Rice bran oil rescues cognitive decline in D-galactose-induced aging mice by inhibiting Aβ accumulation and Tau hyperphosphorylation induced oxidative stress and neuroinflammation

Alzheimer's disease (AD) is a neurodegenerative disorder associated with brain aging， and the accumulation of amyloid-beta (Aβ) and hyperphosphorylated Tau proteins are key pathological features. Currently， drugs for the treatment of AD are mainly single-targeted， but the complex pathogenesis of AD makes it difficult to achieve the desired results. Therefore， the development of multitargeted therapies is crucial for future interventions. Rice bran oil (RBO) has been recognized as an edible oil with several health benefits， but its effects on AD caused by brain aging remain underexplored. In this study， the effects of RBO on memory dysfunction in D-galactose (D-gal) mice and its molecular mechanisms were investigated via in vivo and in silico methods from the perspective of AD pathologies. Our results suggested that compounds in RBO could modulate the activities of β-site amyloid precursor protein cleaving enzyme 1 (BACE1)， mitogen-activated protein kinase 3 (MAPK3)， matrix metalloproteinase 3 (MMP3)， and intercellular adhesion molecule 1 (ICAM1)， leading to inhibition of Aβ accumulation and Tau protein hyper-phosphorylation. Moreover， RBO reduced Aβ-induced oxidative stress by inhibiting the activity of mouse double minute 2 homolog (MDM2) and cyclic adenosine monophosphate response element binding protein binding protein (CREBBP)， and attenuated neuroinflammation by inhibiting the activity of nitric oxide synthase 2 (NOS2) and reducing Aβ accumulation and Tau protein hyperphosphorylation. Additionally， α-linolenic acid in RBO exhibited inhibitory effects on D-gal-induced apoptosis in PC12 cells through modulation of NOS2， MDM2， ICAM1， and phospho-extracellular signal-regulated kinase 1/2 (p-ERK1/2). Similarly， stigmastanol inhibited apoptosis in D-gal-induced PC12 cells through the regulation of NOS2. Thus， RBO can be considered as a potential functional food to attenuate AD owing to its multicomponent and multitarget effects.

Dietary parboiled rice(PR)has a low risk of disease,but little is known about the contribution of PR to the prevention of *** potential role and underlying mechanisms of PR in hyperlipidemia were evaluated in this *** C57BL/6J mice were fed with a normal diet,high-fat diet(HFD)containing refined rice(HFDRR)or PR(HFDPR).It was found that PR intervention improved lipid accumulation in *** data analysis revealed that 27 genes were up-regulated(mostly involved in lipid breakdown)and 86 genes were down-regulated(mostly involved in inflammatory responses)in the HFDPR group compared to the HFDRR *** 15 differentially expressed genes(DEGs)were validated by quantitative real-time PCR(RT-qPCR),while protein interaction network showed that protein tyrosine phosphatase receptor type C(PTPRC)has a central *** gut microbiota of mice was also altered after different dietary treatments,with higher ratio of Firmicutes and Bacteroidetes,increased abundances of Ruminococcaceae,Lachnospiraceae,Christensenellaceae,Porphyromonadaceae,Rikenellaceae and Prevotellaceae,and decreased abundances of Lactobacillaceae,Peptostreptococcaceae,Erysipelotrichaceae and Actinobacteria in the HFDRR *** addition,it was observed that PPAR signaling pathway may act as a bridge between DEGs and differential gut *** results suggested that PR can prevent hyperlipidemia by modulating liver genes and gut microbiota.

