<i>In Vitro</i>Priming of Sugarcane Varieties (RB966928 and RB867515)

Sugarcane, as a glycophyte, shows sensitivity to saline soils at various stages of its growth. The aim of this study was to evaluate the in vitro priming response in two sugarcane varieties (RB966928 and RB867515). Micropropagated plants, from meristems, received priming treatments by adding the salts (NaCl and KCl) in different concentrations (0.0;12.5;25.0 and 50.0 mM), in the MS medium. Subsequently, the plants were cultivated in rooting medium without addition of salts, acclimatized and submitted to gradient ex vitro saline stress with 20 → 40 and 60 mM, of each salt, for 30 days. The analyzed variables were dry matter of shoot and root, number of tillers and estimation of chlorophyll content. The experiment was carried out in a 2 × 2 × 4 factorial arrangement, in a completely randomized experimental design. Twenty replicates were used throughout the experiment. Data were submitted to analysis of variance and regression and the means were compared by Tukey’s test, at a 5% probability level. The priming treatments presented a significant effect, with triple interaction, in the chlorophyll index. In the treatment with NaCl, the variety RB966928 showed an increase in the chlorophyll index with the increase of treatment levels, up to an optimal limit of 31.47 mM. On the other hand, the variety RB867515 showed decreasing in chlorophyll index. In contrast, in KCl treatment, the variety RB867515 presented the increase at the chlorophyll index with the maximum point of 25 mM. For the variables, shoot dry matter (SDM) and root dry matter (RDM) there was a significant difference (p 0.05 and p 0.01, respectively) only between the varieties. The variety RB966928 presented higher SDM and RDM in relation to the variety RB867515. Studies are recommended with increasing the duration of the priming treatments and more detailed study of the culture throughout its productive cycle.

Esophageal stenosis with sloughing esophagitis:A curious manifestation of graft-vs-host disease

We report a case of a 56-year-old woman with a history of allogenic bone marrow transplantation for two years,complaining with dysphagia and weight loss. Upper endoscopy revealed esophageal stenosis and extensive mucosa sloughing. Biopsies confirmed the diagnosis of graft-vs-host disease(GVHD). Balloon dilation,corticosteroids and cyclosporin resulted in marked clinical improvement. Gastrointestinal tract is involved in the majority of patients with chronic GVHD. Esophageal manifestations are rare and include vesiculobullous disease,ulceration,esophageal webs,casts or strictures. Sloughing esophagitis along with severe stenosis requiring endoscopic dilation has never been reported in this context.

Application of RNA interference in triatomine （Hemiptera： Reduviidae） studies

Triatomines （Hemiptera： Reduviidae） are obligate hematophagous insects. They are of medical importance because they are vectors of Trypanosoma cruzi, the causative agent of Chagas disease in the Americas. In recent years, the RNA interference （RNAi） technology has emerged as a practical and useful alternative means of studying gene function in insects, including triatomine bugs. RNAi research in triatomines is still in its early stages, several issues still need to be elucidated, including the description of the molecules involved in the RNAi machinery and aspects related to phenotype evaluation and persistence of the knockdown in different tissues and organs. This review considers recent applications of RNAi to triatomine research, describing the major methods that have been applied during the knockdown process such as the double-stranded RNA delivery mechanism （injection, microinjection, or ingestion） and the phenotype characterization （mRNA and target protein levels） in studies conducted with the intent to provide greater insights into the biology of these insects. In addition to the characterization of insect biomolecules, some with biopharmacological potential, RNAi may provide a new view of the interaction between triatomine and trypanosomatids, enabling the development of new measures for vector control and transmission of the parasite.

