In situ measurement of ECm rheology and microheterogeneity in embedded and overlaid 3D pancreatic tumor stroma co-cultures via passive particle tracking

Tumor growth is regulated by a diverse set of extraellular infuences,including paracrine crosstalk with stromal partners,and biophysical interactions with surrounding cells and *** elucidating the role of physical force and the mechanical properties of the extracellular matrix(ECm)itself as regulators of tumor growth and invasion have been greatly catalyzed by the use of in ritro three-dimensional(3D)tumor *** systems provide the ability to systematically isolate,manipulate,and evaluate impact of stromal components and extracellular mechanics in a platform that is both conducive to imaging and biologically ***,recognizing that mechanoregulatory crosstalk is bi-directional and fully utilizing these models requires complementary methods for in situ measurements of the local mechanical ***,in 3D tumor/fbroblast co-culture models of pancreatic canocer,a discase characterized by its prominent stromal involvement,we evaluate the use of particle-tracking microrhoology to probe dynamic mechanical *** videos of fuorescently labeled polystyrene micro-spheres embedded in ollagen I ECm,we measure spatiotemporal changes in the Brownian motion of probes to report local ECm shear modulus and *** approach reveals st ffening of collagen in fibroblast co-cultures relative to cultures with cancer cells only,which exhibit degraded ECm with heterogeneous *** further show that these effects are dependent on culture geometry with contrast ing behavior for embedded and overlay *** addition to potential application to screening stroma targeted therapeutics,this work also provides insight into how the compoition and plating geometry impact the mechanical propertios of 3D cell cultures that are increasingly widey used in cancer biology.

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.

Theγ-ray emission from the W51 complex is widely acknowledged to be attributed to the interaction between the cosmic rays(Crs)accelerated by the shock of supernova remnant(SNr)W51C and the dense molecular clouds in the adjacent star-forming region,***,the maximum acceleration capability of W51C for Crs remains *** on observations conducted with the Large High Altitude Air Shower Observatory(LHAASO),we report a significant detection ofγrays emanating from the W51 complex,with energies from 2 to 200 *** LHAASO measurements,for the first time,extend theγ-ray emission from the W51 complex beyond 100 TeV and reveal a significant spectrum bending at tens of *** combining the"π^(0)-decay bump"featured data from Fermi-LAT,the broadbandγ-ray spectrum of the W51 region can be well-characterized by a simple pp-collision *** observed spectral bending feature suggests an exponential cutoff at~400 TeV or a power-law break at~200 TeV in the Cr proton spectrum,most likely providing the first evidence of SNrs serving as Cr accelerators approaching the PeV ***,two young star clusters within W51B could also be theoretically viable to produce the most energeticγrays observed by *** findings strongly support the presence of extreme Cr accelerators within the W51 complex and provide new insights into the origin of Galactic Crs.

On the Sapropelic Coals from the Upper Silesian Coal Basin (Poland)

This paper include results of petrographical, chemical, fluorescent and pyrolytic examinations of sapropelites of coal beds (Namurian A, B, C) of the Upper Silesian Coal Basin (Poland) which have been recently found during mining. The results obtained allowed to prove the genetic-petrographic differencies, to estimate the rank, degree of alteration of the organic substance. results of examination have shown that cannel and humic coals were formed of similar organic substances but in different dispositional conditions, which caused mainly changes in petrographic composition and in smaller extent, in chemical and technological properties. Boghead has different properties caused hy different environmental conditions and dissimilar ogranic matter.

Pulmonary disease in cystic fibrosis (CF) is characterized by a chronic neutrophil-dominated inflammation of lung tissue. Inasmuch as some amino acids (AA) play a pivotal role in various aspects of neutrophil metabolism,the aim of this study was to investigate a possible alteration of neutrophil AA metabolism and to evaluate its relation (if any) with the genotype. We performed plasma and neutrophil AA analysis in 26 CF patients with known genotype,10 patients with non-CF bronchiectasis,and 20 normal subjects. The CF group showed a significant decrease of free intracellular neutrophil glutamine (Gln) content compared with controls and the non-CF bronchiectasis group. In the latter group,levels of neutrophil Gln were significantly lower compared with the controls. Amino acid plasma concentration in non-CF bronchiectasis showed a decrease of Gln and taurine compared with controls. Neutrophil Gln content showed values significantly lower in CF patients with severe mutations (class I,II,and III mutations) compared with mild mutations (class IV and V mutations). results of our study add further evidence for intrinsic neutrophil alterations that could play an important role in the pathogenesis of chronic pulmonary disease in CF patients.

远程缺血预适应（remote ischemic preconditioning,rIPC）对经皮冠状动脉介入治疗（percutaneous coronary intervention,PCI）术后造影剂肾病（contrast-induced nephropathy,CIN）的潜在保护作用尚待确定。该研究为双盲、随机、安慰剂对照的多中心研究,纳入年龄〈85岁,肾清除率为30～60mL/（min·1.73m^2）的有临床适应证的患者,除了直接PCI外均按1∶1给予rIPC或标准治疗。主要终点是CIN的发生率。次要终点是围手术期心肌梗死（peri-procedural myocardial infarction,PmI）的发生率。
