Impact of ovariectomy and estradiol-17β(E_(2))replacement on the brain steroid levels of the Indian stinging catfish Heteropneustes fossilis

In the present study,effects of ovariectomy(Ovx)and estradiol-17β(E2)replacement on the brain steroid levels were investigated in the resting and prespawning phases of the female catfish Heteropneustes fossilis.The study showed that Ovx resulted in significant decreases in plasma levels of estradiol-17β(E2)and its precursors,and the effect varied with the reproductive stage and duration.Our study showed that E2 supplementation reversed the effect of Ovx in 3-week Ovx fish group,either restored or increased according to the dose.In contrast,E2 levels increased in the brain after Ovx in the resting stage and produced a biphasic effect of significant decreases on week 1 and 2,and significant increases on week 3,4 and 5 in the pre-spawning stage.The brain E2 levels significantly inhibited in E2 supplementation groups.The testosterone(T)levels showed biphasic effects,an initial decrease(up to 3 weeks Ovx)and an increase later(week 4,5)in the resting stage.In the pre-spawning stage,the T levels increased significantly after Ovx.The E2 supplementation significantly inhibited the T levels,more severely in the low dose group in the resting and pre-spawning stages.Brain 17-hydroxyprogesterone(17-OHP4)levels increased(week 1,2,3)but decreased on week 4 and 5 of Ovx in the resting stage and increased throughout Ovx in the pre-spawning stage except in the 5th week(though compared to other steroids,level was very low;almost negligible).The E2 replacement significantly decreased the steroid levels further down compared to the Ovx control group.Brain progesterone(P4)and pregnenolone(P5)levels decreased initially(week 1 and 2 Ovx)and recovered later.The E2 replacement significantly decreased P4 levels in the Ovx fish in the pre-spawning stage and in the lower dose group in the resting stage.The P5 levels remained inhibited after the E2 supplementation in the resting stage but unchanged or increased it in the pre-spawning stage.Brain cortisol(F)levels were inhibited initially(1 and 2 weeks and later increased after OVX in the rest

The synthesis of bioactive steroids is catalyzed by an array of enzymes of diverse properties and actions.In the present study,seasonal dynamics and kinetics of key steroidogenic enzymes,17α-hydroxylase(Cyp17a),3β-hydroxysteroid dehydrogenase(Hsd3b),20α-hydroxysteroid dehydrogenase(Hsd20a)and 20β-hydroxysteroid dehydrogenase(Hsd20b)were investigated in the female catfish Heteropneustes fossilis.Further,the effects of the estrogen metabolite 2-hydroxyestradiol-17β(2-OHE2)and human chorionic gonadotropin(hCG)on activities of the above enzymes,cytochrome P450 aromatase(Cyp19a1),and steroid products including testosterone and cortisol were determined.The enzymes under investigation showed significant seasonal variations across the annual ovarian cycle with low activity in the gonad resting phase.The enzymes Hsd3b and Cyp17a showed high activity during early oogenesis but the activities of Hsd20a and Hsd20b were higher towards late oogenesis in the spawning phase.Hsd3b and Cyp17a elicited high apparent Km values(low substrate affinity)and high apparent Vmax in the vitellogenic phase compared to the postvitellogenic phase.Hsd20a did not elicit any significant differences in the kinetic parameters between the two phases.Hsd20b showed high apparent Km values(low substrate affinity)and high Vmax in the postvitellogenic phase.The incubation of ovarian slices with 2-OHE2 for 24 h increased dose-dependently Hsd3b,Cyp17a,Hsd20a and Hsd20b activities,similar to hCG.The levels of the corresponding C21 steroid products,progesterone(P4),17α-hydroxyprogesterone(17-OHP4),17,20α-dihydroxy-4-pregnen-3-one(17,20α-DP)and 17,20β-dihydoxy-4-pregnen-3-one(17,20β-DP),and cortisol were elevated.However,2-OHE2 decreased significantly the C19 and C18 steroids,testosterone and E2 levels,and Cyp19a activity.The co-incubation with hCG and 2-OHE2 produced a synergistic effect on the enzyme activities except that of CYP19a.The co-incubation reversed the inhibitory effect of 2-OHE2.The data show that 2-OHE2 exerts a dual role on steroidogenesis,sti

Heavy metal pollution of river water and eco-friendly remediation using potent microalgal species

Pollution of rivers is mainly caused by anthropogenic activities such as discharge of effluent from industrial facilities,maintenance of sewage/effluent treatment plants,and dumping of solid waste on river banks.This study dealt with the pollution issues of the Cooum River in the well-known city of Chennai in South India.Water samples from 27 locations were collected and analyzed for 12 elements,including Ba,B,and Al,as well as heavy metals such as Pb,Cr,Mn,Fe,Co,Ni,Cu,Zn,and Cd.The samples showed levels of these elements that exceeded World Health Organization recommendations.Pearson correlation analysis revealed the inter-dependency among elements,and the contribution of each element based on factor loadings showed its percentage contribution compared to others.Water samples from six significant locations were chosen for remediation with three algae:Chlorella vulgaris,Scenedesmus dimorphus,and Phormedium sp.The uptake of pollutants led to the continuous growth of algae during the incubation period of 15 d,effectively removing heavy metals from the river water.The increasing levels of algal counts and the chlorophyll a content confirmed the algal growth during the incubation period,followed by a declining stage after the incubation period.The scanning electron microscopic images of algae before and after the remediation showed no remarkable modification of morphological patterns.This study showed that the uptake of heavy metals using algae is an effective water pollution remediation measure,making the process practicable in the field on a large scale in the near future.