Background and aims: Dexamethasone is a common glucocorticoid medication with adverse effects that can cause muscle atrophy， but no drug intervention has been approved or recommended for this condition. KF-8 is a rice bran-derived anti-oxidant peptide that extends the life span of Caenorhabditis elegans. Methods: We established a C. elegans model of dexamethasone-induced myopathy to evaluate the potential therapeutic effects of KF-8 in this model. C. elegans muscle function was assessed in terms of locomotory behaviors including crawling， swimming， burrowing， pharyngeal pumping， and head swing. Muscle actin filament integrity was evaluated using fluorescence imaging. The molecular mechanisms of KF-8 were investigated using transcriptome sequencing， qRT-PCR， RNA interference， and western blot analysis. Results: Dexamethasone disrupted actin filaments in the striated muscles of the body wall and inhibited C. elegans crawling， swimming， burrowing， pharyngeal pumping， and head swing. KF-8 reversed the actin filament disruption and locomotor dysfunction induced by dexamethasone. Transcriptome sequencing， pathway enrichment， and qRT-PCR analyses revealed that KF-8 regulated the locomotion-related genes W04G5.10， vha-12， and ddr-1， as well as age-1 (the catalytic subunit ortholog of PI3K)， and akt1. RNA interference， conducted using a genetically engineered E. coli HT115 strain as a food source， confirmed age-1 as a key regulator of locomotor function of C. elegans. Further mechanistic studies with C2C12 myotubes showed that KF-8 regulated the IRS-PI3K-Akt pathway， the master regulator of protein synthesis and degradation. Conclusion: Together， these findings suggest that KF-8 protects against dexamethasone-induced myopathy in C. elegans by regulating locomotion-related genes and the IRS-PI3K-Akt pathway.

以3种晶型的淀粉即大米淀粉(A型)、马铃薯淀粉(B型)、豌豆淀粉(C型)为原料,探究大豆蛋白对不同晶型淀粉理化特性的影响。采用扫描电镜、傅里叶红外光谱仪、快速黏度分析仪和差示量热扫描仪、模拟体外消化等手段分析处理前、后淀粉的结构、功能及消化特性。结果表明,大豆蛋白对不同晶型的淀粉作用后,O-H伸缩振动的吸收峰向更高方向偏移,T0、Tp的值均增加,糊化温度均升高。与大豆蛋白作用后,大米淀粉的RDS含量下降至37.56%,马铃薯淀粉的RDS含量下降至7.48%,豌豆淀粉的RDS含量下降至9.23%。整体表现为颗粒聚集程度加强,热稳定性提高,结构稳定性增强和消化性降低。不同晶型淀粉间结构的差异性为:A型淀粉表现出结晶区有序性更高,不易老化的特性;B型淀粉表现出更易糊化,消化性降低幅度更大的特性;C型淀粉表现为复合后形成类似“丝状”的结构。上述结果说明,蛋白对不同晶型淀粉作用后对特性的改善起到了作用,且不同晶型淀粉之间的表现存在差异。

Brown rice:a missing nutrient-rich health food

Brown rice(BR)is a traditional health food being rich in various active substances, which have effective preventive and therapeutic effects on many diseases. In this review, we systemically summarized the efficacy of BR on cardiovascular diseases(CVDs), hyperlipidemia, hypertension, diabetes, obesity, osteoporosis, neurodegenerative diseases, cancer, immune disorders, inflammation, anxiety, liver and stomach damage, etc. Underlying mechanisms have also been analyzed, which include regulating lipid synthesis and metabolism, improving insulin levels and increasing glucose metabolism, anti-oxidation, promoting the immune system, transcription and non-transcriptional regulation, blocking cell cycle, thereby improving disease symptoms and reducing its recurrence rate. Generally, BR has been almost disappeared in modern society, but it is promising to exploit the potential to become a functional food. Further research on the nutritional value, variety differences, storage problems, taste and molecular mechanism of BR will be conducive to help meticulously design clinical intervention trials and ultimately improve the treatment and prevention of various diseases.

目的:提高疏水性生物活性物质的水溶性和稳定性。方法:利用大米蛋白酶解物(RPH)和黄原胶(XG),以疏水性模型活性物质姜黄素(Cur)为输送对象,构建了RPH-XG-Cur纳米输送体系,测定并分析纳米粒子的包封效果、稳定性、抗氧化性以及形成机制。结果:RPH-XG-Cur纳米粒子的姜黄素包封率为78.40%,粒径74.78 nm,多分散指数0.36,Zeta电位-27.93 mV,添加黄原胶可显著提高纳米粒子的包封性能以及pH和离子强度稳定性;傅里叶红外光谱和荧光光谱分析表明氢键、静电相互作用和疏水相互作用均参与纳米粒子的形成。结论:RPH-XG纳米体系具有作为疏水性生物活性物质纳米输送载体的应用潜力。

