Ammonia and greenhouse gas distribution in a dairy barn during warm periods

This research aimed to quantify concentrations of ammonia(NH3),carbon dioxide(CO_(2))and methane(CH_(4)),estimate emissions,and analyze the factors influencing them during warm periods in an open dairy barn equipped with two cooling systems in a Mediterranean climate *** distribution within the barn was observed to vary both vertically and horizontally,with the highest gas concentrations observed in the central area of the ***_(3),CH_(4)and CO_(2)ranged in 1.7–7.4,7–18,560–724μg·g^(–1),*** ventilation through openings and the operation of cooling systems induced changes in indoor microclimate conditions,influencing cow behavior and,consequently,gas *** concentrations were the highest at air velocities below 0.5 m·s^(–1).The highest concentration of NH_(3)was observed when the temperature-humidity index(THI)was>72 and≤78;and CO_(2)and CH_(4)concentrations were the highest with THI≥72 and decreased with THI≤***_(3)concentrations when barn management included three daily milkings were higher than those measured when barn management was based on two daily milkings,and lower for CH_(4)and CO_(2).NH_(3)and CH_(4)emissions were the highest during barn cleaning,while the lowest NH_(3)emissions occurred during activity of the cows(i.e.,feeding,walking).

BACKgROUND Autoimmunity has emerged as a probable disease modifier in patients with clinically diagnosed type 2 diabetes mellitus(T2DM),that is,patients who have insulin resistance,obesity,and other cardiovascular risk factors,suggesting that the presence of glutamic acid decarboxylase(anti-gAD65),islet antigen 2(anti-IA2),and zinc transporter 8(anti-Zn8T)antibodies could have deleterious effects on beta cell function,causing failure and earlier requirement for insulin *** To evaluate anti-gAD65,anti-IA2 and anti-Zn8T as predictors of early insulin requirement in adolescents with a clinical diagnosis of *** This was a case–control study in patients with clinically diagnosed with T2DM(68 cases and 64 controls with and without early insulin dependence respectively),male and female,aged 12–18 ***,blood pressure,glucose,insulin,C-peptide,glycated hemoglobin A1c,and lipid profiles were *** was used to measure anti-gAD65,anti-IA2,and anti-Zn8T *** statistics,Pearson'sχ2 test,Student's t test,and logistic regression was performed.Pge was 15.9±1.3 years,and there was a disease evolution time of 4.49±0.88 *** presence of anti-gAD65,anti-IA2,and anti-Zn8T positivity was found in 29.5%,18.2%,and 15.9%,*** the groups by early or no insulin dependence showed that the group with insulin had a higher frequency of antibody positivity:anti-gAD65 odds ratio(OR):2.42(1.112–5.303,P=0.026);anti-IA2:OR:1.55(0.859–2.818,P=0.105);and anti-Zn8T:OR:7.32(2.039–26.279,P=0.002).CONCLUSION Anti-gAD65 positivity was high in our ***-gAD65 and anti-Zn8T positivity showed a significantly depleted beta cell reserve phenotype,leading to an increased risk of early insulin dependence.

Background: We currently have international and national guidelines regarding the assessment and monitoring of clinical signs and humane endpoints in animals used in teaching and research, which make the performance of these activities mandatory for any experiment and professional working in this area. Assigning the severity of a research experiment is the result of an analysis of records of observations of the animal’s behavior, and clinical signs. The aim of this study was to describe the importance of carrying out a severity assessment associated with clinical and behavioral monitoring of rodents and rabbits during experimentation to maintain the welfare of these animals undergoing scientific research. Methods: The literature search was carried out using the following terms: “Monitoring”;“Humane endpoints”;“Animal welfare”, “Rodents”;“Rabbits”, and as connectors “and”;“or”, in the following databases: PubMed;LILACS/BIREME and SciELO. Results: A total of 987 articles were identified in the databases, and 20 of these studies were included in this review. Conclusions: Humane endpoint protocols and procedure severity tables are of the utmost importance, both from an ethical point and to refine the results of research conducted on laboratory animals. They should be drawn up jointly by the teams responsible for the project and the maintenance of the animals during the research period, and the data obtained should be published so that the scientific community can have access to it, helping to disseminate these practices, as well as helping to draw up new procedures. Monitoring and evaluating the welfare and clinical condition of animals undergoing scientific research procedures is the responsibility of the professors, researchers, veterinarians, and animal facility coordinators. Transcriptomic responses to drought stress in the Patagonian southern beech Nothofagus alpina