Bethanechol Remains the Preferred Drug in Suspected Underactive Bladder: Findings from Nation-Wide Prescription Analysis

Background: Bethanechol chloride, a cholinergic agonist, is often prescribed in suspected underactive bladders to improve detrusor contractility, despite its limited clinical efficacy. We investigated the usage pattern of bethanechol in actual practice with the understanding that it would enable the physicians to make an informed decision on the coherent use of bethanechol. Methods: A nation-wide survey was carried out to obtain the responses of the urologists. Out of the 755 urologists approached, 630 survey responses were considered for analysis. Results: Usage of bethanechol was advocated as very common [318 (50.48%)], common [200 (31.75%)], not so common [107 (16.98%)], and rare [5 (0.79%)] in postoperative urinary retention, where it was preferred either exclusively [255 (40.48%)] or along with alpha blockers [247 (39.21%)]. Predilection to use alpha-blocker [247 (39.21%)], alpha-blocker plus naloxone [4 (0.64%)], naloxone [1 (0.16%)], alpha-blocker plus bethanechol plus naloxone [1 (0.16%)] was also observed. It was also preferred individually in pathologies causing urinary retention such as benign prostatic hyperplasia [125 (19.84%)], diabetic neuropathy [82 (13.02%)], neurological diseases [69 (10.95%)], senile bladder [14 (2.22%)], drugs [13 (2.06%)], and infective/inflammatory conditions [6 (0.95%)]. Other [321 (50.95%)] physicians opted to prescribe bethanechol in two or more of the enumerated indications. Bethanechol was prescribed orally as 25 mg thrice daily [441 (70.00%)], 50 mg thrice daily [86 (13.65%)], 25 mg four times daily [59 (9.37%)], and many “strongly agree” and “agree” that its sustained release formulation may offer better treatment compliance [565 (89.68%)], safety [548 (86.99%)], and efficacy [544 (86.35%)]. Conclusion: Bethanechol was the most prescribed drug for the management of postoperative urinary retention and other pathologies suspected to cause underactive bladder.

Optimal allocation of solar photovoltaic distributed generation in electrical distribution networks using Archimedes optimization algorithm

This paper proposes to resolve optimal solar photovoltaic(SPV)system locations and sizes in electrical distribution networks using a novel Archimedes optimization algorithm(AOA)inspired by physical principles in order to minimize network dependence and greenhouse gas(GHG)emissions to the greatest extent possible.Loss sensitivity factors are used to predefine the search space for sites,and AOA is used to identify the optimal locations and sizes of SPV systems for reducing grid dependence and GHG emissions from conventional power plants.Experiments with composite agriculture loads on a practical Indian 22-bus agricultural feeder,a 28-bus rural feeder and an IEEE 85-bus feeder demonstrated the critical nature of optimally distributed SPV systems for minimizing grid reliance and reducing GHG emissions from conventional energy sources.Additionally,the voltage profile of the network has been enhanced,resulting in significant reductions in distribution losses.The results of AOA were compared to those of several other nature-inspired heuristic algorithms previously published in the literature,and it was observed that AOA outperformed them in terms of convergence and redundancy when solving complex,non-linear and multivariable optimization problems.

Microbes and their products play key roles in complementing chemical fertilizers and plant protection chemicals by eliciting defence mechanisms in crop plants,thereby providing immunity and resistance against diverse stresses.Among the different environmental technologies used to mitigate climate change,several microbiological interventions are promising,among which biofilms,both naturally-existing and laboratory-developed,and their application have gained momentum recently.Microbial biofilms are aggregations of microbial cells in a self-generated polymeric matrix,which are produced by several genera of bacteria,yeasts,cyanobacteria,and fungi as a mode of reproductive fitness,thereby advantageous in successful proliferation,even in extreme environments.The propensity of microbial biofilms to grow in diverse niches and adapt to biotic and abiotic stresses illustrates their immense potential as biofertilizers and disease suppression options in various crops across diverse ecologies.In the inhospitable habitats of deserts and denudated land,biofilms play an important role in preventing soil erosion and sustaining vegetation,microflora,and fauna.Biofilms represent a mode of growth for several microbes with plant growth-promoting and biocontrol potential,which are important in seed establishment and the facilitation of exchanging nutrients and metabolites from the environment.In this review,we discuss the prospects of microbial biofilms as a green option in agriculture in general and,more specifically,their potential in mitigating climate change.

