Transplantation of human placental chorionic plate-derived mesenchymal stem cells for repair of neurological damage in neonatal hypoxic-ischemic encephalopathy

Neonatal hypoxic-ischemic encephalopathy is often associated with permanent cerebral palsy,neurosensory impairments,and cognitive deficits,and there is no effective treatment for complications related to hypoxic-ischemic encephalopathy.The therapeutic potential of human placental chorionic plate-derived mesenchymal stem cells for various diseases has been explored.However,the potential use of human placental chorionic plate-derived mesenchymal stem cells for the treatment of neonatal hypoxic-ischemic encephalopathy has not yet been investigated.In this study,we injected human placental chorionic plate-derived mesenchymal stem cells into the lateral ventricle of a neonatal hypoxic-ischemic encephalopathy rat model and observed significant improvements in both cognitive and motor function.Protein chip analysis showed that interleukin-3 expression was significantly elevated in neonatal hypoxic-ischemic encephalopathy model rats.Following transplantation of human placental chorionic plate-derived mesenchymal stem cells,interleukin-3 expression was downregulated.To further investigate the role of interleukin-3 in neonatal hypoxic-ischemic encephalopathy,we established an in vitro SH-SY5Y cell model of hypoxic-ischemic injury through oxygen-glucose deprivation and silenced interleukin-3 expression using small interfering RNA.We found that the activity and proliferation of SH-SY5Y cells subjected to oxygen-glucose deprivation were further suppressed by interleukin-3 knockdown.Furthermore,interleukin-3 knockout exacerbated neuronal damage and cognitive and motor function impairment in rat models of hypoxic-ischemic encephalopathy.The findings suggest that transplantation of hpcMSCs ameliorated behavioral impairments in a rat model of hypoxic-ischemic encephalopathy,and this effect was mediated by interleukin-3-dependent neurological function.

Metallenes are an emerging class of two-dimensional(2D)material with outstanding potential in electrocatalysis.Herein,we present a new PdMoSb trimetallene produced by a facile wet-chemistry procedure and tested for the alcohol oxidation reaction.PdMoSb shows an extremely high Pd utilization and superior performance toward ethanol,methanol,and glycerol electro-oxidation compared with PdMo and commercial Pd/C catalysts.Experimental results and density functional theory calculations reveal that the enhanced activity relies not only on the high surface area that characterizes the ultrathin 2D metallene structure,but also on the particular electronic configuration of Sb.Sb facilitates OH−adsorption in the reactive-intermediate pathway and strongly enhances the CO tolerance in the poisoning-intermediate pathway for alcohol oxidation.The excellent alcohol oxidation performance of PdMoSb trimetallene demonstrates the high potential of multimetallenes in the field of electrocatalysis.

A single silk-and multiple pollen-expressed PMEs at the Ga1 locus modulate maize unilateral cross-incompatibility

The Gametophyte factor1(Ga1) locus in maize confers unilateral cross-incompatibility(UCI), and it is controlled by both pollen and silk-specific determinants. Although the Ga1 locus has been reported for more than a century and is widely utilized in maize breeding programs, only the pollen-specific ZmGa1P has been shown to function as a male determinant;thus, the genomic structure of the Ga1 locus and all the determinants that control UCI at this locus have not yet been fully characterized.Here, we used map-based cloning to confirm the determinants of UCI at the Ga1 locus and maize pan-genome sequence data to characterize the genomic structure of the Ga1 locus. The Ga1 locus comprises one silk-expressed pectin methylesterase gene(PME, ZmGa1F) and eight pollenexpressed PMEs(ZmGa1P and ZmGa1PL1-7).Knockout of ZmGa1F in Ga1/Ga1 lines leads to the complete loss of the female barrier function. The expression of individual ZmGa1PL genes in a ga1/ga1 background endows ga1 pollen with the ability to overcome the female barrier of the Ga1 locus. These findings, combined with genomic data and genetic analyses, indicate that the Ga1 locus is modulated by a single female determinant and multiple male determinants, which are tightly linked.The results of this study provide valuable insights into the genomic structure of the Ga2 and Tcb1 loci and will aid applications of these loci in maize breeding programs.

Cisplatin-appended BODIPY for near infraredⅡfluorescent and photoacoustic imaging-guided synergistic phototherapy and chemotherapy of cancer

Combining phototherapy and chemotherapy has been considered a promising modality for cancer therapy due to their synergistic effect.Herein,we developed three D-π-A-structured boron dipyrromethenes(BODIPYs)(named as B-B,B-C,and B-C-Pt).Due to their enlargedπ-conjugated structure and high intramolecular charge transfer effect,the synthesized BODIPYs had photothermal conversion capability,and their absorption and fluorescence spectra were red-shifted.The cisplatin-appended BODIPY(B-C-Pt)exhibited good singlet oxygen(^1O_(2))generation ability and near infrared(NIR)absorption and fluorescence(λ_(Abs)=748 nm,λ_(Em)=947 nm).After being encapsulated by distearoyl phosphoethanolamine polyethyleneglycol 2000(DSPE-PEG-2000),which could inhibit the H-aggregation of B-C-Pt,the absorption and fluorescence of the obtained B-C-Pt nanoparticles(NPs)were red-shifted to 762 and 985 nm,respectively.The^1O_(2)quantum yield and photothermal conversion efficiency of the B-C-Pt NPs were 4.0%and 40.6%,respectively.Moreover,B-C-Pt NPs had chemotherapeutic efficacy due to the presence of cisplatin.In vitro and in vivo studies further demonstrated that B-C-Pt NPs had synergistic therapeutic efficacy.Together,B-C-Pt NPs could be employed in NIRⅡfluorescent and photoacoustic imaging-guided synergistic phototherapy and chemotherapy for cancer treatment.

