The whole-genome survey of Acer griseum,its polymorphic simple sequence repeats development and application

Background:Acer griseum Pax is an endangered species endemic to China with both ornamental and economic value.However,the lack of information on its genome size and characteristics hinders further work at the genome level.Methods:This paper applied bioinformatics methods to predict the characteristics and patterns of the A.griseum genome,providing an important basis for formulating its whole-genome sequencing scheme.This study also characterized the simple sequence repeats(SSRs)of A.griseum,laying the foundation for the development and application of genome-wide SSR markers.In this study,PE150 sequencing was performed by the BGI MGISEQ platform,and the sequence files were analyzed by the K-mer method to estimate the characteristic information by GCE software.Results:The genome size was finally determined to be 739.63 Mb,its heterozygosity ratio was 1.33%,and the repetition ratio was 65.68%.A total of 825,960 SSR loci were identified in the assembled genome sequence,and primers were successfully designed for 526,020 loci.To verify the effectiveness of these primers,100 pairs of primers were randomly selected and synthesized,and 81 pairs successfully amplified the target fragments.Fourteen pairs of primers with good polymorphism were selected for principal component analysis of 31 A.griseum individuals from two populations,showing favorable heterozygosity and PIC values.According to the findings,these SSRs might identify genetic variations based on geographic areas.Conclusion:It is suggested that Illumina+PacBio assembly strategy should be used for whole-genome sequencing due to the high heterozygosity rate and high repetition rate of the genome.In addition,the SSR primers designed in batches in this study laid a foundation for the in-depth study of population structure and population maintenance mechanism of A.griseum,which is helpful for the effective conservation and sustainable utilization of this germplasm resource.

Plasma-exfoliated g-C_(3)N_(4) with oxygen doping: tailoring photocatalytic properties

Heteroatom doping and defect engineering have been proposed as effective ways to modulate the energy band structure and improve the photocatalytic activity of g-C_(3)N_(4). In this work, ultrathin defective g-C_(3)N_(4) was successfully prepared using cold plasma. Plasma exfoliation reduces the thickness of g-C_(3)N_(4) from 10 nm to 3 nm, while simultaneously introducing a large number of nitrogen defects and oxygen atoms into g-C_(3)N_(4). The amount of doped O was regulated by varying the time and power of the plasma treatment. Due to N vacancies, O atoms formed strong bonds with C atoms, resulting in O doping in g-C_(3)N_(4). The mechanism of plasma treatment involves oxygen etching and gas expansion. Photocatalytic experiments demonstrated that appropriate amount of O doping improved the photocatalytic degradation of rhodamine B compared with pure g-C_(3)N_(4). The introduction of O optimized the energy band structure and photoelectric properties of g-C_(3)N_(4). Active species trapping experiments revealed ·O_(2)^(–) as the main active species during the degradation.

The Dianchi golden-line barbel,Sinocyclocheilus grahami(Regan,1904),is one of the“Four Famous Fishes”of Yunnan Province,China.Given its economic value,this species has been artificially bred successfully since 2007,with a nationally selected breed(“S.grahami,Bayou No.1”)certified in 2018.For the future utilization of this species,its growth rate,disease resistance,and wild adaptability need to be improved,which could be achieved with the help of molecular markerassisted selection(MAS).In the current study,we constructed the first chromosome-level genome of S.grahami,assembled 48 pseudo-chromosomes,and obtained a genome size of 1.49 Gb.We also performed QTL-seq analysis of S.grahami using the highest and lowest bulks(i.e.,largest and smallest size)in both a sibling and random population.We screened two quantitative trait loci(QTLs)(Chr3,14.9-39.1 Mb and Chr17,4.1-27.4 Mb)as major growth-related locations.Several candidate genes(e.g.,map2k5,stat1,phf21a,sox6,and smad6)were also identified,with functions related to growth,such as cell differentiation,neuronal development,skeletal muscle development,chondrogenesis,and immunity.These results built a solid foundation for in-depth MAS studies on the growth traits of S.grahami.

经皮穴位电刺激治疗肺癌术后早期睡眠障碍随机对照研究

目的探讨经皮穴位电刺激(TEAS)对肺癌患者术后早期睡眠障碍的影响。方法采用随机对照研究方法,筛选2023年3—7月期间空军军医大学唐都医院肺癌手术患者100例,纳入患者84例,采用随机数字表法分为电刺激组(TEAS组)与对照组(CON组),每组42例。TEAS组从麻醉诱导前30 min至手术结束给予经皮穴位电刺激,穴位选用双侧神门(HT7)、内关(PC6)、三阴交(SP6);对照组(CON组)只粘贴电极片连接电刺激仪,但不接通电源。通过记录两组术前24 h、术后24 h、术后72 h及出院前的匹兹堡睡眠指数量表(PSQI)和阿森斯失眠量表(AIS)评估睡眠情况,记录各时间点的疼痛视觉模拟评分(VAS)等;采用多因素Logistic回归方法,分析影响术后睡眠障碍的主要因素。结果TEAS组术后24 h、术后72 h及出院前PSQI、AIS总分及VAS评分均低于CON组(P<0.05);TEAS组术后24 h睡眠障碍发生率(58.8%)低于CON组(80.5%)。切口疼痛、不能翻身为术后睡眠障碍的危险因素(OR分别为19.584、9.745),TEAS为保护因素(OR 0.228),差异均有统计学意义(P<0.01,P<0.05)。结论TEAS可一定程度改善肺癌术后早期睡眠障碍。切口疼痛、不能翻身为术后睡眠障碍的危险因素,TEAS为术后睡眠障碍的保护因素。

Effects of Acupotomy on Immobilization-Induced Gastrocnemius Contracture and Fibrosis in Rats via Wnt/β-Catenin Signaling

