Decreased expression of gap junction delta-2 (GJD2) messenger RNA and connexin 36 protein in form-deprivation myopia of guinea pigs

Background:More than ten genome-wide association studies have identified the significant association between the gap junction delta-2 (GJD2) gene and myopia.However,no functional studies have been performed to confirm that this gene is correlated with myopia.This study aimed to observe how this gene changed in mRNA and protein level in the form-deprivation myopia (FDM) animal model.Methods:Four-week-old guinea pigs were randomly divided into two groups:control and FDM groups (n =12 for each group).The right eyes of the FDM group were covered with opaque hemispherical plastic lenses for 3 weeks.For all the animals,refractive status,axial length (AL),and corneal radius of curvature were measured at baseline and 3 weeks later by streak retinoscope,A-scan ultrasonography,and keratometer,respectively.Retinal GJD2 mRNA expression and connexin 36 (Cx36) levels in FDM and control groups were measured by quantitative real-time PCR and Western blot analyses,respectively.Those results were compared using independent t test,Mann-Whitney U test,or paired t test.A significance level of P < 0.05 was used.Results:Three weeks later,the FDM group (form-deprived eyes) showed about a myopic shift of approximately-6.75 (-7.94 to -6.31) D,while the control group remained hyperopic with only a shift of-0.50 (-0.75 to 0.25) D (Z =-3.38,P < 0.01).The AL increased by 0.74 (0.61-0.76) and 0.10 (0.05-0.21) mm in FDM and control groups,respectively (Z =-3.37,P < 0.01).The relative mRNA expression of GJD2 in the FDM group decreased 31.58% more than the control group (t =11.44,P < 0.01).The relative protein expression of CX36 on the retina was lowered by 37.72% in form-deprivation eyes as compared to the controls (t=17.74,P < 0.01).Conclusion:Both the mRNA expression of GJD2 and Cx36 protein amount were significantly decreased in the retina of FDM guinea pigs.This indicates that Cx36 is involved in FDM development,providing compensating evidence for the results obtained from genome-wide association studies.

Pregnancy-associated breast cancer(PABC),including breast cancer during pregnancy(BCP)and postpartum breast cancer(PBC),has become a critical issue for clinicians in consideration of both maternal and offspring safety during diagnosis and treatment.The recommendations in this guideline are mainly based on currently available high-quality prospective studies,retrospective cohort studies,meta-analyses,and guidelines and consensus documents published by internationally recognized or national societies or associations.Clinicians should carefully analyze the potential benefits and risks of each treatment before recommending whether to terminate the pregnancy or lactation,balance the advantages and disadvantages of the treatments,and respect the wishes of patients.

Highly efficient electroreduction of CO_(2) by defect single-atomic Ni-N_(3) sites anchored on ordered micro-macroporous carbons

Microporous supports typically fail to fully expose active sites for electrolytes and CO_(2) molecules,and this usually results in low current density for the electrocatalytic CO_(2) reduction reaction(CO_(2)RR).To overcome the biggest obstacle and facilitate commercial applications,defective single-atomic Ni-N_(3) sites anchored to ordered micro-macroporous N-doped carbon(Ni-N/OMC)have been prepared by the pyrolysis of the Ni-ZIF-8@PS(ZIF=zeolitic imidazolate framework)and are intended to provide enhanced CO_(2)RR with a current density at an industrial level.This Ni-ZIF-8@PS is constructed of nickel-based ZIF-8 embedded in the three-dimensional(3D)highly ordered polystyrene spheres(PS).The 3D ordered micro-macroporous architecture of Ni-N/OMC could facilitate the mass transfer of substrates to the accessible defective single-atomic Ni-N_(3) sites through micropores(0.6 nm)and macropores(~200 nm)interconnected by 50 nm channels.In a flow cell,Ni-N/OMC exhibits almost 100.0%CO Faraday efficiency(FECO)between−0.2 and−1.1 V vs.RHE and an industrial level CO partial current density of 208 mA cm^(−2).It has a turnover frequency of 1.5×10^(5) h^(−1) at−1.1 V vs.RHE in 1 M KOH electrolyte,which exceeds that of most reported nickel-based electrocatalysts.This excellent CO_(2)RR performance for Ni-N/OMC makes it a state-of-the-art electrocatalyst for CO_(2)RR.Theoretical calculations show that the defective Ni-N_(3) site can lower the energy of*COOH formation compared with that of the Ni-N4 site,thereby accelerating CO_(2)RR.Ni-N/OMC can also be utilized as a cathodic catalyst in Zn-CO_(2) battery,exhibiting high CO selectivity in the discharge process and excellent stability.This work paves a pathway to rational design of highly efficient electrocatalysts with 3D hierarchically ordered micro-macroporous architecture for CO_(2)RR towards industrial production and commercial applications.

