＜正＞ Multiple alphal-adrenoceptors seem to contract rat tail artery. AlphalA- predominates and either alpha1B-,alpha1D-,or alpha1L-is the second population. We characterized alphal-adrenoceptors with selective agonists/antagonists in tail artery of young Wistar rat. Tail artery was exposed to A61603 （N-[5-（4.5- dihy-dro- 1H- imidazol - 2 - yl） - 2 - hydroxy - 5. 6. 7. 8 - tetrahydronaphthalen- 1 - yl] methanesulfonamide） or to phenylephrine （alphalA- and alphal- agonists ）, and to prazosin （alphal- antagonist）, the alphal A-antagonists 5 - methy-lurapidil,RS 100329 （5-methyl - 3 - [3 - [4 - [2 - （2,2,2,-trifluoroethoxy） phenyl] - 1-piperazinyl] propyl] - 2,4-（1H）-pyrimidinedione）,RS 17053 （N- [2（2-cyclopropylmethoxy） ethyl] - 5-chloro-a, a-dimethyl - 1H-indole - 3-ethylamine）, and the alpha1D-antagonist BMY 7378 （8-[2-[4-（2-methoxyphenyl） - 1-piperazinyl] ethyl] - 8 - azaspiro[4.5] decane - 7,9 -dione） . A61603 showed 100 fold higher affinity over PHE. Prazosin,RS100329,5-MU,and RS17053 displaced agonists with high affinity indicating alphal A-adrenoceptors while PHE activated alphal B-adrenoceptors since BMY 7378 had low affinity.

AIM To determine a potential relationship between serum undercarboxylated(uc OC) concentration and cardiovascular risk factors in type 2 diabetes(T2D) patients and healthy subj.cts(HS).METHODS A cross-sectional study was conducted on 140 subj.cts classified into two groups, 70 with T2D and 70 HS. Medical history and physical examination with anthropometric measurements were obtained from all subj.cts. Body fat percentage was determined by bioelectrical impendency analysis. Serum uc OC concentration was determined by enzyme immunoassay,while serum levels of insulin and hsC RP were obtained using high sensitivity enzyme-linked immunosorbent assay. Insulin resistance was determined using the homeostasis model assessment-IR. Lipid profile [triglycerides,total cholesterol(TC), high-density lipoproteins(HDL-c),low density lipoproteins(LDL-c), very low-density lipoproteins] was determined by spectrophotometry and standard formulas when applicable. RESULTS The T2D patient group showed significantly higher values of waist circumference, waist-to-hip ratio, systolic blood pressure(SBP), diastolic blood pressure(DBP),current smoking, and alcohol use when compared to the HS group(P < 0.05). We observed a significantly lower serum ucO C concentration in T2D than in HS(1.5 ± 1.4vs 2.3 ± 1.8, P < 0.05). In the whole study population,ucO C concentration was inversely correlated with body mass index(BMI)(r =-0.236, P < 0.05), fasting plasma glucose(r =-0.283, P < 0.01) and HDL-c(r =-0.255,P < 0.05); and positively correlated with LDL-c/HDL-c ratio(r = 0.306, P < 0.05) and TC/HDL-c ratio(r =0.284, P < 0.05). In the T2D group, serum uc OC concentration was inversely correlated with BMI(r =-0.310, P < 0.05) and body-fat percentage(r =-0.311,P < 0.05), and positively correlated with DBP(r = 0.450,P < 0.01). Kaurenoic acid (KA), a kaurane-type diterpene extracted from leaves of Mikania hirsutissima, was previously reported as an inhibitor of vascular contractility mainly by blocking extracellular Ca2+ influx. The compound is known for several other biological activities such as antiparasitic, antispasmodic and antibacterial activity. The aim of the present study was to investigate the effect of KA on Aspergillus nidulans. KA (0.3 mM) showed fungistatic activity against A. nidulans with visible hyphal elongation and morphology damage. These effects were reverted by CaCl2 addition showing that KA interferes with intracellular Ca2+ gradient in A. nidulans. This is the first report on the mechanism of action of KA involving calcium levels by altering the elongation of fungi hyphae.

Mollusks of the Rocky Intertidal Zone at Three Sites in Oaxaca,Mexico

The community of mollusks at three rocky intertidal zone in Oaxaca, Mexico was analyzed. Two samplings per site were conducted. At each sampling the inspected area was 10 m2. The sampling unit was 1 m2. 5862 specimens were examined. 68 mollusks species were identified. The families better represented in species richness were: FISSURELLIDAE, MURICIDAE, LOTIIDAE, MYTILIDAE, CHAMIDAE, ISCHNOCHITONIDAE y CHITONIDAE. The species that had the greatest density were Lottia discors, Siphonaria palmata, Lottia acutapex, Chormoytilus palliopunctatus, Brachidontes adamsianus, Chton articulatus e Ischnochiton muscarius. The species with the widest distribution represent 11.76%. With irregular distribution was found in 58.82% of the species. The gastropod Crucibulummonticulus, the bivalve C. palliopunctatus and the polyplacophoran C. articulatus had the greatest size. The value of diversity index was H’ = 4.29 bits/ind. and j. = 0.72. The mollusks community in the rocky intertidal zone study sites is characterized by small size, because they rarely exceed seventy millimeters. The values of H’ and j. indicate that the mollusk community in the study area has a high diversity and high uniformity, corresponding to mature and stable communities in a tropical region.

