Differential distribution of gene polymorphisms associated with hypercholesterolemia,hypertriglyceridemia,and hypoalphalipoproteinemia among Native American and Mestizo Mexicans

BACKGROUND Dyslipidemias are metabolic abnormalities associated with chronic diseases caused by genetic and environmental factors.The Mexican population displays regional differences according to ethnicity with an impact on the type of dyslipidemia.AIM To define the main dyslipidemias,the frequency of lipid-related risk alleles,and their association with hyperlipidemic states among different ethnic groups in West Mexico.METHODS In a retrospective study,1324 adults were selected to compare dyslipidemias and lipid-related gene polymorphisms.Demographic,clinical,and laboratory data were collected.A subgroup of 196 normal weight subjects without impaired glucose was selected for the association analyses.Genotyping was determined by allelic discrimination assay.RESULTS Hypercholesterolemia was the most prevalent dyslipidemia(42.3%).The frequency of the risk alleles associated with hypoalphalipoproteinemia(ABCA1)and hypercholesterolemia(APOE,LDLR)was higher in the Native Americans(P=0.047).In contrast,the Mestizos with European ancestry showed a higher frequency of the risk alleles for hypertriglyceridemia(APOE2,MTTP)(P=0.045).In normal weight Mestizo subjects,the APOB TT and LDLR GG genotypes were associated risk factors for hypercholesterolemia(OR=5.33,95%CI:1.537-18.502,P=0.008 and OR=3.90,95%CI:1.042-14.583,P=0.043,respectively),and displayed an increase in low-density lipoprotein cholesterol levels(APOB:β=40.39,95%CI:14.415-66.366,P=0.004;LDLR:β=20.77,95%CI:5.763-35.784,P=0.007).CONCLUSION Gene polymorphisms and dyslipidemias showed a differential distribution.Regional primary health care strategies are required to mitigate their prevalence considering the genetic and environmental features which could have important implications for personalized medicine within the new era of precision medicine.

Hepatitis C virus clearance and less liver damage in patients with high cholesterol, low-density lipoprotein cholesterol and APOE ε4 allele

BACKGROUND Cholesterol is related to improvements in the rate of sustained virological response and a robust immune response against the hepatitis C virus(HCV).APOE gene polymorphisms regulate cholesterol levels modifying the course of the HCV infection.The relationship between cholesterol,APOE alleles,and the outcome of HCV infection has not been evaluated in the admixed population of Mexico.AIM To investigate the role of APOE-ε2,-ε3,and-ε4 alleles and the metabolic profile in the outcome of HCV infection.METHODS A total of 299 treatment-na?ve HCV patients were included in this retrospective study.Patients were stratified in chronic hepatitis C(CHC)(n=206)and spontaneous clearance(SC)(n=93).A clinical record was registered.Biochemical tests were assessed by dry chemistry assay.APOE genotypes were determined using a Real-Time polymerase chain reaction assay.RESULTS Total cholesterol,low-density lipoprotein cholesterol(LDL-c),triglycerides,and hypercholesterolemia were higher in SC than CHC patients as well as the frequency of the APOEε4 allele(12.4%vs 7.3%).SC patients were overweight(54.8%).Theε4 allele was associated with SC(OR=0.55,95%CI:0.31-0.98,P=0.042)and mild fibrosis(F1-F2)in CHC patients(OR 0.091,95%CI 0.01-0.75,P=0.020).LDL-c≥101.5 mg/dL(OR=0.20,95%CI:0.10-0.41,P<0.001)and BMI≥26.6 kg/m2(OR=0.37,95%CI:0.18-0.76,P<0.001)were associated with SC status;while ALT≥50.5 IU/L was negatively associated(OR=5.67,95%CI:2.69-11.97,P<0.001).CONCLUSION In SC patients,the APOEε4 allele and LDL-c conferred a protective effect in the course of the HCV infection in the context of excess body weight.

The mechanisms that regulate disease progression during hepatitis C virus(HCV)infection and the response to treatment are not clearly identified.Numerous studies have demonstrated that a strong host immune response against HCV favors HCV clearance.In addition,genetic factors and metabolic machinery,particularly cholesterol modulation,are involved in HCV infection.It is likely that the interplay between all of these factors contributes to the outcome of HCV infection.In recent years,the world has experienced its largest epidemic of obesity.Mexico and the United States are the leading sufferers from this epidemic at the global level.Obesity is associated with the development ofnumerous pathologies including hypercholesterolemia which is one of the eight most important risk factors for mortality in Mexico.This may be related to the course of HCV infection in this population.Here,we focus on the urgent need to study the progression of HCV infection in relation to ethnic characteristics.Discoveries are discussed that hold promise in identifying immune,metabolic and genetic factors that,in conjunction,could be therapeutic targets or predictors of the progression of HCV infection.