Objective: To determine whether acupotomy ameliorates immobilization-induced muscle contracture and fibrosis via Wnt/β-catenin signaling pathway. Methods: Thirty Wistar rats were randomly divided into 5 groups(n=6) by a random number table, including control, immobilization, passive stretching, acupotomy, and acupotomy 3 weeks(3-w) groups. The rat model of gastrocnemius contracture was established by immobilizing the right hind limb in plantar flexion for 4 weeks. Rats in the passive stretching group received passive stretching at gastrocnemius, a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Rats in the acupotomy and acupotomy 3-w groups received acupotomy once and combined with passive stretching at gastrocnemius a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Additionally, rats in the acupotomy 3-w group were allowed to walk freely for 3 weeks after 10-day therapy. After treatment, range of motion(ROM), gait analysis [i.e., paw area, stance/swing and maximum ratio of paw area to paw area duration(Max dA/dT)], gastrocnemius wet weight and the ratio of muscle wet weight to body weight(MWW/BW) were tested. Gastrocnemius morphometric and muscle fiber cross-sectional area(CSA) were assessed by hematoxylin-eosin staining. Fibrosis-related mRNA expressions(i.e., Wnt 1, β-catenin, axin-2, α-smooth muscle actin, fibronectin, and types Ⅰ and Ⅲ collagen) were measured using real-time quantitative polymerase chain reactions. Wnt 1, β-catenin and fibronectin concentrations were measured by enzyme-linked immunosorbent assay. Types Ⅰ and Ⅲ collagen in the perimysium and endomysium were analyzed using immunofluorescence. Results: Compared with the control group, ROM, gait function, muscle weight, MWW/BW and CSA were significantly decreased in the immobilization group(all P <0.01), while protein levels of types Ⅰ and Ⅲ collagen, Wnt 1, β-catenin, fibronectin and mRNA levels of fibrosis-related genes were obviously increased(all P<0.01). Printable,flexible ceramic fiber paper based on electrospinning

Based on the fact that it is challenging for the polymer flexible circuit substrates to meet the requirements of serving in high-temperature environments,this work proposed the idea of using printable ceramic fiber paper as a high-temperature flexible circuit substrate.A ceramic fiber paper with all ceramic components had been developed via electrospinning,solving the problems of low strength and severe strength drop at high temperatures of traditional ceramic fiber paper.The tensile strength of the prepared ceramic fiber paper is 2.63 MPa,and the reliable service temperature is 1200℃.Its bulk density is about 1.5 times that of traditional ceramic fiber paper.It can be printed with patterns by commercial inkjet printers like ordinary printing paper and has excellent printability.The feasibility of ceramic fiber paper as a flexible circuit substrate was verified by constructing a simple circuit.When the fiber paper is significantly bent,the circuit still forms a complete path,which proves that it has a strong application potential for high-temperature flexible circuit substrate and is expected to promote the development of flexible electronic devices serving at extreme high-temperature environments.

Enhancing flotation recovery of residual carbon from gasification waste by mixing hydrophobic powder with diesel as collector

Coal gasification fine slag(CGFS)is a solid waste containing residual carbon and ash generated during the coal gasification process,and the separation of the two components is the essential way to realize its environmental pollution reduction and resource value increase.Froth flotation is the preferred method for separating CGFS,but there is a barrier of low carbon recovery in this process due to the extensive adsorption of collector by the well-developed pores on residual carbon.In this study,a sufficiently simple yet innovative collector,a mixture of hydrophobic powder and diesel,was proposed in an attempt to break the bottleneck.Flotation experiments with common diesel and this novel collector were performed respectively,and FTIR,XPS,and SEM-EDX were employed to analyze the collector action mechanism.Flotation results revealed that the novel collector could significantly improve the residual carbon recovery;test results demonstrated that the novel collector could increase the hydrophobic functional group content on the fine slag surface,and the hydrophobic powders in this novel collector mainly appeared at the pore openings of the flotation concentrate.The essence of the mechanism is that the hydrophobic powders play a dual role of blocking pores and providing adsorption sites,thus facilitating the spreading of diesel on the carbon surface and promoting its floatability.The study can provide creative ideas for the efficient disposal of coal gasification waste.

A line-feature based SLAM algorithm is presented in this paper to resolve the conflict between the requirements of computational complexity and information-richness within the point-feature based SLAM algorithm, All operations required for building and maintaining the map, such as model-setting, data association, and state-updating, are described and formulated. This approach has been programmed and successfully tested in the simulation work, and results are shown at the end of this paper.

AIM:To explore the role of positron emission tomographycomputed tomography(PET-CT)examination in the diagnosis and treatment of ocular adnexal mucosa associated lymphoid tissue lymphoma(OAML).METHODS:The general clinical data,postoperative PET-CT results,treatment regimens,and the prognosis of 21 histopathologically confirmed OAML patients between October 2017 and September 2021 were collected.Among the 21 patients,five patients underwent surgical treatment alone,13 patients underwent surgical treatment combined with radiotherapy,and three patients underwent surgical treatment combined with chemotherapy.RESULTS:The follow-up period ranged from 8 to 79mo,with four cases of recurrence and no deaths.Through PETCT examination,two patients exhibited both local ocular metabolic elevation and systemic metastasis,and one of these patients had cervical lymph node metastasis,while the other had submandibular and parotid gland metastasis.Nine patients showed only local ocular metabolic elevation,while 10 patients had no abnormal metabolic activity locally.CONCLUSION:PET-CT examination plays a crucial role in detecting residual lesions and recurrence following tumor resection,aiding in precise disease staging,and facilitating the development of personalized treatment plans,ultimately improving patient prognosis.
