Arachis retusa Krapov. et W. C. Gregory et Valls is endemic in the West-central region of Brazil, occurring In areas endangered by human actions. The establishment of in vitro preservation methods for wild species of Arachis is an alternative to seed banks for germplasm storage, multiplication and distribution. The risk of genetic changes Induced by tissue culture and the monitoring of the genetic stability of the biological material before, during and after storage must be considered In the context of conservation. Random amplified polymorphlc ONA （RAPO） and amplified fragment length polymorphism （AFLP） fingerprinting were used to evaluate the genetic stability of in vitro plants originated from cotyledons and embryo axes of A. retusa. Cotyledons originated shoots through direct organogenesls and embryo axes displayed muItishoot formation Induced by 110 mmol/L and 8.8 mmol/L BAP, respectively. Ninety genomlc regions （loci） generated from RAPO and 372 from AFLP analyses were evaluated. All amplified fragments detected by both techniques in plants derived from the two explant types were monomorphic. The results Indicate that the recovered shoots are genetically stable at the assessed genomic regions.

AIm: To detect aneusomic changes with respect to chromosome 11 copy number in esophageal precancers and cancers wherein the generation of cancer-specific phenotypes is believed to be associated with specific chromosomal aneuploidies. mETHODS: We performed fluorescence in situ hybridization (FISH) on esophageal tissue paraffin sections to analyze changes in chromosome 11 copy number using apotome-generated images by optical sectioning microscopy. Sections were prepared from esophageal tumor tissue, tissues showing preneoplastic changes and histologically normal tissues (control) obtained from patients referred to the clinic for endoscopic evaluation. RESULTS: Our results demonstrated that aneusomy was seen in all the cancers and preneoplastic tissues, while none of the controls showed aneusomic cells. There was no increase in aneusomy from precancers to cancers. CONCLUSION: Our results suggest that evaluation of chromosome 11 aneusomy in esophageal tissue using FISH with an appropriate signal capture-analysis system, can be used as an ancillary molecular marker predictive of early neoplastic changes. Future studies can be directed towards the genes on chromosome 11,which may play a role in the neoplastic transformation of esophageal precancerous lesions to cancers.

Background： Despite the rapid growth in the incidence of acute myocardial infarction （AmI） in China, there is limited information about patients＇ experiences after AmI hospitalization, especially on long-tern1 adverse events and patient-reported outcomes （PROs）. methods： The China Patient-centered Evaluative Assessment of Cardiac Events （PEACE）-Prospective AmI Study will enroll 4000 consecutive Am I patients from 53 diverse hospitals across China and follow them longitudinally for 12 months to docunlent their treatment, recovery, and outcomes. Details of patients＇ medical history, treatment, and in-hospital outcomes are abstracted from medical charts. Comprehensive baseline interviews are being conducted to characterize patient demographics, risk factors, presentation, and healthcare utilization. As part of these interviews, validated instruments are administered to measure PROs, including quality of life, symptoms, mood, cognition, and sexual activity. Follow-up interviews, measuring PROs, medication adherence, risk factor control, and collecting hospitalization events are conducted at 1, 6, and 12 months after discharge. Supporting documents for potential outcomes are collected for adjudication by clinicians at the National Coordinating Center. Blood and urine samples are also obtained at baseline, 1 - and 12-month follow-up. In addition, we are conducting a survey of participating hospitals to characterize their organizational characteristics. Conclusion： The China PEACE-Prospective AmI study will be uniquely positioned to generate new information regarding patient＇s experiences and outcomes alter AmI in China and seiwe as a foundation for quality improveinent activities.

Platelet-rich plasma for muscle injuries: A systematic review of the basic science literature

BACKGROUND Platelet-rich plasma(PRP) is an increasingly used biologic adjunct for muscle injuries, as it is thought to expedite healing. Despite its widespread use, little is known regarding the mechanisms by which PRP produces its efficacious effects in some *** To clarify the effects of PRP on muscular pathologies at the cellular and tissue levels by evaluating the basic science *** A systematic review of Pubmed/mEDLINE and EmBASE databases was performed using the Preferred Reporting Items for Systematic Reviews and metaAnalyses(PRISmA) guidelines and checklist. Level III in vivo and in vitro studies examining PRP effects on muscles, myocytes and/or myoblasts were eligible for inclusion. Extracted data included PRP preparation methods and study *** Twenty-three studies were included(15 in vivo, 6 in vitro, 2 in vitro/in vivo). Only one reported a complete PRP cytology(platelets, and red and white blood cell counts). Five in vitro studies reported increased cellular proliferation, four reported increased gene expression, and three reported increased cellular differentiation. Five in vivo studies reported increased gene expression, three reported superior muscle regeneration, and seven reported improved histological quality of muscular *** The basic science literature on the use of PRP in muscle pathology demonstrates that PRP treatment confers several potentially beneficial effects on healing in comparison to controls. Future research is needed to determine optimal cytology,dosing, timing, and delivery methods of PRP for muscle pathologies.

The emergence of antibiotic-resistant bacterial strains and the weaponization of rare bacterial strains pose a potential threat of pandemic disease. Those involved in the treatment and control of infectious diseases have called for the development of a device capable of rapidly, simultaneously, and safely investigate a myriad of culture conditions. In response to this need, a microliter incubator array system is described and results from a proof-of-concept study using yeast cells to determine optimal growth conditions is presented.

