The application of cutting fluid is significantly increased in the machining sector to improve ***,the inherent characteristics of cutting fluids on ecology,environment,and society shift the interest of researchers to work on environmentally friendly cooling conditions such as cryogenic ***,the effect of cutting speed and feed rate on the machining performance of the AISI‑L6 tool steel is investigated under cryogenic cooling ***,the L9 Taguchi based grey relational analysis(Gra)is conducted to investigate the essential machining indices such as cutting energy,surface roughness,tool wear,and material removal rate(MRR).The results indicate that the cutting speed of 160 m/min and feed rate of 0.16 mm/r are the optimum parameters that significantly improves the machining performance of AISI‑L6 tool steel.

Objective： The treatment of multi- fragmentary, intraarticular fractures of the distal humerus is difficult, even in young patients with bone of good quality. Small distal fragment, diminished bone mineral quality and increased trauma-associated joint destruction make stable joint reconstruction more problematic. The anatomically preshaped locking plates allow angular stable fixation for these complex fractures. We evaluated functional results of patients treated with open reduction and internal fixation with distal humerus locking plates for complex distal hu- merus fractures. Methods： Forty-three consecutive patients with ar- ticular fractures of the distal humerus were treated by open reduction and internal fixation with AO distal humerus plate system and locking reconstruction plates. Forty patients were available for the final outcome analysis. According to AO/ASIF classification, there were 2 cases of type A2, 4 cases of type A3, 1 case of type B1, 1 case of type B2, 14 cases of type C1, 7 cases of type C2 and 11 cases of type C3. Open reduction with triceps splitting technique was used in all patients. The clinical and radiographic follow-up was performed and outcome measures included pain assessment,range of motion, and Mayo elbow performance score. Results： Forty patients were available for the final outcome analysis. There were 29 males and 11 females with an average age of 38.4 years （18-73 years）. Clinical and ra- diological consolidation of the fracture was observed in all cases at an average of 11.6 weeks （9-14 weeks）. The average follow-up was 12 months （10-18 months）. Using the Mayo elbow performance score the results obtained were graded as excellent or good results in 33 patients （82.5%）. One pa- tient had superficial infection, and 4 had myositis ossificans. The prediction and optimization of surface roughness values remain a critical concern in nano-fluids based minimum quantity lubrication (NFMQL) turning of titanium (grade-2) ***,we discuss an application of response surface methodology with Box-Cox transformation to determine the optimal cutting parameters for three surface roughness values,i.e.,ra,Rq,and Rz,in turning of titanium alloy under the NFMQL *** surface roughness prediction model has been established based on the selected input parameters such as cutting speed,feed rate,approach angle,and different nano-fluids *** the multiple regression technique is used to find the relationship between the given responses and input ***,the experimental data were optimized through the desirability function *** findings from the current investigation showed that feed rate is the most effective parameter followed by cutting speed,different nano-fluids,and approach angle on ra and Rq values,whereas cutting speed is more effective in the case of Rz under NFMQL ***,the predicted results are comparatively near to the experimental values and hence,the established models of RSM using Box-Cox transformation can be used for prediction satisfactorily.

ras proteins are signal-transducing GTPases that cycle between inactive GDP-bound and active GTP-bound *** is a prolific signaling molecule interacting with a spectrum of effector molecules and acting through more than one signaling *** ras-effector proteins contain a ras-associating(ra)domain through which these associate with ras in a GTP-dependent *** ra domain is highly conserved among the members of the growth factor receptor-bound(Grb)7 family of proteins which includes Grb7,Grb10 and *** laboratory has reported an unusual observation that ra domain of Grb14 binds to the C-terminal nucleotide binding site of cyclic nucleotide gated channel(CTRCNGA1)and inhibits the channel *** modeling of the CTR-CNGA1 displays 50%---70%tertiary structural similarity towards ras *** named this region as ras-like domain(RLD).The interaction between ra-Grb14 and RLD-CNGA1 is mediated through a simple protein-protein interaction temporally and spatially regulated by light and *** is interesting to note that Grb14 binds to GTPase-mutant rab5,a ras-related small GTPase whereas Grb10 binds only to GTP-bound form of active rab5 but not to GTPase-defective mutant *** results suggest that Grb14 might have been evolved later in the evolution that binds to both ras and nucleotide binding proteins such as *** studies also suggest that eukaryotic CNG channels could be evolved through a gene fusion between prokaryotic ion channels and cyclic nucleotide binding proteins,both of which might have undergone several sequence variations for functional adaptation during evolution.

