Comprehensive molecular characterization to predict immunotherapy response in advanced biliary tract cancer:a phase II trial of pembrolizumab

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

Anti-inflammatory effects of natural volatile organic compounds from Pinus koraiensis and Larix kaempferi in mouse model

Natural volatile organic compounds(VOCs) extracted from conifers such as P. koraiensis and L. kaempferi have long been studied for their anti-oxidant, anti-proliferative, and anti-inflammatory effects. To evaluate the anti-inflammatory effects of VOCs from P. koraiensis and L. kaempferi, lipopolysaccharide(LPS) was administered to generate a mouse model for inflammation by the nasal route to the lungs and intraperitoneally to the whole body. VOCs of P. koraiensis and L. kaempferi were exposed to the mice by standardized wood panels with closed system. Increased levels of serum IgE and PGE2 were observed after exposure to dexamethasone and VOCs. We further determined the expression levels of inflammatory cytokine mRNA in the LPS-induced inflammation model by the reverse transcription quantitative polymerase chain reaction. Furthermore, the levels of cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β, and interleukin-13 were determined in peripheral blood mononuclear cells. Those inflammatory cytokines and the key enzyme for inflammation cyclooxygenase-2 expression in PBMCs were strongly reversed by dexamethasone and VOCs. Lung tissues after nasal LPS exposure showed increased cytokine mRNA expressions which were suppressed by treatment with dexamethasone and VOCs. Furthermore, the damage induced by LPS was attenuated by dexamethasone and VOCs. In conclusion, the results from the present study indicate that VOCs of P. koraiensis and L. kaempferi have a therapeutic potential in the treatment or prevention of local and systemic inflammation due to their immunosuppressive effects.

AIM To investigate the expression and prognostic role of programmed death ligand-1(PD-L1) in locally advanced esophageal squamous cell carcinoma(ESCC).METHODS A total of 200 patients with ESCC who underwent radical esophagectomy with standard lymphadenectomy as the initial definitive treatment in Seoul National University Hospital from December 2000 to April 2013 were eligible for this analysis. Tissue microarrays were constructed by collecting tissue cores from surgical specimens, and immunostained with antibodies directed against PD-L1, p16, and c-Met. Medical records were reviewed retrospectively to assess clinical outcomes. Patients were divided into two groups by PD-L1 status, and significant differences in clinicopathologic characteristics between the two groups were assessed. RESULTS Tumor tissues from 67 ESCC patients(33.5%) were PDL1-positive. Positive p16 expression was observed in 21 specimens(10.5%). The H-score for c-Met expression was ≥ 50 in 42 specimens(21.0%). Although PDL1-positivity was not significantly correlated with any clinical characteristics including age, sex, smoking/alcoholic history, stage, or differentiation, H-scores for c-Met expression were significantly associated with PDL1-positivity(OR = 2.34, 95%CI: 1.16-4.72, P = 0.017). PD-L1 expression was not significantly associated with a change in overall survival(P = 0.656). In contrast, the locoregional relapse rate tended to increase(P = 0.134), and the distant metastasis rate was significantly increased(HR = 1.72, 95%CI: 1.01-2.79, P = 0.028) in patients with PD-L1-positive ESCC compared to those with PD-L1-negative *** PD-L1 expression is positively correlated with c-Met expression in ESCC. PD-L1 may play a critical role in distant failure and progression of ESCC.

利用差分合成孔径雷达干涉测量技术(D-InSAR)获取长白山地区的形变场.结果显示1995～1998年期间,位于长白山东南侧的间白山火山存在6～12 cm的视线向形变。而长白山天池火山处于平静期,没有明显形变.利用2002～2003年的GPS和水准获取的形变数据,分别采用Mogi单源、双源模型反演了长白山地区火山的岩浆囊参数.其中双源模型拟合效果较为理想,两个点源一个位于长白山天池老火山口下方7.9 km处,另一个位于间白山火山下方5.5 km处.对双源模型反演得到的岩浆囊参数进行适当调整,拟合得到与InSAR形变场基本吻合的结果.上述研究结果表明长白山地区火山活动存在时间上的间歇性和空间上的迁移性,为进一步研究长白山地区火山活动机制提供了参考和依据.

