To keep the concept of a safe food supply to the consumers, animal feed industries world over are showing an increasing interest in the direct-fed microbials（DFM） for improved animal performance in terms of growth or productivity. This becomes all the more essential in a situation, where a number of the residues of antibiotics and/or other growth stimulants reach in milk and meat with a number of associated potential risks for the consumers. Hence, in the absence of growth stimulants, a positive manipulation of the rumen microbial ecosystem to enhance the feedstuff utilization for improved production efficiency by ruminants has become of much interest to the researchers and entrepreneurs. A few genera of live microbes（i.e., bacteria, fungi and yeasts in different types of formulations from paste to powder） are infrequently used as DFM for the domestic ruminants. These DFM products are live microbial feed supplements containing naturally occurring microbes in the rumen. Among different DFM possibilities, anaerobic rumen fungi（ArF） based additives have been found to improve ruminant productivity consistently during feeding trials. Administration of ArF during the few trials conducted, led to the increased weight gain, milk production, and total tract digestibility of feed components in ruminants. Anaerobic fungi in the rumen display very strong cell-wall degrading cellulolytic and xylanolytic activities through rhizoid development, resulting in the physical disruption of feed structure paving the way for bacterial action. significant improvements in the fiber digestibility were found to coincide with increases in ArF in the rumen indicating their role. Most of the researches based on DFM have indicated a positive response in nutrient digestion and methane reducing potential during in vivo and/or in vitro supplementation of ArF as DFM. The dominant geodynamic processes that underpin the formation and evolution of Earth’s early crust remain enigmatic calling for new information from less studied ancient cratonic ***,we present U-Pb ages and Hf isotopic compositions of detrital zircon grains from^2.9 Ga old quartzites and magmatic zircon from a 3.505 Ga old dacite from the Iron Ore Group of the singhbhum craton,eastern *** detrital zircon grains range in age between 3.95 Ga and 2.91 *** with the recently reported Hadean,Eoarchean xenocrystic(up to 4.24 Ga)and modem detritus zircon grains from the singhbhum craton,our results suggest that the Eoarchean detrital zircons represent crust generated by recycling of Hadean felsic crust formed at^4.3-4.2 Ga and^3.95 *** observe a prominent shift in Hf isotope compositions at^3.6-3.5 Ga towards super-chondritic values,which signify an increased role for depleted mantle and the relevance of plate *** Paleo-,Mesoarchean zircon Hf isotopic record in the craton indicates crust generation involving the role of both depleted and enriched mantle *** infer a short-lived suprasubduction setting around^3.6-3.5 Ga followed by mantle plume activity during the Paleo-,Mesoarchean crust formation in the singhbhum *** singhbhum craton provides an additional repository for Earth’s oldest materials.

Development and Validation of a UPLC Method for the Determination of Docetaxel and Its related substances in Pharmaceutical Dosage Forms, an Antineoplastic Agent

A novel, simple, and sensitive Ultra Performance Liquid Chromatography (UPLC) method was developed and validated for the quantification of process-related impurities and degradants, as well as the assay of Docetaxel. The stability-indicating capability of the method was demonstrated through forced degradation studies and a comprehensive mass balance evaluation. Chromatographic separation was achieved using an ACQUITY UPLC BEH C18 column (100 × 2.1 mm, 1.7 µm), with gradient elution. The mobile phase A comprised a mixture of water, methanol, and acetonitrile (500:300:200, v/v/v), while mobile phase B was acetonitrile and water (800:200, v/v). The flow rate was set at 0.4 mL/min, with detection at 232 nm using a photodiode array detector. The method exhibited excellent performance, with a tailing factor of 1.10 for Docetaxel. The method was rigorously validated for precision, accuracy, linearity, LOD, LOQ, ruggedness, specificity, and robustness. Forced degradation studies confirmed the method’s suitability for stability analysis. stability testing on the drug substance was conducted following ICH guidelines.

A rapid, straightforward, sensitive, efficient, and cost-effective reverse-phase high-performance liquid chromatographic method was employed for the simultaneous determination of sorbitol, sodium Lactate, and Chlorides in a drug solution for infusion. sorbitol, sodium lactate, and Chloride are all officially recognized in the UsP monograph. Assay methods are provided through various techniques, with titrations being ineffective for trace-level quantification. Alternatively, IC, AAs, and ICP-Ms, though highly accurate, are costly and often unavailable to most testing facilities. When considering methods, it’s important to prioritize both quality control requirements and user-friendly techniques. A simple HPLC simultaneous method was developed for the quantification of Chlorides, sorbitol, and sodium Lactate with a shorter run time. The separation utilized a shimpack sCr-102(H) ion exclusion analytical column (7.9 mm × 300 mm, 7 μm), with a flow rate of 0.6 mL per min. The column compartment temperature was maintained at 40°C, and the injection volume was set at 10 μL, with detection at 200 nm. All measurements were conducted in a 0.1% solution of phosphoric acid. The analytical curves demonstrated linearity (r > 0.9999) in the concentration range of 0.79 to 3.8 mg per mL for sodium Lactate (sL), 0.16 to 0.79 mg per mL for sodium Chloride (sC), and 1.5 to 7.2 mg per mL for sorbitol. Validation of the developed method followed the guidelines of the International Conference on Harmonization (ICH Q2B) and UsP. The method exhibited precision, robustness, accuracy, and selectivity. In accelerated stability testing over 6 months, no significant variations were observed in organoleptic analysis and pH. Consequently, the developed method is deemed suitable for routine quality control analyses, enabling the simultaneous determination of sodium Lactate, sodium Chloride, and sorbitol in pharmaceutical formulations and infusions.