Tibeto-Burman(TB)people have endeavored to adapt to the hypoxic,cold,and high-UV high-altitude environments in the Tibetan Plateau and complex disease exposures in lowland rainforests since the late Paleolithic period.However,the full landscape of genetic history and biological adaptation of geographically diverse TB-speaking people,as well as their interaction mechanism,remain unknown.Here,we generate a whole-genome meta-database of 500 individuals from 39 TB-speaking populations and present a comprehensive landscape of genetic diversity,admixture history,and differentiated adaptative features of geographically different TB-speaking people.We identify genetic differentiation related to geography and language among TB-speaking people,consistent with their differentiated admixture process with incoming or indigenous ancestral source populations.A robust genetic connection between the Tibetan-Yi corridor and the ancient Yellow River people supports their Northern China origin hypothesis.We finally report substructure-related differentiated biological adaptative signatures between highland Tibetans and Loloish speakers.Adaptative signatures associated with the physical pigmentation(EDAR and SLC24A5)and metabolism(ALDH9A1)are identified in Loloish people,which differed from the high-altitude adaptative genetic architecture in Tibetan.TB-related genomic resources provide new insights into the genetic basis of biological adaptation and better reference for the anthropologically informed sampling design in biomedical and genomic cohort research.

Dear Editor,Inflammatory bowel diseases(IBD)such as ulcerative colitis(UC)and Crohn's disease are affecting increasing numbers of patients worldwide.1 These chronic diseases significantly impact patients'quality of life as they are incurable and disabling.1 Patients with IBD have had an increasing rate of hospitalizations,half of all UC patients are hospitalized at least once during their lifetime,and readmissions after a UC-related hospitalization are common.

Most basal-like breast cancers(BLBCs)have a poor prognosis and high crossover with triple-negative breast cancers(TNBCs).However,approximately 50%of TNBC patients develop chemoresistance.^(1) Dexamethasone reportedly induces Krüppel-like factor 5(KLF5)and can cause docetaxel and cisplatin resistance in TNBC.A super-enhancer can maintain the transcription of KLF5.

Single-cell immune landscape of measurable residual disease in acute myeloid leukemia

Measurable residual disease(MRD) is a powerful prognostic factor of relapse in acute myeloid leukemia(AML). We applied the single-cell RNA sequencing to bone marrow(BM) samples from patients with(n=20) and without(n=12) MRD after allogeneic hematopoietic stem cell transplantation. A comprehensive immune landscape with 184,231 cells was created. Compared with CD8+T cells enriched in the MRDnegative group(MRD-＿CD8), those enriched in the MRD-positive group(MRD+＿CD8) showed lower expression levels of cytotoxicity-related genes. Three monocyte clusters(i.e., MRD+＿M) and three B-cell clusters(i.e., MRD+＿B) were enriched in the MRD-positive group. Conversion from an MRD-positive state to an MRD-negative state was accompanied by an increase in MRD-＿CD8 clusters and vice versa. MRDenriched cell clusters employed the macrophage migration inhibitory factor pathway to regulate MRD-＿CD8 clusters. These findings revealed the characteristics of the immune cell landscape in MRD positivity, which will allow for a better understanding of the immune mechanisms for MRD conversion.

The paper discusses the progress and commercialization of binders for energy storage applications,such as batteries.It explains the role of binders in holding together active materials and current collectors,and highlights the challenges associated with conventional organic solvents in binders.The potential of aqueous binders is introduced as a cost-effective and environmentally friendly alternative.The advantages and limitations of different types of binders are discussed,and the importance of binder selection for optimal battery performance is emphasized.The current state of commercialization of binders is reviewed,and the need for collaboration between researchers,manufacturers,and policymakers to develop and promote environmentally friendly and cost-effective binders is emphasized.The paper concludes by outlining future directions for research and development to further improve the performance and commercialization of binders,while addressing limitations such as lack of standardization,high cost,and long-term stability and reliability.

针对目前水环境中抗生素污染严重的问题,使用简单的溶剂热法制备了CuBi_(2)O_(4)/Bi_(2)WO_(6)(CBWO)Z型异质结催化剂.扫描电子显微镜分析结果表明,其结构为棒状和纳米片状.能量色散元素图谱显示,Cu,W,Bi和O元素均匀分散在CBWO-60中;使用X射线衍射和傅里叶变换红外光谱探究了催化剂的晶体结构和化学键、官能团组成;BET表征结果证明,CBWO-60具有较高的比表面积.X射线光电子能谱(XPS)证明Cu^(+)和Cu^(2+)共存,促进了芬顿(Fenton)反应的循环进行,XPS结合能的位移证明了异质结中CuBi_(2)O_(4)和Bi_(2)WO_(6)之间具有强的电子相互作用,而不是物理混合;使用紫外⁃可见漫反射光谱和价带-X射线光电子能谱分析了异质结的能带结构;利用光致发射光谱、电化学阻抗谱和瞬态光电流响应谱探究了催化剂的电荷转移情况.在该系列催化剂中,CBWO-60在光电类芬顿(PEF-like)体系中对环丙沙星(CIP)的降解效率最高,90 min时,降解效率为98.0%.同时,溶液初始pH在2~6范围时,体系始终能够维持有效的CIP去除效率,与传统芬顿体系相比,该体系的pH应用范围得到了有效拓宽.在PEF-like体系中,CBWO-60对喹诺酮类、磺胺类和四环素类抗生素均表现出较强的降解能力,充分证明了CBWO-60的普适性.CBWO-60在连续5次循环实验后,对CIP仍保持87.8%的降解率,并且反应后催化剂的晶体结构没有发生改变.根据高效液相色谱⁃质谱的结果,提出了CIP降解的5种可能途径.
