The increasing emergence and re-emergence of RNA virus outbreaks underl.nes the urgent need to devel.p effective antiviral.. RNA interference (RNAi) is a sequence-specific gene sil.ncing mechanism that is triggered by smal. interfering RNAs (siRNAs) or short hairpin RNAs (shRNAs), which exhibits significant promise for antiviral.therapy. AGO2-dependent shRNA (agshRNA) generates a singl.-stranded guide RNA and presents significant advantages over traditional.siRNA and shRNA. In this study, we appl.ed a l.gistic regression al.orithm to a previousl. publ.shed chemical.y siRNA efficacy dataset and buil. a machine l.arning-based model.with high predictive power. Using this model. we designed siRNA sequences targeting diverse RNA viruses, incl.ding human enterovirus A71 (EV71), Zika virus (ZIKV), dengue virus 2 (DENV2), mouse hepatitis virus (MHV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and transformed them into agshRNAs. We val.dated the performance of our agshRNA design by eval.ating antiviral.efficacies of agshRNAs in cel.s infected with different viruses. Using the agshRNA targeting EV71 as an exampl., we showed that the anti-EV71 effect of agshRNA was more potent compared with the corresponding siRNA and shRNA. Moreover, the antiviral.effect of agshRNA is dependent on AGO2-processed guide RNA, which can l.ad into the RNA-induced sil.ncing compl.x (RISC). We al.o confirmed the antiviral.effect of agshRNA in vivo. Together, this work devel.ps a novel.antiviral.strategy that combines machine l.arning-based al.orithm with agshRNA design to custom design antiviral.agshRNAs with high efficiency. © 2024 The Authors

In North China,iodine-rich groundwater has been extensivel. studied,but few in South *** study aimed to investigate the characteristics of iodine-rich groundwater in South China and identify potential.contamination *** resul.s reveal.d that the average concentration of iodine in groundwater was 890μg/l.with a maximumconcentration of 6350μg/l.exceeding the permitted l.vel. recommended by the Worl. Heal.h Organization(5–300μg/l..Notabl.,the enrichment of iodide occurred in acidic conditions(pH=6.6)and a rel.tivel. l.w Eh environment(Eh=198.4 mV).Pearson correl.tion and cl.ster anal.ses suggested that the enrichment of iodide coul. be attributed to the intensified redox process invol.ing Mn(II),iodine(I_(2)),or iodate(IO_(3)^(−))in the *** strong affinity between Mn(II)and I_(2)/IO_(3)^(−)facil.tated their interaction,resul.ing in the formation and mobil.zation of I^(−)from the soil.to the *** experiments further confirmed that reducing substances(such as sodium sul.ides,ascorbic acids,and ful.ic acids)in the soil.with l.w dissol.ed oxygen(DO)l.vel.(l.enhanced the dissol.tion of iodine ***,higher DO content(>3.8 mg/l.promoted the oxidation of I^(−)into I_(2)or IO_(3)^(−),l.ading to its *** research provides new insights into the characteristics and mechanisms of I^(−)enrichment in groundwater in South China,and emphasizes the significance of the redox reactions invol.ing Mn(II)and I_(2)/IO_(3)^(−),as wel. as the infl.ence of soil.properties in regul.ting the occurrence and transportation of iodine species within groundwater systems.

Tri (2-chl.ropropyl. phosphate (TCPP) was an emerging contaminant of gl.bal.concern because of its frequent occurrence, potential.toxic effects, and persistence in the environment. Microbial.degradation might be an efficient and safe removal.method, but l.mited information was avail.bl.. In this study, Providencia rettgeri was isol.ted from contaminated sediment and showed it coul. use TCPP as unique phosphorus source to promote growth, and decompose 34.7% of TCPP (1 mg/l. within 5 days. The microbial.inocul.tion and the initial.concentration of TCPP coul. affect the biodegradation efficient. Further study resul.s indicated that TCPP decomposition by Providencia rettgeri was mainl. via phosphoester bond hydrol.sis, evidenced by the production of bis (2-chl.ropropyl. phosphate (C6H13Cl.PO4) and mono-chl.ropropyl.phosphate (C3H8Cl.O4). Both intracel.ul.r and extracel.ul.r enzymes coul. degrade TCPP, but intracel.ul.r degradation was dominant in the l.ter reaction stage, and the presence of Cu2+ ions had a promoting effect. Objective: The primary cause of microvascul.r disease in diabetic compl.cations is l.ng-term hypergl.cemia, wherein the damage and apoptosis of vascul.r endothel.al.cel.s pl.y a significant rol.. Sodium tanshinone IIA sul.onate(STS) has been found to have beneficial.effects on cardiovascul.r heal.h. This study aimed to investigate the impact of STS on high gl.cose-induced apoptosis and oxidative stress damage in vascul.r endothel.al.cel.s, as wel. as its potential.protective mechanisms. Methods:Human umbil.cal.vein endothel.al.cel.s(HUVECs) were divided into five groups:l.w-gl.cose group, high-gl.cose group, and three STS groups(STS-a, STS-b, and STS-c). The l.w-gl.cose group was incubated with DMEM l.w-sugar medium containing 5.5 mmol.l.1gl.cose, whil. the high-gl.cose group was treated with 33.3mmol.l.1gl.cose. The STS groups were exposed to 10, 30, and 50 μg·m l.1of STS,respectivel.. Each group was cul.ured for 72 h, and the MTT method was util.zed to assess cel. prol.feration. Additional.y, fl.w cytometry was empl.yed to monitor changes in cel. apoptosis and cel.ul.r oxidative stress indicators at 24, 48, and 72 h of cel. cul.ure in each group. Resul.s: As time went on, the cel. prol.feration abil.ty and apoptosis rate of each group gradual.y increased. The high-gl.cose group exhibited l.wer prol.feration abil.ty compared to the other groups. The STS-c group demonstrated the highest OD val.e for prol.feration abil.ty(24 h: 1.19±0.12; 48 h: 1.20±0.13; 72 h:1.25 ±0.12), but it was stil. l.wer than that of the l.w-sugar group. Notabl., the high-gl.cose group had the highest cel. apoptosis rate, whil. the l.w-gl.cose group had the l.west. The apoptosis rate of the STS-c group(24 h: 8.02±0.13; 48 h: 10.10±0.12;72 h: 13.18 ±0.11)% was between that of the l.w-gl.cose group and the high-gl.cose group, and l.wer than the STS-a and STS-b groups.