A novel and stretchable carbon-nanotube/Ni@TiO_(2):W photocatalytic composite for the complete removal of diclofenac drug from the drinking water

We present the structural,morphological and photocatalytic properties of stretchable composites made with carbon nanotubes (CNTs),silicon rubber and Ni@TiO_(2):W nanoparticles(TiWNi NPs) with average size of 37±2 nm.Microscopy images showed that the TiWNi NPs decorated the surface of the CNT fibers,which are oriented in a preferential direction.TiWNi NPs presented a mixture of anatase/rutile phases with cubic structure.The performance of the TiWNi powders and stretchable composites was evaluated for the photocatalytic degradation of diclofenac (DCF) anti-inflammatory drug under ultraviolet-visible light.The results revealed that the maximum DCF degradation percentages were 34.6%,91.9%,97.1%,98.5%and 100%for the CNT composite (stretched at 0%),TiWNi powders,CNT+TiWNi(stretched at 0%),CNT+TiWNi (stretched at 50%) and CNT+TiWNi (stretched at 100%),respectively.Thus,stretching the CNT+TiWNi composites was a good strategy to enhance the DCF degradation percentage from 97.1%to 100%,since stretching created additional defects (oxygen vacancies) that acted as electron sink,delaying the electron-hole recombination,and favors the DCF degradation.Raman/absorbance measurements confirmed the presence of such defects.Moreover,the reactive oxygen species (ROS) were determined by the scavenger’s experiments and found that the main ROS were the·OH and O_(2)-radicals,which attacked the DCF molecules,causing their degradation.The results of this investigation confirmed that the stretchable CNT/TiWNi-based composites are a viable alternative to remove pharmaceutical contaminants from water and can be manually separated from the decontaminated water,which is unviable using photocatalytic powders.

目的旨在通过系统综述,确定针对复杂慢性病患者增强自我管理的干预措施,以改善慢性呼吸困难,为护理人员促进该类人群的自我管理提供依据。方法2023年12月,系统检索Medline、Scopus、Web of Science、CINAHL、Cochrane图书馆和乔安娜布里格斯研究所(Joanna Briggs Institute,JBI)循证实践数据库。对纳入研究进行数据提取和综合,研究人员采用Rayyan文献管理软件筛选标题及摘要,并依据JBI研究质量评估工具进行方法学质量评估。结果共纳入18项研究。这些研究探讨了针对慢性呼吸困难的各种干预措施,包括药物干预(氧气疗法和吸入疗法)和非药物方法(教育计划、呼吸练习、液体摄入管理、身体感知技术、同伴支持、情商培训和网络应用程序的使用)。这些干预措施减轻了患者情绪困扰,可以使患者在自我管理方面变得独立自主。结论促进复杂慢性病患者对慢性呼吸困难的自我管理需要采取综合方法,但目前研究中没有充分考虑到针对患者行为改变的干预措施,需要进一步开展相关研究完善干预措施。

Background Diffusion tensor imaging （DTI） permits quantitative examination within the pyramidal tract （PT） by measuring fractional anisotropy （FA）. To the best of our knowledge, the inter-variability measures of FA along the PT remain unexplained. A clear understanding of these reference values would help radiologists and neuroscientists to understand normality as well as to detect early pathophysiologic changes of brain diseases. The aim of our study was to calculate the variability of the FA at eleven anatomical landmarks along the PT and the influences of gender and cerebral hemisphere in these measurements in a sample of young, healthy volunteers. Methods A retrospective, cross-sectional study was performed in twenty-three right-handed healthy volunteers who underwent magnetic resonance evaluation of the brain. Mean FA values from eleven anatomical landmarks across the PT （at centrum semiovale, corona radiata, posterior limb of internal capsule （PLIC）, mesencephalon, pons, and medulla oblongata） were evaluated using split-plot factorial analysis of variance （ANOVA）. Results We found a significant interaction effect between anatomical landmark and cerebral hemisphere （F （10, 32）=4.516, P=0.001; Wilks＇ Lambda 0.415, with a large effect size （partial η2=0.585））. The influence of gender and age was non-significant. On average, the midbrain and PLIC FA values were higher than pons and medulla oblongata values; centrum semiovale measurements were higher than those of the corona radiata but lower than PLIC. Conclusions There is a normal variability of FA measurements along PT in healthy individuals, which is influenced by regions of interest location （anatomical landmarks） and cerebral hemisphere. FA measurements should be reported for comparing same-side and same-landmark PT to help avoid comparisons with the contralateral PT; ideally, normative values should exist for a clinically significant age group. Repellent and Insecticidal Activity of Naphthoquinones from the Heartwood of Tectona grandis on Incisitermes marginipennis(Latreille)

