High-density mapping suggests cytoplasmic male sterility with two restorer genes in almond×peach progenies

Peach(Prunus persica)and almond(Prunus dulcis)are two sexually compatible species that produce fertile ***,a highly polymorphic species,is a potential source of new genes for peach that has a strongly eroded gene *** we describe the genetics of a male sterile phenotype that segregated in two almond(‘Texas’)×peach(‘Earlygold’)progenies:an F2(T×E)and a backcross one(T1E)to the‘Earlygold’***-density maps were developed using a 9k peach SNP chip and 135 simple-sequence *** highly syntenic and collinear maps were obtained:one for the F2(T×E)and two for the backcross,T1E(for the hybrid)and E(for‘Earlygold’).A major reduction of recombination was observed in the interspecific maps(T×E and T1E)compared to the intraspecific parent(E).The E map also had extensive monomorphic genomic regions suggesting the presence of large DNA fragments identical by *** data for the male sterility character were consistent with the existence of cytoplasmic male sterility,where individuals having the almond cytoplasm required the almond allele in at least one of two independent restorer genes,rf1 and rf2,to be *** restorer genes were located in a 3.4 Mbp fragment of linkage group 2(rf1)and 1.4 Mbp of linkage group 6(rf2).Both fragments contained several genes coding for pentatricopeptide proteins,demonstrated to be responsible for restoring fertility in other *** implications of these results for using almond as a source of novel variability in peach are discussed.

Influence of La3+ ion in the yttria matrix in improving the microhardness of infrared transparent nano LaxY2-xO3 sintered via hybrid heating

In the present work, hybrid sintering technique which couples the resistive heating and microwave heating is employed to sinter infrared transparent La0.15Y1.85O3 to 99.2% of the theoretical density for the first time to the best of our knowledge. The presence of La3+ in the yttria matrix improves the hardness properties to a greater extent without affecting the transmittance properties, but there is a deterioration in the thermal properties of the sample. So we have limited our studies to La0.15Y1.85O3 which shows better optical, thermal, and hardness properties. The pellets fabricated from the ultra-fine nano powder with average particle size of ~12 nm synthesized by combustion technique and sintered at 1430 °C with an average grain size of 0.22 μm show ~80.1% transmittance in the UV–visible region and 81% in mid infrared region. For a comparative study of the optical, mechanical, and thermal properties, two other variants of sintering strategies namely conventional sintering and microwave sintering are also employed. A comprehensive analysis on the hardness reveals that the hardness of the pellets sintered via hybrid heating is 9.73 GPa and is superior to the pellets sintered using the other two techniques. The thermal conductivity of the sample is also analyzed in detail. The results clearly indicate that the La0.15Y1.85O3 ultra-fine nano powder synthesised by the single-step combustion method and sintered via microwave hybrid heating shows better transmittance properties without compromising the mechanical properties, and can be used very effectively for the fabrication of infrared transparent windows and domes.

Study of Trace Ions in Wet Deposition of an Industrial Site in Monterrey’s Metropolitan Area, Mexico

The chemistry of rainwater has been subject to numerous investigations during the last two decades due to the increase of environmental problems caused by the acid deposition. The present study focuses on one aspect of air pollution, the chemical composition of wet deposition of an industrial zone. The sampling period was from march to December 2009. The station was located on the roof of the Chemistry School at the University of Nuevo León, an area in the north of Monterrey where the majority of pollution is generated. Twenty-five wet precipitation samples were collected with an automatic sampler and analyzed for pH, ions (, , Cl-, Ca2+, Mg2+, Na+, k+) and conductivity. The results show that the average pH is higher than 5.6;the characteristics of the rainwater studied are of alkaline nature due to the values of pH found;there is an evident anthropogenic source contributing to the alkaline of rain water. In order to find possible association between ions and consequently the possible sources of pollutants correlation study was applied using the program SPSS v.12. Good correlations were found between ions and along with Ca2+ and Mg2+. The local extraction industry and surrounding aerosols might be causing of alkaline rain which may be due to the neutralization effect of particulate matter. This study represents a continuation of the studies of rainwater chemistry in the Northeast of Mexico.

Screening of Protease Inhibitory Activity in Aqueous Extracts of marine Invertebrates from Cuban Coast

Protease inhibitors have been isolated from many variable sources;however, the need to identify and characterize new molecules has increased with the discovery of new therapeutic targets and the lack of specificity of already identified compounds with inhibitory activity. The goal of this work was to search for inhibitory activity against four proteolytic enzymes already recognized as therapeutic targets: human neutrophil elastase, dipeptidyl peptidase IV, subtilisin from Bacillus licheniformis and cathepsin k in selected marine invertebrates from the Caribbean Sea. A systematic screening was carried out with selected aqueous extracts belonging to 20 species from seven different phyla: Annelida, Bryozoa, Chordata, Cnidaria, Equinodermata, Mollusca and Porifera, all collected at the coast of Havana (Cuba). All extracts showing initial inhibitory activity were characterized in terms of IC50 values and specific inhibitory activity (SIA). Model enzymes were used in the case of human neutrophil elastase (porcine pancreatic elastase) and cathepsin k (papain) for the screening and all positive results were confirmed by testing toward the therapeutic targets. Ten extracts were identified showing inhibitory activity against human neutrophil elastase, for which the most promising values were obtained for Nerita peloronta. Only one extract, Bunodosoma granulifera, showed inhibitory activity against dipeptidyl peptidase IV with rather poor values of IC50 and SIA. Seven extracts showed inhibitory activity against B. licheniformis subtilisin with very good IC50 and SIA values for Lissodendorix isodyctialis, Cenchritis muricatus, and N. peloronta. Finally, eight extracts were positive for cathepsin k with almost similar parameters values among them. We evaluated fruit availability, phytochemical characteristics and mineral content of r. rubiginosa L. rose hips, at two experimental sites in San Luis province: Potrero de los Funes-El Volcán (PF-V) and Estancia Grande (EG). Plant density was 70 plants/ha (1 ha = 10,000 m2) in PF-V and 50 plants/ha in EG. Fruit availability (DF) in fresh fruits (FF) was 4.39 kg (PF-V) and 3.10 kg (EG) during march, and decreased during June. Ascorbic acid in FF increased during ripening period, peaking on late April, early May, and diminished 75% - 85% in dried fruits. Total soluble solids in FF ranged from 32.55 to 55.20 °Brix. Elements Ba, Ca, Fe, k, Zn and Cu, determined by inductively coupled plasma optical emission spectrometry (ICP-OES), were significant for human health. Dried fruits maintain a high level of ascorbic acid, a characteristic added to content of mineral elements, particularly calcium content, providing an important nutritional value to r. rubiginosa rose hips.