Summary： TS （Turner syndrome） is a genetic disease which affects women caused by complete or partial absence of one of the X chromosomes. This condition is related to an increase in autoimmune diseases incidence like, IBD （inflammatory bowel disease）. Material and Methods： a case report of a 31 years old woman with T S hospitalized with two months of prostration, fever, rashes, nausea, vomit and diarrhea. Investigations have shown colonic mucosa inflammation, besides chronic colitis with cryptic microabscess in the anatomopathological evaluation, suggesting IBD as etiology. Treatment with mesalazine was effective in symptoms improvement. Conclusions： TS is a condition frequently associated with autoimmune diseases, including diseases of the gastrointestinal tract, such as IBDs. Recognition and early treatment of this condition leads to an improvement in the quality of life of these patients and reduces the chances of future complications.

Low Genetic Polymorphism at the Cytochrome C Oxidase I in Silkworm Strains of the Brazilian Germplasm Bank

Nucleotide sequences have been used to distinguish species and specimens for many years. More recently, the use of a partial sequence of 650 bp of the cytochrome c oxidase I, COI mitochondrial gene, has been proposed for species identification, known as DNA barcodes. In this work, a short sequence of the DNA barcode is described—approximately 250 bp, named as “DNA mini-barcode”—to molecularly identify different silkworm strains maintained at the unique public Germplasm Bank of Bombyx mori, at the Universidade Estadual de Maringá, UEM, Brazil. Analysis revealed no significant differences among the silkworm strains. The phylogenetic tree obtained by the neighbor-joining method and K2P distance, in which specimens of B. mandarina were used as outgroup, clustered all the specimens of B. mori in a unique clade. Genetic variability detect within B. mori was low or nonexistent. In conclusion, the partial region of 250 bp of the mitochondrial gene COI herein analyzed may not be efficient to discriminate silkworm strains from the UEM Germplasm Bank of Bombyx mori.

Tuberculosis is one of the 10 leading causes of death in the world. Its symptoms include fever, malaise, weakness, weight loss, chest pain, cough, expectoration, shortness of breath and sepsis, which is its most frequent complication. Due to these symptoms, many patients with tuberculosis require admission to the Intensive Care Unit, where they usually are placed on mechanical ventilation. Tuberculosis is more prevalent in the population in situations of social and economic vulnerability. The main factors that interfere with adherence to treatment and the prognosis of these patients are the patient’s support network and their socioeconomic status. We present the following report of a long-time smoker patient, with chronic kidney disease and previous treatment for tuberculosis, who was brought to the emergency room due to lowering of the sensorium, and was immediately placed on mechanical ventilation. The tests suggested sepsis with a respiratory focus, so a search for Alcohol-Acid-Resistant Bacillus was carried out, with a positive result. The patient was transferred to the Intensive Care Unit due to tachycardia and acute respiratory failure. The patient was a smoker with a high tobacco load, chronic kidney disease, in addition to precarious economic, educational and self-care conditions. This report shows that patients with tuberculosis and its complications can be stabilized through known pharmacological treatment. However, the most effective measure to interrupt the transmission of the disease remains the early diagnosis and the provision of adequate treatment, with guidelines aimed at the cessation of harmful habits, such as smoking.

The occurrence of antimicrobials in the aquatic environment and drinking water has raised the question of their impact on the environment and public health. Animal production is one of the most expressive activities of Brazilian agribusiness. In order to ensure the productivity and competitiveness of the sector, the use of drugs for therapeutic and prophylactic purposes is a common practice. Due to the continuous release of antimicrobials into the environment, the aim of this study was to compare the frequency of detection of tetracyclines and sulfonamides in surface water collected from rural areas in Lidice District of Rio Claro, in the State of Rio de Janeiro, Brazil. An investigative study was conducted with 24 river water samples analyzed by high-performance liquid chromatography coupled to tandem mass spectrometry and the aim of this study was to determine residues of sulfonamides and tetracyclines based on the USEPA method 1694. The results indicated the presence of sulfamethoxazole and oxytetracycline concentrations at the ng·L-1 level. The applied method showed overall good performance with recoveries above 57%, method detection limits ≤ 7.17 ng·L-1, method quantification limits ≤ 23.90 ng·L-1 and good linearity.

