Rapamycin and 3-methyladenine regulate apoptosis and autophagy in bone-derived endothelial progenitor cells

Background Mammalian target of rapamycin （mTOR） is involved in a caspase independent form of programmed cell death called autophagy. The aim of this research was to investigate the effects of rapamycin and 3-methyladenine （3-MA） on autophagy, proliferation, apoptosis, and cell-cycle parameters of rat bone marrow-derived endothelial progenitor cells （EPCs）. Methods Mononuclear cells isolated from rat bone marrow were treated with rapamycin （0.01, 0.1, 1, or 10 pg/L） or 3-MA （1.25, 2.5, 5, or 10 mmol/L） for 24 hours. Expression of the autophagy marker protein LC3-11 was analyzed by Western blotting. Apoptosis and cell-cycle progression were analyzed by flow cytometry. Cell proliferation was measured using the MTT assay. Results Rapamycin treatment of EPCs induced apoptosis and autophagy and inhibited proliferation and cell-cycle progression in a dose-dependent manner. Treatment with 5 mmol/L 3-MA promoted cell proliferation; in contrast, treatment with 10 mmol/L 3-MA promoted apoptosis and induced S-phase arrest. Conclusions Rapamycin treatment of EPCs induced apoptosis and autophagy. Low concentrations of 3-MA had no significant effect on the proliferation and apoptosis of EPCs; The 5 mmol/L group promoted cell proliferation, but had no effect on the apoptosis; the 10 mmol/L group inhibited the proliferation and promoted apoptosis through the cell cycle.

With the vigorous development of artificial intelligence(AI),intelligence applications based on deep neural networks(DNNs)have changed people’s lifestyles and production ***,the large amount of computation and data generated from the network edge becomes the major bottleneck,and the traditional cloud-based computing mode has been unable to meet the requirements of realtime processing *** solve the above problems,by embedding AI model training and inference capabilities into the network edge,edge intelligence(EI)becomes a cutting-edge direction in the field of ***,collaborative DNN inference among the cloud,edge,and end devices provides a promising way to boost ***,at present,EI oriented collaborative DNN inference is still in its early stage,lacking systematic classification and discussion of existing research *** by it,we have comprehensively investigated recent studies on EI-oriented collaborative DNN *** this paper,we first review the background and motivation of ***,we classify four typical collaborative DNN inference paradigms for EI,and analyse their characteristics and key ***,we summarize the current challenges of collaborative DNN inference,discuss future development trends and provide future research directions.

AIM： To quantify the circulating DNA in plasma from patients with hepatocellular carcinoma （HCC） and to evaluate its prognostic value. METHODS： Blood samples were collected from 79 patients with HCC before operation, 20 patients with liver cirrhosis, and 20 healthy volunteers. Circulating DNA was extracted from plasma and quantified. The association between circulating DNA level and prognosis of HCC patients was evaluated. RESULTS： Compared with the healthy volunteers （17.6 ± 9.5 ng/mL）, a significant higher circulating DNA level was found in the patients with HCC （47.1 ± 43.7 ng/ mL, P = 0.000） or with liver cirrhosis （30.0 ± 13.3 ng/ mL, P = 0.002）. The circulating DNA level was closely associated with tumor size （P = 0.008） and TNM stage （P = 0.040）, negatively associated with the 3-year diseasefree survival （DFS）（P = 0.017） and overall survival （OS）（P = 0.001）. CONCLUSION： Large or invasive tumor may release more circulating DNA, and higher level of circulating DNA may be associated with poor prognosis of HCC patients.

Stochastic parallel gradient descent based adaptive optics used for a high contrast imaging coronagraph

An adaptive optics （AO） system based on a stochastic parallel gradient descent （SPGD） algorithm is proposed to reduce the speckle noises in the optical system of a stellar coronagraph in order to further improve the contrast. The principle of the SPGD algorithm is described briefly and a metric suitable for point source imaging optimization is given. The feasibility and good performance of the SPGD algorithm is demonstrated by an experimental system featured with a 140-actuator deformable mirror and a Hartrnann-Shark wavefront sensor. Then the SPGD based AO is applied to a liquid crystal array （LCA） based coronagraph to improve the contrast. The LCA can modulate the incoming light to generate a pupil apodization mask of any pattern. A circular stepped pattern is used in our preliminary experiment and the image contrast shows improvement from 10^-3 to 10^-4.5 at an angular distance of 2A/D after being corrected by SPGD based AO.

