Extracellular vesicles(EVs)have recently emerged as a promising delivery platform for CRISPR/Cas9 ribonucleoproteins(RNPs),owing to their ability to minimize off-target effects and immune ***,enhancements are required to boost the efficiency and safety of Cas9 RNP enrichment within *** response,we employed the Fc/Spa interaction system,in which the human Fc domain was fused to the intracellular domain of PTGFRN-Δ687 and anchored to the EV ***,the B domain of the Spa protein was fused to the C domain of cargos such as Cre or *** to the robust interaction between Fc and Spa,this method enriched nearly twice the amount of cargo within the *** loaded with spCas9 RNP targeting the HSV1 genome exhibited significant inhibition of viral replication in vitro and in ***,following neuron-targeting peptide RVG modification,the in vivo dosage in neural tissues substantially increased,contributing to the clearance of the HSV1 virus in neural tissues and exhibiting a lower off-target *** findings establish a robust platform for efficient EV-based SpCas9 delivery,offering potential therapeutic advantages for HSV1 infections and other neurological disorders.

鄂尔多斯活动地块边界带第四纪晚期构造活动特征及强震孕育机制

鄂尔多斯地块是位于中国内地中心位置的典型活动地块,由于受西南部青藏地块和东部太平洋板块远程作用的影响,地块各边界带构造活动特征和变形具有明显的特殊性和差异性。文章总结了多年来围绕鄂尔多斯活动地块周缘边界带开展的活动断裂定量、地震孕育机制及活动特征等的已有研究结果,对地块周缘断裂活动特征及强震孕育机制进行系统总结。鄂尔多斯活动地块不同边界带断裂第四纪晚期活动特征的不同是强震孕育环境差异的最直接的证据。鄂尔多斯活动地块西边界构造变形样式复杂,受青藏高原向北东挤压扩展的影响,西边界南段断裂以走滑、逆走滑和逆冲为主要特征。而向北到西边界北段,以右旋走滑为主要特征;以青藏高原最新扩展前缘的三关口-牛首山断裂为界,北部的银川盆地表现为典型的断陷盆地,边界断裂有右旋走滑特征,地震活动多以正走滑型为主。北边界的河套盆地以北侧的正断层为其控盆构造,历史和古地震多集中在北侧边界断裂上。南边界的渭河盆地构造特征相对较为复杂,由两组正断层组成,历史大地震多发生在盆地南缘,盆地中北部有中强地震发生。东边界的山西地堑系由多个裂谷型盆地斜列组成,历史大地震表现为南强北弱,北部盆地受张-渤构造带的影响,盆地走向和断层运动性质均发生了明显变化,多具备发生7级左右地震的构造条件。综合认为,断裂活动特征典型的鄂尔多斯活动地块周缘各边界带,未来强震多发生在大地震离逝时间长的地震空区/空段,或是构造带的转换和交汇区。

Dextral strike-slip of Sanguankou-Niushoushan fault zone and extension of arc tectonic belt in the northeastern margin of the Tibet Plateau

The kinematic characteristics of the Sanguankou-Niushoushan fault（SGK-NSSF） are of great significance to the understanding of the extension of the arc tectonic belt in the northeastern margin of the Tibet Plateau. Using field surveys and various data collection methods, including large-scale geological mapping, measurement of typical topographies, and dating of sedimentary strata, it was determined that the SGK-NSSF exhibits obvious dextral strike-slip characteristics and thus is not a sinistral strike-slip fault, as believed by previous researchers. The results of this study show that the geological boundaries for the Paleozoic, Mesozoic, and Cenozoic eras were all dextrally dislocated by the fault, with the faulted displacements being similar. The maximum strike-slip displacement of the fault, after elimination of topographic effects, was found to be 961±6 m. The Sanguankou fault at the northern section exhibits obvious characteristics of more recent activities, with a series of small gullies having undergone synchronized dextral writhing after traversing the fault. The average horizontal slip rate of the fault since the late Quaternary was determined to be approximately 0.35 mm/a. The pre-existing fold structures formed during the late Pliocene were dislocated by the fault and became ex situ, indicating that dextral strike-slip of the fault could not have occurred prior to the late Pliocene. The maximum displacements and average slip rates were used to estimate the onset time of the dextral strike-slip activities of the fault as being after 2.7 Ma. In this study, the understanding of previous researchers concerning the extension in the northeastern margin of the Tibet Plateau was combined with analyses of the successive relationships between fold deformations and fault activities. During the whole service lifetime of aircraft structures with composite materials,impacts are inevitable and can usually cause severe but barely visible *** the occurrences of impact are random and unpredictable,it is a hotspot direction to develop an online impact monitoring system that can meet strict limitations of aerospace applications including small size,light weight,and low power ***(PZT)sensor,being able to generate impact response signals with no external power and cover a large-scale structure with only a small amount of them,is a promising ***,for real systems,networks with multiple nodes are normally required to monitor large-scale structures in a global way to identify any impact localization confliction,yet the existing studies are mostly evaluated with single nodes instead of ***,in this paper,based on a new low-power node designed,a Bluetooth-based digital impact monitoring PZT sensor network is proposed for the first time with its global confliction-solving impact localization *** of the system as a network are researched and analyzed on a complex real aircraft wing box for a global confliction-solving impact localization,showing a satisfying high accuracy.

