Twenty-four-hour movement guidelines during middle adolescence and their association with glucose outcomes and type 2 diabetes mellitus in adulthood

Purpose:The aim of the present study was to determine the association between adherence to the 24-h movement guidelines during middle adolescence and glucose outcomes(glycated hemoglobin and fasting glucose)and type 2 diabetes mellitus(T2DM)in adulthood,14 and 22 years later.Methods:We analyzed data from apparently healthy adolescents aged 12-18 years who participated in WavesⅠandⅡ(1994-1996,n=14,738),WaveⅣ(2008-2009,n=8913),and Wave V(2016-2018,n=3457)of the National Longitudinal Study of Adolescent to Adult Health(Add Health)in the United States.Physical activity,screen time,and sleep duration were measured using questionnaires,and the 24-h guidelines were defined as:5 or more times moderate-to-vigorous physical activity per week,≤2 h per day of screen time,and 9-11 h of sleep for 12-13 years and 8-10 h for 14-17 years.Capillary and venous whole blood was collected and analyzed to determine glycated hemoglobin and fasting glucose for WavesⅣandⅤ,respectively.Results:Only 2.1%of the adolescents met all the 3 guidelines,and 37.8%met none of them.In both wavesⅣandⅤ,adolescents who met physical activity and screen time guidelines had lower odds of T2DM in adulthood than those who did not meet any of these guidelines(WaveⅣ;prevalence ratio(PR)=0.57,95%confidence interval(95%CI):0.21-0.89;Wave V:PR=0.43,95%CI:0.32-0.74).Only for Wave V did adolescents who met all 3 guidelines have lower odds of T2DM at follow-up compared with those who did not meet any of these guidelines(PR=0.47,95%CI:0.24-0.91).Also,for each increase in meeting one of the 24-h recommendations,the odds of T2DM decreased by 18%(PR=0.82,95%CI:0.61-0.99)and 15%(PR=0.85,95%CI:0.65-0.98)in adulthood for WavesⅣandⅤ,respectively.Conclusion:Promoting all 24-h movement guidelines in adolescence,especially physical activity and screen time,is important for lowering the potential risk of T2DM in adulthood.

Downregulation of miRNA-21 and cancer stem cells after chemotherapy results in better outcome in breast cancer patients

Epigenetic modifications have been observed as a decline in miRNA-21 expression and breast cancer stem cell(CSC)population after 3 cycles of standard chemotherapy.The epigenetic response(miRNAs expression)and CSCs are also correlated in patients with Breast Cancer.In patients who tolerated chemotherapy well,miRNA-21(non-coding RNA)expression decreased significantly after three cycles of chemotherapy.The miRNA-21 expression in breast cancer tissue was quantified by quantitative PCR(real-time PCR)using the standard protocol.In addition,breast CSCs(CD44+/CD24-)were also decreased in these patients.The miRNA-21 regulates cell division,proliferation,and autophagy of cancerous cells(as it targets phosphatase and tensin homolog/AKT/transcription factor EB/programmed cell death 4/autophagy-related protein 5 and chemotherapy also produces similar effects),thereby contributing to these benefits.Therefore,when all of the targets on genes have been explored by mimic miRNA,chemotherapy combined with anti-miRNA21 therapy may prove useful in the care of cancer patients.

A computer vision system for early detection of anthracnose in sugar mango(Mangifera indica)based on UV-A illumination

The present work describes the development of a computer vision system for the early detection of anthracnose in sugar mango based on Ultraviolet A illumination(UV-A).Anthracnose,a disease caused by the fungus Colletotrichum sp,is commonly found in the fruit of sugar mango(Mangifera indica).It manifests as surface defects including black spots and is responsible for reducing the quality of the fruit.Consequently,it decreases its commercial value.In more detail,this study poses a system that begins with image acquisition under white and ultraviolet illumination.Furthermore,it proposes to analyze the Red,Green and Blue color information(R,G,B)of the pixels under two types of illumination,using four different methods:RGB-threshold,RGB-Linear Discriminant Analysis(RGBLDA),UV-LDA,and UV-threshold.This analysis produces an early semantic segmentation of healthy and diseased areas of the mango image.The results showed that the combination of the linear discriminant analysis(LDA)and UV-A light(called UV-LDA method)in sugar mango images allows early detection of anthracnose.Particularly,this method achieves the identification of the disease one day earlier than by an expert with respect to the scale of anthracnose severity implemented in this work.

