Temperature Gradient Gel Electrophoresis as a Valuable Accessory Tool for Assessment of Dysbiosis in Crohn’s Disease

Escherichia coli and other Proteobacteria are augmented and several other bacteria are diminished in Crohn’s (CD) disease patients’ intestine. This imbalance in bacterial species composition—termed dysbiosis—seems to be determinant of CD manifestation. Since a great part of intestinal bacteria are not cultivable, detection of CD dysbiosis is accomplished by molecular tools, involving sequences analysis of the 16SrRNA gene (16SrDNA) present in the patient’s clinical samples, which can be done by sequencing or electrophoresis in denaturing gels of 16SrDNA amplicons. By analyzing, by temperature gradient gel electrophoresis (TGGE) and next generation sequencing of 16SV6-V8rDNA amplicons present in gram negative cultures from four distinct clinical samples of a control subject and a CD patient, this study demonstrates that both techniques were able to detect E. coli overgrowth and reduction in species richness in CD and that TGGE can discriminate sequences collectively labeled as “unclassified” in 16SrDNA databases. Although TGGE per se does not identify the sequences, the discriminatory power that it confers represents valuable accessory information to next generation DNA sequencing (NGS), and as such must be used as a NGS complementary tool.

<i>In Vitro</i>Priming of Sugarcane Varieties (RB966928 and RB867515)

Sugarcane, as a glycophyte, shows sensitivity to saline soils at various stages of its growth. The aim of this study was to evaluate the in vitro priming response in two sugarcane varieties (RB966928 and RB867515). Micropropagated plants, from meristems, received priming treatments by adding the salts (NaCl and kCl) in different concentrations (0.0;12.5;25.0 and 50.0 mM), in the MS medium. Subsequently, the plants were cultivated in rooting medium without addition of salts, acclimatized and submitted to gradient ex vitro saline stress with 20 → 40 and 60 mM, of each salt, for 30 days. The analyzed variables were dry matter of shoot and root, number of tillers and estimation of chlorophyll content. The experiment was carried out in a 2 × 2 × 4 factorial arrangement, in a completely randomized experimental design. Twenty replicates were used throughout the experiment. Data were submitted to analysis of variance and regression and the means were compared by Tukey’s test, at a 5% probability level. The priming treatments presented a significant effect, with triple interaction, in the chlorophyll index. In the treatment with NaCl, the variety RB966928 showed an increase in the chlorophyll index with the increase of treatment levels, up to an optimal limit of 31.47 mM. On the other hand, the variety RB867515 showed decreasing in chlorophyll index. In contrast, in kCl treatment, the variety RB867515 presented the increase at the chlorophyll index with the maximum point of 25 mM. For the variables, shoot dry matter (SDM) and root dry matter (RDM) there was a significant difference (p 0.05 and p 0.01, respectively) only between the varieties. The variety RB966928 presented higher SDM and RDM in relation to the variety RB867515. Studies are recommended with increasing the duration of the priming treatments and more detailed study of the culture throughout its productive cycle.

Initial Growth and Gas Exchanges of Plants of Colored Cotton Submitted to Saline Stress

The objective of the research was to evaluate the effects of saline stress on the initial growth and physiological parameters of three varieties of cotton plants, being two cultivars of colored fiber (BRS Topázio and BRS Verde) and one of white fiber (BRS 286). The experiment was conducted under greenhouse conditions. After chemical depletion with sulfuric acid, five seeds were seeded in polyethylene pots of 20 kg, leaving only one plant per pot after thinning. The experimental design was completely randomized, with a factorial scheme of 3 × 5, with five replications, totaling 75 plots, being three cotton varieties and five saline stress treatments—1.1;3.1;5.1;7.1;9.1 dS m−1. The evaluations occurred at 15 and 30 days after the beginning of the treatments (DAT) for growth variables (plant height, stem diameter, number of leaves and leaf area) and gas exchange (stomatal conductance, net photosynthesis rate and transpiration. The variety that was most resistant to the salt stress levels studied was BRS 286, followed by BRS Topazio and BRS Verde.

