Central swallowing in normal adults using functional magnetic resonance imaging

BACKGROUND： While brain-imaging studies in healthy adults have indicated that multiple cortical regions are involved in swallowing, these functional imaging techniques have not been extensively applied to the complete understand neurophysiology of swallowing in China. A full understanding of normal swallowing neurophysiology is important for improving functional outcomes for dysphagia due to neurologic disorders or damage with increasing age. Thus the interpretations of the functional contributions of various brain areas in swallowing should be scientifically researched. OBJECTIVE： To identify the activation and characteristic of swallowing center in healthy adults using functional magnetic resonance imaging. DESIGN, TIME AND SETTING： An uncontrolled neuroimaging study was performed at the Outpatient Clinic, Department of Radiology, West China Hospital of Sichuan University between March and November 2008. PARTICIPANTS： Ten healthy right-handed volunteers, aged over 20 years with a mean age of （34.2 ＋ 8.1） years, a range of 25-45 years and including five males and five females participated. A medical history was obtained from all potential subjects and all subjects were free of systemic diseases and neurological disorders. METHODS： The healthy volunteers were examined with event-related functional magnetic resonance imaging of blood oxygenation level-dependent while laryngeal swallow-related movements were recorded. Subjects were scanned during voluntary saliva swallowing and water bolus swallowing activation tasks. Data was processed using the General Linear Model. A voxel by voxel group comparison was performed using random effect analysis. Any cluster with a corrected P 〈 0.05 for spatial extent was considered significant. MAIN OUTCOME MEASURES： The cerebral cortical activation maps of voluntary swallowing of saliva and swallowing of water bolus in healthy adults were observed. Non-invasive tracing in vivo can be used to observe the migration and distnbution of grafted stem cells, and can provide experimental evidence for treatment. This study utilized adenovirus-carrying enhanced green fluorescent protein （AD5/F35-eGFP） and superparamagnetic iron oxide （SPIO）-Iabeled bone marrow mesenchymal stem cells （BMSCs）. BMSCs, double-labeled by AD5/F35-eGFP and SPIO, were transplanted into rats with spinal cord injury via the subarachnoid space. MRI tracing results demonstrated that BMSCs migrated to the injured spinal cord over time （T2 hypointensity signals）. This result was verified by immunofluorescence. These results indicate that MRI can be utilized to trace in vivo the SPIO-labeled BMSCs after grafting.

Regional homogeneity abnormalities in patients with tensiontype headache:a resting-state fMRI study

Tension-type headache（TTH） is the most prevalent type of primary headache. Many studies have shown that the pathogenesis of primary headache is associated with fine structural or functional changes. However, these studies were mainly based on migraine. The present study aimed to investigate whether TTH patients show functional disturbances compared with healthy subjects. We used restingstate functional magnetic resonance imaging（f MRI） and regional homogeneity（Re Ho） analysis to identify changes in the local synchronization of spontaneous activity in patients with TTH. Ten patients with TTH and 10 age-, gender-, and education-matched healthy controls participated in the study. After demographic and clinical characteristics were acquired, a 3.0-T MRI system was used to obtain restingstate f MRIs. Compared with healthy controls, the TTH group exhibited significantly lower Re Ho values in the bilateral caudate nucleus, the precuneus, the putamen, the left middle frontal gyrus, and the superior frontal gyrus. There was no correlation between mean Re Ho values in TTH patients and duration of TTH, number of attacks, duration of daily attacks, Visual Analogue Scale score, or Headache Impact Test-6 score. These results suggest that TTHpatients exhibit reduced synchronization of neuronal activity in multiple regions involved in the integration and processing of pain signals.

Asthma is a serious health problem that involves not only the respiratory system but also the central nervous *** studies identified either regional or network alterations in patients with asthma,but inconsistent results were obtained.A key question remains unclear:are the regional and neural network deficits related or are they two independent characteristics in asthma?Answering this question is the aim of this *** collecting resting-state functional magnetic resonance imaging from 39 patients with asthma and 40 matched health controls,brain functional measures including regional activity(amplitude of low-frequency fluctuations)and neural network function(degree centrality(DC)and functional connectivity)were calculated to systematically characterize the functional *** exhibited regional abnormities in the left angular gyrus,right precuneus,and inferior temporal gyrus within the default mode *** abnormalities involved both the sensorimotor network and visual network with key regions including the superior frontal gyrus and occipital *** DC in the lingual gyrus was correlated with the degree of airway *** study elucidated different patterns of regional and network changes,thereby suggesting that the two parameters reflect different brain characteristics of *** findings provide evidence for further understanding the potential cerebral alterations in the pathophysiology of asthma.

