Exosomes derived from pulmonary metastatic sites enhance osteosarcoma lung metastasis by transferring the miR-194/215 cluster targeting MARCKS

Osteosarcoma,a prevalent primary malignant bone tumor,often presents with lung metastases,severely impacting patient survival *** vesicles,particularly exosomes,play a pivotal role in the formation and progression of osteosarcoma-related pulmonary ***,the communication between primary osteosarcoma and exosome-mediated pulmonary lesions remains obscure,with the potential impact of pulmonary metastatic foci on osteosarcoma progression largely *** study unveils an innovative mechanism by which exosomes originating from osteosarcoma pulmonary metastatic sites transport the miR-194/215 cluster to the primary tumor *** transportation enhances lung metastatic capability by downregulating myristoylated alanine-rich C-kinase substrate(MARCKS)*** this phenomenon,in this study we employ cationic bovine serum albumin(CBSA)to form nanoparticles(CBSA-anta-194/215)via electrostatic interaction with antagomir-miR-194/*** nanoparticles are loaded into nucleic acid-depleted exosomal membrane vesicles(anta-194/215@Exo)targeting osteosarcoma lung metastatic *** with bioengineered exosome mimetics(anta-194/215@Exo)not only impedes osteosarcoma progression but also significantly prolongs the lifespan of tumor-bearing *** findings suggest that pulmonary metastatic foci-derived exosomes initiate primary osteosarcoma lung metastasis by transferring the miR-194/215 cluster targeting MARCKS,making the miR-194/215 cluster a promising therapeutic target for inhibiting the progression of patients with osteosarcoma lung metastases.

ALKBH5 is a master regulator of N6-methyladenosine(m6A)modification,which plays a crucial role in many biological ***,we show that ALKBH5 is required for breast tumor ***,PRMT6 directly methylates ALKBH5 at R283,which subsequently promotes breast tumor ***,arginine methylation of ALKBH5 by PRMT6 increases LDHA RNA stability via m6A demethylation,leading to increased aerobic ***,PRMT6-mediated ALKBH5 arginine methylation is confirmed in PRMT6-knockout ***,these findings identify a PRMT6-ALKBH5-LDHA signaling axis as a novel target for the treatment of breast cancer.

A novel fracture-cavity reservoir outcrop geological knowledge base construction method considering parameter collection and processing, mutual transformation of data-knowledge, application and update

This study endeavors to formulate a comprehensive methodology for establishing a Geological Knowledge Base(GKB) tailored to fracture-cavity reservoir outcrops within the North Tarim Basin. The acquisition of quantitative geological parameters was accomplished through diverse means such as outcrop observations, thin section studies, unmanned aerial vehicle scanning, and high-resolution cameras. Subsequently, a three-dimensional digital outcrop model was generated, and the parameters were standardized. An assessment of traditional geological knowledge was conducted to delineate the knowledge framework, content, and system of the GKB. The basic parameter knowledge was extracted using multiscale fine characterization techniques, including core statistics, field observations, and microscopic thin section analysis. Key mechanism knowledge was identified by integrating trace elements from filling, isotope geochemical tests, and water-rock simulation experiments. Significant representational knowledge was then extracted by employing various methods such as multiple linear regression, neural network technology, and discriminant classification. Subsequently, an analogy study was performed on the karst fracture-cavity system(KFCS) in both outcrop and underground reservoir settings. The results underscored several key findings:(1) Utilization of a diverse range of techniques,including outcrop observations, core statistics, unmanned aerial vehicle scanning, high-resolution cameras, thin section analysis, and electron scanning imaging, enabled the acquisition and standardization of data. This facilitated effective management and integration of geological parameter data from multiple sources and scales.(2) The GKB for fracture-cavity reservoir outcrops, encompassing basic parameter knowledge, key mechanism knowledge, and significant representational knowledge, provides robust data support and systematic geological insights for the intricate and in-depth examination of the genetic

Synergistically constructed lamination-like network of redox-active polyimide and MXene via π-π interactions for aqueous NH_(4)^(+) storage

As a nonmetallic charge carrier,ammonium ion(NH_(4)^(+))has garnered significant attention in the construction of aqueous batteries due to its advantages of low molar mass,small hydration size and rapid diffusion in aqueous *** are a kind of potential electro-active materials for aqueous NH_(4)^(+)***,traditional polymer electrodes are typically created by covering the bulky collectors with excessive additives,which could lead to low volume capacity and unsatisfactory ***,a nanoparticle-like polyimide(PI)was synthesized and then combined with MXene nanosheets to synergistically construct an additive-free and self-standing PI@MXene composite ***,the redox-active PI nanoparticles are enclosed between conductive MXene flakes to create a 3D lamination-like network that promotes electron transmission,while theπ-πinteractions existing between PI and MXene contribute to the enhanced structural integrity and stability within the composite *** such,it delivers superior aqueous NH_(4)^(+)storage behaviors in terms of a notable specific capacity of 110.7 mA·h·cm^(–3) and a long lifespan with only 0.0064%drop each ***,in-situ Raman and UV–Vis examinations provide evidence of reversible and stable redox mechanism of the PI@MXene composite electrode during NH_(4)^(+)uptake/removal,highlighting its significance in the area of electrochemical energy storage.

