In vivo measurement of NADH fluorescence lifetime in skeletal muscle via -ber-coupled time-correlated single photon counting

Nicotinamide adenine dinucleotide(NADH)is a cofactor that serves to shuttle electrons during metabolic processes such as glycolysis,the tricarboxylic acid cycle,and oxidative phosphorylation(OXPHOS).NADH is autofluorescent,and itsfluorescence lifetime can be used to infer metabolic dynamics in living ***-coupled time-correlated single photon counting(TCSPC)equipped with an implantable needle probe can be used to measure NADH lifetime in vivo,enabling investigation of changing metabolic demand during muscle contraction or tissue *** study illustrates a proof of concept for point-based,minimally-invasive NADHfluorescence lifetime measurement in *** muscle loss(VmL)injuries were created in the left tibialis anterior(TA)muscle of male Sprague Dawley *** lifetime measurements were collected before,during,and after a 30 s tetanic contraction in the injured and uninjured TA muscles,which was subsequently-t to a biexponential decay model to yield a metric of NADH utilization(cytoplasmic vs protein-bound NADH,the A11/A22 ratio).On average,this ratio was higher during and after contraction in uninjured muscle compared to muscle at rest,suggesting higher levels of free NADH in contracting and recovering muscle,indicating increased rates of *** injured muscle,this ratio was higher than uninjured muscle overall but decreased over time,which is consistent with current knowledge of inflammatory response to injury,suggesting tissue regeneration has *** data suggest that-ber-coupled TCSPC has the potential to measure changes in NADH binding in vivo in a minimally invasive manner that requires further investigation.

Telomeric DNA breaks in human induced pluripotent stem cells trigger ATr-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-defined in differentiated cells,how stem cells sense and respond to telomere dysfunction,in particular telomeric double-strand breaks(DSBs),is poorly ***,we report the DNA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPSCs)in response to telomere-internal *** engineer human iPSCs with an inducible TrF1-FokI fusion protein to acutely induce DSBs at *** this model,we demonstrate that TrF1-FokI DSBs activate an ATr-dependent DNA damage response,which leads to p53-independent cell cycle arrest in *** CrISPr–Cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TrF1-FokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(Hr).In contrast to Hr-based telomere maintenance in mouse embryonic stem cells,where Hr causes ZSCAN4-dependent extension of telomeres beyond their initial lengths,Hr-based repair of telomeric breaks is sufficient to maintain iPSC telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TrF1-FokI *** findings suggest a previously unappreciated role for Hr in telomere maintenance in telomerase-positive iPSCs and reveal distinct iPSC-specific responses to targeted telomeric DNA damage.

Uncovering the hierarchical clusters in the heat-affected zone of an electron beam weldedα/βtitanium alloy joint

Using high-resolution transmission Kikuchi diffraction(TKD)and transmission electron microscopy(TEm),we examined the hierarchical clusters that form in situ in the heat-affected zone(HAZ),which are com-monly referred to as“ghost”structures,of bimodal titanium alloy Ti-5Al-2Sn-2Zr-4mo-4Cr(wt%,TC17).The ghost structures are enriched with Al elements but poor in mo and Cr compared to the surroundingβ*** results show that the ghost structure in middle-HAZ mainly consists ofα_(L)laths with a high-angle grain boundary,which exhibits the classic Burgers orientation relationship(BOr)with the host matrix,while it encircles theα_(P)grains in *** the ghost structure is evidenced to form via in-complete martensitic *** results further confirm that the ghost structure is composed ofαL and tinyβ_(L)laths with BOr,with the former being enriched with Al and poor with Cr and mo,while the latter is the ***,twoα_(L)variant clusters with a check-mark morphology are fre-quently observed viewed along[0001]_(αL)//[110]_(βL)directions,which are dominated by the crystallographic and geometrical relationships betweenαandβ*** on the microstructural characterization,it is hypothesized that the ghost structure is transformed from the initialα_(P)phase,due to the coupling ef-fect of high thermal stress(which induces the formation of a large number of dislocations)and element diffusion caused by sudden temperature increase and plunge cooling in the HAZ during the welding pro-cess.