The origin and biosynthesis of the naphthalenoid moiety of juglone in black walnut

Several members of the Juglandaceae family produce juglone,a specialized 1,4-naphthoquinone(1,4-NQ)natural product that is responsible for the notorious allelopathic effects of black walnut(Juglans nigra).Despite its documented ecological roles and potential for being developed as a novel natural product-based herbicide,none of the genes involved in synthesizing juglone have been *** on classical labeling studies,we hypothesized that biosynthesis of juglone’s naphthalenoid moiety is shared with biochemical steps of the phylloquinone ***,using comparative transcriptomics in combination with targeted metabolic profiling of 1,4-NQs in various black walnut organs,we provide evidence that phylloquinone pathway genes involved in 1,4-dihydroxynaphthoic acid(DHNA)formation are expressed in roots for synthesis of a compound other than *** experiments using axenic black walnut root cultures revealed that stable isotopically labeled L-glutamate incorporates into juglone resulting in the same mass shift as that expected for labeling of the quinone ring in *** together,these results indicate that in planta,an intermediate from the phylloquinone pathway provides the naphthalenoid moiety of ***,this work shows that juglone can be de novo synthesized in roots without the contribution of immediate precursors translocated from aerial *** present study illuminates all genes involved in synthesizing the juglone naphthoquinone ring and provides RNA-sequencing datasets that can be used with functional screening studies to elucidate the remaining juglone pathway *** of the generated knowledge is expected to inform future metabolic engineering strategies for harnessing juglone as a novel natural product-based herbicide.

＜正＞ The present study tested the hypothesis that extracellular O2. ? derived from NADPH oxidase （NOX） serves as an autocrine to activate physiological response in coronary arterial myocytes （CAms）. By simultaneously monitoring intracellular（[O2.-]i） and extracellular O2.-([O2.-]o levels,m1 - receptor agonist oxotremorine （OXO） was found to stimulate a more rapid increase in [O2.-]O than [O2.-]i in CAms. Addition of SOD outside cells blocked OXO- induced increases in both [O2.-]i and [O2.-]o. Silencing NOX subunit. Nox 1 by siRNAblocked increases in both [O2.-]i and [O2.-]o,but silencing Nox 4 only attenuated increase in [O2.-]*** ESR spectrometry. OXO was found to increase [O2.-]o by 35 %, which was blocked by Nox 1 siRNA （by 74%）. This ml - activation of NOX stimulated SOD - blockable Ca2+ release in CAms （△[Ca2+]i=821+++++67 nm）. NOX was also shown to be activated by NO donor, spermine NONOate （by 67 % ）. These results suggest that NOX - derived [O2.-]o exerts an autocrine action to stimulate intracellular Ca2+ release and that NOX can be activated by NO. which may counteract the action of excessive NO around CAms.

Resection of Vaginal Septum in Obstructed Hemivagina and Ipsilateral Renal Anomaly: Utility of Three Dimensional Intraoperative Ultrasound

Background: Obstructed hemivagina and ipsilateral renal anomaly (OHVIRA) syndrome is a rare congenital condition of the female urogenital tract, presenting intraoperative challenges. We demonstrate the utility of three dimensional intraoperative ultrasound to better delineate anatomy and aid in optimal resection of the vaginal septum. Case: A 12-year-old female was referred to pediatric and adolescent gynecology for irregular periods and evaluation of her gynecologic organs. Imaging studies confirmed OHVIRA syndrome. She underwent uncomplicated vaginal septum resection, guided by three dimensional intraoperative ultrasound. Summary and Conclusion: Intraoperative ultrasound can better delineate challenging anatomy. We show that three dimensional intraoperative ultrasound offers significant advantages over traditional two dimensional ultrasound and is a supplement to MRI, which may be helpful in complex anatomical cases like OHVIRA syndrome.

Factors Associated with Need for Drainage of Pleural Effusion after Diaphragm Surgery

Background: Diaphragm surgery is common with advanced ovarian malignancies. The purpose of this study is to determine associated factors with the need for drainage of pleural effusion after diaphragm surgery. methods and materials: A retrospective chart review was undertaken in all women undergoing debulking surgery for stage IIIc/IV ovarian cancer from 2007-2009. Results: One hundred and eight patients were found to be eligible for the study, but 73 were the primary focus of this paper due to having undergone debulking surgery, including diaphragmatic surgery, from 2007-2009. All 73 had ablation with the argon beam coagulator, 7 had a full thickness resection, and 7 had extensive peritoneal peel. Five patients had preoperative effusions. Only 7 patients required chest drainage postoperatively. Pre-operative ascites correlated closely with postoperative effusion (p-value = 0.031) but not with drainage (p-value = 0.068). The mean age of patients requiring drainage was significantly older (73 years) than that of patients who did not require drainage (60 years) (p-value = 0.002). Conclusion: Older patients undergoing diaphragm surgery are more likely to require chest tube or thoracentesis due to concurrent symptoms. Pre-operative ascites correlates closely with the development of postoperative effusion after diaphragm surgery.
