(ALKBH5).RNA sequencing analysis showed that Sal downregulated the Hippo signaling pathway.The m~6A reader YTH domain-containing family protein 2(YTHDF2)promoted the degradation of m~6A-containing Yes-associated protein 1(YAP1)mRNA,which is a downstream key effector in the Hippo signaling pathway.Additionally,downregulation of YAP1 promoted autophagy,indicating that the mutual regulation between YAP1 and autophagy can lead to neurotoxicity.These findings reveal the role of Sal on m~6A RNA methylation and suggest that Sal may act as an RNA methylation inducer mediating dopaminergic neuronal death through YAP1 and autophagy.Our results provide greater insights into the neurotoxic effects of catechol isoquinolines compared with other studies and may be a reference for assessing the involvement of RNA methylation in the pathogenesis of Parkinson's disease.

With the rapid development of information technology,IoT devices play a huge role in physiological health data detection.The exponential growth of medical data requires us to reasonably allocate storage space for cloud servers and edge nodes.The storage capacity of edge nodes close to users is limited.We should store hotspot data in edge nodes as much as possible,so as to ensure response timeliness and access hit rate;However,the current scheme cannot guarantee that every sub-message in a complete data stored by the edge node meets the requirements of hot data;How to complete the detection and deletion of redundant data in edge nodes under the premise of protecting user privacy and data dynamic integrity has become a challenging problem.Our paper proposes a redundant data detection method that meets the privacy protection requirements.By scanning the cipher text,it is determined whether each sub-message of the data in the edge node meets the requirements of the hot data.It has the same effect as zero-knowledge proof,and it will not reveal the privacy of users.In addition,for redundant sub-data that does not meet the requirements of hot data,our paper proposes a redundant data deletion scheme that meets the dynamic integrity of the data.We use Content Extraction Signature(CES)to generate the remaining hot data signature after the redundant data is deleted.The feasibility of the scheme is proved through safety analysis and efficiency analysis.

Domain name generation algorithm(DGA)classification is an essential but challenging problem.Both feature-extract-ing machine learning(ML)methods and deep learning(DL)models such as convolutional neural networks and long short-term memory have been developed.However,the performance of these approaches varies with different types of DGAs.Most features in the ML methods can characterize random-looking DGAs better than Word-looking DGAs.To improve the classification performance on word-looking DGAs,subword tokenization is employed for the DL mod-els.Our experimental results proved that the subword tokenization can provide excellent classification performance on the word-looking DGAs.We then propose an integrated scheme that chooses an appropriate method for DGA classification depending on the nature of the DGAs.Results show that the integrated scheme outperformed existing ML and DL methods,and also the subword DL methods.

The integration of interfacial solar steam generation and photocatalytic degradation technology has pro-vided a promising platform to simultaneously produce freshwater and degrade pollutants.However,con-structing low-cost,multi-functional evaporators for treating Cr(VI)-polluted water remains challenging,and the synergistic mechanism on Cr(VI)reduction is fuzzy.Herein,we propose the combined strategy of ball milling and solution mixing for the sustainable production of Bi-MoF microrod from waste poly(ethylene terephthalate),and construct Bi-MoF-based solar evaporators for simultaneous photo-Fenton Cr(VI)reduction and freshwater production.Firstly,the evaporator comprised of Bi-MoF microrod and graphene nanosheet possesses high light absorption,efficient photothermal conversion,and good hydro-philic property.Attributing to the advantages,the hybrid evaporator exhibits the evaporation rate of 2.16 kg m^(-2) h^(-1) and evaporation efficiency of 87.5%under 1 kW m^(-2) of irradiation.When integrating with photo-Fenton reaction,the Cr(VI)reduction efficiency is 91.3%,along with the reaction kinetics of 0.0548 min^(-1),surpassing many advanced catalysts.In the outdoor freshwater production and Cr(VI)reduction,the daily accumulative water yield is 5.17 kg m^(-2) h^(-1),and the Cr(vI)reduction efficiency is 99.9%.Furthermore,we prove that the localization effect derived from the interfacial solar-driven evap-oration enhances H_(2)O_(2) activation for the photo-Fenton reduction of Cr(VI).Based on the result of density functional theory,Bi-MOF microrod provides rich active centers for H_(2)O_(2) activation to produce active sites such as e-or-O_(2).This study not only proposes a new strategy to construct multi-functional solar evaporators for freshwater production and catalytic reduction of pollutants,but also advances the chem-ical upcycling of waste polyesters.

The infamous type Ⅳ failure within the fine-grained heat-affected zone (FGHAZ) in G115 steel weldments seriously threatens the safe operation of ultra-supercritical (USC) power plants.In this work,the traditional thermo-mechanical treatment was modified via the replacement of hot-rolling with cold rolling,i.e.,normalizing,cold rolling,and tempering (NCT),which was developed to improve the creep strength of the FGHAZ in G115 steel weldments.The NCT treatment effectively promoted the dissolution of preformed M_(23)C_(6)particles and relieved the boundary segregation of C and Cr during welding thermal cycling,which accelerated the dispersed reprecipitation of M_(23)C_(6) particles within the fresh reaustenitized grains during post-weld heat treatment.In addition,the precipitation of Cu-rich phases and MX particles was promoted evidently due to the deformation-induced dislocations.As a result,the interacting actions between precipitates,dislocations,and boundaries during creep were reinforced considerably.Following this strategy,the creep rupture life of the FGHAZ in G115 steel weldments can be prolonged by 18.6%,which can further push the application of G115 steel in USC power plants.

