A New Genus of Paleonisciformes from the Early Cretaceous Longjiang Formation in Heilongjiang Province, China

We report a new genus and species of Paleonisciformes-Cretolepis dongbeiensis gen. et sp. nov.,from the Early Cretaceous Longjiang Formation in Qiqihar City, Heilongjiang Province. The new materials can be distinguished from other Paleonisciformes genera by possessing the following characteristics: a sclerotic ring composed of more than five small bones; three pieces of suborbitals; one dermohyomandibular;a jaw hanging apparently inclined anteriorly; long and tall horizontal branch of the preopercular, a short vertical branch, with the angle formed by the two branches slightly higher than 90°; a triangular dorsal fin in the middle point between the pelvic and anal fins. The discovery of Mesozoic Paleonisciformes in northeastern China adds to the biodiversity and geographic distribution of Paleonisciformes.

Genetically modified pigs represent a great promise for generating models of human diseases and producing new *** of genetically edited pigs using somatic cell nuclear transfer(SCNT)or zygote cytoplasmic microinjection is a tedious process due to the low developmental rate or mosaicism of the founder(FO).Herein,we developed a method termed germinal vesicle oocyte gene editing(GVGE)to produce non-mosaic porcine embryos by editing maternal alleles during the GV to MII *** of Cas9 mRNA and X-linked Dmd gene-specific gRNA into GV oocytes did not affect their developmental *** MII oocytes edited during in vitro maturation(IVM)could develop into blastocysts after parthenogenetic activation(PA)or in vitro fertilization(IVF).Genotyping results indicated that the maternal gene X-linked Dmd could be efficiently edited during oocyte *** to81.3% of the edited IVF embryos were non-mosaic Dmd gene mutant *** conclusion,GVGE might be a valuable method for the generation of non-mosaic maternal allele edited FO embryos in a short simple step.

Cytosine and adenine base editors are promising new tools for introducing precise genetic modifications that are required to generate disease models and to improve traits in pigs. Base editors can catalyze the conversion of C→T(C>T) or A→G(A>G) in the target site through a single guide RNA. Injection of base editors into the zygote cytoplasm can result in the production of offspring with precise point mutations, but most F0 are mosaic, and breeding of F1 heterozygous pigs is time-intensive. Here, we developed a method called germinal vesicle oocyte base editing(GVBE) to produce point mutant F0 porcine embryos by editing the maternal alleles during the GV to MⅡ transition. Injection of cytosine base editor 3(BE3) mRNA and X-linked Dmdspecific guide RNAs into GVoocytes efficiently edited maternal Dmd during in vitro maturation and did not affect the maturation potential of the oocytes. The edited MⅡ oocytes developed into blastocysts after parthenogenetic activation(PA) or in vitro fertilization(IVF). However, BE3 may reduce the developmental potential of IVF blastocysts from 31.5%±0.8% to 20.4%±2.1%. There 40%–78.3% diploid PA blastocysts had no more than two different alleles, including up to 10% embryos that had only C>T mutation alleles. Genotyping of IVF blastocysts indicated that over 70% of the edited embryos had one allele or two different alleles of Dmd. Since the male embryos had only a copy of Dmd allele, all five(5/19) F0 male embryos are homozygous and three of them were Dmd precise C>T mutation. Nine(9/19) female IVF embryos had two different alleles including a WT and a C>T mutation. DNA sequencing showed that some of them might be heterozygous embryos. In conclusion, the GVBE method is a valuable method for generating F0 embryos with maternal point mutated alleles in a single step.

Human papillomaviruses （HPVs） including *** （HR） and low-risk （LR） subtypes have distinguishable variation on both genotypes and phenotypes. The co- infection of multiple HR-HPVs, headed by HPV16, is common in cervical cancer in female. Recently accu- mulating reports have focused on the interaction be- tween virus and host, particularly the role of human microRNAs （miRNAs） in anti-viral defense by targeting viral genome. Here, we found a well-conserved target site of miRNAs in the genomes of most HR-HPVs, not LR-HPVs, by scanning all potential target sites of human miRNAs on 24 HPVs of unambiguous subtypes of risk. The site is targeted by two less common human miR- NAs, miR-875 and miR-3144, and is located in E6 onco- gene open reading frame （ORF） and overlap with the first alternative splice exon of viral early transcripts. In validation tests, miR-875 and miR-3144 were identified to suppress the target reporter activity markedly and inhibit the expression of both synthetically exogenous E6 and endogenous E6 oncogene. High level of two miRNAs can inhibit cell growth and promote apoptosisin HPV16-positive cervical cancer cells. This study pro- vides a promising common target of miRNAs for most HR-HPVs and highlights the effects of two low ex- pressed human miRNAs on tumour suppression.

