CircTHSD4 promotes the malignancy and docetaxel (DTX) resistance in prostate cancer by regulating miR-203/HMGA2 axis

Objective:Circular ribose nudeic acids(circRNAs)are implicated in tumor progression and drug resistance of prostate cancer(PCa).The current work explored the function of circ_0005203(aircTHSD4)in the malignancy and docetaxel(DTX)resistance of PCa.Methods:circTHSD4 expression within PCa as well as matched non-carcinoma samples was measured through real time reverse transcription quantitative polymerase chain reaction(RT-qPCR).In addition,a subcellular fraction assay was conducted to determine circTHSD4 subcellular localization within PCa cells.In addition,we performed a Western blot(WB)assay to detect high mobility.group A2 protein(HMGA2)levels.Besides,functional associations of two molecules were investigated through dual luciferase reporter assay.Cell Counting Kit(CCK)-8,colony formation together with Transwell assay was conducted to assess malignant phenotypes of PCa cells,whereas flow cytometry was performed to determine cell apoptosis.Furthermore,a xenograft mouse model was constructed to verify the effect of circTHSD4 on the carcinogenesis of PCa cells.Results:According to RT-qPCR results,circTHSD4 was up-regulated within PCa tissues and cells,which predicted the dismal prognostic outcome of PCa cases.circTHSD4 silencing within PCa cells markedly suppressed cell growth,migration,and colony fomation.circTHSD4 silencing remarkably elevated PCa cell apoptosis and carcinogenesis within the xenograft model.Further,circTHSD4 silencing enhanced docetaxel(DTX)sensitivity in PCa cells.Furthermore,we demonstrated that circTHSD4 modulated the malignancy of PCa cells by regulating HMGA2 expression through sponging miR 203.Conclusion:Together,our findings suggest that cirCTHSD4 overexpression could promote the malignant phenotype and DTX resistance in PCa through the regulation of the miR 203/HMGA2 axis.

Energy-saving design and optimization of pressure-swing-assisted ternary heterogenous azeotropic distillations

A huge amount of energy is always consumed to separate the ternary azeotropic mixtures by distillations.The heterogeneous azeotropic distillation and the pressure-swing distillation are two kinds of effective technologies to separate heterogeneous azeotropes without entrainer addition.To give better play to the synergistic energy-saving effect of these two processes,a novel pressure-swing-assisted ternary heterogeneous azeotropic distillation(THAD)process is proposed firstly.In this process,the ternary heterogeneous azeotrope is decanted into two liquid phases before being refluxed into the azeotropic distillation column to avoid the aqueous phase remixing,and three columns'pressures are modified to decrease the flowrates of the recycle streams.Then the dividing wall column and heat integration technologies are introduced to further reduce its energy consumption,and the pressureswing-assisted ternary heterogeneous azeotropic dividing-wall column and its heat integration structure are achieved.A genetic algorithm procedure is used to optimize the proposed processes.The design results show that the proposed processes have higher energy efficiencies and lower CO_(2)emissions than the published THAD process.

Posttranslational modifications(PTMs)of proteins,particularly acetylation,phosphorylation,and ubiquitination,play critical roles in the host innate immune response.PTMs’dynamic changes and the crosstalk among them are complicated.To build a comprehensive dynamic network of inflammation-related proteins,we integrated data from the whole-cell proteome(WCP),acetylome,phosphoproteome,and ubiquitinome of human and mouse macrophages.Our datasets of acetylation,phosphorylation,and ubiquitination sites helped identify PTM crosstalk within and across proteins involved in the inflammatory response.Stimulation of macrophages by lipopolysaccharide(LPS)resulted in both degradative and non-degradative ubiquitination.Moreover,this study contributes to the interpretation of the roles of known inflammatory molecules and the discovery of novel inflammatory proteins.

The mortality rate of the recent global pandemic corona virus disease 2019(COVID-19)is currently as high as 7%.The SARS-CoV-2 virus is the culprit behind COVID-19.SARS-CoV-2 is an enveloped single-stranded RNA virus,the genome encodes four types of the structural proteins:S protein,E protein(envelope protein),M protein(matrix protein)and N protein(nucleocapsid protein).In COVID-19,monoclonal antibodies have played a significant role in diagnosis and treatment.This article briefly introduced the development of monoclonal antibodies targeting on S protein and N protein,which represents the main direction of monoclonal antibody drugs used in the diagnosis and treatment of COVID-19.Meanwhile,the traditional Chinese medicine also plays important role in the fight against COVID-19 by regulating human immunity.The article introduced the use of traditional Chinese medicine in fighting against COVID-19.

