Effects of 2,3,7,8-TcDD and benzo[a]pyrene on modulating vitellogenin expression in primary culture of crucian carp (carassius auratus) hepatocytes

Vitellogenin (Vtg) is the precursor of yolk protein. Its expression and secretion are estrogen-regulated and are crucial for oocyte maturation. An in vitro xenoestro- gen screening model was established by measuring Vtg in- duction in cultured primary hepatocytes from crucian carp. Vtg production was detected by biotin-avidin sandwich ELISA method while Vtg and cytochrome P4501A1 (cYP1A1) mrNA induction were measured by semi- quan- titative Pcr-primer dropping technique. Vtg and Vtg mrNA were dose-dependently induced by diethylstilbestrol (DES, 0.2—200 ng/mL) in hepatocytes of crucian carp. co-treatment of the DES-induced hepatocytes with either 2,3,7,8-TcDD (TcDD, 0.1—4 pg/mL) or benzo[a]pyrene (B[a]P, 5—1000 ng/mL) resulted in a reduction of Vtg pro- duction and an increment of cYP1A1 mrNA expression both in a dose dependent manner, indicating the anti-estro- genic effects of the compounds. However, at lower tested concentrations, TcDD (0.1, 0.2 pg/mL), B[a]P (5 ng/mL) seemed to have a potentiating effect on Vtg expression and secretion, although by their own these compounds had no observable estrogenic effect on Vtg induction. Tamoxifen (a selective estrogen receptor modulators, 1 nmol/L—1 μmol/L), and β-naphtho-flavone (β-NF, an aryl hydrocarbon receptor inducing compounds, 2.5—1000 ng/mL) also were employed to study the possible interactions in DES-induced Vtg ex- pression. In co-treatment of the DES-induced hepatocytes with β-NF or tamoxifen, the decrease in Vtg production did parallel induction of cYP1A1 for β-NF, but tamoxifen inhib- ited Vtg induction did not parallel induced cYP1A1 expres- sion in all test concentrations. On the contrary, it was found that in co-treatment of the TcDD-induced hepatocytes with DES, TcDD induced cYP1A1 mrNA production was inhib- ited by DES also. These results implicated a possible cross talk between estrogen receptor- and aryl hydrocarbon re- ceptor-mediated pathways in the hepatocytes.

In real world situations, most scheduling problems occur neither as complete off-line nor as complete on-line models. Most likely, a problem arises as an on-line model with some partial information. In this article, we consider such a model. We study the scheduling problem P(n1,n2), where two groups of jobs are to be scheduled. The first job group is available beforehand. As soon as all jobs in the first group are assigned, the second job group appears. The objective is to minimize the longest job completion time (makespan). We show a lower bound of 3/2 even for very special cases. Best possible algorithms are presented for a number of cases. Furthermore, a heuristic is proposed for the general case. The main contribution of this paper is to discuss the impact of the quantity of available information in designing an on-line algorithm. It is interesting to note that the absence of even a little bit information may significantly affect the performance of an algorithm.

Stutzerimonas have been extensively studied due to their remarkable metabolic and physiological ***,research on its phages is currently *** this study,we isolated a novel double-stranded DNA(dsDNA)phage,vB_SstM-PG1,from the marine environment that infects Stutzerimonas stutzeri *** dsDNA genome is 37204 bp long with a G/c content of 64.14%and encodes 54 open reading *** phage possesses a tail packaging structure that is different from known Stutzerimonas stutzeri phages and exhibits structural protein characteristics similar to those of temperate *** addition,two genes of toxin-antitoxin system,including YdaS_antitoxin and HEPN_SAV_6107,were found in the vB_SstM-PG1 genome and play important roles in regulating host growth and *** phylogenetic tree and comparative genomic analysis,it has been determined that vB_SstM-PG1 is not closely related to any phages previously identified in the GenBank ***,it has a connection with enigmatic,uncultured ***,the vB_SstM-PG1 virus exhibits an average nucleotide identity of over 70%with six uncultivated viruses identified in the IMG/Vr v4 *** significant finding has resulted in the identification of a novel viral genus known as Metabovirus.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATcc *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

Tribological evaluation of thermoplastic polyurethane-based bearing materials under water lubrication:Effect of load,sliding speed,and temperature

Polymers are widely used in bearing *** the case of water-lubricated stern tube bearings,thermoplastic polyurethane(TPU)-based composites are used due to their excellent wear resistance,corrosion resistance,and tunable mechanical *** tribological performance,however,depends on operating *** this work,TPU was blended with carbon fiber,graphene platelet,and ultra-high molecular weight polyethylene(UHMWPE).Friction tests of TPU based-composites against copper countersurface were carried out in water to mimic the actual operating conditions of the *** of the resulting contacts were in the boundary lubrication regime,in which friction was attributed to both contact mechanics of asperities as well as water *** results show that the viscoelasticity of TPU has a considerable impact on its tribological *** lubrication at 50°c promotes the softening of polymer surface material during sliding,resulting in higher fluctuation in the coefficient of friction and wear *** is attributed to the reduced thermomechanical *** addition,Schallamach waviness is observed on worn *** tribological properties of TPU are significantly improved by the inclusion of carbon fiber,graphene platelet,and *** formation of graphene transfer-layers and UHMWPE transfer film reduces friction and wear loss,while the inclusion of carbon fiber enhances wear resistance due to improved mechanical properties and load bearing capacity.
