Modelling Animal Activity as Curves: An Approach Using Wavelet-Based functional Data Analysis

Temporal activity patterns in animals emerge from complex interactions between choices made by organisms as responses to biotic interactions and challenges posed by external factors. Temporal activity pattern is an inherently continuous process, even being recorded as a time series. The discreteness of the data set is clearly due to data-acquisition limitations rather than a true underlying discrete nature of the phenomenon itself. Therefore, curves are a natural representation for high-frequency data. Here, we fully model temporal activity data as curves integrating wavelets and functional data analysis, allowing for testing hypotheses based on curves rather than on scalar and vector-valued data. Temporal activity data were obtained experimentally for males and females of a small-bodied marsupial and modelled as wavelets with independent and identically distributed errors and dependent errors. The null hypothesis of no difference in temporal activity pattern between male and female curves was tested with functional analysis of variance (fANOVA). The null hypothesis was rejected by fANOVA and we discussed the differences in temporal activity pattern curves between males and females in terms of ecological and life-history attributes of the reference species. We also performed numerical analysis that shed light on the regularity properties of the wavelet bases used and the thresholding parameters.

Chlorhexidine (CHX) is regarded as one of the most successful antiplaque agents in controlling the formation of dental biofilm. Nevertheless, molecular mechanisms of their effects in Streptococcus mutans are largely unknown. In this work, the effects of sub-lethal and lethal concentrations of chlorhexidine (CHX) on planktonic or biofilm-organized Streptococcus mutans cells were investigated in dose- and time-dependent manner. The Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) for planktonic cells and biofilm conditions were determined by standard methods. Quantitative PCR (qPCR) was used to quantify the relative levels of glucosyltransferase B (gtfB), gtfC and gtfD transcription of S. mutans in the presence of CHX. The CHX activity in the initial biofilm structure and morphological alterations in planktonic cells were examined by Scanning Electron Microscopy (SEM). The results indicate that CHX increased expression of gtfC and gtfD in planktonic S. mutans cells and CHX reduced the expression of gtfB, gtfC, and gtfD in biofilms. High concentrations of CHX resulted in several wilted S. mutans planktonic cells with spilled intracellular material, while decreased cells’ chain length and matrix was found when the initial biofilm was exposed to increasing concentrations of CHX. CHX’s effects against bacteria depend on the type of growth organization and the concentration and time of exposure to the drug. At sub-lethal concentrations, CHX affects the expression of glucosyltransferases, which may have anticariogenic effect.

Effective use of fibroTest to generate decision trees in hepatitis C

AIM： To assess the usefulness of fibroTest to forecast scores by constructing decision trees in patients with chronic hepatitis C.METHODS： We used the C4.5 classification algorithm to construct decision trees with data from 261 patients with chronic hepatitis C without a liver biopsy. The fibroTest attributes of age, gender, bilirubin, apolipoprotein, haptoglobin, α2 macroglobulin, and γ-glutamyl transpeptidase were used as predictors, and the fibroTest score as the target. for testing, a 10-fold cross validation was used.RESULTS： The overall classification error was 14.9% （accuracy 85.1%）. fibroTest＇s cases with true scores of fO and f4 were classified with very high accuracy （18/20 for fO, 9/9 for fO-1 and 92/96 for f4） and the largest confusion centered on f3. The algorithm produced a set of compound rules out of the ten classification trees and was used to classify the 261 patients. The rules for the classification of patients in fO and f4 were effective in more than 75% of the cases in which they were tested.CONCLUSION： The recognition of clinical subgroups should help to enhance our ability to assess differences in fibrosis scores in clinical studies and improve our understanding of fibrosis progression,

将用电磁方法悬浮起来的一对环构成领导者–跟随者系统实现对光滑参考轨线的跟踪要求.这一任务通过设计具有对扰动进行在线估计与补偿功能的自抗扰控制器(ADRC)实现.本文设计方法与传统ADRC的主要不同在于领导者和跟随者的跟踪控制器设计都是基于平坦输出的线性化近似系统.超出线性化近似有效区域的大的高度偏差所导致的未知非线性则被视为扰动,这个扰动借助于线性扩张观测器进行在线估计并通过线性反馈控制器进行消除.实验结果检验了本文所提方法的有效性.