Laser micromachining of CNT/Fe/Al_2O_3 nanocomposites

CNT/Fe/Al2O3 mixed powders were synthesized from Fe/Al2O3 nanopowders using thermal CVD for the homogeneous dispersion of carbon nanotubes CNTs. CNTs consisted of MWNT, and the diameter was approximately 20-30 nm. After sintering, CNTs were homogenously located throughout Al2O3 grain boundary and were buckled. A femto-second laser installed with special optical systems was used for micromachining of the nanocomposites. The relationship between material ablation rate and energy fluence was theoretically investigated and compared with experimental results from cross-sectional SEM analysis. The nanocomposites which have higher content of CNT show a fairly good machining result due to its higher thermal conductivity and smaller grain size as well as lower light transmittance.

A fluid-structure interaction method based on the arbitrary Lagrangian-Eulerian method and a dynamic mesh method was developed to simulate the dynamics of a rigid particle in shear *** the method,the governing equations for the fluid flow and particle motion were sequentially solved in a two-way coupling *** mesh system was deformed or re-meshed by the dynamic mesh *** method was employed to simulate the dynamics of a single particle suspended in a flow channel and the dynamics of the particle were *** simulation results show that the angular velocity is not only a function of the inclination angle,is but also influenced by the aspect ratio yielding a hysteresis,while the angular velocity obtained from the Keller-Scalak model is a function only of the inclination angle and does not show a *** present simulations clearly demonstrate that the Fluid-Structure Interaction（FSI） module is very stable,accurate and robust.

Performance of a New Ion Source for KSTAR Tokamak Plasma Heating

In the experimental campaign of 2010 and 2011 on KSTAR, the NBI-1 system was equipped with one prototype ion source and operated successfully, providing a neutral beam power of 0.7-1.6 MW to the tokamak plasma. The new ion source planned for the 2012 KSTAR campaign had a much more advanced performance compared with the previous one. The target performance of the new ion source was to provide a neutral deuterium beam of 2 MW to the tokamak plasma. The ion source was newly designed, fabricated, and assembled in 2011. The new ion source was then conditioned up to 64 A/100 keV over a 2-hour beam extraction and performance tested at the NB test stand （NBTS） at the Korea Atomic Energy Research Institute （KAERI） in 2012. The measured optimum perveance at which the beam divergence is a minimum was about 2.5μP, and the minimum beam divergent angle was under 1.0° at 60 keV. These results indicate that the 2.0 MW neutral beam power at 100 keV required for the heating of plasma in KSTAR can be delivered by the installation of the new ion source in the KSTAR NBI-1 system.

A Furan-Substituted Polymeric Hole-Transporting Material for Energy Level Regulation and Less Planarity in Colloidal Quantum Dot Solar Cells

For efficient colloidal quantum dot(CQD)solar cells(CQD-SCs),thiol-passivated p-type CQDs are generally used as a hole-transporting material(HTM);however,there are issues with the control of optoelectrical properties,low thiol passivation rate,and poor morphology with a power conversion efficiency(PCE)of approximately 11%.Although polymeric HTMs have been introduced to address these issues,maximizing efficiency and achieving green-solvent processability and thermal stability for commercialization is ***,we synthesize a novel benzodifuran(BDF)-based HTM(asy-ranPBTBDF)showing an electron-deficient state,low steric hindrance,and low planarity compared to those of a typical benzodithiophene(BDT)-based HTM(asy-ranPBTBDT).BDF properties lead to deep high occupied molecular orbital(HOMO)levels,closeπ-πstacking,excellent solubility,and amorphous properties related to efficiency,green-solvent processability,and thermal *** these benefits,the asy-ranPBTBDF-based CQD-SC showed enhanced open-circuit voltage(Voc)(0.65 V)and PCE(13.29%)compared to those of the asy-ranPBTBDT-based device(0.63 V and 12.22%)in toxic processes with *** asy-ranPBTBDF-based CQD-SC showed a PCE of 12.51%in a green-solvent process with 2-methylanisole and improved thermal stability at 80℃(83.8%retaining after 24 h)owing to less lateral crystallization than the asy-ranPBTBDT-based device(60.8%retaining after 24 h).