High-dose infliximab for treatment of pediatric ulcerative colitis:A survey of clinical practice

AIM:To assess attitudes and trends regarding the use of high-dose infliximab among pediatric gastroenterologists for treatment of pediatric ulcerative colitis(UC).METHODS:A 19-item survey was distributed to subscribers of the pediatric gastroenterology(PEDSGI) *** were submitted anonymously and results compiled in a secure ***:A total of 113 subscribers(88% based in the United States) responded(101 pediatric gastroenterology attendings and 12 pediatric gastroenterology fellows).There were 46% in academic medical institutions and 39% in hospital-based *** majority(91%) were treating >10 patients with UC;13% were treating >100 patients with UC;91% had prescribed infliximab(IFX) 5 mg/kg for UC;72% had prescribed IFX 10 mg/kg for *** a 5-point Likert scale,factors that influenced the decision not to increase IFX dosing in patients with UC included:"improvement on initial dose of IFX"(mean:3.88) and "decision to move to colectomy"(3.69).Lowest mean Likert scores were:"lack of guidelines or literature regarding increased IFX dosing"(1.96) and "insurance authorization or other insurance issues"(2.34)."Insurance authorization or other insurance issues" was identified by 39% as at least somewhat of a factor(Likert score ≥ 3) in their decision not to increase the IFX *** 10 mg/kg was more commonly used for the treatment of pediatric UC among responders based in the United States(75/100) compared to non-United States responders(6/13,P = 0.047).Induction of remission was reported by 78% of all responders and 81% reported maintenance of remission with IFX 10 mg/*** responder reported one death with IFX 10 mg/***:IFX 10 mg/kg is more commonly used in the United States to treat pediatric *** and safety data are required to avoid insurance barriers for its use.

Nociception is an important physiological process that detects harmful signals and results in pain perception. In this review, we discuss important experimental evidence involving some TRP ion channels as molecular sensors of chemical, thermal, and mechanical noxious stimuli to evoke the pain and itch sensations. Among them are the TRPA1 channel, members of the vanilloid subfamily （TRPV1, TRPV3, and TRPV4）, and finally members of the melastatin group （TRPM2, TRPM3, and TRPMS）. Given that pain and itch are pro-survival, evolutionarily-honed protective mechanisms, care has to be exercised when developing inhibitory/modulatory com- pounds targeting specific pain/itch-TRPs so that physio- logical protective mechanisms are not disabled to a degree that stimulus-mediated injury can occur. Such events have impeded the development of safe and effective TRPV1- modulating compounds and have diverted substantial resources. A beneficial outcome can be readily accom- plished via simple dosing strategies, and also by incorpo- rating medicinal chemistry design features during compound design and synthesis. Beyond clinical use, where compounds that target more than one channel might have a place and possibly have advantageous features, highly specific and high-potency compounds will be helpful in mechanistic discovery at the structure-function level.

We recently found that growth factor receptor-bound(Grb)protein 14 is a novel physiological modulator of photoreceptor specific cyclic nucleotide-gated channel alpha subunit(CNGA1).Grb14 promotes the CNG channel closure through its ras-associating(ra)*** the current study we show that this ra domain-mediated inhibition of rod CNG channel is electrostatic in ***14 competes with cGMP for the CNGA1 binding pocket and electrostatically interacts with Arg^(559) through a negatively chargedβ-turn at its ra ***,the three Glu residues(180–182)in Grb14 are absolutely critical for electrostatic interaction with the cGMP binding pocket and resultant *** study also demonstrates that substitution of Lys^(140) for Ala or in combination with polyglutamte mutants of Grb14 results in a significantly reduced binding with *** results suggest that in addition to Glu^(180–182) and Lys^(140),other residues in Grb14 may be involved in the electrostatic interaction with *** ra domain is highly conserved among the members of Grb7 family of proteins,which includes Grb7,Grb10 and ***,only Grb14 is able to modulate the channel activity,but not Grb7 and *** together,it suggests the existence of a divergence in ra domains among the members of the Grb7 family.

Consistent climatic perturbations have increased global environmental concerns, especially the impacts of abiotic stresses on crop productivity. Rice is a staple food crop for the majority of the world’s population. Abiotic stresses, including salt, drought, heat, cold and heavy metals, are potential inhibitors of rice growth and yield. Abiotic stresses elicit various acclimation responses that facilitate in stress mitigation. Plant hormones play an important role in mediating the growth and development of rice plants under optimal and stressful environments by activating a multitude of signalling cascades to elicit the rice plant’s adaptive responses. The current review describes the role of plant hormone-mediated abiotic stress tolerance in rice, potential crosstalk between plant hormones involved in rice abiotic stress tolerance and significant advancements in biotechnological initiatives including genetic engineering approach to provide a step forward in making rice resistance to abiotic stress.