The effect of temperature on the electrical conductivity （σ） and Seebeck coefficient （S） of n-type vapor grown carbon nanofibers （CNFs）and poly（vinylidene fluoride）(PVDF) melt-mixed with 15 wt%of those CNFs is *** 40°C,the CNFs show stable n-type character (S=-4.8μV·K-1) with anσof ca.165 S·m-1,while the PVDF/CNF composite film shows anσof ca.9 S·m-1and near-zero S (S=-0.5μV·K-1).This experimental reduction in S is studied by the density functional tight binding （DFTB） method revealing a contact electron transfer from the CNFs to the PVDF in the ***,in the temperature range from 40°C to 100°C,theσ（T） of the CNFs and PVDF/CNF film,successfully described by the 3D variable range hopping （VrH） model,is explained as consequence of a thermally activated backscattering *** the contrary,the S（T）from 40°C to 100°C of the PVDF/CNF film,which satisfactorily matches the model proposed for some multi-walled carbon nanotube （mWCNT）doped mats;however,it does not follow the increase in S（T） found for *** these findings are presented with the aim of discerning the role of these n-type vapor grown carbon nanofibers on theσand S of their melt-mixed polymer composites.

Organic-inorganic hybrid perovskites have gained great attention as promising photocatalysts for hydrogen ***,due to their poor stability in water,the use of aqueous hydrohalic acid solutions is specifically required for an efficient hydrogen ***,three novel photoactive lead-free hybrid perovskites based on bismuth and triazolium cations(denoted as IEF-15,IEF-16,and IEF-17(IEF stands for ImDEA energy frameworks))were synthesized and fully characterized(structural,compositional,optical,etc.).Further,these solids were proposed as photocatalysts for the challenging gas phase overall water splitting(OWS)***,IEF-16 thin films exhibited a remarkable photocatalytic activity in both H_(2) and O_(2) evolution,as a consequence of its appropriate bandgap and energy-band alignment,achieving hydrogen evolution rates ofμmol·g_(H_(2))^(-)1846 and 360 after 24 h under ultraviolet-visible(UV-vis)irradiation or simulated solar irradiation,*** study additionally highlights the remarkable structural and photochemical stability of IEF-16 under different operational conditions(*** volume,irradiation and temperature),paving the way for green hydrogen production from OWS using perovskite-based photocatalysts.

Assessing the resilient Behavior of recycled mix-granulate with repeated Load CBr Testing

A new characterization technique,a repeated load CBr test,was used to evaluate the resilient properties recycled mix-granulate unbound base material. This is a mixture of crushed concrete and crushed masonry,so recycled demolition waste,this is used on a wide scale in the Netherlands as road base material. The repeated load CBr test,done with standard CBr testing facility,was conducted to estimate the resilient modulus. For comparison and assessment purposes the tests were carried out with the same material conditions (composition,grading,moisture and compaction degree) as have been applied in earlier triaxial testing in the road and railway Engineering Laboratory,Delft University of Technology. The results indicate that the repeated load CBr test yields a good estimate of the stress dependent equivalent modulus for the sample material as bulk,which can be used in mechanistic pavement analysis in the absence of triaxial test results.

methionine(met)is an essential and first limiting amino acid in the poultry diet that plays a significant role in chicken embryonic development and *** present study examined the effect of in ovo injection of DL-met and L-met sources and genotypes on chicken embryonic-intestinal development and *** eggs of the two genotypes,TETrA-SL layer hybrid(TSL)—commercial layer hybrid and Hungarian Partridge colored hen breed(HPC)—a native genotype,were randomly distributed into four treatments for each *** treatment groups include the following:1)control non-injected eggs(NoIn);2)saline-injected(SaIn);3)DL-met injected(DLm);and 4)L-met injected(Lm).The in ovo injection was carried out on 17.5 d of embryonic development;after hatching,eight chicks per group were sacrificed,and the jejunum was extracted for *** results showed that both DLm and Lm groups had enhanced intestinal development as evidenced by increased villus width,villus height,and villus area(Pmpared to the *** DLm group had significantly reduced crypt depth,glutathione(gSH)content,glutathione S-transferase 3 alpha(gST3),occludin(OCLN)gene expression and increased villus height to crypt depth ratio in the TSL genotype than the Lm group(Pgenotype has overexpressed insulin-like growth factor 1(IgF1)gene,tricellulin(mD2),occludin(OCLN),superoxide dismutase 1(SOD1),and gST3 genes than the TSL genotype(Pgs showed that in ovo injection of met enhanced intestinal development,and function,with genotypes responding differently under normal *** also influenced the expression of intestinal antioxidants,tight junction,and growth-related genes.

