Celastrol promotes apoptosis of breast cancer MDA-MB-231 cells by targeting HSDL2

Objective:Celastrol is a pentacyclic triterpenoid extracted from the traditional Chinese medicinal herb,Tripterygium wilfordii.This study aims to provide a scientific basis for the rational development and use of celastrol in breast cancer.Method:A quantitative chemical biology approach was used to investigate the protein targets and molecular mechanisms of celastrol in breast cancer cells.Results:Low-concentration celastrol exerted an anti-tumor effect by directly binding to hydroxysteroid dehydrogenase-like 2(HSDL2)and inhibiting its expression.Moreover,the expression of the pro-apoptotic protein,Bcl-2-associated X(BaX),increased,the level of the anti-apoptotic protein,B-cell lymphoma-2(Bcl-2),decreased,and the rate of apoptosis increased.After the transfection of cells with si-HSDL2,the apoptosis rate was similar to that observed after the administration of celastrol.However,apoptosis was reversed by the overexpression of HSDL2.Furthermore,our mass spectrometry(MS)data indicated a relationship between HSDL2 and the mitogen-activated protein kinase(MAPK)signaling pathway.We also found that the expression of HSDL2 was directly related to the degree of extracellular signal-regulated kinase(ERK)phosphorylation.Conclusion:Celastrol may promote apoptosis by suppressing the HSDL2/MAPK/ERK signaling pathway.

Low-Carbon Dispatching for Virtual Power Plant with Aggregated Distributed Energy Storage Considering Spatiotemporal Distribution of Cleanness Value

The scale of distributed energy resources is increasing,but imperfect business models and value transmission mechanisms lead to low utilization ratio and poor responsiveness.To address this issue,the concept of cleanness value of distributed energy storage(DES)is proposed,and the spatiotemporal distribution mechanism is discussed from the perspectives of electrical energy and cleanness.Based on this,an evaluation system for the environmental benefits of DES is constructed to balance the interests between the aggregator and the power system operator.Then,an optimal low-carbon dispatching for a virtual power plant(VPP)with aggregated DES is constructed,where-in energy value and cleanness value are both considered.To achieve the goal,a green attribute labeling method is used to establish a correlation constraint between the nodal carbon potential of the distribution network(DN)and DES behavior,but as a cost,it brings multiple nonlinear relationships.Subsequently,a solution method based on the convex envelope(CE)linear re-construction method is proposed for the multivariate nonlinear programming problem,thereby improving solution efficiency and feasibility.Finally,the simulation verification based on the IEEE 33-bus DN is conducted.The simulation results show that the multidimensional value recognition of DES motivates the willingness of resource users to respond.Meanwhile,resolving the impact of DES on the nodal carbon potential can effectively alleviate overcompensation of the cleanness value.

Bread wheat(Triticum aestivum)is an important crop and serves as a significant source of protein and calories for humans,worldwide.Nevertheless,its large and allopolyploid genome poses constraints on genetic improvement.The complex reticulate evolutionary history and the intricacy of genomic resources make the deciphering of the functional genome considerably more challenging.Recently,we have developed a comprehensive list of versatile computational tools with the integration of statistical models for dissecting the polyploid wheat genome.Here,we summarize the methodological innovations and applications of these tools and databases.A series of step-by-step examples illustrates how these tools can be utilized for dissecting wheat germplasm resources and unveiling functional genes associated with important agronomic traits.Furthermore,we outline future perspectives on new advanced tools and databases,taking into consideration the unique features of bread wheat,to accelerate genomic-assisted wheat breeding.