Optical parameters induced by phase transformation in rF magnetron sputtered TiO2 nanostructured thin films

Pure TiO2 thin films were deposited onto quartz substrates using a ceramic TiO2 target at an elevated substrate temperature of 573 k by rF magnetron sputtering, and an analysis of structural, optical and photoluminescence characteristics of the films upon phase transformation is reported in this paper. Structural investigations using X-ray diffraction revealed that the as-deposited film was amorphous in nature. Thermal annealing for 2 h at 873 k in air resulted in the formation of anatase phase, and a phase transformation to rutile was observed at 1073 *** increase in grain size and an improvement in crystallinity were also observed on annealing. rod- like rutile crystallites were observed in the SEM images of the film annealed at 1273 k. As-deposited films and films annealed up to 1073 k were highly transparent in the visible region with a transparency 480%. Optical band gap of the films decreased upon thermal annealing which is attributed to phase transformation from amorphous to anatase and then to rutile. Optical parameters such as refractive index, optical conductivity and optical dielectric constant increased with increase in annealing temperature. Since rutile is the optically active phase, the superior refractive index of the film annealed at 1073 k along with its high transparency in visible region suggests the application of this film in antireflective coatings. Photoluminescence emission of maximum intensity was observed for the film annealed at 873 k, which exhibits anatase phase. Intense blue emission observed in this film makes it suitable for use in optoelectronic display devices.

Morphometric, Physicochemical, Thermal, and rheological Properties of rice (Oryza sativaL.) Cultivars Indica ×Japonica

The anther culture technical was applied to produce haploid lines of rice (Oryza sativa L.). The hybrids (k/ A92VM061, k/A92VM067, k/A92VM0611, k/A92VM719, k/A92VM720 and k/A92VM721) were obtained in order to generate new varieties from Indica and Japonica cultivars. Morphometric parameters of the grains were evaluated by image analysis. Flours were prepared from the whole rice grains and physicochemical, thermal and rheological properties, X-ray diffraction pattern and evaluation of color using the CIELAB system were assessed. The hybrids lines showed long (061, 611, 721), medium (719 and 720) and short (067) grains. The rice samples presented lipids (2.6% - 3.2%), protein (11% - 15%), total dietary fiber (8.4% - 10.2%), total starch (65% - 74%) and apparent amylose (5% - 32%) contents. Gelatinization temperature (Tp) was found in the range of 66.1℃ - 79.4℃ with enthalpy (ΔH) value between 3.4 - 8.1 J/g. The retrogradation parameters (temperature and ΔH) were lower than those for gelatinization in all samples. The rice samples presented A-type X-ray diffraction pattern. rice pastes showed a non-Newtonian behavior and the brightness (L*) characterize the color of the samples. Hybrid rice grains presented morphometric properties more similar to Japonica than Indica variety. rice hybrid had higher protein content than Indica variety. Apparent amylose, viscosity and gelatinization temperature varied significantly among hybrids and varieties.

Antibacterial Activity of Bougainvillea Glabra, Eucalyptus Globulus, Gnaphalium Attenuatum, and Propolis Collected in Mexico

Bougainvillea glabra C., Eucalyptus globules Labill., and Gnaphalium attenuatum DC., as well as propolis, have been used in Mexican traditional medicine as a remedy to treat respiratory illnesses. There are few biological reports of such material collected in Mexico, despite the high demand for them as raw material for popular and industrial uses. The antibacterial activity of the plants and propolis studied here were evaluated against a panel of bacteria using three different methodologies: agar disc diffusion, macro and micro dilution methods. E. globules and G. attenuatum extracts showed the strongest active values (P > 0.05) in the agar disc diffusion method with a range of 8 - 22 mm inhibition zone, MIC values ranging from 25 - 250g/mL, and MBC values of 25 - 500g/mL obtained by macro and micro dilution methods. B. glabra extracts were active against E. coli, S. typhi, k. pneumoniae, S. aureus and S. agalactiae, with 8 - 13 mm inhibition zone, MICs ranging from 500 - 3000g/mL, and MBCs of 1000 - 3000g/mL. The propolis extract turned out to be active against E. coli, k. pneumoniae, S. aureus and S. agalactiae, with values of 7 - 12 mm, MICs of 1000 - 2000g/mL, and MBCs of 2000 - 2500g/mL. E. globules and G. attenuatum extracts were the most active in the three methodologies assayed. This is also the first time that the antibacterial activity of G. attenuatum has been experimentally demonstrated. The microdilution method showed to be more sensitive, less expensive and minor time-consuming technique compared with the other two.
