Non-small cell lung cancer(NSCLC)is often characterized by an underlying mutation in the epidermal growth factor receptor(EGFR),contributing to aggressive metastatic *** 2-cyano-3,11-dioxo-18 beta-olean-1,12-dien-30-oate(CdOdA-Me),a glycyrrhetinic acid derivative,reportedly improves the therapeutic response to erlotinib(ERL),an EGFR tyrosine kinase *** the present study,we performed a series of studies to demonstrate the efficacy of CdOdA-Me(2μM)in sensitizing HCC827 R(ERL-resistant)cells to ***,we first established the selectivity of ERL-induced drug resistance in the HCC827 R cells,which was sensitized when ERL was combined with CdOdA-Me(2μM),shifting the IC50 from 23.48μM to 5.46μ***,whole transcriptomic microarray expression data demonstrated that the combination of ERL+CdOdA-Me elicited 210 downregulated genes(0.44%of the whole transcriptome(WT))and 174 upregulated genes(0.36%of the WT),of which approximately 80%were unique to the ERL+CdOdA-Me *** effects centered on losses to cell cycle progression transcripts,a reduction of minichromosome maintenance complex components(MCM2-7),all key components of the Cdc45·MCM2-7 GINS(CMG)complex,and replicative helicases;these effects were tantamount to the upregulation of processes associated with the nuclear factor erythroid 2 like 2 translational response to oxidative stress,including sulfiredoxin 1,heme oxygenase 1,and stress-induced growth inhibitor ***,these findings indicate that the synergistic therapeutic effects of ERL+CdOdA-Me on resistant NSCLC cells are mediated via the inhibition of mitosis and induction of oxidative stress.

The genotoxic effects of quinalpho s(QP) were evaluated in the erythrocyte s of Cyprinus carpio using the comet,micronucleus,and nuclear abnormality *** 96 h LCvalue of quinalpho s,estimated by static renewal system,was 10.9 μL/*** the basis of the LCvalue,two sublethal concentrations(1.09 and 2.18 μL/L) of the compound were used together with cyclophosphamide(5 mg/L) as the positive *** fish were exposed for 35 d,and sampling was done at the end of 7,14,21,28,and 35 *** re sults reveal a significant dose-dependent increase in the frequencies of dNA strand breaks,micronucleus,and nuclear *** responses were also observed in fish exposed to the positive genotoxin *** collective findings demonstrated that quinalphos exerts a genotoxic effect on *** results further confirmed that the comet,micronucleus,and nuclear abnormality assays are useful tools in determining the potential genotoxicity of pesticides towards aquatic organisms.

Effect of angiotensin converting enzyme gene I/d polymorphism in South Indian children with nephrotic syndrome

Nephrotic syndrome is one of the most common childhood kidney diseases. It is mostly found in the age group of 2 to 8 years. Around 10%-15% of nephrotic syndrome cases are non-responders of steroid treatment(SRNS).Angiotensin converting enzyme(ACE)(I/d) gene association studies are important for detecting kidney disease and herein we assessed the association of ACE(I/d) polymorphism with nephrotic syndrome in South Indian children. We recruited 260 nephrotic syndrome(162 boys and 98 girls) and 218(140 boys and 78 girls) control subjects. ACE I/d polymorphism was analyzed by PCR using genotype allele specific primers. In ACE(I/d), we did not find significant association for the ungrouped data of nephrotic syndrome children and the control subjects. Kidney biopsies were done in 86 nephrotic syndrome cases(minimal change disease, n = 51;focal segmental glomerulosclerosis, n = 27;diffuse mesangial proliferation, n = 8). We segregated them into the minimal change disease/focal segmental glomerulosclerosis groups and observed that the ACE’d’ allele was identified with borderline significance in cases of focal segmental glomerulosclerosis and the ’Ⅰ’ allele was assessed as having very weak association in cases of minimal change disease. ’Ⅱ’ genotype was weakly associated with minimal change disease. Gender specific analysis revealed weak association of’Id’ genotype with female nephrotic syndrome in females. dominant expression of dd genotype was observed in males with nephrotic syndrome. Our finding indicated that ACE(I/d) has moderate association with focal segmental glomerulosclerosis. However, due to the limited number of biopsy proven focal segmental glomerulosclerosis subjects enrolled, further studies are required to confirm these results.

Floristic Composition and Change in Species diversity over Long Temporal Scales in Upper Bhotekoshi Hydropower Project Area in Nepal

Forest and vegetation of particular ecosystem is generally influenced by external factors especially from development activities. Nepal has been endowed with an immense variety of forest resources with its unique biodiversity. The current study is conducted in the temperate forest of central Nepal to compare the state of vegetation recorded during 1995 with the present condition. To identify floristic composition, species distribution and diversity, same location and same size of quadrates were selected in natural forest and planted or regenerated area as in previous study. Measurement and analytical tools for forest and vegetation reveled a total of 19 species and 341 individuals in 0.14 ha regenerated area and 18 species, 17 genera and 260 individuals in 0.16 ha, natural forest areas. However, in 1995, total 31 individuals of 20 woody species, 18 genera and 15 families were recorded within the sampling plots of 0.16 ha of natural forest. Moreover, Alnus nepalensis is found dominating regenerated area and Caryopteris odorata in natural forests whereas in 1995, Litsea chartacea and Maesa chisia were recorded dominant. The Shannon’s diversity H’ = 2.908, E = 0.970 and Var H’ = -0.00 and Simpsons d = 0.027 recorded in 1995 is found to be with higher values when compared with the analysis of present study. Furthermore, in comparison to 1995 study, this study showed lower basal area and timber volume with declining species diversity. Although, the natural forest is found with higher species composition compared to rehabilitated forest, species of herbs and shrubs were found extensively growing.