为探寻安全、高效的黄曲霉新型抑制物,采用顶空固相微萃取-气质联用(HS-SPME-GC-MS)技术对枯草芽孢杆菌(Bacillus subtilis)PW2所产具有抗霉活性的挥发性有机化合物(volatile organic compounds,VOCs)进行了成分鉴定,并利用平板对扣法从鉴定出的成分中筛选具有强抗霉活性的目标化合物,通过平板对扣法和96孔板梯度稀释法测定目标化合物对黄曲霉的最小抑菌体积分数(MIC),使用黄曲霉毒素B_(1)(aflatoxin B_(1),AFB_(1))ELISA检测试剂盒分析了目标化合物对黄曲霉产AFB_(1)的抑制作用,使用扫描电子显微镜(scanning electron microscope,SEM)观察受试黄曲霉孢子形态的变化和采用分光光度法测定受试黄曲霉细胞膜麦角甾醇相对含量,以探讨抗霉机理。结果显示,从PW2所产VOCs中共鉴定出41种组分,并从中筛选出异辛醇为目标化合物;平板对扣法和96孔板梯度稀释法测得异辛醇对黄曲霉的MIC分别为0.169μL/mL和6.25μL/mL;PDB培养基中异辛醇体积分数分别为3.13、6.25、12.50μL/mL时,均能有效抑制AFB_(1)的产生,且随异辛醇体积分数的升高,抑制效果增强;异辛醇处理可导致黄曲霉孢子表面凹陷和孢子破裂,并且影响细胞膜中麦角甾醇的相对含量,造成细胞膜受损。该研究可为开发以异辛醇为功能成分的黄曲霉抑制剂提供理论参考。

提取方法对米糠纤维素理化性质的影响

以脱脂米糠为原料提取米糠不溶性膳食纤维(IDF),采用氢氧化钠-亚氯酸钠法、超声辅助氢氧化钠-亚氯酸钠法、氢氧化钠-过氧化氢法、超声辅助氢氧化钠-过氧化氢法等四种方法提取米糠纤维素,并比较4种提取方法对米糠纤维素结构特性和功能特性的影响。结果表明,超声辅助氢氧化钠-亚氯酸钠法提取纤维素具有最高纯度(88.69%)和结晶度(59.30%),且有较好热稳定性、持水性、持油性和葡萄糖吸附能力。与氢氧化钠-亚氯酸钠法相比,氢氧化钠-过氧化氢法提取的纤维素的平均粒径小,持水性和持油能力弱,而胆酸盐吸附能力和淀粉酶抑制活性更强。FT-IR图谱表明氢氧化钠-过氧化氢法提取的纤维素中含有一定量的木质素和半纤维素。SEM分析表明氢氧化钠-亚氯酸钠法提取的米糠纤维素表面更加光滑,超声辅助处理可以显著提高了纤维素的纯度以及持水性和持油性,但对纤维素的平均粒径、热稳定性无显著影响。

为探讨在混合式教学模式下建立基础理论、实验技能与创新实践能力培养一体化的食品微生物学教学方法,本文结合食品微生物学的课程特点和以往教学中有待改进的问题,将混合式教学过程中的课前、课堂和课后“三环”教学与布鲁姆教育目标的记忆、理解、应用、分析、评价和创造6个层次相结合,建立了“三环六阶”逐阶深入教学法,并采取了明确教学主线、开拓创新思维和加强课程思政教学等辅助举措来保障教学效果。“三环六阶”教学方法和辅助举措的实施,突出了学生在教学过程中的主体地位,提高了课堂教学效率,调动了学生自主学习理论知识和自觉开展实验研究的积极性,锻炼了学生的综合素质和创新实践能力。该教学方法为探索在网络信息化时代建设符合以成果为导向理念(outcome-based education,OBE)和新工科发展要求的理论、实践一体化的教学模式提供借鉴。