To the Editor:Upfront autologous stem cell transplantation(ASCT)has been an integral part of the management of young and eligible older patients with newly diagnosed multiple myeloma(NDMM)for over three decades.Tandem stem cell transplantation was defined as two planned sequential ASCTs within 3 to 6 months.More than two decades after its introduction,trials exploring the utility of tandem transplants have yielded mixed results.The role of tandem ASCT in multiple myeloma(MM)remains unclear and controversial.In the EMN02/HOVON95 study.

Reduction characteristics of molybdenum trioxide with aluminum and silicon

Thermodynamics for reduction of molybdenum oxides by aluminum and silicon were calculated, and the results show that reduction reaction is feasible at a certain temperature region. Compared to the presence of CaO or CaCO3, reduction products of molybdenum trioxide with aluminum and silicon at various temperatures were detected by X-ray diffraction （XRD）. Results show that molybdenum trioxide is reduced by aluminum or silicon step by step, and the intermediate product is MOO2. At 1000 ℃, molybdenum trioxide could be reduced to metal Mo by aluminum, and in the presence of CaO, metal Mo as the reduction product appears even at 800 ℃. In contrast, silicon could barely reduce molybdenum trioxide to metal Mo even at 1200℃. In the presence of CaO or CaCO3, reducibility of silicon increases significantly, and the reduction products are metal Mo and MoSi2. Altogether, CaO or CaCO3 performs two major roles in reduction process： restraining sublimation of MoO3 and decreasing the temperature of reducing MoO3 to metal Mo.

Feedback maximization of reliability of MDOF quasi integrable-Hamiltonian systems under combined harmonic and white noise excitations

We studied the feedback maximization of reliability of multi-degree-of-freedom (MDOF) quasi integrable-Hamiltonian systems under combined harmonic and white noise excitations. First, the partially averaged It equations are derived by using the stochastic averaging method for quasi integrable-Hamiltonian systems under combined harmonic and white noise excitations. Then, the dynamical programming equation and its boundary and final time conditions for the control problems of maximizing the reliability is established from the partially averaged equations by using the dynamical programming principle. The nonlinear stochastic optimal control for maximizing the reliability is determined from the dynamical programming equation and control constrains. The reliability function of optimally controlled systems is obtained by solving the final dynamical programming equation. Finally, the application of the proposed procedure and effectiveness of the control strategy are illustrated by using an example.

A light-regulated synthetic ion channel constructed by an azobenzene modified hydraphile

Biological ion channels are key molecules for cellular regulation and communication. To mimic the structure and functions of nature ion channels, a new class of light-regulated transmembrane ion channels was reported based on tri（macrocycle） hydraphile and azobenzene photoswitch （hydraphile 1 ）. The liposome-based proton transport assays showed that hydraphile 1 exhibited excellent transmem- brane activity （Y）, and Ymax arrived 0.7 at 40 μmol/L. The successful isomerization of azohenzene moiety was confirmed and qualified by UV and NMR spectra. Upon alternative irradiation of 365 nm UV light and 450 nm visible light, the transmembrane activity of hydraphile I was regulated between 0.35 and 0.5, reversibly. All the obtained results have demonstrated the promise of developing excellent synthetic ion channels with ion gating properties based on simple molecular design.