Effect of Ultraviolet Radiation on Hsp70 Protein Expression in HaCaT Cells

Ultraviolet radiation by its wavelength is divided into: UVA, UVB and UVC. Only UVA and UVB manage to penetrate the ozone layer, but due to anthropological activities, all of them are capable of interacting with humans to a greater or lesser extent, and can generate adverse effects such as cellular stress when interacting with intra-and extracellular biomolecules. The skin is the first organ in contact with UV radiation, and the stress it generates can be analyzed by the expression of a bioindicator of cellular damage such as Hsp70. Therefore, the objective of the project was: to determine the effect of UVA, UVB and UVC radiation on HaCaT epithelial cells, by analyzing the expression of Hsp70. Materials and methods: HaCaT cells were cultured in vitro, which were irradiated with UVA, UVB and UVC light at different doses, to subsequently determine the degree of Hsp70 expression by Immunodetection by PAGE-SDS and Western Blot. Results: Basal expression of Hsp70 was observed in no irradiated HaCaT cells. When HaCaT cells were irradiated with UVA, UVB, UVC, an increase in this Hsp70 protein was observed. With UVA, a higher degree of expression was observed at a time of 30 minutes of irradiation. With UVB the highest expression shifted to a time of 20 minutes. With UVC, overexpression was observed after 10 minutes. Conclusion: UV radiation generates cellular stress on HaCaT cells, evaluated by the stress bioindicator Hsp70. According to the wavelength of UV radiation, those that have a shorter wavelength have a greater potential for cellular damage, such as UVC.

Hemodynamic instability following intravenous dexmedetomidine infusion for sedation under brachial plexus block: Two case reports

BACKGROUND Dexmedetomidine(DMED)is frequently used as a sedative in several medical fields.The benefits of DMED include enhanced quality of regional anesthesia,prolonged analgesia,and postoperative opioid-sparing when administered intravenously or perineurally in combination with regional anesthesia.Severe hemodynamic complications,such as profound bradycardia and hypotension,can occur after DMED administration in critically ill patients or overdosage;however,there are few reports of complications with DMED administration following brachial plexus block(BPB).CASE SUMMARY We present two cases of hemodynamic instability that occurred following the initial loading of DMED under supraclavicular BPB.A healthy 29-year-old man without any medical history showed profound bradycardia after receiving a loading dose of DMED 0.9μg/kg for 9 min.DMED administration was promptly stopped,and after receiving a second dose of atropine,the heart rate recovered.A 62-year-old woman with a history of cardiomyopathy became hypotensive abruptly,requiring the administration of inotrope and vasopressors after receiving a reduced loading dose of 0.5μg/kg for 10 min.Half of the recommended loading dose of DMED was administered due to the underlying heart dysfunction.Decrea-sed blood pressure was maintained despite the intravenous administration of ephedrine.With continuous infusion of dopamine and norepinephrine,the vital signs were maintained within normal ranges.Inotropic and vasopressor support was required for over 6 h after the initial loading dose of DMED.CONCLUSION DMED administration following BPB could trigger hemodynamic instability in patients with decreased cardiac function as well as in healthy individuals.

Effects of mechanical vibration on the physical,metallurgical and mechanical properties of cast A308(LM21)aluminum alloy

This study investigated the microstructure,physical,and mechanical properties of die-cast A308 alloy subjected to mechanical vibration during solidification.Different frequencies(0,20,30,40,and 50 Hz)at constant amplitude(31μm)were employed using a power amplifier as the power input device.X-ray diffraction,optical microscopy,and scanning electron microscopy were used to examine the morphological changes in the cast samples under stationary and vibratory conditions.Metallurgical features of the castings were evaluated using Image J software.The average values of metallurgical features,including primaryα-Al grain size,dendrite arm spacing,average area of eutectic silicon,aspect ratio,and percentage porosity,reduced by 34%,59%,56%,22%,and 62%,respectively,at 30 Hz frequency compared with stationary casting.Mechanical tests of the cast samples showed that the yield strength(YS),ultimate tensile strength(UTS),percentage elongation(%EL),and microhardness(HV)increased by 8%,13%,17%,and 16%,respectively,at 30 Hz frequency compared with stationary casting.The fractured surface of the tensile specimens exhibited mixed-mode fracture behavior because of brittle facets,cleavage facets,ductile tearing,and dimple morphologies.The presence of small dimples showed that plastic deformation occurred before fracture.

Comparative study between intramedullary interlocking nailing and minimally invasive percutaneous plate osteosynthesis for distal tibia extra-articular fractures

Purpose:Treatment of distal tibia fractures poses significant challenge to orthopedic surgeon because of poor blood supply and paucity of soft tissue coverage.There is considerable controversy regarding the superior option of treatment for distal tibia fracture between the minimally invasive percutaneous plate osteosynthesis(MIPPO)technique and intramedullary interlocking(IMIL)nailing for extra-articular distal tibia fractures.The aim of our study is to compare the functional outcome between the two treatment methods.Methods:This was the prospective comparative study of 100 patients with distal third tibia fractures divided into two groups.The first group of patients were treated with MIPPO technique while the second group of patients were managed by IMIL nailing.Patients were followed up in outpatient department to assess the functional outcomes,malunion,delayed union,nonunion,superficial and deep infection between the two groups.Statistical analyses were performed using the SPSS software(version 16.0).Results:Average malunion(degrees)in the MIPPO group was 5(3—7)±1.41 vs.10.22(8—14)±2.04 in the IMIL group(p=0.001).Similarly postoperative knee pain in the IMIL group was 10%vs.2%in the MIPPO group(p=0.001).In terms of superficial infection and nonunion,the results were 8%vs.4%and 2%vs.6%for the MIPPO and IMIL group,respectively(p=0.001).Conclusion:Both procedures have shown the reliable method of fixation for distal extra-articular tibia fractures preserving the soft tissue,bony vascularity and fracture hematoma that provide a favourable biological environment for fracture healing.Considering the results of the study,we have slightly more preference for the MIPPO technique.

