Progressive deterioration of physical work capacity in congestive heart failure (CHF) is often attributed to ongoing skeletal muscle atrophy and abnormalities in muscle metabolism. The purpose of the present study was to investigate if mutations in the p53 gene thought to be a promotor of apoptosis are involved in intrinsic apoptotic abnormalities in skeletal muscle of patients (pts) with CHF. Percutaneous needle biopsy from the m. vastus lateralis were obtained from 19 pts with CHF (Lv EF 25%±10%). Single strand confirmation polymorphism analysis of polymerase chain reation products (PCR SSCP analysis) was used for detection of mutations in exon 5 8 of the p53 gene in skeletal Heart Center, University Leipzig, Germany (Yu JT, Adams v, Fiehn E, Schuler G and Hambrecht R) Institut of Pathology, University Freiburg, Germany (Ye J and Riede U)muscle cells. Four of 19 muscle specimens (21%) showed mobility shifts. To characterize the nuleotide sequence alterations specimens were examined further by direct sequence analysis of PCR product. Two of four specimens showing a band shift in the SSCP analysis exhibited a mutated p53 sequence. Sequence analysis revealed that these alteratons were point mutation exon 8 (14482, G→A) and deletion in exon 5 (13143 13157). A high frequency of p53 mutations was detected in skeletal muscle cells of patients with chronic heart failure. These findings suggest a role for apoptosis in the progression of intrinsic skeletal muscle abnormalities and consequently of exercise intolerance in chronic heart failure.

为系统、深入地研究中国西部盆(盆地)、山(山脉)、原(高原)的壳幔结构与深部动力学过程,2003年我们提出并领导实施了“羚羊计划”(ANTILOPE-Array Network of Tibetan International Lithospheric Observation and Probe Experiments),在青藏高原先后完成了羚羊-I(ANTILOPE-I)到羚羊-Iv(ANTILOPE-Iv)4条二维宽频带台阵剖面,而在青藏高原东西构造结则实施了羚羊-v和羚羊-vI两个三维宽频带台阵探测。另外,我们将前期在准噶尔盆地、天山造山带、塔里木盆地、阿尔金造山带和柴达木盆地开展的九条综合地球物理观测剖面也纳入羚羊计划的总体框架中来。通过“羚羊计划”的实施,我们在中国西部(包括西北部的环青藏高原盆山体系以及西南部的青藏高原)取得了大量的、高质量的、综合的第一手观测数据,获得了中国西部盆、山、原精细的壳幔结构,系统地揭示了中国西部盆山原的深部地球动力学过程。主要结论总结如下:确定了准噶尔盆地基底的结构与属性,优化了盆地的基底构造格架;建立了天山造山带“层间插入削减”新的陆内造山模式,揭示了印欧碰撞在天山岩石圈缩短44%的去向以及由洋陆俯冲到陆陆碰撞俯冲的转换机制;揭示了塔里木盆地、阿尔金造山带和柴达木盆地的盆山接触关系;获得了塔里木盆地顺时针旋转的深部几何学、运动学和动力学证据;确定了青藏高原之下印度板块与欧亚板块的碰撞边界;发现目前的青藏高原由南部的印度板块、北部的欧亚板块和夹持于二者之间的巨型破碎区——西藏“板块”构成,首次确定了各自的岩石圈底边界;修正了高原变形的两个端员模型;建立了深部构造对地表地形的制约关系;系统地揭示了印度板块沿喜马拉雅造山带俯冲的水平距离与俯冲角度的变化规律与控制因素。“羚羊计划”以其巨大的观测网络与综合地球物理探测技术,采用地球物理学、地质学、地球化学等不同学科相结合的分析方法,揭示了印度板块俯冲、西藏巨型破碎区发育、塔里木板块顺时针旋转、西部水汽通道提前关闭、中国西北部干旱、沙漠化提前这一深部结构、动力学过程及其对地表地形、油气资源和环境变化的制约关系,推动了青藏高原地球系统科学理论的发展。

Mineral and bone disorder(MBD)in chronic kidney disease(CKD)is tightly linked to cardiovascular disease(CvD).In this study,we aimed to compare the prognostic value of nine MBD biomarkers to determine those associated best with adverse cardiovascular(Cv)outcomes and *** 5217 participants of the German CKD(GCKD)study enrolled with an estimated glomerular filtration rate(eG FR)between 30–60 mL·min-1 per 1.73 m2 or overt proteinuria,serum osteoprotegerin(OPG),C-terminal fibroblast growth factor-23(FGF23),intact parathyroid hormone(iP TH),bone alkaline phosphatase(BAP),cross-linked C-telopeptide of type 1 collagen(CTX1),procollagen 1intact N-terminal propeptide(P1NP),phosphate,calcium,and 25-OH vitamin D were measured at *** with missing values among these parameters(n=971)were excluded,leaving a total of 4246 participants for *** a median follow-up of6.5 years,387 non-Cv deaths,173 Cv deaths,645 nonfatal major adverse Cv events(MACEs)and 368 hospitalizations for congestive heart failure(CHF)were *** and FGF23 were associated with all outcomes,with the highest hazard ratios(HRs)for *** the final Cox regression model,adjusted for Cv risk factors,including kidney function and all other investigated biomarkers,each standard deviation increase in OPG was associated with non-Cv death(HR 1.76,95%CI:1.35–2.30),Cv death(HR 2.18,95%CI:1.50–3.16),MACE(HR 1.38,95%CI:1.12–1.71)and hospitalization for CHF(HR 2.05,95%CI:1.56–2.69).Out of the nine biomarkers examined,stratification based on serum OPG best identified the CKD patients who were at the highest risk for any adverse Cv outcome and mortality.

JUNO is a multi-purpose neutrino observatory under construction in the south of *** publication presents new sensitivity estimates for the measurement of the △m_(31)^(2),△m_(21)^(2),sin^(2)θ_(12),and sin^(2)θ_(13) oscillation parameters using reactor antineutrinos,which is one of the primary physics goals of the *** sensitivities are obtained using the best knowledge available to date on the location and overburden of the experimental site,the nuclear reactors in the surrounding area and beyond,the detector response uncertainties,and the reactor antineutrino spectral shape constraints expected from the TAO satellite *** is found that the △m_(21)^(2) and sin^(2)θ_(12) oscillation parameters will be determined to 0.5%precision or better in six years of data *** the same period,the △m_(31)^(2) parameter will be determined to about 0.2%precision for each mass ordering *** new precision represents approximately an order of magnitude improvement over existing constraints for these three parameters.

The Jiangmen Underground Neutrino Observatory(JUNO)is a large liquid scintillator detector designed to explore many topics in fundamental *** this study,the potential of searching for proton decay in the p→νK^(+)mode with JUNO is *** kaon and its decay particles feature a clear three-fold coincidence signature that results in a high efficiency for ***,the excellent energy resolution of JUNO permits suppression of the sizable background caused by other delayed *** on these advantages,the detection efficiency for the proton decay via p→νK^(+)is 36.9%±4.9%with a background level of 0.2±0.05(syst)±0.2(stat)events after 10 years of data *** estimated sensitivity based on 200 kton-years of exposure is 9.6×1033 years,which is competitive with the current best limits on the proton lifetime in this channel and complements the use of different detection technologies.
