AIM To detect hyper-conserved regions in the hepatitis B virus(HBV) X gene(HBX) 5' region that could be candidates for gene *** The study included 27 chronic hepatitis B treatmentnaive patients in various clinical stages(from chronic infection to cirrhosis and hepatocellular carcinoma, both HBeA g-negative and HBeA g-positive), and infected with HBV genotypes A-f and H. In a serum sample from each patient with viremia > 3.5 log IU/m L, the HBX 5' end region [nucleotide(nt) 1255-1611] was PCRamplified and submitted to next-generation sequencing(NGS). We assessed genotype variants by phylogenetic analysis, and evaluated conservation of this region by calculating the information content of each nucleotide position in a multiple alignment of all unique sequences(haplotypes) obtained by NGS. Conservation at the HBx protein amino acid(aa) level was also *** NGS yielded 1333069 sequences from the 27 samples, with a median of 4578 sequences/sample(2487-9279, IQR 2817). In 14/27 patients(51.8%), phylogenetic analysis of viral nucleotide haplotypes showed a complex mixture of genotypic variants. Analysis of the information content in the haplotype multiple alignments detected 2 hyper-conserved nucleotide regions, one in the HBX upstream non-coding region(nt 1255-1286) and the other in the 5' end coding region(nt 1519-1603). This last region coded for a conserved amino acid region(aa 63-76) that partially overlaps a Kunitz-like *** Two hyper-conserved regions detected in the HBX 5' end may be of value for targeted gene therapy, regardless of the patients' clinical stage or HBV genotype.

Management evaluation of Panicum maximum cv. massai (Massai guineagrass) under deferment contributes as a strategic reserve of forage in the Brazilian semi-arid region. The objective of this study was to evaluate biomass production and nutritive value of massai guineagrass in an area of thinned caatinga vegetation in two deferment periods (60 and 90 days after plot-leveling cut) and four use periods (30, 60, 90 and 120 days after the end of the rain season), in a random blocks design arranged in a split-plot (2 × 4), with three replications. Extending the deferment period of massai guineagrass promoted elevation of the biomass components, but compromised the pasture quality due to increasing stem biomass. The massai guineagrass deferred 30 days after raining season started and should use 90 days after raining season ended to ensure a forage supply of appropriate nutritional value. The increase in relative air humidity and the occurrence of rainfall after water restriction stimulate regrowth, resulting in improved nutritional value of the grass.

Allotransplantation is helpful in order to maintain the life in some cases, but immunosuppression causes collateral effects. In this study, we were reporting the results of allografts in Landrace pigs, which were implanted on collagen-generator devices. After 2 weeks of implantation, the devices were carried out to assess allograft viability at 30, 60 and 90 days post implant. The implanted cells were islets of Langerhans and the aim was to prove their presence by immunohistochemistry for insulin and glucagon. The results were positive at 30, 60 and 90 days post implant. It demonstrated that the collagen-generator device permits the cell viability without using anti-rejected drugs or immunomodulating cells.

BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in *** To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic *** Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase f(PNGase f). Mammary neoplasms are the most common tumors in female dogs. They are usually treated using solely surgical mastectomy—which is recognized as unsatisfactory in many cases. Given this, the benefits of chemotherapy in dogs with mammary cancer need to be further explored. Some drugs that can be used for treating canines with mammary tumors may be substrates of uptake and/or efflux transporters such as the ATP-binding cassette (ABC) transporters. Unfortunately, very little is known regarding the pathobiology of such proteins in canine tumors, including mammary cancer. Accordingly, this study was designed to characterize the expression of ABC transporters P-glycoprotein, MRP1, and MRP2 and their relation with clinicopathologic factors in order to allow a better understanding of their influence in canine mammary cancer. P-glycoprotein was expressed in tumors from 55.8% of patients, while MRP1 and MRP2 were expressed in 37.2% and 39.5% of tumors, respectively. P-glycoprotein expression showed to be related with regional lymph node spread (P = 0.0038), as well as with tumor grade (P = 0.0353) and with a shorter survival (P = 0.0245). MRP1 revealed a strong association with a higher histological grade (P f lymph node status using Cox proportional-hazards regression multivariate analysis (P = 0.0216). No relations between MRP2 and clinicopathologic features were observed. We have found that P-glycoprotein and MRP1 are expressed in highly aggressive canine mammary tumors and are related with poor prognosis. Our results suggest that they may play a significant role in the course of canine mammary cancer progression and be promising candidate markers for a validation study on therapy outcome.

flame Thermal Spray (fTS) coatings frequently show some porosity and reduced adherence to substrate, which affect its properties, especially its corrosion resistance. In this work, the corrosion resistance of AISI 1018 carbon steel coated by different methods is compared: electroless nickel (EN) coating, NiCrfeBSi obtained by fTS, duplex coatings of an EN deposit on a layer of NiCrfeBSi obtained by fTS and a layer of NiCrfeBSi on an EN deposit. The coatings were characterized using optical microscopy and scanning electron microscopy techniques, EDS microprobe microanalysis, roughness as well as electrochemical polarization tests to obtain the corrosion rate. The results show the enhancement in the corrosion resistance in saline medium of the duplex coatings, especially of the EN coating on fTS layer.

越来越多的科技论文提到,经过传统热处理的高速钢采用深冷处理可进一步提高其性能。本文着重探讨PM S390 MC高速钢试样经深冷处理后性能的改善,研究了深冷处理对该高速钢回火过程中的膨胀曲线及其抗磨料磨损和抗冲蚀磨损性能的影响,并与仅经常规热处理即淬火和三次高温回火的同种钢进行了对比研究。膨胀试验证明,回火时发生的常见过程取决于钢的初始状态(仅淬火或淬火后深冷处理)。这些测试结果表明,深冷处理并不能完全消除残留奥氏体,尤其是在采用高奥氏体化温度时。摩擦试验结果证明,深冷处理工艺可以显著改善高速钢的抗冲蚀磨损性能,而抗磨料磨损性能则取决于钢的奥氏体化温度。