Plasma Sprayed coatings provide protection against corrosion, erosion and wear. Inconel- 718 is a metallic coating which has good wear and erosion resistance. This is plasma sprayed on to Al6061 substrate and then subjected to slurry erosive wear tests. It is observed that the slurry wear rates of Al6061 decreased on coating the substrate material with Inconel- 718.

This work focuses on the efficiency of the LEd acting as the heat sink containing Phase Change Material(PCM). Three different heat sink configurations(H-1, H-2, and H-3) are used in this study. Input power and the number of fins are altered to find their effect on junction temperatures, luminous flux, and thermal resistance. The junction temperature of heat sink H-3 with PCM decreased by 3.1 % when compared with heat sink devoid of PCM at 10 W. The thermal resistance of the heat sink H-3 is reduced by 18.2 % when compared to its counterpart devoid of PCM at 10 W. The luminous flux of the PCM filled heat sink H-3 is found to increase by 12.15 % against the PCM not filled heat sink H-1 at 10 W. The H-3 heat sink with PCM showed superior performance because of the enhanced natural convection and conduction in bulk PCM with fins, and with added high latent heat capacity of PCM.

Role of Intratympanic Steroids in the Management of Idiopathic Sudden Sensori Neural Hearing Loss— Our Experience of 67 Cases

Objective: To assess the outcomes of intratympanic steroids in the management of idiopathic sudden sensorineural hearing loss. Study design: Retrospective study. Setting: Academic tertiary care medical centre. Materials and Methods: A total of 67 patients with idiopathic sudden sensorineural hearing loss were included in our study. Pre- and post-injection audiograms were taken to assess the outcomes in improvement in hearing. Hearing outcomes were assessed with respect to duration of onset of treatment and age of the patient. Improvement in the speech discrimination score was also assessed. Results: A total of 67 patients were included in our study of which unilateral cases were 59 and bilateral cases were 8. Subjectively 35 patients reported improvement in hearing after the therapy while objectively we found 44 ears [41 patients] had improvement. The mean PTA pre ITS was 62.7 and after ITS it was 56.3. Maximum improvement was noted at 2000 Hz and 1000 Hz. Conclusion: Intratympanic steroids can be offered as a first line therapy for idiopathic sudden sensorineural hearing loss as it is minimally invasive, and can be performed as an office based procedure with no systemic side effects. No major complications have been reported in our study and results have been satisfactory.

Mesenchymal stem cell conditioned medium ameliorates diabetic serum-induced insulin resistance in 3T3-L1 cells

Background: Pharmacological factors used to induce insulin resistance (IR) inin vitro models may not mimic the fullin vivo features of type 2 diabetes mellitus (T2dM). This study aimed to examine the ability of diabetic serum (dS) to induce IR and investigate whether adipose-derived mesenchymal stem cell conditioned medium (AdMSC-CM) reverses dS-induced ***: dS was obtained from newly diagnosed T2dM patients. IR was induced in differentiated 3T3-L1 cells by employing dexamethasone, tumor necrosis factor alpha (TNF-α), palmitate and dS. Glucose uptake (2-[N-[7-nitrobenz-2-oxa-1,3-diazol-4-yl] amino]-2-deoxyglucose(2-NBdG) uptake assay), intracellular levels of reactive oxygen species (ROS), and superoxide radicals (O2-) (fluorescence microscopy and fluorometry) were analyzed in control and experimental samples. mRNA expression of key genes involved in glucose transport and inflammation were analyzed by using reverse transcription polymerase chain reaction (RT-PCR). Pro-inflammatory cytokines and phospho-insulin receptor substrate (IRS) (Ser-307) protein expression were analyzed by fluorescence activated cell sorter analysis. Statistical significance was determined by using one-way ANOVA followed by Tukey’s multiple comparison ***: AdMSC-CM significantly increased the dS-mediated decrease in 2-NBdG uptake (11.01 ± 0.50vs. 7.20 ± 0.30,P d reduced dS-driven ROS (fluorescence count, 6.35 ± 0.46vs. 9.80 ± 0.10,P d O2- (fluorescence count, 3.00 ± 0.10vs. 4.60 ± 0.09,P duction. Groundnut(Arachis hypogaea L.)is widely grown and consumed around the world and is considered to have originated from a single hybridization event between two wild *** utilization of wild germplasm in breeding programs has been restricted by reproductive barriers between wild and cultivated species and technical difficulties in making large numbers of *** to overcome these hurdles have resulted in the development of synthetic amphidiploids,namely ISATGR 278-18(Arachis duranesis×Arachis batizocoi)and ISATGR 5B(Arachis magna×***),which possess several desirable traits,including resistance to foliar diseases that generally cause huge yield losses annually in groundnut growing areas of Asia,America,and *** an objective to improve foliar disease resistance,the primary gene pool was diversified by introgressing foliar disease resistance in five cultivated genotypes(ICGV 91114,ICGS 76,ICGV 91278,JL 24,and dH 86)from synthetic amphidiploids using a backcross breeding *** introgression lines with resistance to two foliar diseases(rust and late leaf spot)were identified with levels of resistance equal to the *** backcross derived lines have shown a wide range of variation for several morphological and agronomic *** lines,after further evaluation and selection,can serve as donors in future breeding programs aimed atdeveloping improved cultivars with desirable agronomic traits,high resilience to biotic/abiotic stresses and a broadened genetic base.