Enterprise risk management holds significant importance in fostering sustainable growth of businesses and in serving as a critical element for regulatory bodies to uphold market order.Amidst the challenges posed by intricate and unpredictable risk factors,knowledge graph technology is effectively driving risk management,leveraging its ability to associate and infer knowledge from diverse sources.This review aims to comprehensively summarize the construction techniques of enterprise risk knowledge graphs and their prominent applications across various business scenarios.Firstly,employing bibliometric methods,the aim is to uncover the developmental trends and current research hotspots within the domain of enterprise risk knowledge graphs.In the succeeding section,systematically delineate the technical methods for knowledge extraction and fusion in the standardized construction process of enterprise risk knowledge graphs.Objectively comparing and summarizing the strengths and weaknesses of each method,we provide recommendations for addressing the existing challenges in the construction process.Subsequently,categorizing the applied research of enterprise risk knowledge graphs based on research hotspots and risk category standards,and furnishing a detailed exposition on the applicability of technical routes and methods.Finally,the future research directions that still need to be explored in enterprise risk knowledge graphs were discussed,and relevant improvement suggestions were proposed.Practitioners and researchers can gain insights into the construction of technical theories and practical guidance of enterprise risk knowledge graphs based on this foundation.

Malaria is an ancient infectious disease that threatens millions of lives globally even today.The discovery of artemisinin,inspired by traditional Chinese medicine(TCM),has brought in a paradigm shift and been recognized as the“best hope for the treatment of malaria”by World Health Organization.With its high potency and low toxicity,the wide use of artemisinin effectively treats the otherwise drug-resistant parasites and helps many countries,including China,to eventually eradicate malaria.Here,we will first review the initial discovery of artemisinin,an extraordinary journey that was in stark contrast with many drugs in western medicine.We will then discuss how artemisinin and its derivatives could be repurposed to treat cancer,inflammation,immunoregulation-related diseases,and COVID-19.Finally,we will discuss the implications of the“artemisinin story”and how that can better guide the development of TCM today.We believe that artemisinin is just a starting point and TCM will play an even bigger role in healthcare in the 21st century.

Bailing capsule(百令胶囊) alleviates autoimmune thyroiditis via regulating peroxisome proliferator-activated receptor signaling pathway: a multi-omics analysis

OBJECTIVE: To investigate the efficacy and potential mechanism of Bailing capsule (百令胶囊， BL) anti-autoimmune thyroiditis (AIT). METHODS: Based on the AIT rat model， the effect of BL in alleviating AIT was evaluated by detecting serum thyroid index free triiodothyronine (FT3)， free thyroxine (FT4)， thyroid-stimulating hormone (TSH)， thyroglobulin antibody (TGAb)， thyroid peroxidase antibody (TPOAb)， and inflammatory factors Interferon-gamma (IFN-γ)， Interleukin-4， -10， and -12 (IL-4， IL-10， and IL-12) as well as thyroid tissue Hematoxylin-eosin (HE) staining and ultrastructure observation. The mechanism of BL was explored by combining transcriptome and proteome analysis， and further verified by Western blot (WB). RESULTS: BL effectively reduced serum FT3， FT4， TGAb， and TPOAb levels in AIT rats， restored TSH balance， inhibited the release of pro-inflammatory cytokines IFN-γ and IL-12， promoted the production of anti-inflammatory cytokines IL-4 and IL-10， and significantly reduced IFN-γ/IL-4 and IL-12/IL-10， improved thyroid follicular structure， and protected thyroid tissue from injury. Kyoto Encyclopedia of Genes and Genomes and protein interaction network analysis showed that BL affected the expression of fatty acid-binding protein 4， acyl-CoA synthetase long-chain family member 1， and acyl-CoA dehydrogenase long chain to regulate the peroxisome proliferator-activated receptor signaling pathway， thereby inhibiting the fatty acid metabolism and the inflammatory state of AIT rats. CONCLUSIONS: BL could effectively reduce thyroid inflammation in AIT model rats. The possible BL mechanism was to regulate the peroxisome proliferator-activated receptor signaling pathway and inhibit fatty acid metabolism. This study suggested that BL has the potential to be used in clinical treatment of AIT.

