目的:探讨下调miR-223表达对脓毒症心肌病(SCM)小鼠心肌的保护作用及其机制。方法:按随机数字表法将27只8-10周龄SPF级雄性C57BL/6小鼠分配至SCM模型(0 h,6 h,12 h,18 h,24 h)时相组、Normal组、SCM组、miR-223 antagomir NC组、miR-223 antagomir组,每组3只。腹腔注射脂多糖(lipopolysaccharide,LPS)15 mg/kg构建SCM小鼠模型。miR-223 antagomir NC组与miR-223 antagomir组分别于建模前连续3天鼠尾静脉注射miR-223 antagomir NC、miR-223 antagomir预处理。采用反转录-聚合酶链反应(RT-PCR)研究SCM模型各个时相组小鼠心肌组织miR-223的表达情况。采用苏木素伊红(HE)染色法观察Normal组、SCM组、miR-223 antagomir NC组和miR-223 antagomir组小鼠心肌病理形态变化。采用酶联免疫吸附实验(ELISA)测定Normal组、SCM组、miR-223 antagomir NC组和miR-223 antagomir组小鼠血清cTnI、BNP、CK-MB、IL-6、IL-1β、TNF-α的含量并进行相关性分析。结果:SCM模型时相组小鼠随刺激时间延长,心肌组织miR-223表达水平逐渐升高。与Normal组比较,SCM组、miR-223 antagomir NC组、miR-223 antagomir组小鼠心肌组织出现不同程度损伤;血清心肌损伤标记物cTnI、BNP、CK-MB及炎性因子IL-6、IL-1β、TNF-α的表达水平均上升,差异具有统计学意义(P0.05);miR-223 antagomir组小鼠心肌组织病理损伤程度有所减轻,心肌损伤标记物cTnI、BNP、CK-MB及炎性因子IL-6、IL-1β、TNF-α水平下降,差异具有统计学意义(P<0.05)。相关性分析结果显示小鼠miR-223表达与心肌损伤标记物cTnI、BNP、CK-MB及炎性因子IL-6、IL-1β、TNF-α的表达呈正相关。结论:下调miR-223表达可通过减轻炎症反应对SCM小鼠心肌产生保护作用。

Development of practical and versatile electrophilic difluoromethylating reagent is still a grand challenge so far due to its inherent instability or low ***,we report the design and synthesis of difluoromethyl phenoxathiinium tetrafluoroborate(PT-CF_(2)H^(+)BF_(4)^(-)) as a novel difluoromethylating reagent,which proves to be a bench-stable,general,powerful and versatile reagent with divergent·CF_(2)H,CF_(2)H^(+),and:CF_(2) *** use of this reagent,we demonstrated a vast array of difluoromethyl radical transfer reactions via diverse pathways involving photocatalyst-free visible-light induction,visible-light photoredox catalysis and visible-light mediation single electron transfer of EDA ***,the green and highly effective CF_(2)H^(+) and:CF_(2) transfer reactions were also readily accomplished.

Aqueous potassium-ion batteries(APIBs),recognized as safe and reliable new energy devices,are considered as one of the alternatives to traditional *** MnO_(2),serving as the main cathode,exhibits a lower specific capacity in aqueous electrolytes compared to organic systems and operates through a different reaction *** application of highly conductive graphene may effectively enhance the capacity of APIBs but could complicate the potassium storage *** this study,a MnO_(2) cathode pre-intercalated with K~+ions and grown on graphene(KMO@rGO) was developed using the microwave hydrothermal method for ***@rGO achieved a specific capacity of 90 mA h g^(-1) at a current density of 0.1 A g^(-1),maintaining a capacity retention rate of>90% after 5000 cycles at 5 A g^(-1).In-situ and exsitu characterization techniques revealed the energy-storage mechanism of KMO@rGO:layered MnO_(2)traps a large amount of "dead" water molecules during K~+ions ***,the introduction of graphene enables these water molecules to escape during K~+ ions insertion at the *** galvanostatic intermittent titration technique and density functional theory confirmed that KMO@rGO has a higher K~+ions migration rate than MnO_(2).Therefore,the capacity of this cathode depends on the interaction between dead water and K~+ions during the energy-storage *** optimal structural alignment between layered MnO_(2) and graphene allows electrons to easily flow into the external *** charge compensation forces numerous low-solvent K~+ions to displace interlayer dead water,enhancing the *** unique reaction mechanism is unprecedented in other aqueous battery studies.

