Chip-based flow-through PCr implements the PCr as a continuous process for nucleic acid analytics. The sample is transported in a winding channel through temperature zones required for denaturation, annealing and extension. Main fields of application are the monitoring of continuous processes for rapid identification of contaminants and quality control as well as high throughput screening of cells or microorganisms. A modular arrangement with five heating zones for flow-through PCr is discussed and evaluated. The special heater arrangement allows the implementation of up to 40 cycles on the footprint of a microscope slide, which is placed on top ofa 5 zones heating plate. Liquid/liquid two phase flow of PCr reaction mixture and mineral oil have been applied to create a segmented flow process scheme. In that way, the developed system may provide flow-through PCr as a unit operation for the droplet based microfluidics platform. The single use of disposable devices is commonly preferred due to the sensitivity of the PCr process to contaminations. All-glass microfluidic chips and disposable chip devices, made from polycarbonate as a replication with identically geometry, have been fabricated and tested. For the first time, microchannel geometries with nearly circular profile developed by all-glass technology have been transferred to mass fabrication by injection compression molding. Both devices have been successfully applied for the detection of the tumor suppressor gene p53. Although product yield and selectivity of the amplification process do not depend on the chip material, a well defined, reliable segmented flow regime could only be realized in the all-glass chip.

Accurate prediction of the solids friction factor through horizontal straight pipes is important for the reliable design of a pneumatic conveying system, but it is a challenging assignment to date because of the highly concentrated, turbulent, and complex nature of the gas-solids mixture. Power-station fly ash was transported through different pipeline configurations. Numerical simulation of the dense-phase pneumatic conveying systems for three different solids and two different air flow rates have shown that particle and actual gas velocities and the ratio of the two velocities increases in the flow direction, whereas the reverse trend was found to occur for the solids volumetric concentration. To develop a solids friction-factor model suitable for dense-phase flow, we modified an existing pure dilute-phase model by incorporating sub-models for particle and actual gas velocities and impact and solids friction factor. The solids friction-factor model was validated by using it for scale-up predictions for total pipeline pressure drops in longer and larger pipes and by comparing experimental and predicted pneumatic conveying characteristics for different solids flow rates. The accuracy of the prediction was compared with a recently developed two-layer-based model. We discussed the effect of incorporating the particle and actual gas velocity terms in the solids friction-factor model instead of superficial air velocity.

High-throughput deep-sequencing technology and bioinformatics analysis of the small rNA(srNA)population isolated from plants allows universal virus detection and complete virome reconstruction for a given *** the present srNA deep-sequencing analysis of virus-infected wheat samples in the Czech republic,samples were firstly tested for barley yellow dwarf viruses(BYDVs),wheat streak mosaic virus(WSMV)and wheat dwarf virus(WDV)using ELISA,rT-PCr and *** srNA sequencing of these samples yielded more than^60 million single-end 50-bp reads with high confidence for nine field samples of ***,16.5%of reads were virus-specific and 83.5%were mapped to the *** 21-nt reads(-7.7E+06 reads)were found than 24-nt(~6.20E+06 reads)or 22-nt(-4.30E+06 reads)*** novo assembly of the high-quality contigs revealed the presence of three earlier repoted viruses in the Czech republic:BYDVs(31.48%),WSMV(24.23%)and WDV(2666%).We also showed the presence of cereal yellow dwarf virus(14.33%;two species CYDV-rPS and CYDV-rPV family Luteoviridae/Polerovirus)and wheat yellow dwarf virus(WYDV,3.30%;Luteoviridae).Phylogenetic analysis showed CYDV and WYDV grouped separately from ***,several recombination breakpoints were found among the groups of yellow dwarf viruses(BYDVs,CYDV,and WYDV).Using rNA deep sequencing,we confirmed the presence of the three known viruses(BYDVs,WSMV,and WDV)and the first record of two species of CYDV and WYDV in wheat in the Czech republic.

Wheat streak mosaic virus（WSMV）has become a re-emerging pathogen in cereal crops in the Czech *** was first reported in the former Czechoslovakia in the early 1980s,and then no record of the virus was documented until *** incidence of the virus was recorded in recent years in several winter wheat fields and many grass ***,we surveyed the incidence of WSMV in cereal *** results demonstrated the existence of the virus in winter wheat and volunteer wheat during each year of the monitoring period,which spanned from 2013–*** the range of infected samples was low（6.4%of the total tested samples）,a high incidence of well-distributed virus was *** at least six fields,the virus reached severe and potentially epidemic *** accordance with our previous report detailing WSMV infection of native grasses,we tested several grass species commonly grown in the Czech *** found that some grass species acted as experimental hosts and possible reservoirs of the virus;these included Anthoxanthum odoratum（sweet vernal grass）,Arrhenatherum elatius（false oat-grass）,Lolium multiflorum（Italian rye-grass）,Bromus japonicus（Japanese chess）,Echinochloa crus-galli（barnyard grass）,Holcus lanatus（meadow soft grass）and Holcus mollis（creeping soft grass）.Some of these grass species are also important weeds of cereals,which may be the potential source of WSMV infection in cereal *** widely used winter wheat cultivars were tested in the field after artificial inoculation with WSMV to evaluate virus titre by ***,the tested cultivars had a low virus titre,which is associated with mild disease symptoms and may provide a good level of crop resistance to WSMV.
