Current status and potential of morphometric sperm analysis

The spermatozoon is the most diverse cell type known and this diversity is considered to reflect differences in sperm function. How the diversity in sperm morphology arose during speciation and what role the different specializations play in sperm function, however, remain incompletely characterized. This work reviews the hypotheses proposed to explain sperm morphological evolution, with a focus on some aspects of sperm morphometric evaluation; the ability of morphometrics to predict sperm cryoresistance and male fertility is also discussed. For this, the evaluation of patterns of change of sperm head morphometry throughout a process, instead of the study of the morphometric characteristics of the sperm head at different stages, allows a better identification of the males with different sperm cryoconservation ability. These new approaches, together with more studies employing a greater number of individuals, are needed to obtain novel results concerning the role of sperm morphometry on sperm function. Future studies should aim at understanding the causes of sperm design diversity and the mechanisms that generate them, giving increased attention to other sperm structures besides the sperm head. The implementation of scientific and technological advances could benefit the simultaneous examination of sperm phenotype and sperm function, demonstrating that sperm morphometry could be a useful tool for sperm assessment.

Collagen secretion screening in Drosophila supports a common secretory machinery and multiple rab requirements

Collagens are large secreted trimeric proteins making up most of the animal extracellular *** of collagen has been a focus of interest for cell biologists in recent years because collagen trimers are too large and rigid to fit into the COPII vesicles mediating transport from the endoplasmic reticulum（Er） to the Golgi. Collagen-specific mechanisms to create enlarged Er-to-Golgi transport carriers have been postulated, including cargo loading by conserved Er exit site（ErES） protein ***, we report an rNAi screening for genes involved in collagen secretion in Drosophila. In this screening, we examined distribution of GFP-tagged Collagen IV in live animals and found 88 gene hits for which the knockdown produced intracellular accumulation of Collagen IV in the fat body, the main source of matrix proteins in the larva. Among these hits, only two affected collagen secretion specifically：PH4 a EFB and Plod, encoding enzymes known to mediate posttranslational modification of collagen in the Er. Every other intracellular accumulation hit affected general secretion, consistent with the notion that secretion of collagen does not use a specific mode of vesicular transport, but the general secretory pathway. Included in our hits are many known players in the eukaryotic secretory machinery, like COPII and COPI components, SNArEs and rab-GTPase regulators. Our further analysis of the involvement of rab-GTPases in secretion shows that rab1, rab2 and rab X3, are all required at ErES, each of them differentially affecting ErES morphology. Abolishing activity of all three by rep knockdown, in contrast,led to uncoupling of ErES and Golgi. We additionally present a characterization of a screening hit we named trabuco（tbc）, encoding an ErES-localized TBC domain-containing rab-GAP. Finally, we discuss the success of our screening in identifying secretory pathway genes in comparison to two previous secretion screenings in Drosophila S2 cells.

Validation of the Lysine requirement as reference Amino Acid for Ideal In-Feed Amino Acid ratios in Modern Fast Growing Meat-Type Chickens

Nitrogen (N) balance studies were conducted with male growing broiler chickens to reevaluate the lysine (Lys) requirement of a modern broiler strain (ross 308), making use of eight diets with graded crude protein (CP) supply (6% - 34% CP as-fed). Wheat, soy protein concentrate, wheat gluten, fishmeal and crystalline amino acids (AAs) were the protein sources in the experimental diets with Lys as limiting AA. Following an adaptation period of five days, two consecutive excreta collection periods (2 × 5 d) were conducted: 10 - 20 d of age (starter period) and 25 - 35 d of age (grower period). Statistical evaluation of N balance data utilized an exponential modelling approach. Based on different dietary Lys efficiency, Lys requirement data were derived by modelling depending on average body weight (BW) during starter and grower period and targeted body protein deposition (PD), respectively. In addition, the influence of graded feed intake was taken into account. For the starter period at 600 g BW and assumed 10 g daily body PD, Lys requirement data between 741 mg and 823 mg per day were observed. The corresponding Lys in-feed concentration was 1.06% and 1.18%, dependent on supposed Lys efficiency at 70 g daily feed intake. For the grower period (average BW 1800 g), 1272 mg to 1473 mg Lys per day was needed to yield 16.5 g daily PD. The corresponding required Lys in-feed concentration was between 0.85% and 0.94% Lys for 150 g daily feed intake.

Anti-hepatitis B virus(HBV)therapy leads to the emer- gence of mutant viral strains during the treatment of chronic hepatitis B with nucleos(t)ides analogues. The existence of HBV variants with primary antiviral resistance may be important for treatment choice. We studied two patients with chronic HBV infection by sequencing the HBV polymerase *** had adefovir-and tenofovir-related mutations in the viral polymerase,although they had never been treated. These mutations were rtV214A/rtN238T in one patient and rtA194T in the ***,mutations in untreated patients deserve cautious *** data indicate that mutations that can theoretically confer adefovir or tenofovir resistance may emerge in treatmentnaive patients.