Polarimetry plays an important role in the measurement of solar magnetic fields. We devel- oped a high-sensitivity and high-accuracy polarimeter （HHP） based on nematic liquid crystal variable retarders （LCVRs）, which has a compact setup and no mechanical moving parts. The system design and calibration methods are discussed in detail. The azimuth error of the transmission axis of the polarizer as well as the fast axes of the two LCVRs and the quarter-wave plate were determined using dedicated procedures. Linearly and circularly polarized light were employed to evaluate the performance of the HHP. The experimental results indicate that a polarimetric sensitivity of better than 5.7 × 10-3 can be achieved by using a single short-exposure image, while an accuracy on the order of 10-5 can be reached by using a large number of short-exposure images. This makes the HHP a high-performance system that can be used with a ground-based solar telescope for high-precision solar magnetic field investigations.

During embryonic development, pluripotent endoderm tissue in the developing foregut may adopt pancreatic fate or hepatic fate depending on the activation of key developmental regulators. Transdifferentiation occurs between hepato- cytes and pancreatic cells under specific conditions. Hepatocytes and pancreatic cells have the common endodermal progenitor cells. In this study we isolated hepatic stem/progenitor cells from embryonic day (ED) 12-14 Kun-Ming mice with fluorescence-activated cell sorting (FACS). The cells were cultured under specific conditions. The cultured cells deploy dithizone staining and immunocytochemical staining at the 15th, 30th and 40th day after isolation. The results indicated the presence of insulin-producing cells. When the insulin-producing cells were transplanted into alloxan- induced diabetic mice, the nonfasting blood glucose level was reduced. These results suggested that fetal liver stem/ progenitor cells could be converted into insulin-producing cells under specific culture conditions. Fetal liver stem/ progenitor cells could become the potential source of insulin-producing cells for successful cell transplantation therapy strategies of diabetes.

Anatomical variations in wood among four native species of Leguminosae grown in arid areas of China

Morphological features and anatomical variations are described and illustrated in detail for four native species of Legnminosae grown in arid sandy regions in China, which are Hedysarum scoparium *** Mey., Caragana korshiskii Kom., Lespedeza bicolor Turcz. and Ammopiptanthus mongolicus （Maxim. ex Kom.） Cheng f. All species showed similar morphological features： distinct growth ring boundaries, ring to semi-ring-porosity, simple perforation plates, alternate intervessel pitting, nonseptate fibers, paratracheal confluent axial parenchyma, helical thickenings and heterocellular rays. However, the vessel arrangement and their quantitative features were different. A. mongolicus had smaller vessel diameters and larger vessel fi＇equency, while the values in the other three species were similar, but bigger than those in A. mongolicus. The variation of vessel lengths and fiber lengths along a horizontal direction showed an irregular tendency. There were significant differences in both fiber lengths and vessel element lengths among trees and within trees, except for A. mongolicus. The relationships between anatomical features of secondary xylem and the adaptability of these species to desert environments are also discussed.

Identification and Promoter Activity Analysis of Porcine miR-181 and miR-1

Since its discovery a decade ago, microRNA has been identified as one of the major regulatory gene families in eukaryotic cells. Many functions of microRNAs have been revealed both in flora and fauna in recent years, but the transcriptional regulation of microRNA genes is not well-understood. In the present study, a series of primers were designed in the 2 000 nt upstream regions of porcine miR-181 and miR-1 and then the sequences were cloned into pGL3-basic vector to test their transcriptional activity. Dual-luciferase reporter assays showed that, the activity of 5＂-flanking sequence of miR-181 started on construct -51, decreasing with the length of the fragment up to -444. The upstream 590 bp confer maximal transcriptional activity and the basal promoter activity is located within the -82 to ＋16 bp region. For miR-1, the activity starts on construct -50, decreasing with the length of the fragment up to -1 254 in despite of a bit of fluctuation, and the basal promoter activity is located within the -50 to ＋47 bp region. Furthermore, some putative regulation elements of both miR-181 and miR-1 were located. In addition, tissue distribution revealed that miR-181 is expressed at a relatively low level.

A dispersion-flattened photonic crystal fiber with normal dispersion is designed for generating flat wideband supercontanuum, and the supercontinuum generation in this fiber is numerically analyzed. The results show that by appropriately designing the photonic crystal fiber, it can achieve flattened dispersion in the normal dispersion region. It is found that a fiber characterized by a flattened dispersion with a small normal dispersion is suitable for a flat wideband supercontinuum generation. In the process of spectral broadening, self-phase modulation effect plays a dominant role. By filtering the supercontinuum, pulses with different central wavelength over a wide spectral range can be obtained. The pulse width is determined by the bandwidth of the filter.