Cone-beam computed tomography(CBCT)is mostly used for position verification during thetreatrment ***,severe image artifacts in CBCT hinder its direct use in dosecalculation and adaptive radiation therapy re-planning for proton *** this study,animproved U-Net neural network named CBAM-U-Net was proposed for CBCT noise reductioninproton therapy which is a GBCT denoised U-Net network with convolutional block *** datasets contained 20 groups of head and neck *** CT images wereregistered to CBCT images as ground *** original CBCT denoised U-Net network,sCTU-Net,was trained for model performance *** synthetic CT(SCT)images generated byCBAM-U-Net and the original sCTU-Net are called CBAM-SCT and U-Net-SCT images,*** HU accuracies of the CT,GBCT,and SCT images were compared using fourmetrics:mean absolute error(MAE),root mean square error(RMSE),peak signal-to-noise ratio(PSNR),and structure simila rity index measure(SSIM).The mean values of the MAE,RMSE,PSNR,and SSIM of CBAM-SCT images were 23.80 HU,64.63 HU,52.27 dB,and 09919,respectively,which were superior to those of the U-Net-SCT *** evaluate dosimetricaccuracy,the range accuracy was compared for a single-energy proton *** r-index passrates ofa 4 cm X 4 cm scanned field and simple plan were calculated to compare the effects of thenoise reduction capabilities of the original U-Net and CBAM-U-Net on the dose ***-U-Net reduced noise more effectively than sCTU-Net,particularly in *** proposed a CBAM-U-Net model for CBCT noise reduction in proton *** tothe excellent noise reduction capabilities of CBAM-U-Net,the proposed model provided relativelyexplicit information rega rding patient ***,it maybe be used in dose calculation andadaptive treatment planning in the future.

The specialized equipment utilized in long-line tunnel engineering is evolving towards large-scale,multifunctional,and complex *** vibration caused by the high-frequency units during regular operation is supported by the foundation of the units,and the magnitude of vibration and the operating frequency fluctuate in different engineering contexts,leading to variations in the dynamic response of the *** high-frequency units yield significantly diverse outcomes under different startup conditions and times,resulting in failure to meet operational requirements,influencing the normal function of the tunnel,and causing harm to the foundation structure,personnel,and property in severe *** article formulates a finite element numerical computation model for solid elements using three-dimensional elastic body theory and integrates field measurements to substantiate and ascertain the crucial parameter configurations of the finite element *** proposing a comprehensive startup timing function for high-frequency dynamic machines under different startup conditions,simulating the frequency andmagnitude variations during the startup process,and suggesting functions for changes in frequency and magnitude,a simulated startup schedule function for high-frequency machines is created through *** into account the selection of the transient dynamic analysis step length,the dynamic response results for the lower dynamic foundation during its fundamental frequency crossing process are *** validation checks if the structural magnitude surpasses the safety threshold during the critical phase of unit startup traversing the structural resonance *** design recommendations for high-frequency units’dynamic foundations are provided,taking into account the startup process of the machine and ensuring the safe operation of the tunnel.

Constructing controllable thermal conduction networks is the key to efficiently improve thermal conductivities of polymer *** this work,graphite oxide(GO)and functionalized carbon nanotubes(f-CNTs)are combined to prepare“Line-Plane”-like hetero-structured thermally conductive GO@f-CNTs fillers,which are then performed to construct controllable 3D GO@f-CNTs thermal conduction networks via selfsacrificing template method based on oxalic ***,thermally conductive GO@f-CNTs/polydimethylsiloxane(PDMS)composites are fabricated via casting *** the size of oxalic acid is 0.24 mm and the volume fraction of GO@f-CNTs is 60 vol%,GO@f-CNTs/PDMS composites present the optimal thermal conductivity coefficient(λ,4.00 W·m^(-1)·K^(-1)),about 20 times that of theλof neat PDMS(0.20 W·m^(-1)·K^(-1)),also much higher than theλ(2.44 W·m^(-1)·K^(-1))of GO/f-CNTs/PDMS composites with the same amount of randomly dispersed ***,the obtained GO@f-CNTs/PDMS composites have excellent thermal stability,whoseλdeviation is only about 3%after 500 thermal cycles(20-200℃).

The growth and breakup processes of raindrops within a cloud influence the rain intensity and the sizes of raindrops on the *** Doppler velocity spectrum acquired by a vertically pointing radar(VPR)contains information on atmospheric turbulence and the size classification of falling *** this study,the four types of Convective Cells(CC)during precipitation events with more than 700 mm of precipitation in southern China are *** characteristics of four types of CCs correspond to the isolated convection,the early stage,the mature stage,and the decline stage of organizational convection,in that *** analysis using retrieval of vertical air motion(Vair)and raindrop evolution in clouds from Doppler velocity spectra collected by C-band VPR revealed the growth and breakup of falling raindrops with dynamic *** raindrops appear in the early stages and are accompanied by ice particles,which are impacted by the falling path᾽s *** aggregation,which is primarily related to the alternation of updraft and downdraft,accounts for the mature stage᾽s high efficiency of surface *** CCs in the decline stage originate from the shallow uplift in the weak and broad downdraft under conditions of enough water *** updraft dominates the stage of isolated *** of convective cells could be more accurately represented in model evaluations.
