Clinicopathological significance of p16, cyclin D1, Rb and MIB-1 levels in skull base chordoma and chondrosarcoma

Objective: To investigate the expression of p16, cyclin D1, retinoblastoma tumor suppressor protein (Rb) and MIB-1 in skull base chordoma and chondrosarcoma tissues, and to determine the clinicopathological significance of the above indexes in these ***: A total of 100 skull base chordoma, 30 chondrosarcoma, and 20 normal cartilage tissue samples were analyzed by *** expression levels of p16, cyclinD1,Rb and MIB-1 proteins were assessed for potential correlation with the clinicopathological ***: As compared to normal cartilage specimen (control), there was decreased expression of p16, and increased expression of cyclin D1, Rb and MIB-1 proteins, in both skull base chordoma and chondrosarcoma ***-1 LI levels were significantly increased in skull base chordoma specimens with negative expression of p16, and positive expression of cyclin D1 and Rb (P ＜ 0.05).Significantly elevated MIB-1 LI was also detected in skull base chondrosarcoma tissues, while there was negative expression of p16, cyclin D1 and Rb (P ＜ 0.05).In skull base chordoma, p16 negatively correlated with cyclin D1 and Rb, while cyclin D1 positively correlated with ***, p16, cyclin D1, Rb, or MIB-1 expression showed no correlation with age, gender, or pathological classification of patients with skull base chordoma (P ＞ 0.05).However, p16 and MIB-1 levels correlated with the intradural invasion, and expression of p16, Rb and MIB-1 correlated with the number of tumor foci (P ＜ 0.05).Further, the expression of p16 and MIB-1 appeared to correlate with the prognosis of patients with skull base ***: The abnormal expression of p16, cyclin D1 and Rb proteins might be associated with the tumorigenesis of skull base chordoma and chondrosarcoma.

Endoscopic Approach to Remove Intra-extracranial Tumors in Various Skull Base Regions： 10-year Experience of a Single Center

Background：Some problems have been found in the usually adopted combined approach for the removal of intra-extracranial tumors in skull ***,we described a pure endoscopic transnasal or transoral approach （ETA） for the removal of intra-extracranial tumors in various skull base ***：Retrospectively,clinical data,major surgical complications,pre-and postoperative images,and follow-up information of a series of 85 patients with intra-extracranial tumors in various skull base regions who were treated by surgery via ETA in our skull base center during the past 10 years were reviewed and ***：Gross total tumor removal was achieved in 80/85 cases （94.1%） in this *** 37 cases with tumors in anterior skull base and all 14 cases with tumors in jugular foramen received total tumor *** and three cases with tumors in clivus received total and subtotal tumor removal,*** and subtotal tumor removal was performed for 16 cases and 2 cases in lateral skull base,*** complications in this study included：cerebrospinal fluid leakage （n =3）,meningitis （n =3）,and new cranial nerve deficits （n =3;recovered in 3 months after surgery）.In the follow-up period of40-151 months （median：77 months）,seven patients （8.8%） out of the 80 cases of total tumor removal experienced ***：Complete resection of intra-extracranial growing tumors in various skull base regions can be achieved via the pure ETA in one stage in selected *** procedure for radical removal of tumors is feasible and safe.

Regulation the electronic density of solid-state electrolyte by donor–acceptor(D–A)system can achieve highly-selective Li^(+)transportation and conduction in solid-state Li metal *** study reports a high-performance solid-state electrolyte thorough D–A-linked covalent organic frameworks(COFs)based on intramolecular charge transfer *** other reported COFbased solid-state electrolyte,the developed concept with D–A-linked COFs not only achieves electronic modulation to promote highly-selective Li^(+)migration and inhibit Li dendrite,but also offers a crucial opportunity to understand the role of electronic density in solid-state Li metal *** introduced strong electronegativity F-based ligand in COF electrolyte results in highlyselective Li^(+)(transference number 0.83),high ionic conductivity(6.7×10^(-4)S cm^(−1)),excellent cyclic ability(1000 h)in Li metal symmetric cell and high-capacity retention in Li/LiFePO_(4)cell(90.8%for 300 cycles at 5C)than substituted C-and N-based *** is ascribed to outstanding D–A interaction between donor porphyrin and acceptor F atoms,which effectively expedites electron transferring from porphyrin to F-based ligand and enhances Li^(+)***,we anticipate that this work creates insight into the strategy for accelerating Li^(+)conduction in high-performance solid-state Li metal batteries through D–A system.

