CD4^+CXCR5^+ T cells activate CD27^+IgG^+ B cells via IL-21 in patients with hepatitis C virus infection

BACKGROUND： Chronic hepatitis C virus （HCV） infection causes the skewing and activation of B cell subsets, but the characteristics of IgG＋ B cells in patients with chronic hepa- titis C （CHC） infection have not been thoroughly elucidated. CD4＋CXCR5＋ follicular helper T （Tfh） cells, via interleukin （IL）-21 secretion, activate B cells. However, the role of CD4＋CXCR5＋ T cells in the activation ofIgG＋ B cells in CHC patients is not clear. METHODS： The frequency of IgG＋ B cells, including CD27-IgG＋ B and CD27＋IgG＋ B cells, the expression of the activation markers （CD86 and CD95） in IgG＋ B cells, and the percentage of circu- lating CD4＋CXCR5＋ T cells were detected by flow cytometry in CHC patients （n=70） and healthy controls （n=25）. The con- centrations of serum IL-21 were analyzed using ELISA. The role of CD4＋CXCR5＋ T cells in the activation of IgG＋ B cells was investigated using a co-culture system. RESULTS： A significantly lower proportion of CD27＋IgG＋ B cells with increased expression of CD86 and CD95 was observed in CHC patients. The expression of CD95 was negatively correlated with the percentage of CD27＋IgG＋ B cells, and it contributed to CD27＋IgG＋ B cell apoptosis. Circulating CD4＋CXCR5＋ T cells and serum IL-21 were significantly increased in CHC patients. Moreover, circulating CD4＋CXCR5＋ T cells from CHC patients induced higher expressions of CD86 and CD95 in CD27＋IgG＋ B cells in a co-culture system; the blockade of the IL-21 decreased the expression levels of CD86 and CD95 in CD27＋IgG＋ B ***： HCV infection increased the frequency of CD4＋CXCR5＋ T cells and decreased the frequency of CD27＋IgG＋ B cells. CD4＋CXCR5＋ T cells activated CD27＋IgG＋ B cells via the secretion of IL-21.

Dealloying by which the transition metal is partially or completely leached from an alloy precursor is an effective way to optimize the fundamental effects for further enhancing the electrocatalysis of a ***,to address the deficiencies associated with the commonly used dealloying methods,for example,electrochemical and sulfuric acid/nitric acid treatment,we report an acetic acid-assisted mild strategy to dealloy Cu atoms from the outer surface layers of CuPd alloy nanoparticles to achieve high-efficiency electrocatalysis for oxygen reduction and ethanol oxidation in an alkaline *** leaching of Cu atoms by acetic acid exerts an additional compressive strain effect on the surface layers and exposes more active Pd atoms,which is beneficial for boosting the catalytic performance of a dealloyed catalyst for the oxygen reduction reaction(ORR)and the ethanol oxidation reaction(EOR).In particular,for ORR,the CuPd nanoparticles with a Pd/Cu molar ratio of 2:1 after acetic dealloying show a half-wave potential of 0.912 V(***)and a mass activity of 0.213 AmgPd^(-1) at 0.9 V,respectively,while for EOR,the same dealloyed sample has a mass activity and a specific activity of 8.4 Amg^(-1) and 8.23 mA cm^(-2),respectively,much better than their dealloyed counterparts at other temperatures and commercial Pd/C as well as a Pt/C catalyst.

Association between benign prostatic hyperplasia, body mass index, and metabolic syndrome in Chinese men

Previous studies have showed that men suffering from diabetes mellitus, metabolic syndrome （MetS） and obesity have a higher risk of benign prostatic hyperplasia （BPH）. The present study aimed to examine the association between BPH, obesity, and features of MetS among men of the Hunan area of China. For this cross-sectional study, 904 males （aged 50-59 years） were included. MetS parameters, International Prostate Symptom Score （IPSS）, prostate-specific antigen （PSA） levels, total prostate volume （TPV）, postvoid residual volume （PVR） and maximum urine flow rate （Qmax） were measured. Results showed that MetS was associated with TPV （P = 0.048）, PVR （P = 0.004） and IPSS （P = 0.011）, but not with other indicators of BPH progression such as PSA levels or Qmax. MetS was associated with the voiding symptoms score （P〈 0.05）, but not with the storage symptom score. In addition, body mass index and fasting blood glucose positively correlated with TPV （r= 0.416, P〈 0.001; and r= 0.310, P= 0.011, respectively）. In conclusion, results suggest that MetS is associated with higher prostatic volume, prostate symptom score and voiding symptoms, but not with other features of prostatic hyperplasia such as PSA levels or Qmax. Changes in lifestyle factors, including physical activity and prevention of MetS, might be useful to prevent BPH and its progression, but further studies are needed.

Main observation and conclusion Perovskites(Ba_(0.5)Sr_(0.5))_(1-x)Co_(0.8)Fe_(0.2)O_(3-δ)(x=0.02,0.05,0.1 denoted as BSCF-0.98,BSCF-0.95,BSCF-0.9,respectively)with A-site cation defects are synthesized by simple and efficient sol-gel method and are proved to have better OER catalytic effect than the well-known(Ba_(0.5)Sr_(0.5))_(1-x)Co_(0.8)Fe_(0.2)O_(3-δ)(BSCF)***-0.95 exhibits the best OER catalytic activity in the series *** current density of BSCF-0.95 is about 56%higher than that of BSCF oxide at a potential of 1.7 *** experimental studies have shown that compared with BSCF,BSCF-0.95 oxide has a larger electrochemical surface area(ECSA),a higher content of O_(2)^(2–)species related to surface oxygen vacancies,and faster charge transfer rate,which may be the factors for the enhancement of OER *** theoretical calculation results prove that the center positions of the O 2p-band of perovskite with A-site cation defects are closer to the Fermi level than BSCF oxide,which agrees with the OER performance trend of the material.

