A bHLH transcription factor,CsSPT,regulates high-temperature resistance in cucumber

High-temperature stress threatens the growth and yield of crops. Basic helix-loop-helix(bHLH) transcription factors(TFs) have been shown to play important roles in regulating high-temperature resistance in plants. However, the bHLH TFs responsible for high-temperature tolerance in cucumbers have not been identified. We used transcriptome profiling to screen the high temperature-responsive candidate bHLH TFs in cucumber. Here, we found that the expression of 75 CsbHLH genes was altered under high-temperature stress. The expression of the CsSPT gene was induced by high temperatures in TT(Thermotolerant) cucumber plants. However, the Csspt mutant plants obtained by the CRISPR-Cas9 system showed severe thermosensitive symptoms, including wilted leaves with brown margins and reduced root density and cell *** Csspt mutant plants also exhibited elevated H_(2)O_(2) levels and down-regulated photosystem-related genes under normal ***, there were high relative electrolytic leakage(REC), malondialdehyde(MDA), glutathione(GSH), and superoxide radical(O_(2)^(·-)) levels in the Csspt mutant plants, with decreased Proline content after the high-temperature treatment. Transcriptome analysis showed that the photosystem and chloroplast activities in Csspt mutant plants were extremely disrupted by the high-temperature stress compared with wildtype(WT) plants. Moreover, the plant hormone signal transduction, as well as MAPK and calcium signaling pathways were activated in Csspt mutant plants under high-temperature stress. The HSF and HSP family genes shared the same upregulated expression patterns in Csspt and WT plants under high-temperature conditions. However, most bHLH, NAC, and bZIP family genes were significantly down-regulated by heat in Csspt mutant plants. Research on the Digital Economy's Impact on China's Economic Growth:Based on the Variations in Urbanization Levels Across China's Eastern,Central,and Western Regions and Sectoral Heterogeneity

Establishment of a transformation system in close relatives of wheat under the assistance of TaWOX5

Species closely related to wheat are important genetic resources for agricultural production,functional genomics studies and wheat *** this study,a wheat gene related to regeneration,TaWOX5,was applied to establish the Agrobacterium-mediated transformation systems of Triticum monococcum,hexaploid triticale,and rye(Secale cereale L.)using their immature *** plants were efficiently *** the transformation process,the Agrobacterium infection efficiency was assessed by histochemical staining forβ-glucuronidase(GUS).Finally,the transgenic nature of regenerated plants was verified by polymerase chain reaction(PCR)-based genotyping for the presence of the GUS and bialaphos resistance(bar)genes,histochemical staining for GUS protein,and the QuickStix strip assay for bar *** transformation efficiency of *** genotype PI428182 was 94.4%;the efficiencies of four hexaploid triticale genotypes Lin456,ZS3297,ZS1257,and ZS3224 were 52.1,41.2,19.4,and 16.0%,respectively;and the transformation efficiency of rye cultivar Lanzhou Heimai was 7.8%.Fluorescence in situ hybridization(FISH)and genomic in situ hybridization(GISH)analyses indicated that the GUS transgenes were integrated into the distal or near centromere(proximal)regions of the chromosomes in transgenic *** and hexaploid triticale *** the transgenic hexaploid triticale plants,the foreign DNA fragment was randomly integrated into the AABB and RR ***,the transgene was almost stably inherited in the next generation by Mendel’s *** findings in this study will promote the genetic improvement of the three plant species for grain or forage production and the improvement of cereal species including wheat for functional genomics studies.

An underwater vest containing an antioxidant MXene hydrogel for sensitive recognition of fish locomotion

The perception of fish locomotion is important for understanding their adaptive behaviors and ethological ***,the main strategy used for extracting fish attitudes involves the use of a vision-based monitoring system,which is limited in its range of observation and cannot perform tracking for long ***,we report the use of a wearable tagging electronic device,referred to as an underwater vest,to capture the surrounding flow field disturbances triggered by swimming or momentary postural *** of these goals were achieved by integrating a pair of pseudocapacitive pressure-sensing units and a flexible circuit ***,additional conditions,such as variable hydraulic pressures and minimal changes in fish posture,require high stability and sensitivity of the sensing ***,hybrid hydrogel electrodes were developed through cross-linking MXene with holey-reduced graphene oxide nanosheets and further modification with 1-ethyl-3-methylimidazolium dicyan.mide ionic liquids,which increased the interfacial capacitance and long-term interfacial activity of the ***,the sensing unit exhibited ultrahigh sensitivity(S_(max)~136,207 kPa^(−1))in an aquatic environment for 60 days and superior high-pressure resolution(10 Pa)within a wide working range of 1 ***,an underwater vest integrated with such sensing units clearly distinguished and recorded fish *** believe that the designed device may open avenues in flow field monitoring and ocean current detection and provide new insights into the development of sensitive underwater tagging.

High-temperature thin-film sensors(TFSs)often suffer from inadequate tolerance to elevated *** this study,an innovative approach is presented to fabricate in situ integrated TFSs with a core-shell structure on alloy components using coaxial multi-ink printing *** method replaces traditional layerby-layer(LbL) deposition and LbL sintering processes and achieves simplified one-step *** coaxial TFS includes a conductive Pt core for conducting and sensing and a dielectric shell for electrical isolation and high-temperature *** coaxial Pt resistance grid demonstrates excellent high-temperature stability,with a resistance drift rate of only 0.08%·h^(-1) at 800 ℃,significantly lower than traditional Pt *** employing this method,a Pt thin-film strain gauge(TFSG) is fabricated that boasts remarkable high-temperature electromechanical *** effectively addresses the problem of sensitivity degradation experienced by traditional LbL Pt TFSGs when subjected to high *** demonstrate the system integration potential of the technique by printing and verifying the functionality of a long-path thinfilm resistance grid on turbine blades,which can withstand butane flame up to ~1300℃.These results showcase the potential of core-shell structure of the coaxial TFS for high-temperature applications,providing a novel approach to develop high-performance TFS beyond traditional multilayer structure.

Urgent one-stage endoscopic treatment for choledocholithiasis related moderate to severe acute cholangitis: A propensity scorematched analysis

BACKGROUND During emergency endoscopic retrograde cholangiopancreatography(ERCP),the safety and feasibility of performing one-stage endoscopic treatment for patients with acute cholangitis(AC)due to choledocholithiasis are *** To investigate the safety and feasibility of one-stage endoscopic treatment for moderate to severe *** We enrolled all patients diagnosed with moderate to severe cholangitis due to common bile duct stones from January 2019 to July *** outcomes were compared in this study between patients who underwent ERCP within 24 h and those who underwent ERCP 24 h later,employing a propensity score(PS)*** primary outcomes were intensive care unit(ICU)admission rates,ICU length of stay,and duration of antibiotic *** In total,we included 254 patients and categorized them into two groups based on the time elapsed between admission and intervention:The urgent group(≤24 h,n=102)and the elective group(>24 h,n=152).Ninety-three pairs of patients with similar characteristics were selected by PS *** urgent ERCP group had more ICU admissions(34.4%vs 21.5%,P=0.05),shorter ICU stays(3 d vs 9 d,P<0.001),fewer antibiotic use(6 d vs 9 d,P<0.001),and shorter hospital stays(9 d vs 18.5 d,P<0.001).There were no significant differences observed in adverse events,in-hospital mortality,recurrent cholangitis occurrence,30-d readmission rate or 30-d *** Urgent one-stage ERCP provides the advantages of a shorter ICU stay,a shorter duration of antibiotic use,and a shorter hospital stay.