Background:The new emerging avian influenza A H7N9 virus,causing severe human infection with a mortality rate of around 41%.This study aims to provide a novel treatment option for the prevention and control of H7N9.Methods:H7 hemagglutinin(HA)-specific B cells were isolated from peripheral blood plasma cells of the patients previously infected by H7N9 in Jiangsu Province,China.The human monoclonal antibodies(mAbs)were generated by amplification and cloning of these HA-specific B cells.First,all human mAbs were screened for binding activity by enzyme-linked immunosorbent assay.Then,those mAbs,exhibiting potent affinity to recognize H7 HAs were further evaluated by hemagglutination-inhibiting(HAI)and microneutralizationin vitro assays.Finally,the lead mAb candidate was selected and tested against the lethal challenge of the H7N9 virus using murine models.Results:The mAb 6-137 was able to recognize a panel of H7 HAs with high affinity but not HA of other subtypes,including H1N1 and H3N2.The mAb 6-137 can efficiently inhibit the HA activity in the inactivated H7N9 virus and neutralize 100 tissue culture infectious dose 50(TCID_(50))of H7N9 virus(influenza A/Nanjing/1/2013)invitro,with neutralizing activity as low as 78 ng/mL.In addition,the mAb 6-137 protected the mice against the lethal challenge of H7N9 prophylactically and therapeutically.Conclusion:The mAb 6-137 could be an effective antibody as a prophylactic or therapeutic biological treatment for the H7N9 exposure or infection.

Nonlinear stochastic optimal bounded control of hysteretic systems with actuator saturation

A modified nonlinear stochastic optimal bounded control strategy for random excited hysteretic systems with actuator saturation is proposed. First, a controlled hysteretic system is converted into an equivalent nonlinear nonhysteretic stochastic system. Then, the partially averaged It? stochastic differential equation and dynamical programming equation are established, respectively, by using the stochastic averaging method for quasi non-integrable Hamiltonian systems and stochastic dynamical programming principle, from which the optimal control law consisting of optimal unbounded control and bang-bang control is derived. Finally, the response of optimally controlled system is predicted by solving the Fokker-Planck-Kolmogorov (FPK) equation associated with the fully averaged It? equation. Numerical results show that the proposed control strategy has high control effectiveness and efficiency.

目的:稳定站立和姿态保持是双足机器人正常作业的前提,但各种不确定的外部扰动不可避免。机器人需要通过协调关节运动使其快速恢复到原先的稳定站立状态。创新点:本文提出一种开环bang-bang-bang控制方法,根据扰动情况设计髋关节力矩曲线,可以同时恢复欠驱动双足机器人的平衡状态和身体姿态。与闭环控制相比,本方法恢复更为迅速。方法:首先,将外力扰动情况分为3类:1)外力通过质心,扰动仅改变平衡状态不改变身体姿态;2)外力矩扰动,扰动仅改变身体姿态不改变平衡状态;3)外力不通过质心,扰动同时改变平衡状态和身体姿态(图2)。然后,针对不同扰动情况提出对应的3种开环bang-bang-bang控制方法以及对应的髋关节力矩曲线:1)针对扰动情况1的STB控制(式14),在恢复水平稳定状态的同时不改变原先的身体直立姿态;2)针对扰动情况2的ATB控制(式18),在恢复身体直立姿态的同时不改变水平稳定状态;3)针对扰动情况3的UTB控制(式32),可以同时恢复水平稳定状态以及原先的身体直立姿态。并证明了针对最大可恢复范围内的扰动,本文的控制方法都可以同时恢复双足机器人的平衡状态和身体姿态。最后,通过4组仿真实验验证了本文方法的有效性和相比闭环控制的快速性。结论:针对欠驱动双足机器人站立的扰动恢复问题,提出一种基于髋策略的bang-bang-bang控制方法。该方法可迅速同时恢复机器人的平衡状态及身体姿态。
