High-precision modeling of tide-induced 3-D magnetic field and analysis of geomagnetic satellite orbit requirements

Ground-based magnetic observatories and geomagnetic satellites can observe the induced magnetic field generated by the motion of seawater containing sodium and chlorine ions.Calculating the three-dimensional(3-D)spatial distribution of tide-induced magnetic fields(TIMF)is crucial for inverting the electrical conductivity structure of the oceanic lithosphere.It also serves as an essential basis for designing optimal geomagnetic observatories and satellite orbits.However,existing methods for simulating TIMF suffer from limitations in inaccurately modeling realistic coastlines,heterogeneous land and sea surface properties,and complex deep Earth structures,thereby the interpretational level of TIMF data is reduced.To overcome this issue,we developed a tetrahedral-based finite element method for simulating TIMF,which can efficiently approximate realistic coastlines,heterogeneous land and sea surface properties,and complex deep Earth structures.Firstly,we derived the boundary value problem for the seawater motion-induced electromagnetic field,which was solved using the vector finite element method based on tetrahedral elements.Secondly,using the latest ocean depth and seafloor sediment layer models,we constructed a 3-D conductivity model of the Earth,which includes realistic coastlines,heterogeneous land and sea conductivity distributions.We then computed the TIMF using the M_(2)tidal source as an example and validated our method by comparing it with results obtained from spherical harmonic finite element and integral equation methods.Finally,utilizing the computed high-precision M_(2),N_(2),and O1 TIMF signals,we marked global observatories capable of observing strong M_(2),N_(2),and O1 TIMF signals and predicted alternative stations suitable for tide signal observations.Additionally,we calculated TIMF at heights of 450 and 200 km for the Macao Science Satellite 1 and its subsequent satellites.The results indicate that the amplitude of the tidal-induced magnetic field at 200 km is approximately twice that at 450 km.Th

Molecular cloning,characterization and promoter analysis of LbgCWIN1 and its expression profiles in response to exogenous sucrose during in vitro bulblet initiation in lily

Lily(Lilium spp.) is an important ornamental flower, which is mainly propagated by bulbs. Cell wall invertases(CWINs), which catalyze the irreversibly conversion of sucrose into glucose and fructose in the extracellular space, are key enzymes participating in sucrose allocation in higher plants. Previous studies have shown that CWINs play an essential role in bulblet initiation process in bulbous crops, but the underlying molecular mechanism remains unclear. Here, a CWIN gene of Lilium brownii var. giganteum(Lbg) was identified and amplified from genomic DNA. Quantitative RT-PCR assays revealed that the expression level of LbgCWIN1 was highly upregulated exactly when the endogenous starch degraded in non-sucrose medium during in vitro bulblet initiation in Lbg. Phylogenetic relationship, motif, and domain analysis of LbgCWIN1 protein and CWINs in other plant species showed that all sequences of these CWIN proteins were highly conserved. The promoter sequence of LbgCWIN1 possessed a number of alpha-amylase-, phytohormone-, light-and stress-responsive cis-elements. Meanwhile, β-glucuronidase(GUS) assay showed that the 459 bp upstream fragment from the translational start site displayed maximal promoter activity. These results revealed that LbgCWIN1 might function in the process of in vitro bulblet initiation and be in the response to degradation of endogenous starch.

Mikania micrantha is a fast-growing global invasive weed species that causes severe damage to natural ecosystems and very large economic losses of forest and crop production.It has advantages in photosynthesis,including a similar net photosynthetic rate as C4 plants and a higher carbon fixation capacity.We used a combination of genomics and transcriptomics approaches to study the evolutionary mechanisms and circadian expression patterns of M.micrantha.In M.micrantha,16 positive selection genes focused on photoreaction and utilization of photoassimilates.In different tissues,98.1%of the genes associated with photoresponse had high expression in stems,and more than half of the genes of the C4 cycle had higher expression in stems than in leaves.In stomatal opening and closing,2 genes of carbonic anhydrase(CAs)had higher expression at 18:00 than at 8:00,and the slow anion channel 1(SLAC1)and high-leaf-temperature 1 kinase(HT1)genes were expressed at low levels at 18:00.In addition,genes associated with photosynthesis had higher expression levels at 7:00 and 17:00.We hypothesized that M.micrantha may undergo photosynthesis in the stem and flower organs and that some stomata of the leaves were opening at night by CO_(2)signals.In addition,its evolution may attenuate photoinhibition at high light intensities,and enhance more efficient of photosynthesis during low light intensity.And the tissue-specific photosynthetic types and different diurnal pattern of photosynthetic-related genes may contribute to its rapid colonization of new habitats of M.micrantha.