Enhanced Feature Fusion Segmentation for Tumor Detection Using Intelligent Techniques

In theﬁeld of diagnosis of medical images the challenge lies in tracking and identifying the defective cells and the extent of the defective region within the complex structure of a brain cavity.Locating the defective cells precisely during the diagnosis phase helps toﬁght the greatest exterminator of mankind.Early detec-tion of these defective cells requires an accurate computer-aided diagnostic system(CAD)that supports early treatment and promotes survival rates of patients.An ear-lier version of CAD systems relies greatly on the expertise of radiologist and it con-sumed more time to identify the defective region.The manuscript takes the efﬁcacy of coalescing features like intensity,shape,and texture of the magnetic resonance image(MRI).In the Enhanced Feature Fusion Segmentation based classiﬁcation method(EEFS)the image is enhanced and segmented to extract the prominent fea-tures.To bring out the desired effect the EEFS method uses Enhanced Local Binary Pattern(EnLBP),Partisan Gray Level Co-occurrence Matrix Histogram of Oriented Gradients(PGLCMHOG),and iGrab cut method to segment image.These prominent features along with deep features are coalesced to provide a single-dimensional fea-ture vector that is effectively used for prediction.The coalesced vector is used with the existing classiﬁers to compare the results of these classiﬁers with that of the gen-erated vector.The generated vector provides promising results with commendably less computatio nal time for pre-processing and classiﬁcation of MR medical images.

Cloud computing(CC)is an advanced technology that provides access to predictive resources and data sharing.The cloud environment represents the right type regarding cloud usage model ownership,size,and rights to access.It introduces the scope and nature of cloud computing.In recent times,all processes are fed into the system for which consumer data and cache size are required.One of the most security issues in the cloud environment is Distributed Denial of Ser-vice(DDoS)attacks,responsible for cloud server overloading.This proposed sys-tem ID3(Iterative Dichotomiser 3)Maximum Multifactor Dimensionality Posteriori Method(ID3-MMDP)is used to overcome the drawback and a rela-tively simple way to execute and for the detection of(DDoS)attack.First,the pro-posed ID3-MMDP method calls for the resources of the cloud platform and then implements the attack detection technology based on information entropy to detect DDoS attacks.Since because the entropy value can show the discrete or aggregated characteristics of the current data set,it can be used for the detection of abnormal dataﬂow,User-uploaded data,ID3-MMDP system checks and read risk measurement and processing,bug ratingﬁle size changes,orﬁle name changes and changes in the format design of the data size entropy value.Unique properties can be used whenever the program approaches any data error to detect abnormal data services.Finally,the experiment also veriﬁes the DDoS attack detection capability algorithm.

Ischemic colitis masquerading as colonic tumor:Case report with review of literature

Ischemic colitis can mimic a carcinoma on computed tomographic (CT) imaging or endoscopic examination. A coexisting colonic carcinoma or another potentially obstructing lesion has also been described in 20% of the cases of ischemic colitis. CT scan can differentiate it from colon cancer in 75% of cases. However, colonoscopy is the preferred method for diagnosing ischemic colitis as it allows for direct visualization with tissue sampling. Varied presentations of ischemic colitis have been described as an ulcerated or submucosal mass or as a narrowed segment of colon with ulcerated mucosa on colonoscopy. Awareness and early recognition of such varied presentations of a common condition is necessary to differentiate from a colonic carcinoma, and to avoid unnecessary surgery and related complications.

Single Fully Differential Second Generation Current Conveyor Based Four-Quadrant Analog Multiplier Design and Its Applications

This manuscript presents a new fourquadrant analog multiplier using a recently reported current mode active building block, namely the Fully differential second generation current conveyor(FDCCⅡ).The proposed circuit employs single FDCCⅡ and two NMOSFETs only, thus has simple architecture. It is fully-integrable as no other external passive component has been used. Non-ideal behaviour of the reported configuration has been analysed considering current and voltage tracking errors of the FDCCⅡ. Workability of the derived multiplier is verified with PSPICE(Cadence16.6) simulations using model parameter of TSMC 0.35μm CMOS process and found to be in close agreement with theoretical anticipations. The static power consumption of the circuit is 0.107 mW. The circuit works well with good linearity(nonlinearity error ≤ 0.96%) for the input voltage range of ±0.5 V for a supply voltage of ±1 V and the output is insensitive to temperature variation. Simulation results show that the –3 dB bandwidth of the proposed multiplier is 20.67 MHz and the output referred noise√is less than 9 nV/Hz at 1 kΩ load condition. MonteCarlo analysis has also been performed for the proposed configuration. The applicability of the reported multiplier as amplitude modulator, squarer, and frequency doubler are also demonstrated.