Physical Characterization of Ionic Liquid-Modified Polyvinyl Alcohol and Sodium Thiocyanate Polymer Electrolytes for Electrochemical Double-Layer Capacitor Application

Novel gel polymer electrolytes(GPEs)composed of polyvinyl alcohol(PVA)and sodium thiocyanate were developed via a solution casting technique.An ionic liquid(IL),1-ethyl-3-methyl-imidazolium tricyanomethanide([EMIM][TCM]),was doped into a polymer–salt complex system(PVA+NaSCN)to further enhance the conductivity.IL-doped polymer electrolyte(ILDPE)films were characterized using X-ray diffraction(XRD),polarized optical microscopy(POM),Fourier-transform infrared(FTIR)spectroscopy,and conductivity measurements.XRD was performed to check the degree of crystallinity and amorphicity of the ILDPE films,and the amorphicity of GPEs increased with the increase of the IL content.POM was employed to evaluate the changes in the surface morphology due to the inclusion of salt and IL in the PVA.The compositional nature of the GPE films was examined via FTIR studies.The electrical and electrochemical properties were characterized by cyclic voltammetry and electrochemical impedance spectroscopy.The maximum conductivity for the GPE film was estimated to be 1.10×10^(-5) S/cm for 6%(mass fraction)of IL in the polymer–salt complex.The ionic transference number was approximately 0.97.An electrochemical double-layer capacitor(EDLC)was built from optimized GPE films and reduced graphene oxide-based electrodes.The specific capacitance calculated from the cyclic voltammograms of the EDLC cells was 3 F/g.

Non-invasive cuffless blood pressure and heart rate monitoring using impedance cardiography

Background Continuous blood pressure(BP)monitoring provides additional information about how changes in BP may correlate with daily activities and sleep patterns.Recommendations from the American Heart Association and American College of Cardiology strongly suggest confirming a diagnosis of hypertension with continuous BP monitoring.Non-invasive and non-intrusive detection of haemodynamic parameters is emerging as a norm,based on self-monitoring wearable medical devices.Researchers have carried out several studies using non-invasive and continuous BP measurements as an alternative to conventional cuff-based measurements.In this work,we proposed a novel method for cuffless estimation of BP using impedance cardiography(ICG).Methods We conducted a single-centre,cross-sectional study of 104 subjects(of whom 30 were categorized as controls and the remaining 74 as the disease group)at the Medical College and Hospital,Kolkata.The disease group consisted of patients with confirmed coronary artery disease,while the individuals in the control group were deemed to be healthy.All subjects underwent electrocardiogram recording by on-duty doctors in order to determine their health status.A custom-made device based on the principle of impedance plethysmography was designed to record impedance changes due to subjects’peripheral blood flow.The device was used to record ICG signals.In this study,we developed a novel auto-adaptive algorithm based on ICG signals for non-invasive,cuffless,continuous monitoring of BP and heart rate.Separate mathematical models were developed for all the estimated parameters(BP and heart rate)for both the study groups(control and disease).The developed models were auto-adaptive and did not require subject-specific calibration.Performance indicators including,𝑟2,error percentage,standard deviation,and mean difference were used to quantify the performance of the models.Results The ICG signal recorded by the device was used to extract features and compute the augmentation index.The calculated augmentation

Synthesis and characterization of red-emitting Yb/Ho-CaSiO3 upconversion phosphors

Host material plays an important role in obtaining efficient photon upconversion and downshifting luminescence.Generally, fluoride and oxyfluoride glasses-based materials are used for high-efficiency photon upconversion.However, the poor thermal stability of fluoride glasses and the toxicity of fluorine ions limit their applications. In this report, Yb/Ho-doped CaSiO3wollastonite phosphors have been demonstrated as efficient red-emitting upconversion phosphors. The phosphors have been synthesized by microwave hydrothermal process followed by heat treatment at 1050°C in the air environment. 2M phase of the β-wollastonite has been confirmed by X-ray diffraction and raman spectroscopy while the existence of the Yb and Ho dopants in the CaSiO3lattice has been confirmed by X-ray photoelectron spectroscopy. The synthesized samples showed strong red upconversion emission centered at 662 nm and near-infrared downshifting emissions at 2016 nm upon 980 nm excitation. The emissions were found to depend significantly on the Ho concentration. Temporal evolution of the main emission bands was investigated to show that the energy transfer upconversion from Yb to Ho ions was responsible for the efficient upconversion and downshifting phenomenon.