Background Deciphering the genetic architecture of drought tolerance could allow the candidate genes identification responding to water stress. In the Andean Patagonian forest, the genus Nothofagus represents an ecologically relevant species to be included in different genomic studies. These studies are scarce in South American ecosystems however represent an important source of genomic data in order to interpret future climate-change environment scenarios of these emblematic forests. Here, we achieved the assemblage of the transcriptome of N. alpina while searching for key genes of activated or suppressed metabolic pathways in response to drought *** De novo transcriptome assembly resulted in 104,030 transcripts. Following confirmation of drought conditions, based on reduction of leaf water potential and stomatal conductance, a differential gene expression analysis resulted in 2720 significantly expressed genes(1601 up-regulated and 1119 down-regulated). Enrichment analysis(over-representation analysis and gene set enrichment analysis) resulted in more than one hundred stress-responsive term ontologies(i.e. biological processes) and pathways. Terms such as response to abscisic acid and pathways such as plant hormone signal transduction or starch and sucrose metabolism were over-represented. Protein–protein interaction assessment resulted in networks with significantly expressed top common hub gene clusters(e.g. planttype cell wall biogenesis among down-regulated or ABA-signalling among up-regulated). The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614g.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOCD614g regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Combined estimation of state and feed-back gain for optimal load frequency control is *** frequency control(LFC)addresses the problem of controlling system frequency in response to disturbance,and is one of main research areas in power system operation.A well acknowledged solution to this problem is feedback stabilization,where the Linear Quadratic Regulator(LQR)based controller computes the feedback gain K from the known system parameters and implements the control,assuming the availability of all the state ***,this approach restricts control to cases where the state variables are readily available and the system parameters are ***,by estimating the states continuously from available measurements of some of the states,it can accommodate dynamic changes in the system *** paper proposes the technique of augmenting the state variables with controller *** introduces a non-linearity to the augmented system and thereby the estimation is performed using an Extended Kalman *** results in producing controller gains that are capable of controlling the system in response to changes in load demand,system parameter variation and measurement noise.

Treatment-induced apoptosis of cancer cells is one goal of cancer ***,more heat is generated by mitochondria during apoptosis,especially the uncoupled apoptotic state,^(1,2) compared to the resting *** this case study,we explore these thermal effects by longitudinally measuring temperature variations in a breast lesion of a pathological complete responder during neoadjuvant chemotherapy(NAC).Diffuse Optical Spectroscopic Imaging(DOSI)was employed to derive absolute deep tissue temperature using subtle spectral features of the water peak at 975 nm.^(3)A significant temperature increase was observed in time windows during the anthracycline and cyclophosphamide(AC)regimen but not in the paclitaxel and bevacizumab *** concentration changes generally did not follow temperature,suggesting the measured temperature increases were likely due to mitochondrial uncoupling rather than a direct vascular effect.A simultaneous increase of tissue oxygen saturation with temperature was observed,suggesting that oxidative stress also contributes to *** preliminary,this study indicates longitudinal DOSI tissue temperature monitoring provides information that can improve our understanding of the mechanisms of tissue response during NAC.

New few parameters differential evolution algorithm with application to structural identification

Differential evolution algorithm(DEA) is a stochastic, population-based global optimization method. In this paper, we propose new schemes for both mutation and crossover operators in order to enhance the performances of the standard DEA. The advantage of these proposed operators is that they are "parameters-less", without a tuning phase of algorithm parameters that is often a disadvantage of DEA. Once the modified differential evolutions are presented, a large comparative analysis is performed with the aim to assess both correctness and efficiency of the proposed operators. Advantages of proposed DEA are used in an important task of modern structural engineering that is mechanical identification under external dynamic loads. This is because of the importance of using a "parametersless" algorithm in identification problems whose characteristics typically vary strongly case by case, needing of a continuous set up of the algorithm proposed. This important advantage of proposed optimizers, in front of other identification algorithms, is used to develop a computer code suitable for the automatic identification of a simple supported beam subject to an impact load, that has been tested both using numerical simulations and real standard tests dynamic. The results point out that this algorithm is an interesting candidate for standard applications in structural identification problems.