Marine Priority Region No. 33 (MPR 33) lies in the State of Guerrero, where the National Commission for Knowledge and Use of Biodiversity has declared a lack of information on marine species, Class POLYPLACOPHORA among others. This program was carried out on the rocky intertidal zone of MPR 33 with the obj.ctives to: 1) determine species richness, 2) community composition based on families represented, 3) estimate the density, 4) index of diversity, 5) analyze body proportions and 6) geographic distribution of species. Three surveys on five sites were conducted during 2009 and 2012, using one mt2 quadrant for systematic sampling procedure;collecting all POLYPLACOPHORA specimens found within the quadrants, then proceeding with their preservation. 441 organisms were analyzed where 17 species were identified, on nine genera within six families. Fifteen new species were recorded for the MPR 33. Family ISCHNOCHITONIDAE Dall, 1889 was the best represented in species richness and CHITONIDAE Rafinesque, 1815 in abundance. Density of 8.82 organisms/m2, the H′ index was 2.38 bits/individuals and j. = 0.57. The species Chiton (Chiton) articulatus Sowerby, 1832 showed the highest density, greater size and was found in all five collection sites. Three species, (Chiton (C.) articulatus, Ischnochiton (Ischnochiton) muscarius (Reeve, 1847) and Chiton (Chiton) alboli-neatus Broderip & Sowerby, 1829), were considered wide. Intertidal species richness was associated with habitat stability and wave intensity;the more unstable sites had greater richness while most stable sites lower. Species inventory and understanding the communities’ interactions of the marine fauna of the State of Guerrero are important;therefore these efforts must be continued.

BACKGROUND Obesity is a maj.r health problem due to its high prevalence. The relationship between obesity and cardiovascular disease is unclear. Some studies agree that certain conditions associated with obesity, such as physical inactivity or cardiovascular risk factors, are responsible for cardiovascular risk excess among obese people. Carotid intima-media thickness and carotid plaques(CP) have been associated with cardiovascular adverse events in healthy populations, and recent data suggest a higher prevalence of subclinical carotid atherosclerosis in obese and metabolically unhealthy patients. However, there are no studies correlating subclinical atherosclerosis and adverse events(AE) in obese *** To determine the association between carotid disease and AE in obese patients with negative exercise echocardiography(EE).METHODS From j.nuary 1, 2006 to December 31, 2010, 2000 consecutive patients with a suspicion of coronary artery disease were submitted for EE and carotid ultrasonography. Exclusion criteria included previous vascular disease, left ventricular ej.ction fraction < 50%, positive EE, significant valvular heart disease and inferior to submaximal EE. An AE was defined as all-cause mortality,myocardial infarction and cerebrovascular accident. Subclinical atherosclerosis was defined as CP presence according to Manheim and the American Society of Echocardiography *** Of the 652 patients who fulfilled the inclusion criteria, 226(34.7%) had body mass indexes ≥ 30 kg/m2, and 76 of them(33.6%) had CP. During a mean follow-up time of 8.2(2.1) years, 27 AE were found(11.9%). Mean event-free survival at 1, 5 and 10 years was 99.1%(0.6), 95.1%(1.4) and 86.5%(2.7), respectively. In univariate analysis, CP predicted AE [hazard ratio(HR) 2.52, 95% confidence interval(CI) 1.17-5.46; P = 0.019]. Microalgae biomass has been reported in the literature as one of the most promising sources for obtaining different products of industrial interest such as lipids, fatty acids, carotenoids, proteins and fibers. The lipid fraction of microalgae comprises neutral lipids, free fatty acids and polar lipids. It is of great importance to estimate the composition of the lipid fraction to define the potential for use, either as a raw material for the production of biofuels or for use for nutraceuticals and/or food purposes. The microalgae Scenedesmus sp. cultivated in a photobioreactor, the sky open raceway type, was evaluated for lipid content, identification and quantification of lipid components obtained from different extracts. In the quantification of the lipid content, extraction methods were proposed without chemical treatment (use of solvents only) such as chloroform:methanol (2:1 v/v)—Bligh & Dyer, Ethanol, Ethyl acetate:Hexane (1:1 v/v) and others with chemical treatment such as j.Schmid-Bondzynski-Ratzlaff (acid) and saponification (basic). Antitumor Effect of Cationic INKKI Peptide from Bovine <i>β</i>-Casein on Melanoma B16F10

Cationic peptide with the sequence INKKI 41-45 was isolated from bovine β-casein after tryptic hydrolysis and synthetized. The aim of this work was to evaluate the antiproliferative activity in vitro and antitumor effect in animal model. The in vitro cytotoxicity was evaluated on B16F10 melanoma cells by MTT assay. Detection of apoptosis was measured using the annexin V/PI double staining and cell cycle analysis performed flow cytometry. Caspase-3 activity was analyzed with substrate specific fluorogenic DEVD-MCA. In vivo, antitumor activity was evaluated in B16F10 melanoma tumor-bearing C57BL/6j.mice. The animals were treated with 55 mg/kg INKKI administered into peritumoral region, while control group received saline solution. The following antitumor parameters were examined: tumor volume, number of metastases, tumor delayed time, tumor doubling time. Histological analyses were performed with H & E staining. The results showed that INKKI induced dose-response cytotoxicity selective for B16F10 melanoma cells (IC50 1.7 μM) and did not present cytotoxic effects for FN1 fibroblast cells. INKKI-induced apoptosis detected trough of annexin V/PI assay and it was accompanied with an increase of sub-G1 apoptotic fractions and significant increase of caspase-3 cleavage. The tumor-bearing mice treated with INKKI showed a significant reduction in tumor volume of 72.62% and decreased of metastasis number loci. In addition, INKKI caused a significant delay in tumor growth and prolonged the tumor doubling time. Histological analysis revealed an increased of necrosis areas and reduction of tumor cells in tumor treated with INKKI, it was a many hallmark of its antitumor effects observed from in vivo experiments. In conclusion, we show that INKKI is a peptide that could be considered a new putative candidate development to anticancer therapy drug.