Diffusion tensor imaging reveals brain abnormality patterns in a patient with semantic dementia

BACKGROUND： Previous studies regarding primary progressive aphasia （PPA） have focused on progressive, non-fluent aphasia. Little information is available with regard to the use of diffusion tensor imaging compared with conventional magnetic resonance imaging for the detection of subtle structural abnormalities. OBJECTIVE： To investigate and localize brain abnormalities in a Chinese patient with semantic dementia. DESIGN, TIME AND SETTING： A concurrent, non-randomized, case-controlled, neuroimaging, clinical trial was performed at the Department of Radiology, West China Hospital of Sichuan University in March 2009. PARTICIPANTS： One 75-year-old male patient, who was diagnosed with semantic dementia, and 21 age- and gender-matched healthy volunteers were recruited for the study. METHODS： Diffusion tensor imaging was used to determine mean diffusion （MD） and fractional anisotropy （FA） in the brains of the patient and the 21 healthy subjects. Voxel-based analysis of MD and FA values was performed using statistical parametric mapping. MAIN OUTCOME MEASURES： MD and FA value maps differences between patient and controls. RESULTS： MD was significantly increased in both cerebra, but was predominant on the left side and expanded to outside of the language-related region. Reduced MD was not detected in any of the brains. FA was shown to be decreased in the corpus callosum, but was increased in the basal ganglia. CONCLUSION： The present study provided clear in vivo magnetic imaging evidence of diffuse brain involvement in semantic dementia. Increases in MD were greater than in FA when brain diffusion alterations were detected, which suggested that MD could be a better marker of disease progression.

The T_(1)-T_(2) dual-mode probes for magnetic resonance imaging(MRI)can non-invasively acquire comprehensive information of different tissues or generate self-complementary information of the same tissue at the same time,making MRI a more flexible imaging modality for complicated *** this work,three Gadolinium-diethylene-triaminepentaaceticacid(Gd-DTPA)complex conjugated superparamagnetic iron oxide(SPIO)nanoparticles with different Gd/Fe molar ratio(0.94,1.28 and 1.67)were prepared as T_(1)-T_(2) dual-mode MRI probes,named as SPIO@PEG-GdDTPA0.94,SPIO@PEGGdDTPA1.28 and SPIO@PEG-GdDTPA1.67,*** SPIO@PEG-GdDTPA nanocomposites with 8 nm spherical SPIO nanocrystals showed good Gd3þchelate ***@PEG-GdDTPA0.94 nanocomposites with lowest Gd/Fe molar ratio show no cytotoxicity to Raw 264.7 cells as compared to SPIO@PEG-GdDTPA1.28 and SPIO@***@PEG-GdDTPA0.94 nanocomposites with r1(8.4mM^(-1)s^(-1)),r2(83.2mM^(-1)s^(-1))and relatively ideal r2/r1 ratio(9.9)were selected for T_(1)-T_(2) dual-mode MRI of blood vessels and liver tissue in *** contrast images were obtained for both cardiovascular system and liver in animal studies under a clinical 3 T ***,one can get high-quality contrast-enhanced blood vessel images within the first 2 h after contrast agent administration and acquire liver tissue anatomy information up to 24 ***,the strategy of one shot of the dual mode MRI agent could bring numerous benefits not only for patients but also to the radiologists and clinicians,*** time,lowering side effects and collecting data of different organs sequentially.

Despite the great potential of anti-PD-L1 antibodies for immunotherapy,their low response rate due to an immunosuppressive tumor microenvironment has hampered their *** address this issue,we constructed a cell membrane-coated nanosystem(mB4S)to reverse an immunosuppressive microenvironment to an immuno-supportive one for strengthening the anti-tumor *** this system,Epirubicin(EPI)as an immunogenic cell death(ICD)inducer was coupled to a branched glycopolymer via hydrazone bonds and diABZI as a stimulator of interferon genes(STING)agonist was encapsulated into *** internalization of mB4S,EPI was acidic-responsively released to induce ICD,which was characterized by an increased level of calreticulin(CRT)exposure and enhanced ATP ***,diABZI effectively activated the STING *** with mB4S in combination with an anti-PD-L1 antibody elicited potent immune responses by increasing the ratio of matured dendritic cells(DCs)and CD8+T cells,promoting cytokines secretion,up-regulating M1-like tumor-associated macrophages(TAMs)and down-regulating immunosuppressive myeloid-derived suppressor cells(MDSCs).Therefore,this nanosystem for co-delivery of an ICD inducer and a STING agonist achieved promotion of DCs maturation and CD8+T cells infiltration,creating an immuno-supportive microenvironment,thus potentiating the therapy effect of the anti-PD-L1 antibody in both 4T1 breast and CT26 colon tumor mice.
