Objective:To investigate the in vitro antimicrobial activities of the leaf extract in different solvents viz.,methanol,ethanol and water extracts of the selected plant Ricinus ***:Ager well diffusion method and agar tube dilution method were carried out to perform the antibacterial and antifungal activity of methanol,ethanol and aqueous ***:Metbanol leaf extracts were found to be more active against Gram positive bacteria(Bacillus subtilis:ATCC 6059 and Staphylococcus aureus:ATCC 6538)as well as Gram negative bacteria(Pseudomonas aeruginosa:ATCC 7221 and Klebsiella pneumoniae)than ethanol and aqueous leaf *** activity of methanol and aqueous leaf extracts were also carded out against selected fungal strains as Aspergillus fumigatus and Aspergillus *** as well as aqueous leaf extracts of Ricinus communis were effective in inhibiting the fungal ***:The efficient antibacterial and antifungai activity of Ricinus communis from the present investigation revealed that the methanol leaf extracts of the selected plant have significant potential to inhibit the growth of pathogenic bacterial and fungal strains than ethanol and aqueous leaf extracts.

Determination of Ketoprofen in Human Plasma by RP-HPLC

For in vivo pharmacokinetic studies, it is pre-requisite to quantify drug concentrations in plasma. In the present study a RP-HPLC procedure was developed and validated for the assessment of ketoprofen in human plasma. For this purpose mobile phase consisting of methaol:water (70:30) adjusted to pH 3.3 with phosphoric acid was used, and chromatography was carried out on Discovery HS C18 column, 5 μm (25 cm × 4.6 mm). The flow rate was 1 mL·min-1 and quantitative assessment was performed at 260 nm. The retention time was found to be was found to be accurate and illustrated linearity from 0.2441 to 125 μg·mL-1 with the determination coefficient (r2) of 0.9999, also accuracy and precision were found to be <2 (%RSD). The intraday accuracy for concentrations 62.5 μg·mL-1, 15.625 μg·mL-1, 7.812 μg·mL-1 and 1.953 μg·mL-1 were found to be 99.747%, 99.475%, 98.457% and 99.824% respectively where as for interday accuracy consecutive values for days 1, 2 and 3 were 99.104%, 99.091%, 98.96% and 99.385% in plasma. All validation parameters were assessed and were found to be within the limits. The proposed method was accurate, specific, quick (retention time < 10 min), selective (showed no interference with excipients), cost effective and a good resolution which gave this method an advantage over the different other reported methods for the estimation of ketoprofen in human plasma.

Background: Type 2 diabetes mellitus (T2DM) is a chronic disease that has become a major health care concern, especially in developing countries like Pakistan. Lifestyle modification and appropriate pharmacotherapy are shown to improve blood glucose levels, lipid abnormalities and blood pressure. It is not known how many patients adhere to advice and drugs prescribed. This study aimed to determine adherence to lifestyle and therapeutic advice. Methods: A cross sectional hospital based study was conducted among patients attending the diabetic clinic at the Aga Khan University Hospital, using a structured questionnaire. Adult patients with T2DM and with at least one year duration of diabetes were included in the study. Results: Participants were aged between 32 and 92 years old with a mean age of 55.7 years old (SD ± 10.7). Mean duration of diabetes was 10.7 years old (SD ± 7.7). Majority (94%) of the patients were literate. Around half (47.3%) of the patients have had achieved glycemic target (HbA1c < 7%). Above target glycemic control was more common among patients with ischemic heart disease (68.1%), neuropathy (64.8%) and those on insulin (62.5%). Self-reported non-adherence for blood sugar monitoring (9.5%), physical activity (61.7%), tobacco use (43.4%) and foot care (43.9%) were noted. About 25% of the participants were not fully adherent to dietary advice. None of the patients from our study reported non-adherence to medications. Good adherence to physical activity was found in males with college degree. The highest percentage of tobacco use (33.3%) was reported among businessmen. Conclusion: We noted low adherence to advice for physical activity, tobacco use and SMBG, but a high adherence to prescribed medications and insulin. This was a selected group visiting a teaching hospital. This will need to be studied further in the community and efforts are required to motivate patients.