When microarray gene expression data are used to predict multiple drug resistance(MDR)phenotypes for anticancer drugs,the normalization strategy and the quality of the selected signature genes are usually the main causes of inconsistency among different experiments.A stable statistical drug response prediction model is urgently required in *** this study,the microarray gene expression data of multiple cancer cell lines with MDR was *** each probe-set,the expression value was defined as present/absent(1/0)and was classified into a gene set defined with protein domain organization(PDO).After employing the gene content method of phylogenetic analysis,a phylogenetic model(cell tree)for MDR phenotype prediction was built at the PDO gene set *** results indicate that classification of cancer cell lines is predominantly affected by both the histopa-thological features and the MDR phenotype(paclitaxel and vinblastine).When applying this model to predict the MDR phenotype of independent samples,the phylogenetic model performs better than signature gene *** the utility of our procedure is limited due to sample heterogeneity,it still has potential application in MDR research,especially for hematological tumors or established cell lines.

Advanced natural killer/T cell lymphoma(NKTL)has demonstrated poor prognosis with currently available ***,we report the efficacy of anti-programmed death 1(PD-1)antibody with the P-GEMOX(pegaspargase,gemcitabine,and oxaliplatin)regimen in advanced *** patients underwent six 21-day cycles of anti-PD-1 antibody(day 1),pegaspargase 2000 U/m^(2)(day 1),gemcitabine 1 g/m^(2)(days 1 and 8)and oxaliplatin 130 mg/m^(2)(day 1),followed by anti-PD-1 antibody maintenance every 3 *** death-ligand 1(PD-L1)expression and genetic alterations were determined in paraffin-embedded pretreatment tissue samples using immunohistochemistry and next-generation sequencing(NGS)*** were assessed using ^(18)F-fluorodeoxyglucose positron emission tomography(^(18)FDG-PET)and computed tomography or magnetic resonance *** patients exhibited significant responses,comprising of seven complete remissions and one partial remission(overall response rate:88.9%).After a median follow-up of 10.6 months,6/9 patients(66.7%)remained in complete *** most common grade 3/4 adverse events were anemia(33.3%),neutropenia(33.3%),and thrombocytopenia(33.3%);all of which were manageable and *** produced a high response rate in patients with positive PD-L1 expression(5/6,83.3%).NGS analysis suggested that STAT3/JAK3/PD-L1 alterations and ARID1A mutation were associated with immunochemotherapy *** in DDX3X and alteration in epigenetic modifiers of KMT2D,TET2,and BCORL1 might indicate a poor response to *** conclusion,the anti-PD-1 antibody plus P-GEMOX regimen demonstrated promising efficacy in advanced ***-L1 expression combined with specific genetic alterations could be used as potential biomarkers to predict therapeutic responses to immunochemotherapy.

Exploring the phase transition of high entropy alloys(HEAs)with multiple major elements is of great importance for understanding the underlying physical *** martensitic phase tran-sition has been frequently reported in HEAs,however,nanoscale microstructural phase evolution has not been investigated to the same ***,we have prepared the Ti_(33)Nb_(15)Zr_(25)Hf_(25)O_(2)HEA and investi-gated the strain glass transition and its associated properties using dynamic mechanical analysis and mi-crostructure *** have found that the elastic modulus in Ti_(33)Nb_(15)Zr_(25)Hf_(25)O_(2)HEA deviates from Wachtman’s equation and observed the Elinvar effect in the form of temperature-independent mod-ulus in the temperature range from 150 K to 450 K and frequency-dependence modulus around 220 *** strain glass transition has been evidenced in Ti_(33)Nb_(15)Zr_(25)Hf_(25)O_(2)HEA by the formation and growth of nano-sized domains during in-situ transmission electron microscopy(TEM)cooling,and substantiated by the broken ergodicity during zero-field-cooling/*** strain glass transition is believed to account for the Elinvar effect,where the modulus hardening of nano-sized domains compensates dynam-ically with the modulus softening of the transformable matrix.

Objective:To characterize modified R-CODOX-M/IVAC-based chemotherapy to lower the severe adverse events in Chinese adult patients with sporadic Burkitt ***:We enrolled a retrospective cohort including 123 adult patients with untreated sporadic Burkitt lymphoma from August 2008 to September 2019 at Sun Yat-sen University Cancer *** studied a dose-modified and long-course R-CODOX-M/IVAC regimen utilizing a low dose of 1.0 g/m2/cycle cyclophosphamide,2 g/m2/cycle methotrexate,4,500 mg/m2/cycle ifosfamide,and 4.0 g/m2/cycle ***-nine patients with low risk disease underwent 4–6 cycles of dose-modified R-CODOX-M-based ***-four patients with high risk disease underwent 6–8 cycles of dose-modified alternating R-CODOX-M/IVAC ***:The objective remission was 87.0%.The event-free survival rate and overall survival at 3 years were 81.2%and 92.1%,*** grade 3–4 adverse events included leukopenia(91.9%),anemia(58.5%),thrombocytopenia(73.2%),and febrile neutropenia(48.8%).A total of 26.0%and 37.4%of patients received red blood cell and platelet transfusions,*** observed 4 cases(3.3%)of septic shock after *** treatment-related deaths occurred from severe ***:The modified R-CODOX-M/IVAC chemotherapy regimen was effective for sporadic Burkitt lymphoma in the Chinese population,with a lower toxicity than standard regimens.