Pattern and timing of late Cenozoic rapid exhumation and uplift of the Helan Mountain,China

The Helan Mountain, an intraplate deformation belt in the North China Craton, is located in the northern portion of the China North-South seismic belt, and at the northwestern margin of the Ordos Block. The Cenozoic deformation history of the Helan Mountain is characterized by extension along the eastern Helan Mountain fault (EHSF), resulting in the exhumation and uplift of the Helan Mountain, relative to the rifting of the adjacent Yinchuan Basin. Here we present new apatite fission track (AFT) data from several transects adjacent to the EHSF in the central and northern Helan Mountain. AFT ages from the northern Helan Mountain (Dawukou and Zhengyiguan transects) range from 10 Ma to 89 Ma, whereas AFT ages from the southern Helan Mountain (Suyukou transect) are greater than 71 Ma. The AFT data analysis reveals initiation of rapid uplift and exhumation of the Helan Mountain at 10–12 Ma. Additionally, a plot of the AFT ages versus their mean track length shows a distinctive "boomerang" pattern indicating that the Helan Mountain experienced a discrete phase of accelerated exhumation beginning at 10-12 Ma. Spatially, AFT samples systematically increase in age away from the EHSF and are consistent with late Cenozoic exhumation that was slow in the southwestern Helan Mountain and rapid in the northeastern Helan Mountain, as well more rapid adjacent to the EHSF and slower away from the EHSF. Obviously, the spatial distribution of late Cenozoic exhumation indicates that normal faulting of the EHSF is related to southwestward tilting and rapid exhumation of the Helan Mountain beginning at 10–12 Ma. The uplift and exhumation of the Helan Mountain was a response to the intensive extension of the northwestern margin of the Ordos Block in the late Cenozoic; this occurred under a regional extensional stress field oriented NW-SE along the Yinchuan-Jilantai-Hetao and the Weihe-Shanxi graben systems adjacent to the Ordos Block.

Characteristics of Late Quaternary Activity of the Luhuatai Buried Fault Revealed by Drilling

The Luhuatai fault is one of the important buried tectonics in the Yinchuan basin. Based on the results of shallow seismic exploration, we conducted composite drilling section exploration and dating of the samples from boreholes. Some useful data was obtained, such as the depth of the upper breaking point, the latest activity age, displacement in the late Quaternary, and slip rates, etc. This study shows that the activity is different between the north and south segment along the Luhuatai fault. The north segment is a Holocene fault, while the south segment is a late mid-Pleistocene fault. From north to south along the north segment of Luhuatai fault, the activity has been enhanced, and the faulting is stronger in late Pleistocene than Holocene.

本文首先介绍了无人机摄影测量技术获取数字高程模型(digital elevation model,缩写为DEM)和地貌数据(正射影像)的作业流程,对比分析了三种不同质量密集点云生成的DEM在水平位置和高程上的差异;然后以1709年中卫南M7(1/2)大地震的主体地表破裂带为例,提取其上地震断层的垂直位错量和水平位移量。研究结果显示:高质量密集点云生成的DEM分辨率可达厘米级,且处理时间不需太长,其水平位置和高程与另外两种质量密集点云生成的DEM差异均小于0.100 m;基于高质量密集点云可生成6.33 cm/pix分辨率的DEM,提取1709年中卫南地震地表破裂带上地震断层的垂直位错量为(0.704±0.293) m,水平位移量为5.1 m,与前人的研究结果相吻合,因此可以代表该地震的同震位移,这表明无人机摄影测量技术能够获取地震地表破裂带典型场点的高分辨率地形地貌数据,并基于生成的DEM可进一步提取地震断层的定量参数。

银川盆地东南缘面子山—清水营断裂活动性

研究银川盆地的构造活动特征对于认识青藏、鄂尔多斯和阿拉善3大地块的相互作用具有重要意义。银川盆地东南缘的面子山—猪头岭一带地形高差显著,影像上线性明显,可能发育一条未知的活动断裂。本文沿面子山—清水营一线开展了野外地质调查、探槽开挖和地层年代测试等工作,结果表明:银川盆地东南缘发育面子山—清水营断裂,全长50 km,以猪头岭北为界,分为面子山断层和清水营断层两段。面子山断层以高角度正断为主,断层擦痕显示具有左旋走滑运动,走向北东,跨断层数条冲沟显示具有左旋扭动特征;而清水营断层则表现为逆断层,走向北东东,发育陡坎地貌。该断裂断错了(23.65±1.17) ka BP和(33.16±1.82) ka BP的沉积地层,表明断裂最新活动时代为晚更新世晚期。该断层的发现表明,鄂尔多斯地块的西北边界复杂,构造变形带较宽,且不严格的受控于断陷盆地的边界断裂,其构造变形已扩张到地块一侧,这可能与青藏高原持续向东北向的推挤扩展有关。