Molecular Assessment of Non-Muscle Invasive and Muscle Invasive Bladder Tumors: Mapping of Putative Urothelial Stem Cells and Toll-Like Receptors (TLR) Signaling

Purpose: The main objectives of this study were to characterize and compare the urothelial stem cells (healthy and cancer cells) and TLRs features in the urinary bladder of men without lesionsand with non-muscle-invasive and muscle invasive urothelial tumors. Materials and Methods: Thirty samples of the urinary bladder of 50 to 80-year-old men, with and without diagnosis of malignant urothelial lesions were used. The 30 samples were divided into 3 groups (n = 10 per group): Normal Group;Non-Muscle Invasive Bladder Cancer Group;Muscle Invasive Bladder Cancer Group. The samples were histopathologically and immunohistochemically analyzed. The study was conducted at teaching Hospital of the University of Campinas (UNICAMP). Results: The CD44 and CD133 immunoreactivities were significantly intense in the muscle-invasive cancer group when compared to the other groups. The ABCG2 biomarker demonstrated intense immunoreactivities in both non-muscle and muscle invasive groups, and absent immunoreactivity in the normal group. All groups showed weak CD117 immunoreactivity. Putative Healthy Stem Cells (CD44/CD133/ CD117+) occurred in all groups. Putative Cancer Stem Cells (CD44/CD133/ABCG2+) only occurred in the non-muscle and muscle invasive cancer groups. TLR2 immunoreactivity was significantly lower in the non-muscle invasive cancer group and absent in the muscle invasive cancer group. TLR4 immunoreactivity was significantly lower in both cancer groups. Conclusions: This study leads us to the conclusion that putative cancer stem cell occurrence was sensitive to the decreased in TLR2 and TLR4 immunoreactivities. Also, TLR2 and TLR4 demonstrated their involvement in the regulation of the different biomarkers for putative healthy and cancer urothelial stem cells, probably acting as negative regulators of urothelial carcinogenesis. Watt-level and spectrally flat mid-infrared supercontinuum in fluoroindate fibers

We report on infrared supercontinuum （SC） generation in step-index fluoroindate-based fiber by using an all-fiber laser source. In comparison to widely used ZBLAN fibers for high-power mid-infrared （MIR） SC generation, fluoroindate fibers have multiphoton absorption edges at significandy longer wavelengths and can sustain similar intensities. Recent developments highlighted in the present study allowed the production of fluoroindate fibers with MIR background loss of 2 dB/km, which is similar to or even better than ZBLAN fibers. By using an all-fiber picosecond laser source based on an erbium amplifier followed by a thulium power amplifier, we demonstrate the generation of 1.0 W infrared SC spanning over 2.25 octaves from 1 μm to 5 μm. The generated MIR SC also exhibits high spectral flatness with a 6 dB spectral bandwidth from 1.91 μm to 4.77 μm and an average power two orders of magnitude greater than in previous demonstrations with a similar spectral distribution.

Among degenerative musculoskeletal disorders,osteoarthritis remains one of the main causes of pain and disability in the adult *** available alternatives to alleviate symptoms include conservative treatments such as physical therapy,anti-inflammatory drugs and an educational approach to lifestyle *** use of certain analgesics,such as opiates and corticosteroids offer short-term results but does not address the etiological source of pain and *** addition,prolonged use of such medications can cause additional ***,the demand for regeneration of joint cartilage has led to an alternative approach called"orthobiologics".This alternative is based on cellular and molecular components capable of inducing and promoting tissue *** derived from adipose tissue have been studied as an excellent source of orthobiologics in an attempt to promote joint cartilage ***,the lack of standardization regarding collection and processing protocols presents a challenge for the generalization of study results and determination of *** the best of our knowledge,orthobiologics derived from fat have not yet been ***,this manuscript proposes the HGS classification system which aims to describe certain parameters that are relevant to the quality of organic products regarding harvesting techniques(H),graft type(G),and number of centrifugations(S).The more parameters used would imply greater characterization and complexity of the evaluation of the biological product *** HGS classification may provide a valuable contribution to the understanding of clinical procedures and research results,aiming to ultimately usher in a standardization of optimal practice.