Carbohydrates, Growth and Production of “Roxo de Valinhos” Fig Tree in Initial Development under Irrigation Management

The carbohydrates translocation and consequently growth and production of fig tree (Ficuscarica L.) vary according to the different management on cultivation conditions. The aim of this study was to evaluate the changes in the levels and total carbohydrates accumulation together with growth and “Roxo de Valinhos” fig trees production onimplementation of orchards in initial phase, cultivated with and without irrigation. We adopted a factorial arrangement (2 x 7) with four repetitions distributed in installments (with and without irrigation) subdivided in time (collect time). Destructive analyzes were performed at 40, 80, 120, 160, 200, 240 and 280 days after pruning (DAP) and are measured: stem diameter and branch, stem length and branch, number of leaves, internodes and fruit. Subsequently, the plant parts were sectioned to obtain the leaf area, length and roots volume, fresh and dry matter weight. The number, weight and total productivity of fruits were evaluated. The media of all growth attributes and production characteristics were higher in treatments with water irrigation. The total carbohydrate content was higher at 120 and 160 DAP and the carbohydrates accumulation was increasing for most institutions over the plants development, except for the leaves that showed a decrease in the levels at 160 DAP. The fruits showed greater carbohydrates accumulation in relation to the other evaluated organs.

UPLC-QTOF-MS Analysis of Extracts from the Leaves of <i>Pouteria caimito</i>(Sapotaceae) and Their Antioxidant Activity

This study describes the phytochemical profile and antioxidant activity of an extract from the leaves of Pouteria caimito (Sapotaceae). The extract in ethanol was obtained by maceration at room temperature and subjected to the liquid-liquid partition to obtain fractions in hexane and ethyl acetate. Steroids, triterpenes, saponins, alkaloids and flavonoids were identified by the phytochemical prospection of extracts and fractions from the leaves. The analysis of the ethyl acetate fraction by UPLC-QTOF-MS allowed us to identify eight triterpenes, namely, euscaphic acid (1), hyptadienic acid (2), betulinic acid (3), oleanolic acid (4), ursolic acid (5), 3β-(O-p-coumaroyl)- alphitolic acid (6), 3β-(O-p-coumaroyl)-maslinic acid (7) and 3β-(O-p-coumaroyl)-2-hydroxy-urs-12-en-28-oic acid (8). The ethanol extract and ethyl acetate fraction presented total phenolic contents of 10.6 ± 0.1 and 11.4 ± 0.3 mg GAE g-1, respectively, and considerable antioxidant activity in the DPPH assay with EC50 values of 299.4 ± 1.5 and 391.8 ± 0.9 μg·ml-1, respectively.

Labor Pain through the Eyes of Brazilian Women and Delivery Nurses

Objective: Understanding the perception of women who have recently given birth in a Brazilian hospital regarding labor pain and nursing involvement in the labor process. Methodology: exploratory, descriptive and qualitative study carried out in a Maternity part of the supplementary health system in the Federal District (DF). Data were collected between May and July 2015 through interviews using a semi-structured questionnaire. The sample consisted of 16 mothers and data analysis followed Bardin methodology in three phases. Results: The pain experienced during labor is surrounded by feelings and expectations. Both positive and negative feelings experienced by mothers were identified in the interviews and unanimous opinion was the fact that the second stage is the most painful period as continuous pain is experienced. Some of the participants compared pain with strong menstrual cramps, renal colic and back pain;others, said labor pain was the strongest ever experienced. The immediate contact with the newborn was referred to as a reward for coping with breast pain and suffering. Other factors such as the importance of follow-up, support and coping with the pain and care provided by health staff were also highlighted. Conclusion: empowerment of the mother and family through knowledge is considered an important tool to fight pain and cope with the birth process in a positive and physiological way, which in the long run promotes a cultural change in the population regarding the importance of vaginal birth.

Synthesis and Characterization of LaNixCo1-xO3 Perovskites via Complex Precursor Methods

This work presents a study on the synthesis of LaNixCo1-xO3 perovskites via complex precursor methods. Perovskite oxides with the composition LaNixCo1-xO3 were synthesized by chelating precursor and polymeric precursor methods using nickel and/or cobalt nitrates, lanthanum nitrate, ethylene glycol, citric acid, and EDTA as starting source. The obtained perovskite were characterized by thermogravimetric analysis, infrared spectroscopy, X-ray diffraction and the morphology of the samples were investigated by N2 adsorption experiments and average medium particle size. TG curves and FTIR spectra were particularly useful in establishing of the optimal calcination temperature of the precursor powders. X-ray diffraction patterns revealed the formation of the perovskite structure in all samples prepared by both synthesis method and after calcinations at 700℃. The results showed that the preparation method resulted in oxides with the intended structure. The specific surface area values were influenced by preparation method.
