Lentil(Lens culinaris Medik) is one of the most important food legumes in the world. The use in lentil of molecular marker-assisted breeding is limited, owing to the low availability of polymorphic markers. In the present study, we developed a set of polymorphic intron-spanning markers(ISMs) using a cross-species mapping approach. In this approach, putative unique transcripts(PUTs) of L. culinaris were mapped onto the Medicago truncatula genome, exploiting its closeness with the lentil genome. Spliced alignment of the PUTs resulted in a total of 25,717 alignments, allowing the development of 1703 ISMs. From these, a subset of 105 ISMs were synthesized and validated with a 51% amplification success rate in 32 lentil genotypes. Of these ISMs, 40(74%) were polymorphic and generated 2–11 alleles per locus in a genetically diverse panel of 32 lentil genotypes including wild *** set of polymorphic ISMs along with their functional annotation data will be useful in lentil breeding.

Antineoplastons are peptide and amino acid derivatives that occur naturally in the human body. They inhibit the growth of neoplastic cells without growth inhibition of normal cells. Phenylacetylglutaminate (Pg) is an active ingredient of antineoplastons A10 and AS2-1 (ANP) and is also a metabolic by-product of phenylbutyrate (PB). The formulation of antineoplaston AS2-1 is a 4:1 mixture of phenylacetate (PN) and Pg. Antineoplaston A10 is a 4:1 mixture of Pg and isoPg. This study investigates the molecular mechanism of action of Pg and PN. The Human U87 glioblastoma (gBM) cell line was used as the model system in this study. A total human gene array screen using the Affymetrix Human genome plus 2.0 oligonucleotide arrays was performed using mrNA derived from U87 cells exposed to Pg and PN. Pathway analysis was performed to allow the visualization of effect on metabolic pathways and gene interaction networks. Our preliminary results indicate that Pg and PN interrupt signal transduction in rAS/MAPK/ErK and PI3K/AKT/PTEN pathways, interfere with cell cycle, decrease metabolism and promote apoptosis in human U87 gBM cells. The effect on multiple cellular pathways and targets, suggests that ANP and PB are promising candidates for clinical studies in gBM.

MicrorNAs (mirNAs) are endogenous small rNA regulatory molecules of approximate 20-24 nucleotides that are involved in regulating the intrinsic growth and development of organs in plants and animals as well as in maintaining the integrity of genomes. Past few years have witnessed an increase in research reports on the crucial role of mirNAs in plant stress response. Plant mirNAs regulate gene expression at the post-transcriptional level not only by suppression of mrNA translation but also by direct cleavage of the target mrNAs. This review starts with a brief overview on small rNAs including mirNAs, biogenesis of mirNA and focuses mainly on the various up and down-regulated plant mirNAs under different biotic and abiotic stresses showing advancement of studies about mirNA and their stress regulation pathway. This review explores the emerging role of mirNAs as potential biomarkers in plant stress responses.

Incidence and risk factors for vitreous loss in residents performing manual small-incision cataract surgery

AIM: To assess the incidence of vitreous loss and associated risk factors in residents performing manual small-incision cataract surgery(MSICS). METHODS: The present retrospective record review study was performed on 490 patients who underwent MSICS performed between November 2018 and December 2019 by 7 third-year postgraduate residents. The study group comprised of patients having intraoperative vitreous prolapse. All the surgeries were performed under supervision of a trained assistant. rESULTS: The mean age of the participants at the time of surgery was 68.42±2.05 y. Of the 490 patients, 250 patients were male, and 240 patients were female(P=0.23). A total of 215(43.9%) eyes had mature white cataract, 185(37.8%) eyes had brown cataract, and 90(18.3%) eyes had immature senile cataract. The incidence of intraoperative vitreous loss among residents was 2%(10/490). Vitreous loss occurred during hydrodissection [1/10(10%)], nucleus delivery [3/10(30%)], irrigation and aspiration [5/10(50%)], and intraocular lens insertion [1/10(10%)]. Multivariate stepwise Logistic regression analysis confirmed immature senile cataract [odds ratio(Or)=3.99;P=0.02], irrigation and aspiration of cortical material(Or=3.07;P=0.03), and anterior capsular extension(Or=3.22, P=0.03) as independent risk factors for vitreous loss. CONCLUSION: Immature senile cataract, irrigation and aspiration of cortical material, and anterior capsular extension are independent risk factors for vitreous loss. Our findings may serve as a guide for future trainers or residents learning MSICS.

Floods are one of nature's most destructive disasters because of the immense damage to land,buildings,and human *** is difficult to forecast the areas that are vulnerable to flash flooding due to the dynamic and complex nature of the flash ***,earlier identification of flash flood susceptible sites can be performed using advanced machine learning models for managing flood *** this study,we applied and assessed two new hybrid ensemble models,namely Dagging and random Subspace(rS)coupled with Artificial Neural Network(ANN),random Forest(rF),and Support Vector Machine(SVM)which are the other three state-of-the-art machine learning models for modelling flood susceptibility maps at the Teesta river basin,the northern region of *** application of these models includes twelve flood influencing factors with 413 current and former flooding points,which were transferred in a gIS *** information gain ratio,the multicollinearity diagnostics tests were employed to determine the association between the occurrences and flood influential *** the validation and the comparison of these models,for the ability to predict the statistical appraisal measures such as Freidman,Wilcoxon signed-rank,and t-paired tests and receiver Operating Characteristic Curve(rOC)were *** value of the Area Under the Curve(AUC)of rOC was above 0.80 for all *** flood susceptibility modelling,the Dagging model performs superior,followed by rF,the ANN,the SVM,and the rS,then the several benchmark *** approach and solution-oriented outcomes outlined in this paper will assist state and local authorities as well as policy makers in reducing flood-related threats and will also assist in the implementation of effective mitigation strategies to mitigate future damage.

AIM To assess overall confidence level of trainees in assessing and treating shock, we sought to improve awareness of recurrent biases in clinical decision-making to help address appropriate educational *** Pediatric trainees on a national listserv were offered the opportunity to complete an electronic survey anonymously. Four commonly occurring clinical scenarios were presented, and respondents were asked to choose whether or not they would give fluid, rank factors utilized in decision-making, and comment on confidence level in their decision. rESULTS Pediatric trainees have a very low confidence level for assessment and treatment of shock. Highest confidence level is for initial assessment and treatment of shock involving American College of Critical Care Medicine/Pediatric Advanced Life Support recommendations. Children with preexisting cardiac comorbidities are at high risk ofunder-resuscitation. CONCLUSION Pediatric trainees nationwide have low confidence in managing various shock states, and would benefit from guidance and teaching around certain common clinical situations.