Ruthenium(Ru)has been recognized as a prospective candidate to substitute platinum catalysts in water-splitting-based hydrogen production.However,minimizing the Ru contents,optimizing the water dissociation energy of Ru sites,and enhancing the long-term stability are extremely required,but still face a great challenge.Here,we report on creating tungsten oxide-anchored Ru clusters(Ru-WO_(x))with electron-rich and anti-corrosive microenvironments for efficient and robust seawater splitting.Benefiting from the abundant oxygen vacancy structure in tungsten oxide support,the Ru-WO_(x)exhibits strong Ru-O and Ru-W bonds at the interface.Our study elucidates that the strong Ru-O bonds in Ru-WO_(x)may accelerate the water dissociation kinetics,and the Ru-W bonds will lead to the strong metal-support interaction and electrons transfer fromWto Ru.The optimal Ru-WO_(x)catalysts exhibit a low overpotential of 29 and 218mVat the current density of 10 mA cm^(−2) in alkaline and seawater media,respectively.The outstanding long-term stability discloses that the Ru-WO_(x)catalysts own efficient corrosion resistance in seawater electrolysis.We believe that thiswork offers new insights into the essential roles of electron-rich and anti-corrosivemicroenvironments in Ru-based catalysts and provide a new pathway to design efficient and robust cathodes for seawater splitting.

In this study, the in-situ synthesized ZrB_(2) nanoparticles and rare earth Sc were introduced to enhance the strength and ductility of 7N01 aluminum alloy, via the generation of high-melting and uniform nanodispersoids. The microstructure and mechanical property evolution of the prepared composites and the interaction between ZrB_(2) and Sc were studied in detail. The microstructure investigation shows that the introduction of rare earth scandium(Sc) can promote the distribution of ZrB_(2) nanoparticles, by improving their wettability to the Al melt. Meanwhile, the addition of rare earth Sc also modifies the coarse Al Zn Mg Mn Fe precipitated phases, refines the matrix grains and generates high-melting Al_3(Sc,Zr)/Al_3Sc nanodispersoids. Tensile tests of the composites show that with the combinatorial introduction of ZrB_(2) and Sc, the strength and ductility of the composites are improved simultaneously compared with the corresponding 7N01 alloy, ZrB_(2) /7N01 composite and Sc/7N01 alloy. And the optimum contents of ZrB_(2) and Sc are 3 wt% and 0.2 wt% in this study. The yield strength, ultimate strength and elongation of(3 wt% ZrB_(2) +0.2 wt% Sc)/7N01 composite are 477 MPa, 506 MPa and 9.8%, increased about 18.1%, 12.2%and 38% compared to 7N01 alloy. Furthermore, the cooperation strengthening mechanisms of ZrB_(2) and Sc are also discussed.

Identified as the pathogenic genes of Alzheimer's disease(AD),APP,PSEN1,and PSEN2 mainly lead to early-onset AD,whose course is more aggressive,and atypical symptoms are more common than sporadic AD.Here,a novel missense mutation,APP E674Q(also named“Shanghai APP”),was detected in a Chinese index patient with typical late-onset AD(LOAD)who developed memory decline in his mid-70s.The results from neuroimaging were consistent with AD,where widespread amyloidβdeposition was demonstrated in 18 F-florbetapir Positron Emission Tomography(PET).APP E674Q is close to theβ-secretase cleavage site and the well-studied Swedish APP mutation(KM670/671NL),which was predicted to be pathogenic in silico.Molecular dynamics simulation indicated that the E674Q mutation resulted in a rearrangement of the interaction mode between APP and BACE1 and that the E674Q mutation was more prone to cleavage by BACE1.The in vitro results suggested that the E674Q mutation was pathogenic by facilitating the BACE1-mediated processing of APP and the production of Aβ.Furthermore,we applied an adeno-associated virus(AAV)-mediated transfer of the human E674Q mutant APP gene to the hippocampi of two-month-old C57Bl/6 J mice.AAV-E674Q-injected mice exhibited impaired learning behavior and increased pathological burden in the brain,implying that the E674Q mutation had a pathogenicity that bore a comparison with the classical Swedish mutation.Collectively,we report a strong amyloidogenic effect of the E674Q substitution in AD.To our knowledge,E674Q is the only pathogenic mutation within the amyloid processing sequence causing LOAD.

Understanding the combustion behavior of electric bicycle batteries and unveiling its relationship with fire extinguishing

In this study,a detailed analysis of the combustion behaviors of the lithium iron phosphate(LFP)and lithium manganese oxide(LMO)batteries used in electric bicycles was conducted.This research included quantitative measurements of the combustion duration,flame height,combustion temperature,heat release rate,and total heat release.The results indicated that LMO batteries exhibited higher combustion temperatures of 600–700°C,flame heights of 70–75 cm,a significantly higher heat release rate of40.1 k W(12 Ah),and a total heat release of 1.04 MJ(12 Ah)compared to LFP batteries with the same capacity.Based on these experimental results,a normalized total heat release(NORTHR)parameter was proposed,demonstrating good universality for batteries with different capacities.Utilizing this parameter,quantitative calculations and optimization of the extinguishing agent dosage were conducted for fires involving these two types of batteries,and the method was validated by extinguishing fires for these two types of battery packs with water-based extinguishing fluids.