Assessing Lysine requirement of Growing Chicken by Direct Comparison between Supplementation Technique and “Goettingen Approach”

Validated procedures play an important role to obtain accurate information about individual amino acid requirement data. The aim of the present study was to assess lysine (Lys) requirement of growing chicken both by classical supplementation technique and principles of diet dilution technique as applied with “Goettingen approach”. During the starter period (1 - 21 d), a growth study with male meat type chicken (ross 308) was conducted making use of five graded dietary Lys-levels (3 repetition boxes with 3 birds/box). L-Lys×HCl was gradually added to a diet based on wheat, soybean protein concentrate, wheat gluten and fishmeal to yield 80%, 87.5%, 95%, 102.5% and 110% of the expected requirement level (13 g Lys/kg as fed). Diets were iso-energetic (12.8 MJME/kg) and iso-nitrogenous (21.65% crude protein). Birds were fed on free choice level also to assess the feed intake (FI) effects as important factor on traditional response criteria. Analyzed body composition at start and end of the growth study yielded N deposition (ND) data for further data assessment using exponential approximations depending on dietary Lys content or observed Lys intake. The results indicated significant differences (p response on body weight gain (BWG) and observed dietary protein quality with unexpected consequences for the derived Lys requirement data. According to the independent variable (Lys in % of diet versus daily Lys intake) and aimed level of daily ND, the needed in-feed content of Lys varied between 1.24% and 1.46%. Application of the exponential modelling by “Goettingen approach” overcame these misleading conclusions by modelling the relationship between required Lys intake and observed response data (BWG, ND) taking also into account the expected real feed intake to formulate the needed in-feed concentration.

东南亚地处热带,生物多样性极为丰富,可分为4个热点地区:印度-缅甸区的中南半岛、巽他区(含马来半岛、婆罗洲、苏门答腊岛)、菲律宾区(菲律宾群岛)、华莱士区(苏拉威西岛、爪哇岛、马鲁古群岛、小巽他群岛等)。中南半岛在泥盆纪便已是欧亚大陆的一部分,在印度板块撞击欧亚大陆之后受挤压而出;巽他区来自于冈瓦纳古陆和澳洲古陆;菲律宾群岛部分来自于劳亚古陆的碎片向南漂移,部分来自于太平洋西南岛弧的向北迁移;华莱士区则是劳亚古陆碎片、太平洋西南岛弧以及澳洲古陆北侧碎片的组合。巽他区地处赤道,常年温湿;菲律宾区、华莱士区、中南半岛则都受到不同程度的季风气候决定的干湿季变动。地质历史和季风气候影响程度的不同,奠定了东南亚4个生物多样性热点地区的雏形。华莱士区保存有大量的早期被子植物原始类群如睡莲目(Nymphaeales)和木兰藤目(Austrobaileyales),是现代被子植物起源地和冰期避难所之一。巽他区(婆罗洲)和中南半岛是亚洲热带植物的现代分布中心和"进化前沿",是整个东南亚地区重要的种源;而华莱士区的爪哇岛和小巽他群岛主要是物种迁入和中转的种库。这样的物种形成历史与迁移格局,塑造了东南亚4个生物多样性热点地区物种多样性水平与地理范围的基本格局。巽他区和印度-缅甸区曾在冰期通过陆桥相连,使得东南亚成为周边植物扩散交汇的一个"十字路口"。但是,人们对东南亚生物多样性热点地区的物种长距离扩散规律及植物地理学分区仍存在分歧;东南亚与邻近生物多样性热点地区如新几内亚岛、西高止山脉-斯里兰卡、中国横断山区的历史联系,还尚待深入解析。利用现代分子生物学技术,覆盖整个东南亚地区进行全域取样开展代表性类群的物种迁移与生物地理学研究,有望进一步揭示东南亚生物多样性热点地区的形成过程与演化趋势。

The state-of-the-arts of the study on grid interconnection between Iberian Peninsula and West Maghreb region

With the change of energy structure in Europe and the trend of diversification of energy supply, the interconnection of the power grid in the Mediterranean region has gradually become a hotspot in European energy research. This paper introduces the power supply, load demand, power grid construction and the present situation of multinational interregional interconnection in the Iberian Peninsula of the Tour of Mediterranean in Europe(Portugal and Spain) and the Western Maghreb countries(Morocco and Algeria). And the driving force and influence factors of regional power grid interconnection are also analyzed. According to the MEDGrID project aimed at realizing the Mediterranean power grid interconnection, this paper presents the interconnection schemes of the Iberian Peninsula and the Western Maghreb. Furthermore, the subsequent technical problems need to be solved are also expounded.
