In current study toxicological profile of a commonly used herbal formulation was evaluated that is used extensively for gynecological disorders like menorrhagia, metrorrhagia, leucorrhea, irregular menstrual cycle, pre-menstrual syndrome and post-menopausal bleeding. It was also claimed to strengthen endometrium and ovaries. Since this herbal formulation was been used by a large number of population hence there was a need to assess acute and sub-chronic toxicity. Acute oral toxicity (LD50) was observed in albino mice using standard protocols whereas sub-chronic, hematological and histopathological studies were assessed on 24 albino rabbits after giving herbal formulation for 60 days in two doses (20 and 60 mg/kg) against control groups. The outcomes of present study showed that the drug is safe up to 5000 mg/kg following acute oral toxicity test and no mortality was observed during sub chronic toxicity studies. Results of sub-chronic toxicity did not show any significant changes in biochemical, hematological and histopathological parameters. However, some indicators such as urea, creatinine, hemoglobin, and RBC count were altered, but these changes do not correlate with the histopathological results and may be associated to intra individual variations. Despite the safety of the drug in few animals, clinical trials and more investigations on a large number of animals are essentially needed to establish safety and efficacy of the herbal formulation.

Diuretics are efficaciously used in management of various clinical emergencies like hypertension, heart failure, cirrhosis, hypercalciuria, hematuria and nephrotic syndrome. Cymbopogon jwarancusa is an aromatic perennial grass used in both traditional and Unani system of medicine to eradicate diseases like colds, seasonal fever, asthma, tuberculosis, rheumatic pain, back pain, toothache and nervous disorders. C. jwarancusa essential oils are used in perfumery, soap, detergents, medicines and pharmaceutical industry. Monoterpenes and sesquiterpenes constitute the highest composition in essential oil of C. jwarancusa. The present was designed to compare the diuretic activity of C. jwarancusa after single and multi-doses. Furosemide (20 mg/kg) was used as reference drug and 10% DMSO was used as vehicle. Diuretic activity was noticed by measuring urine volume and calculating diuretic and Lipchitz values. Maximum diuretic response was observed at 500 mg/kg of extract after both single and multi-dose administration. On basis of results it may be concluded that C. jwarancusa may be used as diuretic agent.

Evaluation of the Anti-Hyperlipidemic Activity of Nelumbo nucifera Fruit in Rabbits Fed with High Cholesterol Diet

Recently use of herbal therapies and diet rich in flavonoids and vitamins has augmented significantly to manage minor to modest illnesses. Several anti-hyperlipidemic agents are currently available;however most of them have various unwanted effects. That’s why more people are switching towards safer alternatives, specially derived from plants with fewer side effects. Hence current study was focused to determine the anti-hyperlipidemic potential of Nelumbo nucifera fruit in rabbits fed with high cholesterol diet. This study would surely help in the exploration of a novel agent from the plant source and therefore may recommend the application of traditional medicines in humans. 28 white healthy rabbits of either sex weighing 1000 - 1500 gm were divided into four groups. Control group was given gum tragacanth and 3 groups served as test groups and were given N. nucifera fruit extract 50, 100 and 200 mg/kg for 45 days. Initially animals of all four groups received high cholesterol diet (HCD) 0.125 gm/kg cholesterol in 0.5% corn oil for 30 days. All drugs were given orally. Blood samples were taken thrice from the ear vein of animals, 1st after 24 hours of thirty days of HCD then again after 24 hours of thirty and forty five days dosing of N. nucifera fruit extract. The results showed remarkable cholesterol lowering effects of N. nucifera fruit extract at doses 50, 100 and 200 mg/kg but were particularly dominant at 200 mg/kg at which fruit extract highly significantly reduced TC, TG and LDL-C after 30 and 45 days and increased HDL-C significantly after 30 days and highly significantly after 45 days. Flavonoids, saponins, tannins and procyanidin are significant constituents of N. nucifera fruit which may have contributed in lipid lowering effects of N. nucifera fruit which can contribute in lowering the risk of CVDs and stroke but more preclinical and clinical studies are required to confirm these findings.

Medicinal plants are under continuous analysis worldwide to explore new medication to treat atherosclerosis and cardiovascular diseases. Citrus fruits have displayed utmost antioxidant activity, particularly flavonoids in Citrus paradisi have exhibited strong antioxidant and anti-inflammatory characteristics. This study was intended to examine effects of C. paradisi on blood coagulation and anticoagulation at doses of 0.1, 0.3 and 0.5 ml/kg. Highly significant increase in thrombin time (TT) and substantial decline in fibrinogen (Fb) level was noticed as related to control. There was substantial increase in TT, prothrombin time (PT) and activated partial thromboplastin time (aPTT) and noteworthy decrease in Fb level by warfarin. C. paradisi at 0.3 ml/kg considerably inhibited aggregation of platelet by collagen, adenosine phosphate, arachidonic acid and epinephrine, while there was also substantial rise in Protein C, TAT complex, RBC and hemoglobin concentration. In conclusion, these results suggest that the addition of C. paradisi to the diet of patients at risk of developing cardiovascular events could significantly decrease the morbidity or mortality in these patients due to high contents of phytochemicals like naringin, hesperidin, limonene and other flavonoids.

Analgesic Activities of Methanol Extract of Terminalia chebulaFruit

Pain is a sensation which in many cases represents the only symptom for the diagnosis of several disorders. Throughout ages man has used many different remedies for pain relief, among which medicinal herbs are most common due to their efficacy and relative safety due to their natural origin. In present study methanol extract of Terminalia chebula fruit was evaluated for its analgesic effects in albino mice. The extract was prepared using whole dried powdered fruit with seeds in absolute methanol. It was then subjected to rotary evaporator under reduced pressure to get concentrated extract;different doses (300, 500 and 1000 mg/kg) were assessed for analgesic activity by tail immersion technique and acetic acid induced writhing test. Similar doses of Aspirin were used as standard drug (positive control) for comparison. Results of both tail flick method and acetic acid induced writhing test revealed that T. chebula fruit extract possessed varying degree of analgesic activity significant at 300 mg/kg and highly significant at 500 and 1000 mg/kg in comparison to control. The results were almost similar to standard drug. In acetic acid induced writhing test, maximum inhibition of writhing was observed at 1000 mg/kg where the number of writhes decreased from 14.1 to 5.2 indicating 63.1% inhibition. Above findings suggest that T. chebula fruit extract possesses significant analgesic activity in albino mice with reference to positive and negative control groups;however further studies on a large number of animals with clinical trials are required to confirm safe and effective use of this fruit extract in humans for analgesic purpose.
