Background： Although much attention has been paid to the pharmacokinetics （PKs） of different factor VIII （FVIlI） concentrates in persons with hemophilia A （HA）, limited information is available in young boys with severe HA. In this study, we aimed to assess the PK parameters of FVIII products in boys with severe HA in China. Methods： A total of 36 boys （plasma-derived [pd]-FVIII, n= 15; recombinant [r] FVIII, n = 21） were enrolled between January 2015 and May 2016 in Beijing Children＇s Hospital. PK characteristics of FVIII products were studied according to a reduced 4-sampling time point design （1 h, 9 h, 24 h, and 48 h postinfusion）. Results： The mean FVIII half-life （t1/2） was 10.99 ± 3.45 h （range 5.52-20.02 h）, the mean in vivo recovery （IVR） was 2.01 ± 0.42 IU/dl per IU/kg （range 1.24-3.02 IU/dl per IU/kg） and mean clearance （CL） of FVIII is 4.34 ± 1.58 ml·kg^-1·h ^-1 （range 2.29-7.90 ml·kg^-1·h·^-1）. We also analyzed the influence of several parameters that potentially modulate FVIII PK. The age was closely associated with FVIII half-life （R^2 = 0.32, P 〈 0.01 ）. The t1/2 of FVIII increased by 0.59 h per year. Besides age, yon Willebrand factor antigen （VWF：Ag） also was associated with FVIII half-life （R^2 = 0.52, P 〈 0.01）. Patients with blood Group O had a shorter FVIII halt-life than patients with non-O blood group （9.40 ± 0.68 h vs. 12.3 ± 0.79 h, t = 2.70, P = 0.01）. The FVIII IVR correlated with age （R^2 =0.21, P 〈 0.01） and VWF：Ag level （R^2 = 0.28, P 〈 0.01 ）. CL rates were taster in young patients and in those with low-VWF：Ag levels. CL rates of FVIII are higher in blood Group O versus non-blood Group O persons （5.02 ± 0.38 vs. 4.00 ± 0.32 ml·kg^-1·h^-1 , t = 2.53, P = 0.02）. Conclusions： Chinese boys with severe HA have similar PK values to other ethnic groups and large differences in FVIII PK between individual patients. Age, blood group, and VWF：Ag levels are important determining factors for FVlll CL.

The pursuit of precisely recording and localizing neural activities in brain cortical regions drives the development of advanced electrocorticography (ECoG) devices. Remarkable progress has led to the emergence of micro-ECoG (µECoG) devices with sub-millimeter resolutions. This review presents the current research status, development directions, potential innovations and applications of high-density, high-throughput µECoG devices. First, we summarize the challenges associated with accurately recording single or multiple neurons using existing µECoG devices, including passive multielectrode and active transistor arrays. Second, we focus on cutting-edge advancements in passive µECoG devices by discussing the design principles and fabrication strategies to optimize three key parameters: impedance, mechanical flexibility, and biocompatibility. Furthermore, recent findings highlight the need for further research and development in active transistor arrays, including silicon, metal oxide, and solution-gated transistors. These active transistor arrays have the potential to unlock the capabilities of high-density, high-throughput µECoG devices and overcome the limitations of passive multielectrode arrays. The review explores the potential innovations and applications of µECoG devices, showcasing their effectiveness for both brain science research and clinical applications. © 2024

Satellite-assisted 6G wide-area edge intelligence:dynamics-aware task offloading and resource allocation for remote IoT services

Future 6G networks will integrate non-terrestrial communication with mobile edge computing to enable wide-area edge intelligence, providing ubiquitous communication and computation services for everyone and everything. In this paper, we introduce the intelligent cloud-edge-device architecture into low Earth orbit(LEO) satellite-supported remote Internet of Things(IoT) networks to facilitate latency-sensitive and compute-intensive IoT applications. However, the dynamic spatio-temporal characteristics inherent in LEO satellite networks, such as the high volatility of task traffic and the high mobility of space nodes, pose severe challenges. For efficient task execution, we investigate the joint optimization of cross-region task offloading and cross-domain resource allocation, and innovatively propose a spatio-temporal attention-based proximal policy optimization(STA-PPO) algorithm. Specifically, the temporal attention-based actor network analyzes the timing dependencies of task arrival and makes time-variant decisions. Simultaneously, the spatial attention-based critic network captures the spatial variations of satellite motion and evaluates the topology-related value. We verify the effectiveness of the cloud-edge-device collaboration through extensive simulations. Numerical results demonstrate that the STA-PPO algorithm outperforms benchmarks, showing the lowest system delay and the highest link throughput.

In chemical science, the vertical ionization potential(VIP) is a crucial metric for understanding the electronegativity, hardness and softness of chemical material systems as well as the electronic structure and stability of molecules. Ever since the last century, the model chemistry composite methods have witnessed tremendous developments in computing the thermodynamic properties as well as the barrier heights. However, their performance in realm of the vertical electron processes of molecular systems has been rarely explored. In this study, we for the first time benchmarked the model chemistry composite methods(e.g., CBS-QB3, G4 and W1BD) in comparison with the commonly used Koopmans' s theorem(KT), electron propagator theory(e.g., OVGF, D2, P3 and P3+) and CCSD(T) methods in calculating the VIP for up to 613 molecular systems with available experimental measurements. The large-scale test calculations strongly showed that the CBS-QB3 model chemistry composite technique can be well recommended to calculate VIP from the perspectives of accuracy, economy and applicability. Notably, the VIP values of up to 7 molecules were identified to have the absolute errors of larger than 0.3 e V at all calculation levels,which have strong hints that their VIP experimental values should be re-investigated.