Spatiotemporal evolution of surface urban heat islands: Concerns regarding summer heat wave periods

Many cities face heat wave(HW) events, combined with the existent surface urban heat island(SUHI) effects. This places pressure on human settlements and sustainable development. However, few studies have investigated the SUHI effects from the perspective of HWs. In this study, the summer HWs in Beijing from 2001 to 2021 were calculated, and the evolution of HWs and SUHIs was quantitatively analyzed based on the dynamic nature of the urban-rural boundary. Beijing experienced 27 HWs in the 21 years, including 10 instances in June, four in July, and 13 in August. The SUHI varied during HWs, between 2–3°C in most years. The highest SUHI occurred in 2019, reaching 3.99°C and covering the largest area(10,887 km~2). The fluctuation in HWs and SUHIs generally displayed the same spatiotemporal pattern, and HWs amplified the SUHIs to a certain extent, with the highest correlation coefficient being 0.44. Additionally, impervious surfaces and cropland contributed most to SUHIs,and night light enhanced SUHIs. Observing the co-evolution of HWs and SUHIs will be helpful for ecological maintenance and urban infrastructure optimization and provide theoretical support for reducing heat risk and improving the human settlement environment.

Although biomass-derived carbon(biochar)has been widely used in the energy field,the relation between the carbonization condition and the physical/chemical property of the product remains elusive.Here,we revealed the carbonization condition's effect on the morphology,surface property,and electrochemical performance of the obtained carbon.An open slit pore structure with shower-puff-like nanoparticles can be obtained by finely tuning the carbonization temperature,and its unique pore structure and surface properties enable the Li–O_(2) battery with cycling longevity(221 cycles with 99.8%Coulombic efficiency at 0.2 mA cm^(−2) and controlled discharge–charge depths of 500 mAh g^(−1))and high capacity(16,334 mAh g^(−1) at 0.02 mA cm^(−2)).This work provides a greater understanding of the mechanism of the biochar carbonization procedure under various pyrolysis conditions,paving the way for future study of energy storage devices.

Defects in phospholipase C zeta cause polyspermy and low fertilization after conventional IVF:not just ICSI failure

Phospholipase C zeta(PLC)is a key sperm-borne oocyte-activating factor that triggers Ca^(2+)oscillations and the subsequent block to polyspermy following gamete fusion.Mutations in PLCZ1,the gene encoding PLCζ,cause male infertility and intracytoplasmic sperm injection(ICSI)fertilization failure;and PLCζ expression and localization patterns are significantly correlated with ICSI fertilization rate(FR).However,in conventional in vitro fertilization(cIVF),whether and how sperm PLCζ affects fertilization remain unclear.Herein,we identified one previously reported and two novel PLCZ1 mutations associated with polyspermy in vitro that are characterized by excessive sperm-zona binding and a delay in pronuclei(PN)formation.Immunofluorescence staining and oocyte activation testing revealed that virtually all spermatozoa from patients lacked functional PLCζ and were thus unable to evoke Ca^(2+) oscillations.ICSI with an artificial oocyte activation treatment successfully rescued the polyspermic phenotype and resulted in a live birth.Furthermore,we analyzed PLCζ in an additional 58 males after cIVF treatment in the Reproductive and Genetic Hospital of CiTiC-Xiangya(Changsha,China)between February 2019 and January 2022.We found that the proportion of spermatozoa that expressed PLCζ was positively correlated with both 2PN rate and total FR.The optimal cutoff value below which males were likely to experience low FR(total FR≤30%)after clVF was 56.7%for the proportion of spermatozoa expressing PLC5.Our study expands the mutation and the phenotypic spectrum of PLCZ1 and further suggests that PLCζ constitutes a promising biomarker for identifying low FRs cases in cIVF due to sperm-related oocyte activation deficiency and that sperm PLCζ analysis may benefit the widermale population and not onlymen with IcsI failure.

