采用硅胶、大孔树脂、葡聚糖凝胶、MCI树脂、薄层色谱以及高效液相色谱等色谱分离方法,从鱼腥草(Houttuynia cordata)地上部分95%乙醇提取物的乙酸乙酯萃取部位分离得到10个化合物。运用质谱、核磁共振等波谱技术和文献对比法鉴定了其结构,分别鉴定为去氢催吐萝芙木醇(1)、5,6-环氧-3-羟基-β-紫罗兰酮(2)、黑麦草内酯(3)、pubinernoid A(4)、槲皮苷(5)、槲皮素(6)、阿福豆苷(7)、4-甲氧基苯-1,2-二醇(8)、4-羟基苯甲醛(9)、苯甲酸(10)。化合物1~4为首次从鱼腥草中分离得到的降碳倍半萜。对所有化合物进行了抗植物病原菌活性筛选,结果显示:在40 mg/mL浓度下,化合物5~7对禾谷镰刀菌、褐枝孢菌、番茄灰霉病菌、瓜类球腔菌和棉花枯萎病菌抑菌率介于38%~50%之间。

The homogeneity of the microstructure and composition are critical in determining the properties of rare earthbarium‐cuprate,single grain bulk superconductors[(RE)BCO].The magnitude of the trapped magnetic field achieveable in these technologically important materials,in particular,is influenced heavily by the size and distribution of(RE)_(2)baCuO_(x)(RE‐211)flux pinning inclusions in the bulk microstructure,whereas the size and distribution of silver agglomerates present within the bulk superconducting matrix correlate directly with improved mechanical *** careful engineering,these materials have significant potential for application in range of devices related to energy storage,medicine,electro‐magnetic machinery and microelectronic *** of(RE)BCO bulk superconductors typically involves heating a powder compact above its peritectic decomposition temperature followed by slow cooling to facilitate the growth of a single ***(RE)BCO composition has a different peritectic temperature and growth rate,which,therefore,necessitates different requirements in the heating *** fabrication temperature and growth rate,for example,may have an effect on the RE‐211 and silver distribution,which may,in turn,affect the superconducting properties of the resulting single *** this work we compare the distributions of silver and RE‐211 in the single grain microstructures of YBCOAg,GdBCO‐Ag,EuBCO‐Ag and SmBCO‐Ag bulk superconductors using optical microscopy,scanning electron microscopy and energy dispersive X‐ray *** observe that the distributions are very similar along both the a/b‐axis and c‐axis of these *** suggests that factors other than the maximum temperature used to achieve peritectic decomposition and the rate of single grain growth are particularly influential in determining the properties of the as‐processed samples in the top seeded melt growth *** observation demonstrates there is freedom to us

Well-crystallized MgZnO alloy thin films with hexagonal wurtzite structure were fabricated by sol-gel method. With the band gap increases, the surface roughness and the grain size reduces. It is worth noting that the intensity of the band-edge luminescence of Mg doped films enhances with the increase of the Mg content. The microstructure and photoluminescence mechanism have been discussed based on X-ray diffraction patterns, atomic force microscopy images, ultraviolet-visible absorption spectra, photoluminescence spectra and Fourier transform infrared spectra.

目的构建急性药物中毒性脑病患者重症监护室(intensive care unit, ICU)住院时间延长的预测模型并评价其效能。方法选择重症监护医疗信息集市(MIMIC)-Ⅳ2.2数据库中148例急性药物中毒性脑病患者作为研究对象,收集患者临床资料,根据ICU住院时间分非延长组(≤48 h)与延长组(>48 h)。采用最小绝对收缩和选择算子(LASSO)回归联合Logistic回归筛选变量,构建和绘制列线图。分别采用受试者工作特征曲线下面积(AUC)、Hosmer-Lemeshow校准曲线和决策曲线分析(DCA)评价模型的区分度、校准度及临床适用度。结果患者ICU住院时间1~15 d,其中ICU住院时间延长69例,采取LASSO回归与Logistic回归相结合方法筛选预测变量。结果显示SOFA评分、心率、合并心血管疾病、使用机械通气4个变量为独立危险因素,依据以上预测变量构建和绘制列线图,列线图的AUC为0.837,95%CI 0.774~0.900;Bootstrap内部验证AUC 0.873,95%CI 0.817~0.930,说明该列线图预测模型具有较好的预测能力。校准曲线和Hosmer-Lemeshow检验(χ^(2)=6.392,P=0.603)均显示该模型具有较高的一致性和拟合度;DCA结果表明,患者可从模型中净获益(阈值范围0.05~1.00),具有较好的临床适用性。结论本研究开发的模型性能良好,有助于评估急性药物中毒性脑病患者ICU住院时间的延长风险。