Synthesis of Leucite from Potash Feldspar

Leucite particles were synthesized from feldspar mixed with 0% to 52% potassium nitrate fired from 800 ℃ to 1 200 ℃ by solid state method. The X-ray Diffraction （XRD） patterns show that in the temperature range from 800 ℃ to 1 200 ℃, the leucite can be removed as the single crystalline phase. Kalsilite may be crystallized with leucite at 800 ℃, but can be eliminated after prolonged heating. The scanning electron Microscopy （SEM） images clearly display the that crystals of micrometer scale leucite, and the leucite crystals distribute evenly in the matrix. The Thermal expansion coefficient （TEC） of the samples fabricated is as high as 20.52×10^-6 ℃^-1 measured from 20 ℃ to 500 ℃. The mechanism of transformation from feldspar to leucite was proposed.

The 2.5 m wide-field and high-resolution solar telescope(WeHoST)is currently under developing for solar *** aims to achieve high-resolution observations over a super-wide field of view(FOV)of5′×5′,and a desired resolution of 0.3″.To meet the scientific requirements of WeHoST,the ground-layer adaptive optics(GLAO)with a specially designed wave front sensing system is as the primary *** introduce the GLAO configuration,particularly the wave front sensing *** analytic method,we simulate the performance of both classical AO and GLAO systems,optimize the wave front sensing system,and evaluate GLAO performance in terms of PSF uniformity and correction improvement across whole *** results indicate that,the classical AO will achieve diffraction-limited resolution;the suggested GLAO configuration will uniformly improve the seeing across the full 5′×5′FOV,reducing the FWHM across the axis FOV to less than0.3″(λ≥705 nm,r0≥11 cm),which is more than two times *** specially designed wave front sensor schedule offers new potential for WeHoST’s GLAO,particularly the multi-FOV GLAO and the flexibility to select the detected *** capabilities will significantly enhance the scientific output of the telescope.

Retinoic acid receptor α promotes autophagy to alleviate liver ischemia and reperfusion injury

AIM: To study the role of autophagy and the relationship between retinoic acid receptor α(RARα) and autophagy in liver ischemia and reperfusion(IR) ***: All-trans retinoic acid(ATRA) was administered to mice for two weeks before operation. Reverse transcription-polymerase chain reaction and Western blot were used to detect the expression levels of related factors. To demonstrate the role of RARα,LE540,a RARα inhibitor,was used to treat hepatocytes injured by H2O2 in ***: ATRA pretreatment noticeably diminished levels of serum alanine aminotransferase and as-partate aminotransferase as well as the degree of histopathological changes. Apoptosis was also inhibited,whereas autophagy was promoted. In vitro,RARα was inhibited by LE540,which resulted in decreased autophagy and increased apoptosis. Similarly,the expression of Foxo3 a and p-Akt was downregulated,but Foxo1 expression was ***: This research provides evidence that ATRA can protect the liver from IR injury by promoting autophagy,which is dependent on Foxo3/p-Akt/Foxo1 signaling.

Structural evolutions and electronic properties of Au_nGd(n=6–15)small clusters:A first principles study

Structural, electronic, and magnetic properties of AunGd （n = 6-15） small clusters are investigated by using first principles spin polarized calculations and combining with the ab-initio evolutionary structure simulations. The calculated binding energies indicate that after doping a Gd atom AunGd cluster is obviously more stable than a pure AUn＋l cluster. Au6Gd with the quasiplanar structure has a largest magnetic moment of 7.421 gB. The Gd-4f electrons play an important role in determining the high magnetic moments of AunGd clusters, but in Au6Gd and Aul2Gd clusters the unignorable spin polarized effects from the Au-6s and Au-5d electrons further enhance their magnetism. The HOMO-LUMO （here, HOMO and LUMO stand for the highest occupied molecular orbital, and the lowest unoccupied molecular orbital, respectively） energy gaps of munGd clusters are smaller than those of pure AUn＋l clusters, indicating that AunGd clusters have potential as new catalysts with enhanced reactivity.

Influence of non-uniform ultrasonic vibration on casting fluidity of liquid aluminum alloy

The application of ultrasonic vibration to the casting process can be realized through mould(die)***,the resonant vibration of the mould is always accompanied by a non-uniform vibration distribution at different parts,which may induce a complex liquid flow and affect the casting fluidity during the mould filling *** influence of non-uniform ultrasonic vibration on the fluidity of liquid AlSi9Cu3 alloy was studied by mould vibration with different vibration *** is found that ultrasonic mould vibration can generate two opposite effects on the casting fluidity:the first,ultrasonic cavitation in melt induced by mould vibration promotes the casting fluidity;the second,the non-uniform mould vibration can induce a melt flow toward the weak vibration areas and turbulence there,consequently decreasing the casting *** the melt flow and turbulence are violent enough to offset the promoting effect of cavitation on fluidity,the ultrasonic vibration will finally induce a resultant decrease of casting *** decreasing effect is proportional to the vibration gradient.