Spinal cord injuries impose a notably economic burden on society,mainly because of the severe after-effects they cause.Despite the ongoing development of various therapies for spinal cord injuries,their effectiveness remains unsatisfactory.However,a deeper understanding of metabolism has opened up a new therapeutic opportunity in the form of metabolic reprogramming.In this review,we explore the metabolic changes that occur during spinal cord injuries,their consequences,and the therapeutic tools available for metabolic reprogramming.Normal spinal cord metabolism is characterized by independent cellular metabolism and intercellular metabolic coupling.However,spinal cord injury results in metabolic disorders that include disturbances in glucose metabolism,lipid metabolism,and mitochondrial dysfunction.These metabolic disturbances lead to corresponding pathological changes,including the failure of axonal regeneration,the accumulation of scarring,and the activation of microglia.To rescue spinal cord injury at the metabolic level,potential metabolic reprogramming approaches have emerged,including replenishing metabolic substrates,reconstituting metabolic couplings,and targeting mitochondrial therapies to alter cell fate.The available evidence suggests that metabolic reprogramming holds great promise as a next-generation approach for the treatment of spinal cord injury.To further advance the metabolic treatment of the spinal cord injury,future efforts should focus on a deeper understanding of neurometabolism,the development of more advanced metabolomics technologies,and the design of highly effective metabolic interventions.

Tripterygium glycosides tablet(TGT),the classical commercial drug of Tripterygium wilfordii Hook.F.has been effectively used in the treatment of rheumatoid arthritis,nephrotic syndrome,leprosy,Behcet's syndrome,leprosy reaction and autoimmune hepatitis.However,due to its narrow and limited treatment window,TGT-induced organ toxicity(among which liver injury accounts for about 40%of clinical reports)has gained increasing attention.The present study aimed to clarify the cellular and molecular events underlying TGT-induced acute liver injury using single-cell RNA sequencing(scRNA-seq)technology.The TGT-induced acute liver injury mouse model was constructed through short-term TGT exposure and further verified by hematoxylin-eosin staining and liver function-related serum indicators,including alanine aminotransferase,aspartate aminotransferase,alkaline phosphatase and total bilirubin.Using the mouse model,we identified 15 specific subtypes of cells in the liver tissue,including endothelial cells,hepatocytes,cholangiocytes,and hepatic stellate cells.Further analysis indicated that TGT caused a significant inflammatory response in liver endothelial cells at different spatial locations;led to marked inflammatory response,apoptosis and fatty acid metabolism dysfunction in hepatocytes;activated hepatic stellate cells;brought about the activation,inflammation,and phagocytosis of liver capsular macrophages cells;resulted in immune dysfunction of liver lymphocytes;disturbed the intercellular crosstalk in liver microenvironment by regulating various signaling pathways.Thus,these findings elaborate the mechanism underlying TGT-induced acute liver injury,provide new insights into the safe and rational applications in the clinic,and complement the identification of new biomarkers and therapeutic targets for liver protection.

Spatial Interaction Network Analysis of Crude Oil Trade Relations between Countries along the Belt and Road

Based on the theories and methods of complex network,crude oil trade flows between countries along the Belt and Road(B&R,hereafter)are inserted into the Geo-space of B&R and form a spatial interaction network which takes the countries as nodes and takes the trade relations as edges.The networked mining and evolution analysis can provide important references for the research on trade relations among the B&R countries and the formulation of trade policy.This paper researches and discusses the construction,statistical analysis,top networks and stability of the crude oil trade network between the B&R countries from 2001 to 2020 from the perspectives of Geo-Computation for Social Sciences(GCSS)and spatial interaction.Firstly,evolutions of out-degree,in-degree,out-strength and in-strength of the top 10 countries in the crude oil trade network are computed and analyzed.Secondly,the top network method is used to explore the evolution characteristics of hierarchical structures.And finally,the sequential evolution characteristics of the crude oil trade network stability are analyzed utilizing the network stability measure method based on the trade relationship autocorrelation function.The analysis results show that Russia has the largest out-degree and out-strength,and China has the largest in-degree and in-strength.The crude oil trade volume of the top 10 import and export networks between 2001—2020 accounts for over 90%of the total trade volume of the crude oil trade network,and the proportion remains relatively stable.However,the stability of the network showed strong fluctuations in 2009,2012 and 2014,which may be closely related to major international events in these years,which could furtherly be used to build a correlation model between network volatility and major events.This paper explores how to construct and analyze the spatial interaction network of crude oil trade and can provide references for trade relations research and trade policy formulation of B&R countries.