baCKGROUND Severe hyperthyroidism is a life-threatening exacerbation of thyrotoxicosis,characterized by high fever and multiorgan failure. The most common medical treatments are administration of antithyroid drugs and radioactive iodine, and thyroidectomy. In some patients, antithyroid therapy is limited due to serious adverse effects or failure to control disease progression. In some extreme cases,such as thyroid storm, conventional therapy alone does not yield effective and rapid improvement before the development of multiorgan *** SUMMARY This report describes a Chinese patient with severe hyperthyroidism accompanied by multiorgan failure, who was transferred to the medical intensive care unit of our hospital. The patient presented with palpitations, vomiting,diarrhea, and shortness of breath for a week. Laboratory tests showed elevation of thyroid hormones. Hepatic failure occurred with high aminotransferase levels and jaundice. Given her abnormal liver function and medication history, we could not exclude diagnosis of propylthiouracil-induced hepatic ***, she also suffered from heart failure. Therapeutic plasma exchange(commonly known as TPE) and continuous renal replacement therapy(commonly known as CRRT) were used as life-saving therapy, which resulted in notable improvement of clinical symptoms and laboratory *** Combined TPE and CRRT are safe and effective for patients with hyperthyroidism and multiorgan failure.

GNSS观测技术为研究板块运动、地震同震和震后形变以及无震慢滑移等地壳物理现象提供了重要的观测数据,其中的关键步骤是在充分考虑时间序列随机特征的基础上,精准提取时间序列的构造运动信息和非构造运动信息.本文提出一种基于贝叶斯框架的GNSS连续站坐标时间序列建模方法,顾及随机特征精准提取出模型参数的最优解及其误差.首先,引入基于贝叶斯框架的参数解算方法,获得模型参数的最优解及误差;其次,系统分析观测数据噪声随机特征和模型参数随机特征对模型解算结果的影响;最后,将该方法应用于青藏高原东北隅GNSS观测数据构造运动与非构造变形的有效提取.相比基于最小二乘拟合的GNSS连续站坐标时间序列建模传统方法,本文引入基于贝叶斯框架的参数最优解解算的GWMCMC算法,顾及随机特征精准高效提取出GNSS坐标时间序列中测站的构造运动趋势与非构造运动信息,能够更加准确获取参数的最优解及误差.同时相较传统的MCMC算法,GWMCMC算法通过并行计算改进算法的性能并提高了计算效率.本文的研究成果为后续利用构造变形开展地壳形变运动学特征和动力学机制提供数据支撑.

Emerging and recurrent infectious diseases caused by human coronaviruses(HCoVs)continue to pose a significant threat to global public health *** light of this ongoing threat,the development of a broad-spectrum drug to combat HCoVs is an urgently ***,we report a series of anti-pan-coronavirus ssDNA aptamers screened using Systematic Evolution of Ligands by Exponential Enrichment(SELEX).These aptamers have nanomolar affinity with the nucleocapsid protein(NP)of Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and also show excellent binding efficiency to the N proteins of both SARS,MERS,HCoV-OC43 and-NL63 with affinity KD values of 1.31 to 135.36 *** aptamer-based therapeutics exhibited potent antiviral activity against both the authentic SARS-CoV-2 prototype strain and the Omicron variant(ba.5)with EC50 values at 2.00 nM and 41.08 nM,*** protein docking analysis also evidenced that these aptamers exhibit strong affinities for N proteins of pan-coronavirus and other HCoVs(−229E and-HKU1).In conclusion,we have identified six aptamers with a high pan-coronavirus antiviral activity,which could potentially serve as an effective strategy for preventing infections by unknown coronaviruses and addressing the ongoing global health threat.

Currently,human health due to corona virus disease 2019(COVID-19)pandemic has been seriously *** coronavirus severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)spike(S)protein plays a crucial role in virus transmission and several S-based therapeutic approaches have been approved for the treatment of ***,the efficacy is compromised by the SARS-CoV-2 evolvement and *** we report the SARS-CoV-2 S protein receptor-binding domain(RBD)inhibitor licorice-saponin A3(A3)could widely inhibit RBD of SARS-CoV-2 variants,including Beta,Delta,and Omicron ba.1,XBB and ***,A3 could potently inhibit SARS-CoV-2 Omicron virus in Vero E6 cells,with EC50 of 1.016μ*** mechanism was related to binding with Y453 of RBD determined by hydrogen-deuterium exchange mass spectrometry(HDX-MS)analysis combined with quantum mechanics/molecular mechanics(QM/MM)***,phosphoproteomics analysis and multi fluorescent immunohistochemistry(mIHC)respectively indicated that A3 also inhibits host inflammation by directly modulating the JNK and p38 mitogen-activated protein kinase(MAPK)pathways and rebalancing the corresponding immune *** work supports A3 as a promising broad-spectrum small molecule drug candidate for COVID-19.

The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron ba.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both ba.2 and non-VOCD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both ba.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.