Wood is a lignocellulosic material that,because of its organic nature,is a source of nutrients for bacteria,fungi,and insects.Incisitermes marginipennis(Latreille)is an interesting dry wood-boring termite.Because it is an extremely destructive pest,difficult to control due to its cryptic lifestyle,and for its role in the biogeochemical cycle of carbon and other elements;it is a fundamental organism.The use of less toxic and eco-friendly substances is important for protecting domestic,cultural,and historical heritage made of wood or lignocellulosic materials.The aim of this study was to evaluate the biological activity of various naphthoquinones extracted from the heartwood of T.grandis on I.marginipennis.First,heartwood flour was used for Soxhlet-type extractions to isolate and characterize the heartwood compounds by thin-layer chromatography.To identify the heartwood compounds,nuclear magnetic resonance and gas chromatography coupled to a mass spectrometer were used.The information obtained was supported using the Interactive Spectroscopy software.The identified heartwood compounds were lapachol,tectoquinone,deoxylapachol,dehydro-α-lapachone,tectol and tecomaquinone-I.Using nymphal termites of I.marginipennis as a biological model,the isolated and identified molecules were under three different tests(repellent,insecticide and protective)and their biological activity was individually evaluated.The results showed that the molecules obtained have a repellent-protective effect against I.marginipennis,although not in all cases,these molecules have high percentages of mortality and decreased the weight loss of the treated material exposed to I.marginipennis.

Gastric mucosa-associated lymphoid tissue lymphomas and Helicobacter pylori infection:A Colombian perspective

AIM:To assess the significance of chromosome translocation t(11;18)(q21;q21),B-cell lymphoma 10 (BCL-10) protein and Helicobacter pylori (H.pylori) infection in gastric mucosa-associated lymphoid tissue (MALT) lymphoma in Colombia.METHODS:Fifty cases of gastric MALT lymphoma and their respective post-treatment follow-up biopsies were examined to assess the presence of the translocation t(11;18)(q21;q21) as identified by fluorescence in situ hybridization;to detect protein expression patterns of BCL10 using immunohistochemistry;and for evaluation of tumor histology to determine the correlation of these factors and resistance to H.pylori eradication.RESULTS:Infection with H.pylori was confirmed in all cases of gastric MALT lymphoma in association with chronic gastritis.Bacterial eradication led to tumor regression in 66% of cases.The translocation t(11;18)(q21;q21) was not present in any of these cases,nor was there evidence of tumor transformation to diffuse large B-cell lymphoma.Thirty-four percent of the patients showed resistance to tumor regression,and within this group,7 cases,representing 14% of all those analyzed,were considered to be t(11;18)(q21;q21)-positive gastric MALT lymphomas.Protein expression of BCL10 in the nucleus was associated with the presence of translocation and treatment resistance.Cases that were considered unresponsive to therapy were histologically characterized by the presence of homogeneous tumor cells and a lack of plasmacytic differentiation.Responder cases exhibited higher cellular heterogeneity and a greater frequency of plasma cells.CONCLUSION:Both t(11;18)(q21;q21)-positive MALT lymphoma cases and those with nuclear BCL10 expression are considered resistant to H.pylori eradication.It is suggested that chronic antigenic stimulation is not a dominant event in resistant cases.

Wild plants represent relatively unexplored resource of high economic potential,especially as an alternative to developing new crops and,even more relevant,for improving existing crops and contributing to nutrition and health.The wild species M alpighia mexicana(manzanita)has a wide tradition of food,medicinal and ornamental use in Mexico.It is part of the American-origin group of tropical shrubs that produce edible red fruits,such as A cerola,which is considered the most important natural source of vitamin C in the world.Given the role played by M.mexicana in Mexico,and particularly in Barranca del Río Santiago(Santiago River Canyon),we modelled its potential distribution in both geographical areas.We used species'records from databases,local herbaria and records collected by the authors as well as climatic variables representing long term average,variability and extreme conditions of temperature and precipitation.To fit the models we used the modelling algorithm Maxent and selected an adequate configuration by testing a range of model complexity settings.The results indicate a clear species preference for warm-dry tropical forest,most extensively in the Balsas river depression and the central valleys of Oaxaca.The probability of the species presence in the western region was also high,although the probability was also high for smaller surface areas,such as the region of Santiago river canyons,which are covered by warm-dry tropical forests.

HLA B27 as Predisposition Factor to Suffer Age Related Macular Degeneration

To research whether specific alleles HLA class Ⅰ （HLA-A and HLA-B） and class Ⅱ （HLA-DR） are risk factors for the development of exudative type of Age Related Macular Degeneration（ARMD）, HLA antigens are expressed both in normal and affected eyes with ARMD. We designed a prospective case-controlled study. We recruited 75 patients with choroidal neovascuiarization predominantly classic or occult, secondary to ARMD, and treated with photodynamic therapy. Two hundred and fifty patients over 55 years old, without ophthalmologic pathology who went to hospital for an analytical routine check were used as control. The analysis of the data shows a significant difference between two groups. Allele HLA-B27 correlated positively with ARMD （p 〈 0.0113）. However, we didn＇t find alleles negatively associated. Thus HLA-B27 is an allele predisposed to suffer ARMD.