Nanograting-Based Dynamic Structural Colors Using Heterogeneous Materials

A 2.5-dimensional dynamic structural color based on nanogratings of heterogeneous materials was proposed by interweaving a pH-responsive hydrogel with IP-L *** nanogrid structures exhibit brilliant tuneable structural color, high sensitivity, and ultrafast recovery speeds in response to *** 4D printing-based grayscale design approach was proposed for the patterned encoding and array printing of dynamic structural colors, promoting their application in patterned printing, information encryption, and microfluidic chip sensing. Dynamic structural colors can change in response to different environmental stimuli. This ability remains effective even when the size of the species responsible for the structural color is reduced to a few micrometers, providing a promising sensing mechanism for solving microenvironmental sensing problems in micro-robotics and microfluidics. However, the lack of dynamic structural colors that can encode rapidly, easily integrate, and accurately reflect changes in physical quantities hinders their use in microscale sensing applications. Herein, we present a 2.5-dimensional dynamic structural color based on nanogratings of heterogeneous materials, which were obtained by interweaving a pH-responsive hydrogel with an IP-L photoresist. Transverse gratings printed with pH-responsive hydrogels elongated the period of longitudinal grating in the swollen state, resulting in pH-tuned structural colors at a 45 degrees incidence. Moreover, the patterned encoding and array printing of dynamic structural colors were achieved using grayscale stripe images to accurately encode the periods and heights of the nanogrid structures. Overall, dynamic structural color networks exhibit promising potential for applications in information encryption and in situ sensing for microfluidic chips.

The coronavirus disease 2019（COVID-19） outbreak caused by the severe acute respiratory syndrome coronavirus-2（SARS-CoV-2） developed into a global health emergency. Systemic microthrombus caused by SARS-CoV-2 infection is a common complication in patients with COVID-19. Cardiac microthrombosis as a complication of SARS-CoV-2 infection is the primary cause of cardiac injury and death in patietns with severe COVID-19. In this study, we performed single-cell sequencing analysis of the right ventricular free wall tissue from healthy donors,patients who died during the hypercoagulable period of characteristic coagulation abnormality（CAC）, and patients who died during the fibrinolytic period of CAC. We collected 61,187 cells enriched in 24 immune cell subsets and 13 cardiac-resident cell subsets. We found that in the course of SARS-CoV-2 infected heart microthrombus, MYO1EhighRASGEF1Bhighmonocyte-derived macrophages promoted hyperactivation of the immune system and initiated the extrinsic coagulation pathway by activating chemokines CCL3, CCL5. This series of events is the main cause of cardiac microthrombi following SARS-CoV-2 infection. In a SARS-CoV-2 infected heart microthrombus, excessive immune activation is accompanied by an increase in cellular iron content, which in turn promotes oxidative stress and intensifies intercellular competition. This induces cells to alter their metabolic environment, resulting in increased sugar uptake via the glycosaminoglycan synthesis pathway. In addition, high levels of reactive oxygen species generated by elevated iron levels promote increased endogenous malondialdehyde synthesis in a subpopulation of cardiac endothelial cells. This exacerbates endothelial cell dysfunction and exacerbates the coagulopathy process.

NLRC3 alleviates hypoxia/reoxygenation induced inflammation in RAW264.7 cells by inhibiting K63-linked ubiquitination of TRAF6
