Dr.ught Str.ss and Pr.har.est Aflatoxin Contamination in Agr.cultur.l Commodity: Genetics, Genomics and Pr.teomics

Thr.ughout the wor.d, aflatoxin contamination is consider.d one of the most ser.ous food safety issues concer.ing health. Chr.nic pr.blems with pr.har.est afiatoxin contamination occur.in the souther. US, and ar. par.icular.y tr.ublesome in cor., peanut, cottonseed, and tr.e nuts. Dr.ught str.ss is a major.factor.to contr.bute to pr.har.est afiatoxin contamination. r.cent studies have demonstr.ted higher.concentr.tion of defense or.str.ss-r.lated pr.teins in cor. ker.er. of r.sistant genotypes compar.d with susceptible genotypes, suggesting that pr.har.est field condition （dr.ught or.not dr.ught） influences gene expr.ssion differ.ntly in differ.nt genotypes r.sulting in differ.nt levels of ＂end pr.ducts＂： Pr.pathogenesis-r.lated） pr.teins in the matur. ker.els. Because of the complexity of Asper.illus-plant inter.ctions, better.under.tanding of the mechanisms of genetic r.sistance will be needed using genomics and pr.teomics for.cr.p impr.vement. Genetic impr.vement of cr.p r.sistance to dr.ught str.ss is one component and will pr.vide a good per.pective on the efficacy of contr.l str.tegy. Pr.teomic compar.sons of cor. ker.el pr.teins between r.sistant or.susceptible genotypes to Asper.illus flavus infection have identified str.ss-r.lated pr.teins along with antifungal pr.teins as associated with ker.el r.sistance. Gene expr.ssion studies in developing cor. ker.els ar. in agr.ement with the pr.teomic studies that defense-r.lated genes could be upr.gulated or.downr.gulated by abiotic str.sses.

Monitor.ng the Expr.ssion of Maize Genes in Developing Ker.els under.Dr.ug ht Str.ss usingOligo-micr.ar.ay

Pr.har.est aflatoxin contamination of gr.in gr.wn on the US southeaster. Coast Plain is pr.voked and aggr.vated by abiotic str.ss. The pr.mar. abiotic str.ss is dr.ught along with high temper.tur.s. The objectives of the pr.sent study wer. to monitor.gene expr.ssion in developing ker.els in r.sponse to dr.ught str.ss and to identify dr.ught-r.sponsive genes for.possible use in ger.plasm assessment. The maize br.eding line Tex6 was used, and gene expr.ssion pr.files wer. analyzed in developing ker.els under.dr.ught str.ss ver.es well-water.d conditions at the stages of 25, 30, 35, 40, 45 d after.pollination （DAP） using the 70 mer.maize oligo-ar.ays. A total of 9 573 positive ar.ay spots wer. detected with unique gene IDs, and 7 988 wer. common in both str.ssed and well-water.d samples. Expr.ssion patter.s of some genes in sever.l str.ss r.sponse-associated pathways, including abscisic acid, jasmonic acid and phenylalanine ammonia-lyase, wer. examined, and these specific genes wer. r.sponsive to dr.ught str.ss positively. r.al-time quantitative polymer.se chain r.action validated micr.ar.ay expr.ssion data. The compar.son between Tex6 and B73 r.vealed that ther. wer. significant differ.nces in specific gene expr.ssion, patter.s and levels. Sever.l defense-r.lated genes had been downr.gulated, even though some defense-r.lated or.dr.ught r.sponsive genes wer. upr.gulated at the later.stages.

Spatial and tempor.l patter.s of insect damage in r.lation to aflatoxin contamination in a cor. field with plants of unifor. genetic backgr.und ar. not well under.tood. After.pr.vious examination of spatial patter.s of insect damage and aflatoxin in pr.-har.est cor. fields, we fur.her.examined both spatial and tempor.l patter.s of cob- and ker.el- feeding insect damage, and aflatoxin level with two samplings at pr.-har.est in 2008 and 2009. The feeding damage by each of the ear.ker.el-feeding insects （i.e., cor. ear.or./fall ar.ywor. damage on the silk/cob, and discolor.tion of cor. ker.els by stink bugs） and maize weevil population wer. assessed at each gr.d point with five ear.. Sampling data showed a field edge effect in both insect damage and aflatoxin contamination in both year.. Maize weevils tended towar. an aggr.gated distr.bution mor. fr.quently than either.cor. ear.or. or.stink bug damage in both year.. The fr.quency of detecting aggr.gated distr.bution for.aflatoxin level was less than any of the insect damage assessments. Stink bug damage and maize weevil number.wer. mor. closely associated with aflatoxin level than was cor. ear.or. damage. In addition, the indices of spatial-tempor.l association （χ） demonstr.ted that the number.of maize weevils was associated between the fir.t （4 weeks pr.-har.est） and second （1 week pr.-har.est） samplings in both year. on all fields. In contr.st, cor. ear.or. damage between the fir.t and second samplings fr.m the field on the Belflower.Far., and aflatoxin level and cor. ear.or. damage fr.m the field on the Lang Far. wer. dissociated in 2009.