What has become known as Stahl's Theorem in power-engineering circles has been used to justify a convergence guarantee of the Holomorphic Embedding Method(HEM)as it applies to the power-flow problem.In this,the second part of a two-part paper,we examine implications to numerical convergence of the HEM and the numerical properties of a Pade approximant algorithm.We show that even if the point of interest is within the convergence domain,numerical convergence of the sequence of Pade approximants computed with finite precision is not guaranteed.We propose a convergence factor equation that can be used to both estimate the convergence rate and the capacity of the branch cut.We also show that the study of convergence properties of the Pade approximant is the study of the location of the branch-points of the function,which in turn dictate branch-cut topology and capacity and,therefore,convergence rate.

Temporal Integrative Omics Reveals an Increase in Nondegradative Ubiquitylation during Primary Hepatocyte Dedifferentiation

Primary hepatocytes(PHCs)are widely used in various fields,but the progressive deterioration of liverspecific features in vitro significantly limits their application.While the transcriptional regulation and whole cell proteome(WCP)of PHCs have been extensively studied,only a small number of studies have addressed the role of posttranslational modifications in this process.To elucidate the underlying mechanisms that induce dedifferentiation,we carried out parallel quantifications of the transcriptome,WCP,ubiquitinome,and phosphoproteome of rat PHCs after 0,6,12,24,and 48 h of in vitro culture.Our data constitute a detailed proteomic analysis of dedifferentiated PHCs including 2196 proteins,2056 ubiquitinated sites,and 4932 phosphorylated peptides.We revealed a low correlation between the transcriptome and WCP during dedifferentiation.A combined analysis of the ubiquitinome with the corresponding WCP indicated that the dedifferentiation of PHCs led to an increase in nondegradative K27 ubiquitination.Functional analysis of the altered phosphoproteins suggested a significant enrichment in ferroptosis.In all,404 proteins with both ubiquitination and phosphorylation were identified to be involved in critical metabolic events.Furthermore,Ptbph Hnqjd,Hnrnpu,and Srrm2 were identified as hub genes.Taken together,our data provide new insights into proteome dynamics during PHC dedifferentiation and potential targets to inhibit the dedifferentiation process.

目的:利用光学相干断层扫描血管成像(OCTA)探讨不同程度非增殖性糖尿病视网膜病变(NPDR)的表现差异。方法:横断面研究。纳入糖尿病患者77例77眼,分为非DR组(NDR,23眼)和NPDR组(54眼),其中轻度NPDR(20眼)、中度NPDR(20眼)和重度NPDR(14眼)。NDR组与NPDR组的黄斑区中央凹无血管区(FAZ)面积、浅层和深层毛细血管密度(SSP和DSP)、视力(LogMAR)进行比较,不同程度NPDR的视力、FAZ面积、SSP和DSP水平进行比较,FAZ面积、视力、SSP和DSP与病情严重程度进行相关性分析。结果:与NDR组相比,NPDR组的视力值(LogMAR)、黄斑区FAZ面积增大,SSP和DSP降低(P<0.05);不同程度NPDR的视力、FAZ面积、SSP和DSP水平差异显著(P<0.05)。视力(LogMAR)、FAZ面积与病情严重程度呈正相关,SSP和DSP与病情严重程度呈负相关。结论:随着NPDR病情的进展,视力(LogMAR)、FAZ面积增大,SSP和DSP降低。

目的总结DSG2/PKP2/DSP基因变异相关儿童致心律失常型右室心肌病(ARVC)的临床表型及基因型特点,为儿科精准化诊治ARVC提供依据。方法回顾性分析2018年1月至2023年6月在首都医科大学附属北京儿童医院诊治的7例DSG2/PKP2/DSP基因变异致ARVC患儿的临床资料,基因检测采用靶向高通量二代测序法,并通过Sanger进行家系成员验证。结果7例(男3例、女4例)患儿入院年龄中位数为11.8岁。7例患儿心电图及24 h动态心电图均记录到室性心律失常。7例患儿超声心动图提示右心室不同程度扩大,2例合并左心室扩大。心脏磁共振成像显示2例患儿右室壁运动幅度减低,4例患儿双心室室壁运动异常;3例患儿双心室心肌可见延迟强化信号,2例左心室心肌可见延迟强化信号。4例患儿存在DSG2基因变异,其中3例为复合杂合变异,1例为纯合变异;2例患儿分别存在PKP2和DSP基因杂合变异;1例患儿同时存在PKP2和DSP基因杂合变异。7例患儿共检测到9个基因变异位点,其中5个为未报道的新变异。6例经家系验证,5例患儿为家族遗传性变异,1例为自发变异。7例患儿均给予β受体阻滞剂和(或)胺碘酮口服抗心律失常治疗,6例给予抗心力衰竭治疗。随访3~50个月,1例死亡,1例因严重全心功能衰竭行心脏移植术。结论儿童ARVC临床表现差异大,左心室受累患儿预后较差;基因检测有助于早期诊断和风险评估;尽早干预及药物治疗可延缓部分患儿的病情进展;5个新变异丰富了ARVC的基因突变谱。

