YKL40 expression in CD14^+ liver cells in acute and chronic injury

AIm:To demonstrate that CD14 + cells are an important source of the growth factor YKL40 in acute and chronic liver ***:rats were inoculated with one dose of CCl4 to induce acute *** biopsies were obtained at 0,6,12,24,48 and 72 *** chronic damage,CCl4 was administered three days per week for 6 or 8 *** samples were collected,and cellular populations were isolated by liver digestion and purified by cell ***40 mrNA and protein expression were evaluated by realtime polymerase chain reaction and western ***:Acute liver damage induced a rapid increase of YKL40 mrNA beginning at 12 *** peaked at 24 h,with a 26fold increase over basal *** 72 h however,YKL40 expression levels had nearly returned to control *** the other hand,chronic damage induced a sustained increase in YKL40 expression,with 7and 9fold higher levels at 6 and 8 wk,*** pattern of YKL40 expression in different subpopulations showed that CD14+cells,which include Kupffer cells,are a source of YKL40 after acute damage at 72 h[0.09 relative expression units(rEU)]as well as after chronic injury at 6 wk(0.11 rEU).Hepatocytes,in turn,accounted for 0.06 and 0.01 rEU after 72 h(acute)or 6 wk(chronic),*** rest of the CD14cells(including T lymphocytes,B lymphocytes,natural killer and natural killer T cells) yielded 0.07 and 0.15 rEU at 72 h and 6 wk,***40 protein expression in liver was detected at 72 h as well as 6 and 8 wk,with the highest expression relative to controls(11fold;P≤0.05)seen at 6 *** were stimulated by *** demonstrate that under these conditions,these cells showed maximum expression of YKL40 at 12 h,with Pmpared with ***:Hepatic CD14 + cells are an YKL40 mrNA and protein source in acute and chronic liver injury,with expression patterns similar to growth factors implicated in inflammationfibrogenesis.

Background Alteplase improves functional outcomes of patients with acute ischaemic stroke,but its effects on symptomatic infarct swelling,an adverse complication of stroke and the influence of CT hyperdense artery sign(HAS)are *** substudy of the Third International Stroke Trial aimed to investigate the association between HAS and symptomatic infarct swelling and effect of intravenous alteplase on this *** We included stroke patients whose prerandomisation scan was non-contrast ***,masked to clinical information,assessed baseline(prerandomisation)and follow-up(24-48 hours postrandomisation)CT scans for HAS,defined as an intracranial artery appearing denser than contralateral *** infarct swelling was defined as clinically significant neurological deterioration≤7 days after stroke with radiological evidence of midline shift,effacement of basal cisterns or uncal *** Among 2961 patients,HAS presence at baseline was associated with higher risk of symptomatic infarct swelling(Or 2.21;95% CI 1.42 to 3.44).Alteplase increased the risk of swelling(Or 1.69;95% CI 1.11 to 2.57),with no difference between patients with and those without baseline HAS(p=0.49).In patients with baseline HAS,alteplase reduced the proportion with HAS at follow-up(Or 0.67;95% CI 0.50 to 0.91),where HAS disappearance was associated with reduced risk of swelling(Or 0.25,95% CI 0.14 to 0.47).Conclusion Although alteplase was associated with increased risk of symptomatic infarct swelling in patients with or without baseline HAS,it was also associated with accelerated clearance of HAS,which in return reduced swelling,providing further mechanistic insights to underpin the benefits of alteplase.

This article provides a review of tropical cyclone(TC)surface wind estimation from an operational forecasting ***,we provide a summary of operational forecast center practices and historical ***,we discuss current and emerging objective estimates of TC surface winds,including algorithms,archive datasets,and individual algorithm strengths and weaknesses as applied to operational TC surface wind forecast *** review leads to recommendations about required surface coverage—an area covering at least 1100 km from center of TC at a 2-km resolution in the inner-core,and at a frequency of at least once every 6 *** is enough coverage to support a complete analysis of the TC surface wind field from center to the extent of the 34-kt(17 m s^(-1))winds at 6-h *** also suggest future designs of TC surface wind capabilities include funding to ensure near real-time data delivery to operators so that operational evaluation and use are feasible within proposed ***,we suggest that users of archived operational wind radii datasets contact operational organizations to ensure these datasets are appropriate for their needs as the datasets vary in quality through time and space,even from a single organisation.