Background: Skin reactions and musculoskeletal injuries caused by intramuscular injection are an increased risk for skin and soft tissue infections and may culminate in sepsis. Objective: To describe the history of an elderly patient who developed a lesion in her right arm after home administration of intramuscular iron injection in this arm and its outcome. Methods: This is a case report of a patient who participated in a study in an intensive care unit. Results: This study describes a 65-year-old patient with multiple comorbidities who was admitted to the hospital complaining of pain for two months and difficulty in moving her right shoulder associated with skin and soft tissue infections in her right arm after intramuscular iron administration by a relative for the treatment of multifactorial anemia and refractory melena. The patient worsened her general condition and was transferred to the intensive care unit. Despite the therapy instituted, the patient developed sepsis of cutaneous origin, multiple organ dysfunction, and death. Conclusions: The administration of intramuscular medication requires the participation of qualified professionals, such as nurses, mainly within the scope of the Brazilian public health system which commonly comprises patients in a context of vulnerability.

Sensitivity of a Rapid Mix Test with Combined Synthetic Antigens Derived from Mycobacterium Leprae PGL-1 for Diagnosis and Surveillance of Leprosy

Introduction: In the Americas, Brazil contributes 91.63% of the total cases and the state of Pará still has high endemia for leprosy. Objective: To analyze the performance of a rapid test for the diagnosis and epidemiological surveillance of leprosy in endemic areas. Methods: The sample consisted of 70 MB multibacillary leprosy (MB) patients, 63 paucibacillary (PB) patients, and 80 intradomiciliary consanguineous contacts (ICSCO) of patients. A rapid test with a 15-minute reading was applied using two prototypes: prototype 1, double test with trisaccharide antigen (NT-P-BSA) at 1a. line (83.2 ng/test) and disaccharide antigen (ND-O-BSA) at 2a. (83.2 ng/test), both with a flow of 0.08 μL/mm with a 10 μC membrane, anti-IgM conjugate with a flow of 0.040 μL/mm and a Tris-Triton and prototype 2 runner buffer with MIX antigen (trisaccharide + disaccharide) in the same concentrations and conditions of prototype 1. Results: The comparison of the MIX test positivity rate and the disaccharide or trisaccharide doublet test across all samples was statistically significant, demonstrating that the MIX test had higher seropositivity rates compared to the ND-O-BSA or NT-P-BSA. It was demonstrated that the MIX test showed a good performance, with 25.39% of the PB patients negative for the disaccharide and trisaccharide duplet test, but positive for MIX. Conclusions: These data suggest the potential for further optimizing the performance by adding other synthetic antigens to the MIX antigens.

AIM:To propose an alternative model of hepatic encephalopathy(HE) in mice,resembling the human features of the ***:Mice received two consecutive intraperitoneal injections of thioacetamide(TAA) at low dosage(300 mg/kg).Liver injury was assessed by serum transaminase levels(ALT) and liver histology(hematoxylin and eosin).Neutrophil infiltration was estimated by confocal liver intravital *** was evaluated using prolonged prothrombin and partial thromboplastin *** parameters were measured through tail *** levels were quantified in serum and brain ***(EEG) and psychomotor activity score were performed to show brain *** edema was evaluated using magnetic resonance ***:Mice submitted to the TAA regime developed massive liver injury,as shown by elevation of serum ALT levels and a high degree of liver *** intense hepatic neutrophil accumulation occurred in response to TAA-induced liver *** led to mice mortality and weight loss,which was associated with severe ***,TAA-treated mice presented with increased serum and cerebral levels of ammonia,in parallel with alterations in EEG spectrum and discrete brain edema,as shown by magnetic resonance *** agreement with this,neuropsychomotor abnormalities ensued 36 h after TAA,fulfilling several HE features observed in *** this context of liver injury and neurological dysfunction,we observed lung inflammation and alterations in blood pressure and heart rate that were indicative of multiple organ dysfunction ***:In summary,we describe a new murine model of hepatic encephalopathy comprising multiple features of the disease in humans,which may provide new insights for treatment.