目的分析不伴黄斑水肿的非增生型糖尿病视网膜病变(NPDR)的影响因素。方法回顾性病例对照研究。连续纳入2023年5月至8月昆明医科大学第一附属医院确诊的2型糖尿病71例, 其中无糖尿病视网膜病变23例作为对照组;48例不伴黄斑水肿的NPDR作为观察组。观察组包括轻度NPDR者20例, 中度NPDR者16例, 重度NPDR者12例。比较各组之间的性别、年龄、糖尿病病程及糖化血红蛋白(HbA1c)等指标, 使用多变量Logistic回归分析来探讨不伴黄斑水肿的NPDR的影响因素。结果对照组和观察组患者在性别(χ^(2) =2.37, P=0.123)、年龄(t=0.19,P=0.847)、高血压现病史(χ^(2) =1.81, P=0.179)、甘油三酯(Z=0.72, P=0.472)、低密度脂蛋白(Z=0.14, P=0.888)及高密度脂蛋白(Z=0.57, P=0.527)对比, 差异均无统计学意义;而HbA1c[(7.32±0.86)% vs (9.71±1.33)%]、总胆固醇水平[3.63(3.43, 4.02)mmol/L vs 4.53(4.01, 5.31)mmol/L]及糖尿病病程[3.00(2.00, 12.00)年vs 13.50(7.00, 17.00)年]指标对比, 差异有统计学意义(t=9.10, P<0.001;Z=4.74, P<0.001;Z=3.29, P=0.001)。其中, 轻度、中度、重度不伴黄斑水肿的NPDR患者的HbA1c、总胆固醇水平比较, 差异具有统计学意义(F=4.66, P=0.014;F=4.17, P=0.029)。采用多变量Logistic回归方程分析, HbA1c水平(OR=1.91,P=0.011)、总胆固醇水平(OR=2.24, P=0.013)是不伴黄斑水肿的NPDR发生的危险因素。结论 HbA1c水平和总胆固醇水平是不伴黄斑水肿的NPDR的危险因素;不伴黄斑水肿的NPDR的严重程度与HbA1c水平、总胆固醇水平成正相关关系。

Although there has been a high interest in Xylaria over the last 30 years, there have been very few reports on the genus from the Chinese mainland. Two species of Xylaria collected from southern China are reported. Xylaria spinulosa is introduced as a new species based on morphology and sequence data analysis. Xylaria spinulosa differs from other species in the genus mainly by its long thorns covering the surface of the stroma. The morphological description is as follows. Saprobic on dead tree trunk, forming on the host surface. Sexual morph: Stromata upright, usually solitary,unbranched, cylindrical or clavate, fertile at apex, texture soft, internally white, whole stromata up to1.8–2.5 cm long × 0.3–0.5 cm broad, 0.5–1 cm long at stipes. Surface of stromata black, covered with long soft thorn. Stalk well defined and easy to distinguish. Perithecia 0.3–0.5 mm diam., spherical,embedded, closely arranged. Ostioles inconspicuous, not clear. Asci 80–214 × 8–12 μm（157 × 10.2μm, n=20）, unitunicate, 8-spored, cylindrical, long-stipitate, the spore-bearing part 60–170 μm long.Apical apparatus 8.5–11.5 μm high × 5–6 μm broad, blue in Melzer’s reagent, urn-shaped.Ascospores19–23 × 6.5–8.5 μm（21.3 × 7.6 μm, n=30）, dark brown to black, unicellular, ellipsoid to inequilateral, with broadly rounded ends, smooth-walled. Germ slit half spore-length, oblique to sigmoid, lacking sheath and appendage. Xylaria atrosphaerica is a new record for China.