Bacterial vaginosis:a synthesis of the literature on etiology,prevalence,risk factors,and relationship with chlamydia and gonorrhea infections

Bacterial vaginosis(BV) is a common vaginal disorder in women of reproductive age. Since the initial work of Leopoldo in 1953 and Gardner and Dukes in 1955, researchers have not been able to identify the causative etiologic agent of BV. There is increasing evidence, however, that BV occurs when Lactobacillus spp., the predominant species in healthy vaginal flora, are replaced by anaerobic bacteria, such as Gardenella vaginalis, mobiluncus curtisii, m. mulieris, other anaerobic bacteria and/or mycoplasma hominis. Worldwide, it estimated that 20%–30% of women of reproductive age attending sexually transmitted infection(STI) clinics suffer from BV, and that its prevalence can be as high as 50%–60% in high-risk populations(e.g., those who practice commercial sex work(CSW). Epidemiological data show that women are more likely to report BV if they: 1) have had a higher number of lifetime sexual partners; 2) are unmarried; 3) have engaged in their first intercourse at a younger age; 4) have engaged in CSW, and 5) practice regular douching. In the past decade, several studies have provided evidence on the contribution of sexual activity to BV. However, it is difficult to state that BV is a STI without being able to identify the etiologic agent. BV has also emerged as a public health problem due to its association with other STIs, including: human immunodeficiency virus(HIV), herpes simplex virus type 2(HSV-2), Chlamydia trachomatis(CT) and Neisseria gonorrhoeae(NG). The most recent evidence on the association between BV and CT/NG infection comes from two secondary analyses of cohort data conducted among women attending STI clinics. Based on these studies, women with BV had a 1.8 and 1.9-fold increased risk for NG and CT infection, respectively. Taken together, BV is likely a risk factor or at least an important contributor to subsequent NG or CT infection in high-risk women. Characterization of Amino Acid Based molecular micelles with molecular modeling

The enantiomers of chiral drugs often have different potencies, toxicities, and biochemical properties. Therefore, the FDA and other worldwide regulatory agencies require manufactures to test and prove the enantiomeric purity of chiral drugs. Amino acid based molecular micelles (AABmm) have been used in chiral CE separations since the 1990’s because of their low environmental impact and because their properties can easily be tuned by changing the amino acids in the chiral surfactant head groups. Using molecular dynamics simulations to investigate the structures and properties of AABmm is part of an ongoing study focusing on investigating and elucidating the factors responsible for chiral recognition with AABmm. The results will be useful for the proper design and selection of more efficient chiral selectors. The micelles investigated contained approximately twenty covalently linked surfactant monomers. Each monomer was in turn composed of an undecyl hydrocarbon chain bound to a dipeptide headgroup containing of all combinations of L-Alanine, L-Valine, and L-Leucine. These materials are of interest because they are effective chiral selectors in capillary electrophoresis separations. molecular dynamics simulation analyses were used to investigate how the sizes and positions of the headgroup amino acid r-groups affected the solvent accessible surface areas of each AABmm chiral center. In addition, headgroup dihedral angle analyses were used to investigate how amino acid r-group size and position affected the overall headgroup conformations. Finally, distance measurements were used to study the structural and conformational flexibilities of each AABmm headgroup. All analyses were performed in the context of a broader study focused on developing structure-based predictive tools to identify the factors responsible for a) self-assembly, b) function, c) higher ordered structure and d) molecular recognition of these amino acid based molecular micelles.