We report on the structural,magnetic,and magnetocaloric properties of EurhO_(3) *** oxidation states of Eu and rh ions were studied using X-ray photoelectron spectroscopy(XPS).It is found that the Eu ions are mainly in the divalent oxidation state while the rh ions have+4 ***_(3) powders are found to be antiferromagnetic with a second order magnetic transition at Neel temperature(T_(N)=2.9 K).Analysis of the magnetic susceptibility versus temperature data in terms of the Curie=Weiss law:(χ=C/(T-θ_(W))) for T>T_(N),yields θ_(W)=-3.1 K and effective magne tic moment μ_(ff)^(exp),which is close to the theoretical value μ_(eff)^(theo).The magnetic entropy change(-Δ_(Sm)),was determined by em ploying the thermodynam ic maxwell's *** μ_0H=5 T and near T_(N),ΔS_(m)^(max) and relative cooling power(rCP) exhibit large values of 33.7 j/(kg·K) and 238 j/kg,*** large magnitude of-Δ_(Sm) and rCP show that the EurhO_(3) compound could be a potential candidate to be used in cryogenic magnetic refrigeration.

Effect of age on the standardized ileal amino acid digestibility of soybean meal and canola meal in broilers

Standardized ileal digestibility coefficients(SIDC)of nitrogen(N)and amino acids(AA)in two protein sources(soybean meal[SBm]and canola meal[Cm])were investigated at six broiler ages(d 7,14,21,28,35,and 42).Two assay diets were formulated to contain either SBm(413 g/kg)or Cm(553 g/kg)as the sole dietary AA *** dioxide(5 g/kg)was added as an indigestible marker.A total of 696 male broilers at 1 d old were allotted to 12 replicate cages per age *** assay diet was offered to birds for 4 d prior to the ileal digesta collection on d 7(14 birds/cage),14(12 birds/cage),21(10 birds/cage),28(8 birds/cage),35(8 birds/cage)and 42(6 birds/cage),*** apparent digestibility coefficients were standardized using age-specific basal endogenous AA *** the SBm group,though the SIDC of N tended to be influenced(quadratic;P=0.075)by age,no linear or quadratic response of age effect was observed on the average SIDC of indispensable(IAA)and total AA(TAA).An age effect(quadratic;Prved on the average SIDC of dispensable AA(DAA)in SBm with the highest value recorded at d 7,followed by a decrease from d 14 to 28,which increased beyond d *** SIDC of some individual AA(Arg,Thr,Trp,Cys,Pro)were affected(Pr Pratic manner by *** the Cm,the SIDC of N,average SIDC of IAA,DAA and TAA were influenced(quadratic;Pr Prage SIDC of DAA and TAA were higher from d 7 to 14,declined at d 21,and then increased beyond d *** average SIDC of IAA was low between d 7 and 28 and increased *** SIDC of individual AA were affected(linear or quadratic;Pr Prent magnitudes by *** age influence on the SIDC AA was variable,depending on the protein source and *** results demonstrate that age-specific SIDC AA data might need consideration in broiler feed formulations.

Background Pediatric post coronavirus disease 2019(COVID-19)condition(PPCC)is a heterogeneous syndrome,which can significantly affect the daily lives of *** study aimed to identify clinically meaningful phenotypes in children with PPCC,to better characterize and treat this *** Participants were children with physician-diagnosed PPCC,referred to the academic hospital Amsterdam UmC in the Netherlands between November 2021 and march *** factors and information on post-COVID symptoms,comorbidities,and impact on daily life were *** clusters were identified using an unsupervised and unbiased approach for mixed data *** Analysis of 111 patients(aged 3–18 years)revealed three distinct clusters within *** 1(n=62,median age=15 years)predominantly consisted of girls(74.2%).These patients suffered relatively more from exercise intolerance,dyspnea,and smell *** 2(n=33,median age=13 years)contained patients with an even gender distribution(51.5%girls).They suffered from relatively more sleep problems,memory loss,gastrointestinal symptoms,and *** 3(n=16,median age=11 years)had a higher proportion of boys(75.0%),suffered relatively more from fever,had significantly fewer symptoms(median of 5 symptoms compared to 8 and 10 for clusters 1 and 2 respectively),and experienced a lower impact on daily *** This study identified three distinct clinical PPCC phenotypes,with variations in sex,age,symptom patterns,and impact on daily *** findings highlight the need for further research to understand the potentially diverse underlying mechanisms contributing to post-COVID symptoms in children.
