NeCAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NeCAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NeCAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NeCAB2 interacts with predominantly striatal G-protein-coupled receptors, while NeCAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NeCAB proteins exhibit distinct subcellular localizations: NeCAB1 is found in the nucleus and cytosol, NeCAB2 resides in endosomes and the plasma membrane, and NeCAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NeCAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NeCAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. What Makes Homeowners Consider Protective Actions to Reduce Disaster Risk?An Application of the Precaution Adoption Process Model and Life Course Theory

We hypothesize that for disaster risk mitigation,many households,despite being aware of their risk and possible mitigation actions,never seriously consider doing anything about *** mitigation-focused decisions,since there is no equivalent to warning messages,the decision process is likely to evolve over an extended *** explore what activates hurricane mitigation protective action decisions through three research questions:(1)to what extent are homeowners unengaged in protective action decision making?(2)What homeowner characteristics are associated with lack of engagement?And(3)to what extent do different life events trigger engagement in the decision-making process?We use the Precaution Adoption Process Model to conceptualize engagement as distinct from decision making;the broader protective action decision-making literature to explore drivers of engagement;and Life Course Theory to examine potential transitions from unengaged to *** use survey data of homeowners in North Carolina to examine these questions *** suggest that one-third of respondents had never engaged in protective action decisions,that life experiences differ in their occurrence frequency and effect on households’mitigation decisions,and that some events,such as renovating,reroofing,or purchasing a home may offer critical moments that could be leveraged to encourage greater engagement in mitigation decision making.

Three-dimensional printers have revolutionized many scientific fields with its low-cost,accessibility and ease of *** this paper,we show how stereolithography(SLA)based 3D printers can enable realization of innovative 3D optical devices formed through the fusion of metamaterials with geometrical optics or *** utilizes a combination of desktop SLA 3D printer and metal deposition/coating *** this approach,we present innovative metamaterial embedded optical components such as mushroom-type metamaterials,curved wide-angle metamaterial absorbers/reflectors and a frequency selective moth eye hemispherical *** a unique MeGO device formed through the fusion of a frequency selective metamaterial with an optical parabolic reflector has been demonstrated that combines their individual properties in a single *** fabricated MeGO devices operate in the millimeter wave frequency *** and measurement results using terahertz continuous-wave spectrometer validate their functionality and *** improving resolution in 3D printing,MeGO devices will be able to reach Terahertz and optical frequencies in the near future.

Present of wind power is sporadically and cannot be utilized as the only fundamental load of energy *** paper proposes a wind-solar hybrid energy storage system(HeSS)to ensure a stable supply grid for a longer period.A multi-objective genetic algorithm(MOGA)and state of charge(SOC)region division for the batteries are introduced to solve the objective function and configuration of the system capacity,***/Simulink was used for simulation *** optimization results show that for a 0.5 MW wind power and 0.5 MW photovoltaic system,with a combination of a 300 Ah lithium battery,a 200 Ah lead-acid battery,and a water storage tank,the proposed strategy reduces the system construction cost by approximately 18,000 ***,the cycle count of the electrochemical energy storage systemincreases from4515 to 4660,while the depth of discharge decreases from 55.37%to 53.65%,achieving shallow charging and discharging,thereby extending battery life and reducing grid voltage fluctuations *** proposed strategy is a guide for stabilizing the grid connection of wind and solar power generation,capability allocation,and energy management of energy conservation systems.

Current role of palliative interventions in advanced pancreatic cancer

Pancreatic adenocarcinoma is the third leading cause of cancer death in the United States. Unfortunately, at diagnosis, most patients are not candidates for curative resection. Surgical palliation, a procedure performed with the intention of relieving symptoms or improving quality of life, comes to the forefront of management. This article reviews the palliative management of unresectable pancreatic cancer, including obstructive jaundice, duodenal obstruction and pain control with celiac plexus block. Although surgical bypasses for both biliary and duodenal obstructions usually achieve good technical success, they result in considerable perioperative morbidity and mortality, even when performed laparoscopically. The effectiveness of selfexpanding metal stents for biliary drainage is excellent with low morbidity. Surgical gastrojejunostomy for duodenal obstruction appears to be best for patients with a life expectancy of greater than 2 mo while endoscopic stenting has been shown to be feasible with good symptom relief in those with a shorter life expectancy. Regardless of the palliative procedure performed, all physicians involved must be adequately trained in end of life management to ensure the best possible care for patients.

Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other *** to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma *** cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under *** vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell ***,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)***,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between *** the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy *** this review,we first provide a general overview of 3D cell cultures compared to 2D cell *** then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO ***,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.

Merging bound states in the continuum in an open acoustic resonator

Bound states in the continuum （BICs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIC is very sensitive to the structure ***,we report merging acoustic BICs in a single open resonator,robust against the structure *** find that both symmetry-protected BIC and Friedrich-Wintgen BIC are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BICs move toward each other and merge into a single one at a critical *** to an individual BIC,the merged BIC is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BICs in a coupled 3D waveguide-resonator,which are evidenced by the vanishing of the linewidth of Fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.

Retinoid X receptor α downregulation is required for tail and caudal spinal cord regeneration in the adult newt

Some adult vertebrate species,such as newts,axolotls and zebrafish,have the ability to regenerate their central nervous system（CNS）.However,the factors that establish a permissive CNS environment for correct morphological and functional regeneration in these species are not well *** evidence supports a role for retinoid signaling in the intrinsic ability of neurons,in these regeneration-competent species,to regrow after CNS ***,we demonstrated that a specific retinoic acid receptor（RAR）subtype,RARβ,mediates the effects of endogenous retinoic acid（RA）on neuronal growth and guidance in the adult newt CNS after ***,we now examine the expression of the retinoid X receptor RXRα（a potential heterodimeric transcriptional regulator with RARβ）,in newt tail and spinal cord *** show that at 21 days post-amputation（dpa）,RXRαis expressed at temporally distinct periods and in non-overlapping spatial domains compared to RARβ.Whereas RARβprotein levels increase,RXRαproteins level decrease by 21 dpa.A selective agonist for RXR,SR11237,prevents both this downregulation of RXRαand upregulation of RARβand inhibits tail and caudal spinal cord ***,treatment with a selective antagonist for RARβ,Le135,inhibits regeneration with the same morphological consequences as treatment with ***,Le135 treatment also inhibits the normal downregulation of RXRαin tail and spinal cord tissues at 21 *** results reveal a previously unidentified,indirect regulatory feedback loop between these two receptor subtypes in regulating the regeneration of tail and spinal cord tissues in this regeneration-competent newt.

On the effects of spatial resolution on effective distance measurement in digital landscapes

Background:Connectivity is an important landscape attribute in ecological studies and conservation practices and is often expressed in terms of effective *** the cost of movement of an organism over a landscape is effectively represented by a raster surface,effective distances can be equated with the cost-weighted distance of least-cost *** is generally recognized that this measure is sensitive to the grid’s cell size,but little is known if it is always sensitive in the same way and to the same degree and if not,what makes it more(or less)*** conducted computational experiments with both synthetic and real landscape data,in which we generated and analyzed large samples of effective distances measured on cost surfaces of varying cell sizes derived from those *** particular focus was on the statistical behavior of the ratio—referred to as‘accuracy indicator’—of the effective distance measured on a lower-resolution cost surface to that measured on a higher-resolution cost ***:In the experiment with synthetic cost surfaces,the sample values of the accuracy indicator were generally clustered around 1,but slightly greater with the absence of linear sequences(or barriers)of high-cost or inadmissible cells and smaller with the presence of such *** latter tendency was more dominant,and both tendencies became more pronounced as the difference between the spatial resolutions of the associated cost surfaces *** two real satellite images(of different resolutions with fairly large discrepancies)were used as the basis of cost estimation,the variation of the accuracy indicator was found to be substantially large in the vicinity(1500 m)of the source but decreases quickly with an increase in distance from ***:effective distances measured on lower-resolution cost surfaces are generally highly correlated with—and useful predictors of—effective distances measured on higher-resolution cost *** relationship t
