Telomeric DNA breaks in human induced pluripotent stem cells trigger ATr-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-defined in differentiated cells,how stem cells sense and respond to telomere dysfunction,in particular telomeric double-strand breaks(DSBs),is poorly ***,we report the DNA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPScs)in response to telomere-internal *** engineer human iPScs with an inducible TrF1-FokI fusion protein to acutely induce DSBs at *** this model,we demonstrate that TrF1-FokI DSBs activate an ATr-dependent DNA damage response,which leads to p53-independent cell cycle arrest in *** crISPr–cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TrF1-FokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(Hr).In contrast to Hr-based telomere maintenance in mouse embryonic stem cells,where Hr causes ZScAN4-dependent extension of telomeres beyond their initial lengths,Hr-based repair of telomeric breaks is sufficient to maintain iPSc telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TrF1-FokI *** findings suggest a previously unappreciated role for Hr in telomere maintenance in telomerase-positive iPScs and reveal distinct iPSc-specific responses to targeted telomeric DNA damage.

Exploring the effects of coronary artery disease as a preexisting comorbidity on mortality in hospitalized septic patients:a retrospective observation study

Secure and efficient prediction of electric vehicle charging demand usingα^(2)-LSTM and AES-128 cryptography

In recent years,there has been a significant surge in demand for electric vehicles(EVs),necessitating accurate prediction of EV charging *** prediction plays a crucial role in evaluating its impact on the power grid,encompassing power management and peak demand *** this paper,a novel deep neural network based onα^(2)-LSTM is proposed to predict the demand for charging from electric vehicles at a 15-minute time ***,we employ AES-128 for station quantization and secure communication with *** proposed algorithm achieves a 9.2%reduction in both the root Mean Square Error(rMSE)and the mean absolute error compared to LSTM,along with a 13.01%increase in demand *** present a 12-month prediction of EV charging demand at charging stations,accompanied by an effective comparative analysis of Mean Absolute Percentage Error(MAPE)and Mean Percentage Error(MPE)over the last five years using our proposed *** prediction analysis has been conducted using Python programming.

Listening to music manipulates attention to be more externally focused,which has the potential to improve muscular *** study aimed to determine the effect of listening to music on muscle activation during an isometric exercise task,and compare this effect to those of other attentional focus *** healthy subjects(n=35;16 men/19 women)completed an isometric elbow flexion task for 1 min in three randomized and counterbalanced conditions:internal focus(INT),external focus with a simple distraction task(EXT),or listening to music(MUS).Muscle activation of the biceps and triceps brachii and heart rate(Hr)were recorded throughout the exercise *** of perceived exertion(rPE),affective valence,and motivation were measured at the end of each *** was no difference in muscle activation measures among the three *** during MUS was lower than EXT at 15 s([89.4±11.8]beats/min vs.[93.1±12.9]beats/min;p=0.018)and 30 s([90.6±12.4]beats/min vs.[94.2±12.5]beats/min;p=0.026),and lower than INT at 60 s([93.3±13.3]beats/min vs.[96.7±12.0]beats/min;p=0.016).Overall rPE was higher for INT(13.4±2.2)than for MUS([12.6±2.0];p=0.020)and EXT([11.94±2.22];pctive valence was higher for MUS than for INT([2.7±1.4]vs.[2.1±1.5];p=0.011).Manipulating attentional focus did not alter muscle activation for a light-intensity isometric muscular endurance task,though MUS was reported as more positive and requiring less exertion to complete than *** music can therefore be recommended during light-intensity isometric exercise based on the psychological benefits observed.

The accurate identification of dinosaur egg accumulations as nests or clutches is crucial for understanding the reproductive behaviour of these extinct ***,existing methods often rely on the presence of complete eggs and embryo remains,and sedimentological criteria that are only applicable to well-structured *** this study,we introduce an innovative approach to characterize egg accumulations in structureless sediments,where traditional nest structures may not be *** methodology employs a unique combination of sedimentological,taphonomic,geochemical,and geophysical proxies for the study of egg *** applied this approach to the egg accumulation from Paimogo(Jurassic,Portugal),traditionally interpreted as a *** findings reveal that the Paimogo egg assemblage is a secondary deposit,resulting from a flooding event in a fluvial plain that dismantled several allosauroid and crocodylomorph *** eggshell vapor conductance results,coupled with sedimentological evidence,suggest that allosauroid dinosaurs buried their eggs in the dry terrain of overbank areas close to a main channel during the breeding season,likely during the dry season to prevent the embryos from *** research underscores the necessity of multidisciplinary approaches in interpreting egg accumulations and offers a novel methodology for studying these accumulations in structureless *** findings provide new insights into the breeding behaviour and nesting preferences of these extinct organisms,contributing to our understanding of dinosaur ecology.

Holographic dark energy in non-conserved gravity theory

recently, reconsidering the rastall idea T_(μ;νν)^(v)=α_(,μ) through relativistic thermodynamics gives a new form for the scalar field α which led us to construct modern modified theory of gravity debugged ‘non-conserved gravity theory’ Fazlollahi 2023 Euro. Phys. J. c 83 923. This theory unlike other modified theories of gravity cannot directly explain the current acceleration expansion in the absence of the cosmological constant and or existence of other forms of dark energy. Hence, in this study we have reinvestigated holographic dark energy Ρ_(X)~H^(2) in the non-conserved theory of gravity. In this context, the density and pressure of dark energy depend on the non-conserved term and density of the dust matter field. As shown, due to non-conservation effects on large-scale structures, unlike the original holographic model, our model onsets an acceleration epoch for the current Universe satisfies observations. Moreover, the interaction and viscous scenarios are studied for this model.