Background Myelin oligodendrocyte glycoprotein(MOG)antibody-associated disease(MOGAD)is an autoimmune demy-elinating disorder of the central nervous system.Methods Extracted proteins from 34 cerebrospinal fluid(CSF)samples[patients with MOGAD(MOG group,n=12);healthy controls(HC group,n=12);patients with MOG seronegative and metagenomics next-generation sequencing-negative inflammatory neurological diseases(IND group,n=10)]were processed and subjected to label-free quantitative proteomics.Supervised partial least squares-discriminant analysis(PLS-DA)and orthogonal PLS-DA(O-PLS-DA)models were also performed based on proteomics data.Functional analysis of differentially expressed proteins(DEPs)was performed using Gene Ontology,InterPro,and Kyoto Encyclopedia Genes and Genomes.An enzyme-linked immunosorbent assay was used to determine the complement levels in serum from patients with MOGAD.Results Four hundred and twenty-nine DEPs(149 upregulated and 280 downregulated proteins)were identified in the MOG group compared to the HC group according to the P value and fold change(FC).Using the O-PLS-DA model,872 differentially abundant proteins were identified with variable importance projection(VIP)scores>1.Five proteins(gamma-glutamyl hydrolase,cathepsin F,interalpha-trypsin inhibitor heavy chain 5,latent transforming growth factor beta-binding protein 4 and leukocyte-associated immunoglobulin-like receptor 1)overlapping between the top 30 DEPs with top-ranked P value and FC and top 30 proteins in PLS-DA VIP lists were acquired.Functional analysis revealed that the dysregulated proteins in the MOG group were primarily involved in complement and coagulation cascades,cell adhesion,axon guidance,and glycosphingolipid biosynthesis compared to the HC group.Conclusion The proteomic alterations in CSF samples from children with MOGAD identified in the current study might provide opportunities for developing novel biomarker candidates.

Toroidal dipole response in rectangular waveguide:used to generate vector beams and vector vortex beams

Toroidal multipole is a special current distribution that has many different characteristics from electric multipole and magnetic multipole distributions.Because of its special properties,the toroidal dipole is a research hotspot in the field of metamaterials and nanophotonics.However,the low scattering of the toroidal dipole moment makes its excitation a challenging task.At present,there are relatively few studies on its specific engineering applications.In this paper,by slotting in the rectangular cavity,the excitation of an equivalent toroidal dipole is successfully achieved over a wide frequency range of 53-58 GHz.Results indicate that under the action of the toroidal dipole,the TE_(10)mode electromagnetic waves transmitted in the rectangular waveguide are converted into vector beams and are radiated outwards.Further adjusting the spatial distribution of the magnetic dipoles in the toroidal dipoles yields results that indicate that the resonance mode in the slot is still dominated by the magnetic toroidal dipole moment,and the electromagnetic waves radiating outward are vortex beams carrying vector polarization.The scattered energy of each dipole moment inside the antenna is calculated.This calculation verifies that the mass of the vector beam and vector vortex beam is closely related to the toroidal dipole supported by this antenna.The proposed structure can be applied to explorations in vortex filtering,in photon entanglement,and in the photonic spin Hall effect.

Light intensity regulates the sexual behaviors of oriental fruit fly Bactrocera dorsalis under laboratory conditions

The oriental fruit fly,Bactrocera dorsalis(Hendel),is a devastating pest of citrus fruits.After successful mating,adult females insert their eggs into the ripened fruit,resulting in moldy and rotten fruit and causing great economic losses for the citrus industry.In the field,flies initiate copulatory behaviors as twilight approaches,and decreasing light intensity in this period is the normal stimulus for copulation.In this study,ten light intensities ranging from 0–30000 lux were set to identify the typical intensity that strongly regulates the copulation behavior of B.dorsalis.Three light intensities found to regulate the copulation behavior were then selected to verify their effects on adult male wing fanning and female chemotaxis towards 2,3,5-trimethylpyrazine(TMP).At last,strong light and complete darkness were artificially combined in the lab to verify whether they could prevent copulation to inform behavioral manipulation of oriental flies in the future.The results indicated that adult flies generally initiated copulatory behaviors at low light intensity(<1000 lux).Stronger light significantly prevented copulation in proportion to intensity,with nearly no copulation events initiated when light intensity was above 20000 lux.Both male wing fanning and female chemotaxis towards TMP were attenuated as light intensity became stronger.However,at 10000 lux,males still fanned their wings to a certain extent while TMP completely lost its attractiveness to females.In the darkness,adults did not initiate any sexual behaviors,e.g.,copulation,wing fanning,or chemotaxis to TMP.One hour of strong light(10000 lux)combined with continuous darkness completely prevented mating.These results show that light condition is an essential factor for copulatory behaviors in the oriental fruit fly.Researchers could thus manipulate light conditions artificially or disrupt the molecular target in flies’light transduction pathway to develop environmentally-friendly techniques to control this pest.