Photocatalytic ozonation-based degradation of phenol by ZnO—TiO_(2)nanocomposites in spinning disk reactor

Spinning disk reactor(SDR)has emerged as a novel process intensification photocatalytic reactor,and it has higher mass transfer efficiency and photon utilization for the degradation of toxic organic pollutants by advanced oxidation processes(AOPs).In this study,ZnO—TiO_(2)nanocomposites were prepared by solgel method,and coated on the disk of SDR by impregnation-pull-drying-calcination *** performance of catalyst was characterized by X-ray diffraction,scanning electron microscope,X-ray photoelectron spectroscopy,photoluminescence and ultraviolet—visible diffuse reflectance *** ozonation in SDR was used to remove phenol,and various factors on degradation effect were studied in *** results showed that the rate of degradation and mineralization reached 100%and 83.4%under UV light irradiation after 50 min,compared with photocatalysis and ozonation,the removal rate increased by 69.3%and 34.7%,and mineralization rate increased by 56.7%and 62.9%,which indicated that the coupling of photocatalysis and ozonation had a synergistic *** radical capture experiments demonstrated that the active species such as photogenerated holes(h^(+)),hydroxyl radicals(·OH),superoxide radical(·O_(2)-)were responsible for phenol degradation,and·OH played a leading role in the degradation process,while h+and·O_(2)^(-)played a non-leading role.

Mikania micrantha is a fast-growing global invasive weed species that causes severe damage to natural ecosystems and very large economic losses of forest and crop *** has advantages in photosynthesis,including a similar net photosynthetic rate as C4 plants and a higher carbon fixation *** used a combination of genomics and transcriptomics approaches to study the evolutionary mechanisms and circadian expression patterns of *** ***,16 positive selection genes focused on photoreaction and utilization of *** different tissues,98.1%of the genes associated with photoresponse had high expression in stems,and more than half of the genes of the C4 cycle had higher expression in stems than in *** stomatal opening and closing,2 genes of carbonic anhydrase(CAs)had higher expression at 18:00 than at 8:00,and the slow anion channel 1(SLAC1)and high-leaf-temperature 1 kinase(HT1)genes were expressed at low levels at 18:*** addition,genes associated with photosynthesis had higher expression levels at 7:00 and 17:*** hypothesized that *** may undergo photosynthesis in the stem and flower organs and that some stomata of the leaves were opening at night by CO_(2)*** addition,its evolution may attenuate photoinhibition at high light intensities,and enhance more efficient of photosynthesis during low light *** the tissue-specific photosynthetic types and different diurnal pattern of photosynthetic-related genes may contribute to its rapid colonization of new habitats of ***.

Predictive value of preoperative routine examination for the prognosis of patients with pT2N0M0 or pT3N0M0 colorectal cancer

BACKGROUND In recent years,the incidence of colorectal cancer(CRC)has been *** the popularization of endoscopic technology,a number of early CRC has been ***,despite current treatment methods,some patients with early CRC still experience postoperative recurrence and *** To search for indicators associated with early CRC recurrence and metastasis to identify high-risk *** A total of 513 patients with pT2N0M0 or pT3N0M0 CRC were retrospectively enrolled in this *** of blood routine test,liver and kidney function tests and tumor markers were collected before *** were followed up through disease-specific database and telephone *** recurrence,metastasis or death were used as the end point of study to find the risk factors and predictive value related to early CRC recurrence and *** We comprehensively compared the predictive value of preoperative blood routine,blood biochemistry and tumor markers for disease-free survival(DFS)and overall survival(OS)of *** multivariate analysis demonstrated that low platelet count was significantly associated with poor DFS[hazard ratio(HR)=0.995,95% confidence interval(CI):0.991-0.999,P=0.015],while serum carcinoembryonic antigen(CEA)level(HR=1.008,95%CI:1.001-1.016,P=0.027)and serum total cholesterol level(HR=1.538,95%CI:1.026-2.305,P=0.037)were independent risk factors for *** cutoff value of serum CEA level for predicting OS was 2.74 ng/*** the OS of CRC patients with serum CEA higher than the cutoff value was worse than those with lower CEA level,the difference between the two groups was not statistically significant(P=0.075).CONCLUSION For patients with T2N0M0 or T3N0M0 CRC,preoperative platelet count was a protective factor for DFS,while serum CEA level was an independent risk factor for *** that these measures are easier to detect and more acceptable to patients,they may have broader applications.

Epigenetic control on transcription of vernalization genes and whole-genome gene expression profile induced by vernalization in common wheat

Vernalization is necessary for winter wheat to ***,it is unclear whether vernalization is also required for spring wheat,which is frequently sown in fall,and what molecular mechanisms underlie the vernalization response in wheat *** this study,we examined the molecular mechanisms that regulate vernalization response in winter and spring wheat *** this purpose,we determined how major vernalization genes(VRN1,VRN2,and VRN3)respond to vernalization in these varieties and whether modifications to histones play a role in changes in gene *** also identified genes that are differentially regulated in response to vernalization in winter and spring wheat *** found that in winter wheat,but not in spring wheat,VRN1 expression decreases when returned to warm temperature following *** finding may be associated with differences between spring and winter wheat in the levels of tri-methylation of lysine 27 on histone H3(H3K27me3)and tri-methylation of lysine 4 on histone H3(H3K4me3)at the VRN1 *** of winter wheat transcriptomes before and after vernalization revealed that vernalization influences the expression of several genes,including those involved in leucine catabolism,cysteine biosynthesis,and flavonoid *** findings provide new candidates for further study on the mechanism of vernalization regulation in wheat.
