Protective effects of Wuwei Xiaodu Drink against chronic osteomyelitis through Foxp3^(+)CD25^(+)CD4^(+) Treg cells via the IL-2/STAT5 signaling pathway

To explore the effectiveness and safety of a Chinese medicinal decoction Wuwei Xiaodu Drink(WWXDD)in inhibiting chronic osteomyelitis via regulatory T cells *** effective constitutes of WWXDD and osteomyelitis related genes were *** proteins were cross-validated using the Venny *** function and KEGG pathway analysis were performed for target proteins,while pharmacological network was *** bone properties were analyzed by HE staining and the concentrations of immune factors were measured by *** expression of CTLA-4 and Foxp3 mRNA and STAT5,p-STAT5,CTLA-4 and Foxp3 protein were detected using Real-time PCR and Western blot,*** was used to analyze the percentages of cells.A total of 117 genes overlapped between 785 target genes of the active compounds of WWXDD and 912 osteomyelitis related ***-related genes,including IL-6,TNFα,IL-1βand IL-2 showed high connection degree in the drug-compound-disease-target *** function and KEGG pathway analysis revealed that 117 intersection genes mainly enriched in virus infection related pathways,immune related pathways and chemokine signaling ***,the development of chronic osteomyelitis was suppressed in model rats after treatment with ***,the concentrations of IL-2 and CD4^(+)CD25^(+)Foxp3 Treg perr centages together with the levels of p-STAT5,CTLA-4 and Foxp3 were also ***,IL-2 and WWXDD drug-containing serum exhibited opposite effects on regulating IL-2,IL-10,TGF-β1,Foxp3,CTLA4 and *** addition,a STAT5 phosphorylation inhibitor suppressed the expression of Foxp3 and *** can treat chronic osteomyelitis through suppressing the main regulating factors of Tregs and interfere its *** results bring a new solution for chronic osteomyelitis.

Using Copula functions to predict climatic change impacts on floods in river source regions

Exploration of potential biomarkers and therapeutic targets for trauma-related acute kidney injury

Purpose:Acute kidney injury(AKI)is one of the most common functional injuries observed in trauma ***,certain trauma medications may exacerbate renal ***,the early detection of trauma-related AKI holds paramount importance in improving trauma ***:Qualified datasets were selected from public databases,and common differentially expressed genes related to trauma-induced AKI and hub genes were identified through enrichment analysis and the establishment of protein-protein interaction(PPI)***,the specificity of these hub genes was investigated using the sepsis dataset and conducted a comprehensive literature review to assess their *** raw data from both datasets were downloaded using R software(version 4.2.1)and processed with the"affy"package19 for correction and ***:Our analysis revealed 585 upregulated and 629 downregulated differentially expressed genes in the AKI dataset,along with 586 upregulated and 948 downregulated differentially expressed genes in the trauma ***,the establishment of the PPI network and subsequent topological analysis highlighted key hub genes,including CD44,CD163,TIMP metallopeptidase inhibitor 1,cytochrome b-245 beta chain,versican,membrane spanning 4-domains A4A,mitogen-activated protein kinase 14,and early growth response ***,their receiver operating characteristic curves displayed areas exceeding 75%,indicating good diagnostic ***,our findings postulated a unique molecular mechanism underlying trauma-related ***:This study presents an alternative strategy for the early diagnosis and treatment of trauma-related AKI,based on the identification of potential biomarkers and therapeutic ***,this study provides theoretical references for elucidating the mechanisms of trauma-related AKI.

Dynamic mechanical characteristics of deep Jinping marble in complex stress environments

Rapid recovery of desert shrubs is attributed to imbalance of water supply to demand after disturbance in Northwest China

Background Disturbance is a key driver of changes in terrestrial ecosystems, especially in water limited regions. In the desert ecosystem of Northwest China, shrubs are often foraged by camels and this causes the shrubs lost their aboveground tissues. Although the aboveground tissue could recover after the disturbance, the mechanisms of resprouts' recovery after the disturbance are still poorly understood. We investigated the morphological and physiological characteristics and root water uptake sources of resprouts and non-disturbed seedlings of four desert shrub species that were often foraged by *** The results showed that the resprouts grew rapidly after the removal of aboveground tissue. The height and aboveground biomass of resprouts reached the level of 5-year-old seedlings following a growing season. Compared to un-disturbed seedlings, higher leaf water potential, leaf hydraulic conductance and photosynthesis rate were observed in resprouts, indicating that resprouts grew rapidly after disturbance. Furthermore, we found that the root water uptake source did not change after disturbance. This indicated that the roots of resprouts kept intact water uptake capacity after disturbance, which could keep supplying sufficient water to the aboveground tissue. Due to the removal of aboveground tissue of the resprouts, the water demand of aboveground tissue decreased, which induced an imbalance of water supply from roots to demand of aboveground tissues of resprouts. This explained the improved physiological performance and rapid recovery of resprouts after *** The imbalance of water supply to demand contributes to the rapid recovery of desert shrubs after disturbance. The findings provide insight into the eco-physiological mechanism for the maintenance of desert ecosystems that are facing disturbance.

Frozen ground(FG)plays an important role in global and regional climates and environments through changes in land freeze-thaw processes,which have been conducted mainly in different ***,the changes in land surface freeze-thaw processes under climate change on a global scale are still *** on ERA5-Land hourly land skin temperature data,this study evaluated changes in the global FG area,global land surface first freeze date(FFD),last freeze date(LFD)and frost-free period(FFP)from 1950 to *** results show that the current FG areas(1991-2020 mean)in the Northern Hemisphere(NH),Southern Hemisphere(SH),and globe are 68.50×10^(6),9.03×10^(6),and 77.53×10^(6)km^(2),which account for 72.4%,26.8%,and 60.4%of the exposed land(excluding glaciers,ice sheets,and water bodies)in the NH,SH and the globe,respectively;further,relative to 1951-1980,the FG area decreased by 1.9%,8.8%,and 2.8%,*** FG at lower latitudes degrades to intermittently FG,and intermittently FG degrades to non-frozen ground,which caused the global FG boundary to retreat to higher latitudes from 1950 to *** annual FG areas in the NH,SH,and globe all show significant decreasing trends(prend of FFP in the NH(1.34 d per decade)is larger than that in the SH(1.15 d per decade).
